Abstract:An innovative organic small molecule with a D-A structure was synthesized by connecting triphenylamine to BODIPY via a thiophene bridge. Triphenylamine and thiophene units ingeniously modulate the balance between steric hindrance and π-π interactions around the flat aza-BODIPY core. The molecule exhibits near-infrared fluorescence absorption and emits at roughly 1100 nm, featuring a significant Stokes shift. Both the molecule and its nanoparticles demonstrate high stability and achieve a remarkable 35 % photothermal conversion efficiency when conjugated with the P(OEGMA)20-P(Asp)14 copolymer. In vitro assessments show low dark toxicity and outstanding biocompatibility. Moreover, in vivo studies and photothermal therapy in mice indicate substantial tumor shrinkage and reduced recurrence, confirming its potential in cancer treatment. These results highlight the promise of this organic molecule and its nanoparticles for NIR-II imaging-guided photothermal therapy, introducing a novel approach to phototheranostic applications for cancer management.

Abstract:Near-infrared (NIR) responsive nanoparticles are an important platform for multimodal phototherapy. Importantly, the simultaneous NIR-triggered photodynamic (PDT) and photothermal (PTT) therapy is a powerful approach to increase the antitumor efficiency of phototherapic nanoparticles due to the synergistic effect. Herein, a boron dipyrromethene (BODIPY)-based amphiphilic dye with enhanced electron donor-acceptor-donor (D-A-D) structure (BDP-AP) was designed and synthesized, which could self-assemble into stable nanoparticles (BDP-AP NPs) for the synergistic NIR-triggered PDT/PTT therapy. BDP-AP NPs synchronously generated singlet oxygen (1O2) and achieved preeminent photothermal conversion efficiency (61.42%). The in vitro and in vivo experiments showed that BDP-AP NPs possessed negligible dark cytotoxicity and infusive anticancer performance. BDP-AP NPs provide valuable guidance for the construction of PDT/PTT-synergistic NIR nanoagents to improve the efficiency of photoinduced cancer therapy in the future.

Abstract:The synthesis and characterization of a highly photostable bromo-substituted BODIPY dye (I) fused-ring-expanded with thienopyrrole moieties is reported. The results of MTT assays and flow cytometric analyses in living HeLa cells demonstrate that I has a high singlet oxygen quantum yield (ΦΔ = 0.63) and exhibits photocytotoxicity upon irradiation in the NIR region making it potentially suitable for use in PDT.

Abstract:Journal of Porphyrins and Phthalocyanines, Ahead of Print. Boron-dipyrromethene (BODIPY) as a traditional fluorescent dye has received increasing attention because of its high molar extinction coefficient, easy modification, and good light stability. In this work, three BODIPY compounds (BDP-1, BDP-2 and BDP-3) were designed and synthesized by introducing PEG chains and a naphthalimide (NI) group onto the parent structure of BODIPY. Due to the electron-withdrawing properties of NI, the compounds of BDP-2 and BDP-3 formed a typical donor (D)-acceptor (A) structure, which led to the red-shifted absorption and fluorescence spectrum. The synthesized BODIPY exhibited intense green emission, with fluorescence quantum yield of 0.66, 0.29, and 0.19 for BDP-1, BDP-2 and BDP-3 respectively. The decrease in fluorescence of BDP-2 and BDP-3 was probably ascribed to the enhanced intramolecular charge transfer because of the introduction of the NI group. The good biocompatibility and low cytotoxicity of the synthesized BODIPY compounds were verified by the CCK-8 assay. The subcellular localization of the three compounds was further evaluated through a confocal laser scanning microscope. The results showed that these BODIPY derivatives were mainly localized in the lysosomes of cancer cells. Thus, the synthesized BODIPY fluorescent dyes show promising applications in tumor imaging benefiting from their good biocompatibility and strong fluorescence.

Abstract:Fusion thiophene to BODIPY endows the chromophore with more fundamental and special functions. Herein, novel symmetric bisbenzothieno[ b ]-fused BODIPYs (BTBs) were synthesized and developed. These newly prepared BTB dyes possessed absorption maxima in the range of 643–665 nm, and many outstanding properties including large molar extinction coefficients, and high singlet oxygen generation. Theoretical calculations affirmed that the smaller singlet-triplet energy gap and larger spin−orbit coupling result in efficient intersystem crossing, thus increasing the singlet oxygen generation yield. Notably, Me 2 NPh-substituted BTB displayed good reactive oxygen species generation efficiency up to 0.41, and a remarkable photo-cytotoxicity (IC 50 = 83.5 nM) in HeLa cells, which shows potential in low-dose photodynamic therapy.

