Abstract:The dysfunction of coronary microcirculation is an important cause of coronary artery disease (CAD). The index of microcirculatory resistance (IMR) is a quantitative evaluation of coronary microcirculatory function, which provides a significant reference for the prediction, diagnosis, treatment, and prognosis of CAD. IMR also plays a key role in investigating the interaction between epicardial and microcirculatory dysfunctions, and is closely associated with coronary hemodynamic parameters such as flow rate, distal coronary pressure, and aortic pressure, which have been widely applied in computational studies of CAD. However, there is currently a lack of consensus across studies on the normal and pathological ranges of IMR. The relationships between IMR and coronary hemodynamic parameters have not been accurately quantified, which limits the application of IMR in computational CAD studies. In this paper, we discuss the research gaps between IMR and its potential applications in the computational simulation of CAD. Computational simulation based on the combination of IMR and other hemodynamic parameters is a promising technology to improve the diagnosis and guide clinical trials of CAD.

Abstract:Introduction: Coronary microvascular disease (CMD) is increasingly appreciated as a contributor to adverse cardiovascular outcomes, but underlying mechanisms of CMD remain poorly understood. Investigation of CMD in animal models is limited by a lack of techniques to quantify flow in the heart. We assessed the hypothesis that intravital microscopy (IVM) can enable in vivo imaging of red blood cell (RBC) flow profiles in the coronary microcirculation at cellular resolution. Methods: C57BL/6 mice (n=20) were used to develop cardiac IVM protocols. Cardiomyocyte membrane staining was achieved using intravenous injection of a voltage dye (di-2-anepeq). RBC labeling was done by ex-vivo staining of cells with Ter119 followed by transfusion. IVM was performed through a thoracotomy with animals anesthetized and ventilated. Confocal and two-photon microscopy was done using a custom tissue stabilizer and an Olympus FV1000MPE microscope synchronized to physiologic signals using custom hardware and software and cardiac pacing protocols. Images were analyzed in MATLAB to extract myocyte contractile properties, capillary diameter, and capillary flow properties. Results: Using cardiac and respiratory gating, images with cellular resolution could be measured in the beating heart at all phases of the cardiac cycle. This enabled quantification of cardiomyocyte contractile function and capillary diameter and widefield imaging by stitching of several images (Fig. 1A). Tracking of RBCs over serial images allowed quantification of RBC velocity across multiple capillaries simultaneously (Fig. 1B). Conclusion: Cardiac IVM can provide quantification of cardiac function and blood flow at the single cell scale and will provide an exciting tool for investigating CMD in models of heart disease.

Abstract:The mouse is the mammalian model of choice for investigating cardiovascular biology, given our ability to manipulate it by genetic, pharmacologic, mechanical, and environmental means. Imaging is an important approach to phenotyping both function and structure of cardiac and vascular components. This review details commonly used imaging approaches, with a focus on echocardiography and magnetic resonance imaging, with brief overviews of other imaging modalities. In this update, we also emphasize the importance of rigor and reproducibility in imaging approaches, experimental design, and documentation. Finally, we briefly outline emerging imaging approaches but caution that reliability and validity data may be lacking. © 2024 Wiley Periodicals LLC.

Abstract:Introduction: Cerebral microhemorrhages (CMH) are the pathologic substrate of MRI-demonstrable cerebral microbleeds, and are thought to occur when integrity of cerebral microvessels is compromised, leading to deposits of hemosiderin/iron accumulating within brain tissue. The presence of CMH is associated with cognitive decline and ischemic and hemorrhagic stroke. Hypertension (HTN) is a major risk factor for cerebral microbleeds and has been linked to a more significant microbleed burden in patients. Despite the clinical significance of HTN and cerebral microbleeds, our understanding of HTN-induced microbleed formation remains limited, and the vascular origin of CMH remains obscure. Methods: We used a perfusion-based vascular label with tissue clearing to enable three-dimensional visualization of CMH with the surrounding microvascular network. Adult (18-month-old) C57BL/6J mice were administered angiotensin II to induce HTN (n=10) or PBS (n=10). Lectin-DyLight-649 was injected retro-orbitally to bind to the endothelial walls of all blood vessels in a mouse. Brains were cut into 1-mm thick sections, stained with Prussian blue to label CMH, cleared using iDISCO, and imaged via confocal microscopy. MATLAB and neuTube were used to quantify vessel diameters. Results: Using this methodology, we were able to collect three-dimensional transmission images of CMH that were colocalized with three-dimensional fluorescence images of the surrounding microvasculature. The average vessel diameter of the 5 nearest vessels to 17 different CMH was found to be 4.22±0.81 μm, which corresponds to capillary-size vessels. Conclusions: These data support a capillary origin for cerebral microhemorrhages in this mouse model. The capillary origin implies a role for the blood-brain barrier in the development of CMH and cerebral microbleeds in this model of HTN and aging. Three-dimensional imaging has substantial utility for improved understanding of CMH origin.

