Abstract:The analysis of circulating tumor cells (CTCs) allows a noninvasive method of “real-time liquid biopsy” from the blood samples of cancer patients for the diagnosis of early-stage cancer, prognosis, and monitoring therapeutic response. In this study, we develop a simple, inexpensive, and reliable method that utilizes a small molecule peptide, the asparagine-glycine-arginine (NGR), as a capture probe for the selective enrichment and isolation of circulating tumor cells (CTCs). The multiscale TiO2 nanofibers are obtained by electrospinning and calcination. Bovine serum albumin (BSA) is decorated onto TiO2 nanofiber surfaces to inhibit non-target cell adhesion, while NGR peptides are conjugated onto the TiO2-BSA surface through the glutaraldehyde (GA) to specifically capture the target cells. The TiO2-BSA-NGR substrate exhibits a high capture sensitivity and efficiency from the mimical blood samples with PC-3 cancer cells as low as 10 cells/mL. The TiO2 nanofiber substrate can be a promising strategy for the capture and enumeration of CTCs in cancer progression monitoring.Graphic Abstract

Abstract:Circulating tumor cells (CTCs) have become known as minimally invasive multifunctional biomarkers for earlier diagnosis, prognosis, recurrence risk assessment, and therapeutic monitoring in recent years. However, effectively capturing these CTCs is still difficult because of the extremely low abundance of CTCs and the diverse phenotypes of cancer cells. In this study, we present a novel necklace-like polydopamine nanosphere (PDA NS)/alginate composite nanofiber with a hierarchical nanotopographical structure and a surface chemical signal for capturing the CTCs. The height of the nanotopography, which is formed by connecting PDA NSs with nanofibers via electrospinning, can be easily adjusted by changing the size of the PDA NSs. Abstract:Circulating tumor cell (CTC) detection and enumeration have been considered as a noninvasive biopsy method for the diagnosis, characterization, and monitoring of various types of cancers. However, CTCs are exceptionally rare, which makes CTC detection technologically challenging. In the past few decades, much effort has been focused on highly efficient CTC capture, while the activity of CTCs has often been ignored. Here, we develop an effective method for nondestructive CTC capture, release, and detection. Folic acid (FA), as a targeting molecule, is conjugated on magnetic nanospheres through a cleavable disulfide bond-containing linker (cystamine) and a polyethylene glycol (PEG2k) linker, forming MN@Cys@PEG2k-FA nanoprobes, which can bind with folate receptor (FR) positive CTCs specifically and efficiently, leading to the capture of CTCs with an external magnetic field. When approximately 150 and 10 model CTCs were spiked in 1 mL of lysis blood, 93.1 ± 2.9% and 80.0 ± 9.7% CTCs were recovered, respectively. In total, 81.3 ± 2.6% captured CTCs can be released from MN@Cys@PEG2k-FA magnetic nanospheres by treatment with dithiothreitol. The released CTCs are easily identified from blood cells for specific detection and enumeration combined with immunofluorescence staining with a limit of detection of 10 CTC mL-1 lysed blood. Moreover, the released cells remain healthy with high viability (98.6 ± 0.78%) and can be cultured in vitro without detectable changes in morphology or behavior compared with healthy untreated cells. The high viability of the released CTCs may provide the possibility for downstream proteomics research of CTCs; therefore, cultured CTCs were collected for proteomics. As a result, 3504 proteins were identified. In conclusion, the MN@Cys@PEG2k-FA magnetic nanospheres prepared in this study may be a promising tool for early-stage cancer diagnosis and provide the possibility for downstream analysis of CTCs.