Theoretical study on the photophysical properties of thiophene-fused-type BODIPY series molecules in fluorescence imaging and photodynamic therapy

Abstract:As a class of photosensitizers (PSs) with dual functions of photodynamic therapy (PDT) and fluorescence imaging, the relationship between the structure and dual-function of thiophene-fused-type BODIPY dyes has not been studied in depth before. We found that the thiophene-fused-type BODIPY triplet photosensitizer is produced according to the energy level matching rule and the introduction of the thiophene ring significantly reduces the energy gap ΔEST between singlet and triplet states, as revealed by our investigation of the excited state structures and energies of thieno-fused BODIPY dyes. At the same time, a tiny ΔEST also results in a greatly enhanced intersystem crossing (ISC) rate, kISC. The kISC value of MeO-BODIPY, having the highest singlet oxygen quantum yield (ΦΔ), is the largest. Substitution with a strong electron donor N,N-dimethylaminophenyl (DMA) leads to the vertical configuration in the T1 state. The small ΔE (0.0029 eV) between the HOMO and HOMO-1 triggers the photo induced electron transfer (PET) of inhibiting ISC and fluorescence. When thieno-fused BODIPYs react with pyrrole, the increase of π-conjugation and smaller ΔEHOMO-LUMO explain the redshift in emission wavelength of thieno-pyrrole-fused BODIPY. The more planar configuration of the S1 state and the stronger oscillator intensity reflect a higher fluorescence quantum yield (ΦF). The extension of π-conjugation can cause molecules to transition to higher-level singlet excited states (Sn states, n ≥ 1) after absorbing energy and reduce the energy level of the excited state, resulting in multiple channels and favoring 1O2 production for thieno-pyrrole-fused BODIPYs with electron-withdrawing groups at the para-position of the phenyl groups. Due to ΔES0-T1 < 0.980 eV, the substitution of electron-donating groups cannot produce 1O2. In this work, we have revealed the mechanism of ISC and the fluorescence emission process in the thiophene-fused-type BODIPY dye, which has provided a theoretical foundation and guidance for the future design of BODIPY-based heavy-atom-free PSs for molecular applications in PDT.
A Novel BODIPY-based Nano-Photosensitizer with Aggregation-Induced Emission for Cancer Photodynamic Therapy

Abstract:The discovery of aggregation-induced emission (AIE) effect provides opportunities for the rapid development of fluorescence imaging-guided photodynamic therapy (PDT). In this work, a boron dipyrromethene (BODIPY)-based photosensitizer (ET-BDP-O) with AIE characteristics was developed, in which the two linear arms of BODIPY group were linked with triphenylamine to form an electron Donor–Acceptor–Donor (D–A–D) architecture while side chain was equipped with triethylene glycol group. ET-BDP-O was able to directly self-assemble into nanoparticles (NPs) without supplement of any other matrices or stabilizers due to its amphiphilic property. The as-prepared ET-BDP-O NPs had an excellent colloid stability with the size of 125 nm. Benefiting from the AIE property, ET-BDP-O NPs could generate strong fluorescence and reactive oxygen species under light-emitting diode light irradiation (60[Formula: see text]mW/cm[Formula: see text]. After internalized in cancer cells, ET-BDP-O NPs were able to emit bright red fluorescence signal for bioimaging. In addition, the cell viability assay demonstrated that the ET-BDP-O NPs exhibited excellent photo-cytotoxicity against cancer cells, while negligible cytotoxicity under dark environment. Thus, ET-BDP-O NPs might be regarded as a promising photosensitizer for fluorescence imaging-guided PDT in future.

Abstract:The discovery of new photosensitizer drugs with long wavelength Uv–vis absorption, high efficiency and low side-effects is still a challenge in photodynamic therapy. Here a series of novel mes o-substitutedphenyl thieno[3,2-b]thiophene-fused BODIPY derivatives were designed, synthesized and characterized. All these compounds have strong absorption at 640–680 nm and obvious fluorescence emission at 650–760 nm. They exhibited high singlet oxygen generation ability and significant photodynamic efficiency against Eca-109 cancer cells. Compounds II 4 , II 6 , II 9 , II 10 and II 13 could generate intracellular ROS and induce cell apoptosis after laser irradiation, which displayed superior photodynamic efficiency against Eca-109 cells than Temoporfin in vitro and in vivo . Among them, compound II 4 specifically exhibited excellent anti-tumor efficacy, and could be selected as a new drug candidate for PDT.