Abstract:Cerebral small vessel disease has been linked to cognitive, psychiatric and physical disabilities, especially in the elderly. However, the underlying pathophysiology remains incompletely understood, largely due to the limited accessibility of these small vessels in the live brain. Here, we report an intravital imaging and analysis platform for high-resolution, quantitative and comprehensive characterization of pathological alterations in the mouse cerebral microvasculature. By exploiting multi-parametric photoacoustic microscopy (PAM), microvascular structure, blood perfusion, oxygenation and flow were imaged in the awake brain. With the aid of vessel segmentation, these structural and functional parameters were extracted at the single-microvessel level, from which vascular density, tortuosity, wall shear stress, resistance and associated cerebral oxygen extraction fraction and metabolism were also quantified. With the use of vasodilatory stimulus, multifaceted cerebrovascular reactivity (CVR) was characterized in vivo. By extending the classic Evans blue assay to in vivo, permeability of the blood-brain barrier (BBB) was dynamically evaluated. The utility of this enabling technique was examined by studying cerebrovascular alterations in an established mouse model of high-fat diet-induced obesity. Our results revealed increased vascular density, reduced arterial flow, enhanced oxygen extraction, impaired BBB integrity, and increased multifaceted CVR in the obese brain. Interestingly, the 'counterintuitive' increase of CVR was supported by the elevated active endothelial nitric oxide synthase in the obese mouse. Providing comprehensive and quantitative insights into cerebral microvessels and their responses under pathological conditions, this technique opens a new door to mechanistic studies of the cerebral small vessel disease and its implications in neurodegeneration and stroke.

Abstract:Deviation of blood flow from an optimal range is known to be associated with the initiation and progression of vascular pathologies. Important open questions remain about how the abnormal flow drives specific wall changes in pathologies such as cerebral aneurysms where the flow is highly heterogeneous and complex. This knowledge gap precludes the clinical use of readily available flow data to predict outcomes and improve treatment of these diseases. As both flow and the pathological wall changes are spatially heterogeneous, a crucial requirement for progress in this area is a methodology for co-mapping local data from vascular wall biology with local hemodynamic data. In this study, we developed an imaging pipeline to address this pressing need. A protocol that employs scanning multiphoton microscopy was designed to obtain 3D data sets for smooth muscle actin, collagen and elastin in intact vascular specimens. A cluster analysis was developed to objectively categorize the smooth muscle cells (SMC) across the vascular specimen based on SMC density. In the final step in this pipeline, the location specific categorization of SMC, along with wall thickness was co-mapped with patient specific hemodynamic results, enabling direct quantitative comparison of local flow and wall biology in 3D intact specimens.

Abstract:Background: Cerebral cavernous malformation (CCM) is a disease characterized by vascular malformations that primarily develop in the brain. These malformations are prone to leak, and their rupture or thrombotic closure can cause life-threatening hemorrhages and strokes. Mouse models have been instrumental to study the disease, but most cause premature lethality, precluding the investigation of disease mechanisms through intravital microscopy. Current mouse models also do not recapitulate human CCM skin lesions. Methods: Endothelial-specific deletion of Ccm3 via systemic tamoxifen application at postnatal day 4 or 5 prolongs survival and induces vascular malformations in the mouse brain and ear skin. CCM skin lesions can also be induced by topic tamoxifen administration directly to the ear. The thin, flat morphology of the ear skin is ideal for intravital microscopy. Dextran dyes and platelet markers allow to study blood flow and blood clot formation, in living animals in real time. Results: We report that human CCM skin lesions can be recapitulated in a mouse model and that skin lesions share hallmarks of CCM brain lesions. Intravital imaging reveals that CCM skin lesions are slow-flow malformations prone to thrombus formation. Conclusions: Intravital imaging of CCM skin lesions expands the toolkit of CCM research and allows longitudinal studies of lesion growth.