Abstract:Cancer cells metastasize and are transported in the bloodstream, easily reaching any site in the body through the blood circulation. A method designed to assess the number of circulating tumor cells (CTCs) should be validated as a clinical tool for predicting the response to therapy and monitoring disease progression in patients with cancer. Although CTCs are detectable in many cases, they remain unavailable for clinic usage due to their high testing cost, tedious operation and poor clinical relevance. Herein, we developed a regeneratable microchip for isolating CTCs that is available for robust cell heterogeneity assays on-site without the need for a sterile environment. The ivy-like hierarchical roughened zinc oxide (ZnO) nanograss interface was synthesized and directly integrated into the microfluidic devices and enables effective CTC capture and flexible, nontoxic CTC release during incubation in a mildly acidic solution, thus enabling cellular and molecular analyses. The microchip can be regenerated and recycled to capture CTCs with the remaining ZnO without affecting the efficiency, even after countless cycles of cell release. Moreover, microbial infection is avoided during its storage, distribution and even in open-space usage, which ideally appeals to the demands of point-of-care (POC) and home testing and meets to the requirements for blood examinations in undeveloped or resource-limited settings. Furthermore, the findings generated using this platform based on the cocktail of anti-epithelial cell adhesion molecule (EpCAM) and anti-vimentin antibodies indicate that CTC capture was more precise and reasonable for patients with advanced cancer.

Abstract:Circulating tumor cells (CTCs) in the blood which have detached from both the primary tumor and any metastases may be considered as a "liquid biopsy" and are expected to replace tumor biopsies in the monitoring of treatment response and determining patient prognosis. Here, we introduce a facile and efficient CTC detection material made of hydroxyapatite/chitosan (HA/CTS), which is beneficial because of its transparency and excellent biological compatibility. Atomic force microscopy images show that the roughness of the HA/CTS nanofilm (HA/CTSNF) substrates can be controlled by changing the HA:CTS ratio. Enhanced local topographic interactions between nano-components on cancer cell membranes, and the antibody coated nanostructured substrate lead to improved CTC capture and separation. This remarkable nanostructured substrate has the potential for CTC culture in situ and merits further analysis. CTCs captured from artificial blood samples were observed in culture on HA/CTSNF substrates over a period of 14 days by using conventional staining methods (hematoxylin eosin and Wright's stain). We conclude that these substrates are multifunctional materials capable of isolating and culturing CTCs for subsequent studies.

Abstract:Circulating tumor cells (CTCs) play a significant role in cancer diagnosis and personalized therapy, and it is still a significant challenge to efficiently capture and gently release CTCs from clinical samples for downstream manipulation and molecular analysis. Many CTC devices incorporating various nanostructures have been developed for CTC isolation with sufficient capture efficiency, however, fabricating such nanostructured substrates often requires elaborate design and complicated procedures. Here we fabricate a degradable zinc-phosphate-based hierarchical nanosubstrate (HZnPNS), and we demonstrate its excellent CTC-capture performance along with effective cell-release capability for downstream molecular analysis. This transparent hierarchical architecture prepared by a low-temperature hydrothermal method, enables substantially enhanced capture efficiency and convenient imaging. Biocompatible sodium citrate could rapidly dissolve the architecture at room temperature, allowing that 88 ± 4% of captured cells are gently released with a high viability of 92 ± 1%. Furthermore, antiepithelial cell adhesion molecule antibody functionalized HZnPNS (anti-EpCAM/HZnPNS) was successfully applied to isolate CTCs from whole blood samples of cancer patients, as well as release CTCs for global DNA methylation analysis, indicating it will serve as a simple and reliable alternative platform for CTC detection.

Abstract:The method for detection of circulating tumor cells (CTCs) is considered as a promising cancer liquid biopsy technique. However, due to the rarity and heterogeneity, it is still a challenging task to capture CTC with high purity and viability from blood for the following enumeration and molecular analysis. Herein, we fabricated a hydrogel-chitosan nanofiber substrate to effectively capture the target cancer cells, assisted by the poly(carboxybetaine methacrylate) (pCBMA) hydrogel and the anti-epithelial cell adhesion molecule (anti-EpCAM) antibody. The pCBMA hydrogel layer was fabricated by free radical polymerization to provide an antifouling surface, which could help to reduce the non-specific capture of blood cells to improve the capture purity of CTC. Then, chitosan nanofibers were introduced onto the hydrogel layer to provide a nanostructured surface to improve the capture efficiency. Finally, the capture yield of artificial blood samples on the anti-EpCAM modified hydrogel-nanofiber substrate was up to 79.9%, revealing the potential application of the hydrogel-nanofiber substrate on efficient and sensitive capture of rare circulating tumor cells.
Selective capture of circulating tumor cells by antifouling nanostructure substrate made of hydrogel nanoparticles