Abstract:As a non-invasive cancer therapy method, photodynamic therapy (PDT) shows tremendous promise in clinical cancer treatment. Light-activated singlet oxygen production of photosensitizers (PSs) is the prerequisite for cancer PDT, and the use of organic photosensitizers is always limited by visible light-based activation, hydrophilicity, biocompatibility, selectivity and quantum yield of singlet oxygen. Currently, both zinc porphyrin- and BODIPY-based structures have been widely used in the development of PDT PSs. Here, we developed a novel naphthalimide bridged zinc porphyrin/BODIPY molecule (Por-BDP-1) with two poly(ethylene glycol) (PEG) chains, in which D-A structure was constructed between the naphthalimide group and porphyrin group. After self-assembly into nanoparticles, Por-BDP-1 NPs (Diameter: 122.4 nm) could quench fluorescence in 600-700 nm, bind with calf thymus-DNA, and produce singlet oxygen during light-irradiation (laser: 680 nm, 1.0 W/cm2). In addition, Por-BDP-1 NPs effectively killed HeLa cells with a IC50 value = 44.8 mu g/mL and showed a lower dark toxicity under the same conditions. All our results demonstrated that our naphthalimide bridged zinc porphyrin/BODIPY nano-photosensitizer is a promising nanoagent for PDT in the clinic.
Discovery of a Monoiodo Aza-BODIPY Near-Infrared Photosensitizer: in Vitro and in Vivo Evaluation for Photodynamic Therapy

Abstract:Photodynamic therapy (PDT) as a rising platform of the cancer treatment method is receiving increased attention. Through systematic evaluation of halogen substitution on aza-4,4-difluoro-4-bora-3a,4a-diaza-s-indacenes (BODIPY), we have found that monoiodo-derived aza-BODIPYs provided greater efficacy than other halogenated aza-BODIPY PSs. 4 and 15 as monoiodinated aza-BODIPY dyes containing p-methoxyphenyl moiety were identified to be potent NIR aza-BODIPY-type PSs with IC50 values against HeLa cells at a light dose of 54 J/cm2 as low as 76 and 81 nM, respectively. 4 possessed superior phototoxicity, low dark toxicity, and good thermal/photostability and distributed majorly in mitochondria in cells. Apoptosis was verified to be the main cell death pathway, and in vitro reactive oxygen species generation was demonstrated. In vivo whole-body fluorescence imaging and ex vivo organ distribution studies suggested that 4 afforded an excellent PDT effect with a low drug dose under single-time light irradiation and revealed advantages over known PSs of ADPM06 and Ce6.

Abstract:Aromatic annulation on the periphery of BODIPY has proven to be particularly effective in expanding its pi-conjugated system, thereby enabling near-infrared (NIR) absorption and emission. However, achieving this shift in BODIPY's absorption and emission to the NIR region without compromising fluorescence quantum yield remains a significant challenge. Herein, we report a new family of symmetrical dibenzothieno and dibenzothieno[2,3-d]thieno[a]-fused BODIPYs (5a-c and 8a-c) synthesized via a simple four-step route involving regioselective 1,7-bromination to selectively enhance the reactivity specifically at the 1,7 positions. These aromatic heterocyclic [a]-fused BODIPYs dyes exhibit "butterfly-shaped" crystal structures, with a tunable dihedral angle between the indacene plane and its "wings" ranging from 2.47 degrees to 11.49 degrees. The symmetrical [a]-fusion of dibenzothieno and dibenzothieno[2,3-d]thieno rings results in NIR absorption and emission with high fluorescence quantum yields (Phi(F) > 0.8), and effectively narrows the HOMO-LUMO gap by elevating the energy level of the HOMO. The high fluorescence quantum yields and tunable HOMO levels of these aromatic heterocyclic [a]-fused BODIPY dyes make them highly promising as functional chromophores, particularly in the field of bioimaging. This work offers valuable insights for developing NIR fluorescent dyes with high fluorescence quantum yields for use as photosensitizers.