Abstract:The use of genetic engineering to develop important neuropathological mouse models has made cerebrovascular imaging essential for the investigation of numerous brain disorders, especially cerebrovascular disorders, such as aneurysms, arteriovenous malformations, and ischemic and hemorrhagic stroke. New laboratory-based X-ray microimagers exist that provide easy access, reliable operation, and performance previously found only in synchrotron-based instruments. Here, we reported a novel approach using such a system to detect intracerebral hemorrhage and resultant cerebrovascular pathology. Adult male C57BL/6 mice (n = 12) underwent 30 μl autologous blood injection into the right basal ganglia region. After sacrificing the animals and vascular perfusion with Microfil® MV-122 Yellow to opacify vascular and microvascular structures, the brain was post-fixed and partially hydrated for 3D imaging with a MicroXCT-400® at 30 KeV and 2-μm resolution. Tomographic reconstruction of high-resolution microimages was accomplished with Amira® software. High-quality 3D images included cerebrocortical microvessels, the circle of Willis, the sagittal sinus, transverse sinus, and other arterial and venous systems. In the ipsilateral hemisphere, there clearly were early-stage vasodilatation and later-stage neovascularization. Very high-resolution, laboratory-based, X-ray micro-CT contrast imaging can accomplish sensitive quantifications of normal and pathological small cerebrovascular changes, especially in hemorrhagic stroke and subsequent hemorrhage-induced neovascularization.

Abstract:Background: Cerebral microbleeds (CMBs) are age-related vascular lesions, predisposing patients to a lifetime risk of hemorrhagic stroke and cognitive decline. The cellular architecture in the lesions and their genetic alterations have not been elucidated, limiting biomarker development and therapeutic targeting. Herein, we report histopathological and transcriptomic signatures of CMBs in a murine model. Methods: Seven APOE4 + / + ; 5xFAD mice were sacrificed at 8 months and had their brains harvested. Brain tissue was stained with H&E and Prussian Blue for non-heme iron to identify CMBs. Astrocytes (GFAP + ), microglia (CD163 - /Iba1 + ), macrophages (CD163 + /Iba1 + ), B cells (CD20 + ), T cells (CD3 + ), and endothelium (CD31 + ) were stained using multiplexed immunofluorescence. Regions of interest (ROIs) around CMBs and directly adjacent tissue, as well as contralateral control brain tissue, were selected and quantified using QuPath. GeoMx spatial transcriptomics was performed on lesions and contralateral control brain areas in the same mice. The DEG analysis was completed using DESeq2. Results: CMBs had higher astrocyte and microglia counts (normalized to ROI area) than control tissue (p<0.05). Microglia showed reactive, amoeboid morphological changes: hypertrophic cell bodies and shorter processes. T cells had a trend toward higher cell counts in CMBs (p=0.12). Macrophages and B cell counts were similar between CMBs and controls. The spatial transcriptomics identified 19 DEGs between CMBs and control tissue (p<0.1, FDR corrected) related to permeability, cell metabolism, apoptosis, and transcriptional regulation. Conclusions: This is the first report relating the CMB lesional microenvironment to spatial transcriptomic analyses. Further studies shall aim at confirming these findings in human CMB lesions, and their association with circulating molecules. The results pave the way for enhanced mechanistic insight, biomarker discovery and potential therapeutic targeting.