Abstract:<p>The detection and analysis of circulating tumor cells (CTCs) from cancer patients' blood samples present a powerful means to monitor cancer progression. In this work, an antifouling nanostructure substrate made of hydrogel nanoparticles was fabricated for an effective capture of CTCs from the blood samples. The hydrogel nanoparticles were synthesized by zwitterionic sulfobetaine methacrylate (SBMA), methacrylic acid (MAA) and N, N'-methylene bisacrylamide (MBA) through a simple polymerization. SBMA could provide an effective antifouling layer for the substrate to prevent nonspecific cell adhesion, MAA could afford active carboxyl groups for the immobilization of antibody to achieve specific CTC capture, and the nanostructured surface could improve the interaction of the target cells with the antibody modified substrate surface to enhance the capture efficiency of CTCs. Moreover, it was not necessary to further modify the antifouling molecules on the hydrogel nanoparticle substrate's surface, reducing the complexity and difficulty of the substrate preparation. The results showed that about 87% of target cells (MCF-7 cells) were captured on the antibody modified hydrogel nanoparticle substrate. In contrast, the substrate showed little adhesive capacity for the nonspecific cells (K562 cells), and only 0.15% of cells were captured. And 98% of the captured cells kept good cell viability. Finally, 1-32 CTCs/mL were detected from the blood samples of five cancer patients, while no CTC was found in five healthy samples. It is envisaged that the new hydrogel nanostructure substrate is capable of capturing CTCs efficiently and specifically from patient blood samples to be used in cancer treatment.</p>

Abstract:Circulating tumor cell (CTC) detection has become a powerful tool for early diagnosis of metastasis and precise monitoring of cancer progression. While existing microchips have achieved promising progress on CTC capture, facile release and in situ regulation of CTC from devices for downstream analysis have rarely been realized. Herein, we demonstrated a porous nanoflower substrate (PNFS)-embedded microfluidic device that integrated multiple functionalities of effective CTC capture, in situ intracellular delivery, and nondestructive CTC release on a single device. The core of the device, PNFS, consisting of hier- archical Zn-phosphate composite structure on a nanoporous membrane, was produced by converting from micro/nano-fabricated ZnO nanostraws by low -temperature hydrothermal method and could be degraded via sodium citrate solution treatment for 20 min. Cancer cells could be captured on the PNFS with high efficiency up to 93.9 +/- 1.7% after specific antibody conjugations, and be non-destructively released from the PNFS by using biocompatible sodium citrate to rapidly dissolve the PNFS at room tem- perature. In addition, the degradable PNFS was integrated with a microfluidic electroporation device, allowing in situ intracellular molecular delivery into the captured cells for cell modifications, where such reengineered cells could be further released for subsequent applications. This device promises to be a versatile and efficient platform for CTC analysis, with great potential for the futuristic cell -based cancer diagnostics and therapies. (C) 2020 Elsevier Ltd. Abstract:The detection of circulating tumor cells (CTCs), an approach considered to be "liquid biopsy", is crucial in cancer diagnosis, monitoring and prognosis. However, the extremely large number of blood cells challenges the rare CTC isolation and enrichment. In this report, a red blood cell membrane mimetic surface (CMMS) is fabricated on material-independent substrates to repel blood cell adhesion. Meanwhile, tumor cell targeting ligands, folic acid (FA) and an arginine-glycine-aspartic acid (RGD) peptide, are tethered on the CMMS to give the decorated surface (CMMS-FA-RGD) tumor cell capture ability. The CMMS is composed of a mussel-inspired self-adhesive polydopamine layer and a covalently anchored non-fouling or anti-cell-adhesion layer of a phosphorylcholine zwitterion polymer and poly(ethylene glycol) (PEG). The protruding ends of the PEG chains of the anchored CMMS are further coupled with FA and RGD ligands to endow the tumor cells with specific binding. Furthermore, all the components of the step-by-step constructed surfaces are quantitatively controllable for optimizing the non-specific cell repellence and tumor cell binding performances. Thus, the delicately engineered CTC capture surface enhances the HeLa cell enrichment factor to 19 000-fold by repelling the adhesion of >99.999% blood cells, resulting in high capture efficiency (91%) and capture purity (89%) from the spiked whole blood samples. This substrate independent tumor cell capture and blood cell repellent surface modification strategy may provide a facile, versatile and cost-effective technology solution for more efficient cancer diagnosis and targeted therapy.