Abstract:<p>At present, BODIPY and its derivatives are a kind of multifunctional photosensitive organic dyes, because of their small Stokes shift, strong absorption, narrow absorption and emission, low photobleaching, high fluorescence quantum efficiency, polarity, and pH insensitive and stable chemical structure, thus, it can be used in various fields such as medical treatment, energy, environmental protection and so on. This article mainly describes the application of BODIPY and its derivatives in photodynamic therapy, elaborates on its synthesis method, the role of photodynamic therapy, and discusses the factors that affect the location and efficacy of BODIPY photodynamic therapy. For the analysis and summary of BODIPY and its derivatives, we hope that this review can point the way for researchers.</p>

Abstract:The study of a macromolecule derived from DPP and triphenylamine, ( DPP-BisTPA ) by computational chemistry, its synthesis by direct arylation, optical characterization (UV-Vis and fluorescence) and electrochemistry (cyclic voltammetry), as well as its evaluation as a generator of reactive oxygen species indirectly, through the degradation of uric acid. The results obtained by DFT using B3LYP/6-31G (d, p) and TD-DFT using CAM-B3LYP/6-31G (d, p) reveal values of energy levels of the first singlet and triplet excited state that indicate a possible intersystem crossover and the possible generation of reactive oxygen species by a type I mechanism. The compound presents an absorption region within the phototherapeutic window. The electrochemical bandgap is 1.64 eV which suggests a behavior as a semiconductor. DPP-BisTPa were processed as hemispherical nanoparticles with a size around 100 nm, and NPOs were evaluated as a photosensitizer with a ROS generation yield of 4% using a photodynamic therapy flashlight as the light source.

Discovery of Subcellular-Targeted Aza-BODIPY Photosensitizers for Efficient Photodynamic Antitumor Therapy.

Abstract:In this study, we linked classical organelle-targeting groups, such as triphenylphosphonium, pentafluorobenzene, and morpholine, to our previously reported potent monoiodo Aza-BODIPY photosensitizer (BDP-15). They were conveniently prepared and retained the advantages of Aza-BODIPY PS with intense NIR absorption, moderate quantum yield, potent photosensitizing efficiency, and good stability. The in vitro antitumor assessment indicated that mitochondria-targeting and lysosome-targeting groups were more effective than ER-targeting groups. Considering undesirable dark toxicity of triphenylphosphonium-modified PSs, compound 6 containing amide-linked morpholine possessed a favorable dark/phototoxicity ratio (>6900 for tumor cells) and was localized in lysosomes with Pearson's coefficient of 0.91 to Lyso-Tracker Green DND-26. 6 exhibited significantly increased intracellular ROS production and resulted in early/late apoptosis and necrosis to disrupt tumor cells. Moreover, in vivo antitumor efficacy exploration suggested that even under a slightly low dose of light (30 J/cm2) and single-time photoirradiation, 6 retarded tumor growth dramatically and displayed much better PDT activity over BDP-15 and Ce6.

Abstract:Organic dyes with intensive near-infrared (NIR) absorption have shown practical application for fluorescence imaging. Based on Diketopyrrolopyrrole (DPP) and Boron-dipyrromethene (BODIPY), NIR dyad BD and triad BDB with two BODIPY units have been synthesized. There is a great steric hindrance existing between the DPP and BODIPY units; however, triad BDB shows a little more effective intramolecular charge transfer than dyad BD, as result in a stronger absorbance of the longest absorption band. According to theoretical calculations, the longest absorption maxima are contributed by the HOMO-LUMO transition for both BD and BDB. Both dyes were then fabricated into hydrophilic nanoparticles (NPs) via nano-precipitation method, the NIR dyes and the corresponding NPs all exhibit excellent photostability. Furthermore, the NPs show great biocompatibility, low dark cytotoxicity on HeLa cells. While taking Zebrafish as the live-animal model, BD NPs and BDB NPs show good fluorescence imaging property.

Abstract:Objective: To design and synthesize photosensitizers with different substituents and to identify its physicochemical characteritics and photodynamic effect on cancer cells. Methods: Two kinds of BODIPY photosensitizers BPOI and BPCI were synthesized through condensation reaction between aldehyde and reactive hydrogen of pyrrole, followed with electrophilic substitution reaction. Physicochemical properties were characterized by 1H NMR, FT-IR and UV-visible absorption spectra and fluorescence emission spectra. The ability to produce reactive oxygen species was detected by BPDF and DCFH-DA. Photodynamic therapy effect on rat glioma C6 cells in vitro was determined by MTT method. Results: Two kinds of BODIPY photosensitizers BPOI and BPCI were successfully synthesized with different substituents, which were confirmed by 1H NMR, FT-IR. Both materials had low toxicity and could be readily taken up by tumor cells. The ability of synthesized photosensitizers to produce reactive oxygen species was strongly influenced by solvent polarity when the substituent was electron-donating group, while no effect was found when the substituent was electron-withdrawing group. Conclusion: Photosensitizer BPOI with electron-donating substituent produces reactive oxygen species with a slow rate in a highly polar environment, while greatly enhanced this effect in a low polarity environment, which is expected to be used for environmental-selective photodynamic therapy in tumor cells.