Abstract:In the past 2 decades, research on atherosclerotic cardiovascular disease has uncovered inflammation to be a key driver of the pathophysiological process. A pressing need therefore exists to quantitatively and longitudinally probe inflammation, in preclinical models and in cardiovascular disease patients, ideally using non-invasive methods and at multiple levels. Here, we developed and employed in vivo multiparametric imaging approaches to investigate the immune response following myocardial infarction. The myocardial infarction models encompassed either transient or permanent left anterior descending coronary artery occlusion in C57BL/6 and Apoe-/-mice. We performed nanotracer-based fluorine magnetic resonance imaging and positron emission tomography (PET) imaging using a CD11b-specific nanobody and a C-C motif chemokine receptor 2-binding probe. We found that immune cell influx in the infarct was more pronounced in the permanent occlusion model. Further, using 18F-fluorothymidine and 18F-fluorodeoxyglucose PET, we detected increased hematopoietic activity after myocardial infarction, with no difference between the models. Finally, we observed persistent systemic inflammation and exacerbated atherosclerosis in Apoe-/- mice, regardless of which infarction model was used. Taken together, we showed the strengths and capabilities of multiparametric imaging in detecting inflammatory activity in cardiovascular disease, which augments the development of clinical readouts.

Abstract:OBJECTIVE:Arteriogenesis is an important mechanism that contributes to restoration of oxygen supply in chronically ischemic tissues, but remains incompletely understood due to technical limitations. This study presents a novel approach for comprehensive assessment of the remodeling pattern in a complex microvascular network containing multiple collateral microvessels.METHODS:We have developed a hardware-software integrated platform for quantitative, longitudinal, and label-free imaging of network-wide hemodynamic changes and arteriogenesis at the single-vessel level. By ligating feeding arteries in the mouse ear, we induced network-wide hemodynamic redistribution and localized arteriogenesis. The utility of this technology was demonstrated by studying the influence of obesity on microvascular arteriogenesis.RESULTS:Simultaneously monitoring the remodeling of competing collateral arterioles revealed a new, inverse relationship between initial vascular resistance and extent of arteriogenesis. Obese mice exhibited similar remodeling responses to lean mice through the first week, including diameter increase and flow upregulation in collateral arterioles. However, these gains were subsequently lost in obese mice.CONCLUSIONS:Capable of label-free, comprehensive, and dynamic quantification of structural and functional changes in the microvascular network in vivo, this platform opens up new opportunities to study the mechanisms of microvascular arteriogenesis, its implications in diseases, and approaches to pharmacologically rectify microvascular dysfunction.

Abstract:Monitoring hemodynamic responses in the rodent stroke model is important in the study of ischemic stroke. However, it is hard for a single imaging modality to comprehensively explore these changes. In this study, we implemented laser speckle imaging (LSI) and visible-light optical coherence tomography (Vis-OCT) to detect cerebral blood flow (CBF), angiography and oxygen saturation (sO 2 ) in the mouse model of distal middle cerebral artery occlusion (dMCAO). LSI provides full-field, real-time cerebral blood flow (CBF) map to guide the Vis-OCT imaging. Vis-OCT offers angiography and sO 2 measurements. The results showed the detailed CBF and vasculature changes before and after dMCAO. After dMCAO, sO 2 changed little in arteries and arterioles, while sO 2 decreased in veins and venules. We also showed that larger veins had a higher decrease in sO 2 value in consecutive vein branches after dMCAO. It is suggested that the combination of LSI/Vis-OCT provides a promising tool for the rodent stroke model study.

Abstract:Pulmonary vascular dysfunction is characterized by remodeling and loss of microvessels in the lung and is a major manifestation of chronic lung diseases (CLD). In murine models of CLD, the small arterioles and capillaries are the first and most prevalent vessels that are affected by pruning and remodeling. Thus, visualization of the pulmonary arterial vasculature in three dimensions is essential to define pruning and remodeling both temporally and spatially and its role in the pathogenesis of CLD, aging, and tissue repair. To this end, we have developed a novel method to visualize and quantitate the murine pulmonary arterial circulation using microcomputed tomography (µCT) imaging. Using this perfusion technique, we can quantitate microvessels to approximately 6 µM in diameter. We hypothesize that bleomycin-induced injury would have a significant impact on the arterial vascular structure. As proof of principle, we demonstrated that as a result of bleomycin-induced injury at peak fibrosis, significant alterations in arterial vessel structure were visible in the three-dimensional models as well as quantification. Thus, we have successfully developed a perfusion methodology and complementary analysis techniques, which allows for the reconstruction, visualization, and quantitation of the mouse pulmonary arterial microvasculature in three-dimensions. This tool will further support the examination and understanding of angiogenesis during the development of CLD as well as repair following injury.