Abstract:The study developed a simple, effective and inexpensive strategy for capturing, enriching and detecting circulating tumor cells (CTCs) by using folic acid (FA) as the targeting molecule instead of antibodies. This work constructed magnetic halloysite nanotubes (MHNTs) coated with biocompatible beta-cyclodextrin (CD), and conjugated to FA via a PEG-Ad linker, to specifically capture the FA receptor (FR)-overexpressing cancer cells. The capture efficiencies of MHNTs@beta-CD@Ad-PEG-FA for the Skov3, Hela and A549 cancer cells were 96.3%, 97.0% and 95.6% respectively. In addition, the nanoparticles were able to capture very low numbers of the cancer cells (25-500 cells/mL) from PBS and whole blood, as well as selectively capture the cancer cells over normal HEK 293 T cells. Furthermore, the captured cells were viable and grew normally in vitro, indicating the future potential of downstream analyses. This approach can be adapted for different CTCs, once the tumor-specific surface markers are identified and the efficacy of targeting ligands is established. Taken together, FA-conjugated MHNTs nanoparticles are a highly promising tool for isolating CTCs for the diagnosis and treatment of cancer.

Abstract:Here we describe a fluorescent microspheres-based separation and analysis that enables the isolation of circulating tumor cells (CTCs) from whole blood of patients with metastatic cancer and the identification of isolated CTCs in situ without immunostaining. This approach uses antibody-functionalized fluorescent polystyrene (PS) microspheres that can selectively bind to CTCs. The binding of CTCs and fluorescent PS microspheres leads to the formation of complexes of CTCs and fluorescent PS microspheres, thereby the CTCs are size-amplified and labeled simultaneously. A pyramidal microcavity array (PMCA) is fabricated using microfabrication technology to create a precise microfilter structure with a high aspect ratio. The PMCA filter device can effectively isolate microspheres-labeled CTCs, while allow hematologic cells to deform and pass through. Using this approach, CTCs are isolated and identified in 15 of 18 patients with metastatic colorectal cancer. This approach will open new possibilities for CTCs isolation and identification and can serve a versatile platform to facilitate CTCs analysis in diverse biomedical applications.
Engineered Red Blood Cells For Capturing Circulating Tumor Cells With High Performance

Abstract:Filtration of circulating tumor cells (CTCs) in peripheral blood is of proven importance for early cancer diagnosis, treatment monitoring, metastasis diagnosis, and prognostic evaluation. However, currently available strategies for enriching CTCs, such as magnetic activated cell sorting (MACS), face serious problems with purity due to nonspecific interactions between beads and leukocytes in the process of capturing. In the present study, the tumor-targeting molecule folic acid (FA) and magnetic nanoparticles (MNPs) were coated on the surface of red blood cells (RBCs) by hydrophobic interaction and chemical conjugation, respectively. The resulting engineered RBCs rapidly adhered to CTCs and the obtained CTC-RBC conjugates were isolated in a magnetic field. After treatment with RBC lysis buffer and centrifugation, CTCs were released and captured. The duration of the entire process was less than three hours. Cell counting showed that the capture efficiency was above 90% and the purity of the obtained CTCs was higher than 75%. The performance of the proposed method exceeded that of MACS (R) beads (80% for capture efficiency and 20% for purity) under the same conditions. The obtained CTCs could be successfully re-cultured and proliferated in vitro. Our engineered RBCs have provided a novel method for enriching rare cells in the physiological environment.

Abstract:Circulating tumor cells (CTCs) played a significant role in early diagnosis and prognosis of carcinomas, and efficient capture of CTCs was highly desired to provide important and reliable evidence for clinical diagnosis. In present work, we successfully synthesized functional magnetic Fe3O4/P(MMA-AA) composite nanoparticles (FCNPs) inspired by a counterbalance concept for recognition and capture of CTCs. This counterbalance, composed of polyethylene glycol (PEG) suppressing cell adhesion and anti-epithelial-cell-adhesion-molecule (anti-EpCAM) antibody targeting tumor cells, could both enhance the specific capture of tumor cells and reduce unspecific adhesion of normal cells. The study showed that the PEG density on the surface of the FCNPs affected the specificity of the materials, and a density of ca. 15% was efficient for reducing the unspecific adhesion. After incubation with the mixture of HepG2 cells and Jurkat T cells, the FCNPs reached a capture efficiency as high as about 86.5% of the cancer cells, suggesting great potential on detection of CTCs in the diagnoses and prognoses of cancer metastasis.

Abstract:Herein, we developed an inexpensive titanium dioxide (TiO2) nanofiber substrate for efficient and selective capture of circulating tumor cells (CTCs) from mimic patients' samples. The TiO2 nanofiber substrates were fabricated by electrospinning in combination with the calcination process. The surface of nanofiber substrates was modified with the anti-adhesion molecule, bovine serum albumin (BSA) and the nucleolin aptamer AS1411, wherein, aptamer AS1411 specifically binds to the nucleolin protein overexpressed on the membrane surface of cancer cells. The formed TiO2 nanofiber substrates exhibited high efficacy and specificity to capture nucleolin positive cells through synergistic topographic interactions. Using the rare number of cell capture experiments, the capture efficiency of up to 75 % was achieved on the surface of the nanofiber substrate for rare number target cells spiked in the white blood cells (WBCs) from 1 mL whole blood samples. In conclusion, this study highlighted the potential of the TiO2-BSA-biotin-AS1411 nanofiber substrate as a highly efficient platform to realize the selective and specific capture of rare CTCs in the clinical settings.

Abstract:The capture of circulating tumor cells (CTCs) is ofgreat significance in reducing cancer mortality andcomplications. However, the nonspecific binding ofproteins and white blood cells (WBCs) weakens thetargeting capabilities of the capture surfaces, whichcritically hampers the efficiency and purity of thecaptured CTCs. Herein, we propose a liquid-like in-terface design strategy that consists of liquid-likepolymer chains and anti-EpCAM modification pro-cesses for high-purity and high-efficiency capture ofCTCs. The dynamicflexible feature of the liquid-likechains endows the modified surfaces with excellentantiadhesion property for proteins and blood cells.The liquid-like surfaces can capture the target CTCsand show high cell viability due to the environment-friendly surface modification processes. Whenliquid-like surface designs were introduced in thedeterministic lateral displacement (DLD)-patternedmicrofluidic chip, the nonspecific adhesion rate of WBCs was reduced by more thanfivefold comparedto that in the DLD chip without liquid-like interfacedesign, while maintaining comparable capture effi-ciency. Overall, this strategy provides a novel per-spective on surface design for achieving high purityand efficient capture of CTCs.

Abstract:Recovering pure and viable circulating tumor cells (CTCs) from blood has been a challenging task for molecular characterization and functional analysis, which has attracted wide attention these days. Herein, we fabricate a thermoresponsive chitosan nanofiber substrate to effectively capture, purify, and release the target cancer cells, assisted by PNIPAAm brushes and DNA hybridization. The PNIPAAm brushes are designed to enable WBCs to detach from aptamer-PNIPAAm-chitosan-nanofiber (aptamer-P-CNFs) surfaces during the conformational transition. Meanwhile these specific captured CTCs are retained at a high purity. Moreover, effective and intact release of CTCs from the substrates without any foreign agents is realized by complementary sequences efficiently hybridizing with aptamers, and the specific cell release makes CTCs further purified. The present work provides a new strategy in the design of biointerface for recovering target CTCs from whole blood samples with high purity.
A Supersensitive CTC Analysis System Based on Triangular Silver Nanoprisms and SPION with Function of Capture, Enrichment, Detection, and Release