Abstract:Increasing evidence implicates cerebral microvascular dysfunction in the pathophysiology of numerous central nervous system pathologies, including stroke. Understanding the molecular alterations in cerebral microvessels in these conditions will provide original opportunities for scientific investigation at the pre-clinical and clinical levels. In this study, we conducted a novel genome-wide transcriptomic analysis of microvessels in a mouse model of transient focal cerebral ischemia. Using a publicly available human ischemic stroke dataset, we identified shared alterations in our microvessel dataset with implications for human pathophysiology. From this unbiased analysis, we report predicted alterations in inter- and intra-cellular signaling, emphasizing perturbations in genes involved in blood brain barrier function, endothelial cell activation and metabolism. Furthermore, our study unveiled previously unreported gene expression changes associated with altered sphingolipid metabolism. Altogether, our results have identified microvessel-specific transcriptomic changes in a number of translationally relevant pathways that support the targeting of these pathways in preclinical studies. The data shared here provide a resource for future investigation of translationally relevant pathways in ischemic stroke.
Ultrasound Bio-microscopy for Measurement of Coronary Artery Flow and Estimation of Infarct Size in a Mouse Model of Acute Myocardial Infarction

Abstract:Ultrasound bio-microscopy was used to measure hemodynamic changes in the left main coronary artery after myocardial infarction (MI), and its usefulness in estimating infarct size was evaluated. MI was induced by left anterior descending artery ligation. Diastolic peak velocity (V-d), mean flow velocity (V-mean) and the velocity-time integral (VTI) were measured 2 and 6 h after MI. Serum troponin I levels were assayed 2, 6 and 12 h after MI. At 2 h, V-mean and VTI significantly differed between mice that underwent low and high left anterior descending artery ligation; V-d, V-mean and VTI were correlated with infarct size (r = -0.557, -0.693 and -0.672, respectively; all p < 0.01). Infarct size was more strongly correlated with 2-h ultrasound bio-microscopy measurements than with 2-h serum troponin I level. Measurement of coronary artery blood flow by ultrasound bio-microscopy may be useful for early estimation of infarct size in mice. Abstract:Objective Recent advances in tissue clearing and high‐throughput imaging have enabled the acquisition of extended‐volume microvasculature images at a submicron resolution. The objective of this study was to extract information from this type of images by integrating a sequence of 3D image processing steps on Terabyte scale datasets. Methods We acquired coronary microvasculature images throughout an entire short‐axis slice of a 3‐month‐old Wistar–Kyoto rat heart. This dataset covered 13 × 10 × 0.6 mm at a resolution of 0.933 × 0.933 × 1.866 μm and occupied 700 Gigabytes of disk space. We used chunk‐based image segmentation, combined with an efficient graph generation technique, to quantify the microvasculature in the large‐scale images. Specifically, we focused on the microvasculature with a vessel diameter up to 15 μm. Results Morphological data for the complete short‐axis ring were extracted within 16 h using this pipeline. From the analyses, we identified that microvessel lengths in the rat coronary microvasculature varied from 6 to 300 μm. However, their distribution was heavily skewed toward shorter lengths, with a mode of 16.5 μm. In contrast, vessel diameters ranged from 3 to 15 μm and had an approximately normal distribution of 6.5 ± 2 μm. Conclusion The tools and techniques from this study will serve other investigations into the microcirculation, and the wealth of data from this study will enable the analysis of biophysical mechanisms using computer models.

Abstract:The blood and lymphatic vasculature networks are not yet fully understood even in mouse because of the inherent limitations of imaging systems and quantification methods. This study aims to evaluate the usefulness of the tissue-clearing technology for visualizing blood and lymphatic vessels in adult mouse. Clear, unobstructed brain/body imaging cocktails and computational analysis (CUBIC) enables us to capture the high-resolution 3D images of organ- or area-specific vascular structures. To evaluate these 3D structural images, signals are first classified from the original captured images by machine learning at pixel base. Then, these classified target signals are subjected to topological data analysis and non-homogeneous Poisson process model to extract geometric features. Consequently, the structural difference of vasculatures is successfully evaluated in mouse disease models. In conclusion, this study demonstrates the utility of CUBIC for analysis of vascular structures and presents its feasibility as an analysis modality in combination with 3D images and mathematical frameworks.