Abstract:Detection of circulating tumor cells (CTCs) may be applied for diagnosis of early tumors like a liquid biopsy. However, the sensitivity remains a challenge because CTCs are extremely rare in peripheral blood. In this study, we developed a supersensitive CTC analysis system based on triangular silver nanoprisms (AgNPR) and superparamagnetic iron oxide nanoparticles (SPION) with function of capture, enrichment, detection, and release. The AgNPR was encoded with MBA (i.e., 4-mercaptobenzoic acid) and modified with rBSA (i.e., reductive bovine serum albumin) and FA (i.e., folic acid) generating organic/inorganic composite nanoparticle MBA-AgNPR-rBSA-FA, which has the function of surface-enhanced Raman scattering (SERS). The optimized SERS nanoparticles (i.e., MBA3-AgNPR-rBSA4-FA2) can be utilized for CTC detection in blood samples with high sensitivity and specificity, and the LOD (i.e., limit of detection) reaches to five cells per milliliter. In addition, the SPION was also modified with rBSA and FA generating magnetic nanoparticle SPION-rBSA-FA. Our supersensitive CTC analysis system is composed of MBA3-AgNPR-rBSA4-FA2 and SPION-rBSA-FA nanoparticles, which were applied for capture (via interaction between FA and FRα), enrichment (via magnet), and detection (via SERS) of cancer cells from blood samples. The results demonstrate that our supersensitive CTC analysis system has a better sensitivity and specificity than the SERS nanoparticles alone, and the LOD is up to 1 cell/mL. The flow cytometry and LSCM (i.e., laser scanning confocal microscope) results indicate the CTCs captured, enriched, and isolated by our supersensitive CTC analysis system can also be further released (via adding excessive free FA) for further cell expansion and phenotype identification.

Abstract:Circulating tumor cells (CTCs) serve as crucial indicators for tumor occurrence, progression, and prognosis monitoring. However, achieving high sensitivity and high purity capture of CTCs remains challenging. Additionally, in situ capture and synchronous clearance hold promise as methods to impede tumor metastasis, but further exploration is needed. In this study, biomimetic cell membrane-coated magnetic nanoparticles (NPs) were designed to address the issue of nonspecific adsorption of capture probes by the immune system during blood circulation. Membranes from human breast cancer cells (tumor cell membranes, TMs) and leukocytes (white blood cell membranes, WMs) were extracted and fused to form a hybrid membrane (HM), which was further modified onto the surface of porous magnetic NPs loaded with indocyanine green (ICG). The incorporation of TM enhanced the material's target specificity, thus increasing capture efficiency, while WM coating reduced interference from homologous white blood cells (WBCs), further enhancing capture purity. Additionally, in conjunction with our novel inverted microfluidic chip, this work introduces the first use of polymer photonic crystals as the capture interface for CTCs. Besides providing an advantageous surface structure for CTC attachment, the 808 nm photonic bandgap effectively amplifies the 808 nm excitation light at the capture surface position. Therefore, upon capturing CTCs, the ICG molecules in the probes facilitate enhanced photothermal (PTT) and photodynamic (PDT) synergistic effects, directly inactivating the captured CTCs. This method achieves capture efficiency and purity exceeding 95% and permits in situ inactivation post-capture, providing an important approach for future research on impeding tumor metastasis in vivo.

Abstract:Circulating tumor cells (CTCs) have been recognized as significant research target for cancer prognosis and metastasis mechanism. Efficient and all-subpopulations-covered isolation and facile release of CTCs with good viability are pivotal for profound mechanism research and molecular profiling. However, integrated nanoplatform with these two functions is yet to be elucidated. Among existing release techniques including physical, biological, and chemical methods, acid responsive release of CTCs attracts intensive interests due to its straightforward and easily accessible feature. However, extra acidic substance seems indispensable so far, which poses potential hazard to the rare and susceptible CTCs. To address this challenge, polyelectrolyte functionalized CaCO3 coated magnetic nanocrystal clusters are developed as the nanoplatform for multi-subpopulations isolation and pH-sensitive release of CTCs. Only buffer solution components are needed rather than other acidic substance to provide mild acidic environment with pH value of 6.5, facilitate highly efficient release. Moreover, the bioefficacy of CTC capture and mild condition recovery is not compromised by formation of protein corona on the cell-friendly nanoplatform. This nanoplatform exhibits good performance in both mimic blood samples and cancer patients' clinical samples.
