Abstract:Biologically derived hierarchical structural materials not only boast energy‐efficient processing but also exhibit impressive mechanical performance. Silk stands as the gold standard in hierarchical fiber production, leveraging a unique combination of advantages. Nevertheless, the artificial replication of silk poses technical challenges related to precision processing and comprehensive molecular control. To address such issues, this study investigates the hierarchical assembly of solid regenerated silk in an air atmosphere, facilitated by the incorporation of carbon nanotube (CNT) seeding. Results obtained highlight that this CNT seeding facilitates multiscale structure development in response to post‐spin tensile stress. Such CNT bridged structure assembly bypasses some natural processing control variables (pH, ions) and the necessary solvent immersed state for conventional silk post‐drawing. Combining secondary electron hyperspectral imaging and 3D synchrotron X‐ray ptychotomography, this study reports silk protein conversion from a disordered as‐spun state to a longitudinal orientated semi‐crystalline nano structure during drawing. The development of microscale structure during the drawing process is attributed to the presence of CNTs, yielding mechanical properties comparable to, and frequently surpassing, those exhibited by native fibers. These findings collectively propose a framework for exploring novel processing routes and offer a practical means controlling self‐assembly in silk materials.

Abstract:Silkworms have been developed over thousands years to optimize folding and crystallization of fibroin under highly controlled conditions which have resulted in their efficient fiber formation. In this paper, we reconstructed the three-dimensional architecture of the spinneret of a wild silkworm Samia cynthia ricini from approximately 1000 optical micrographs of the semi-thin cross sections. The chitin plates and muscles were observed in the silk press part together with large change in the diameter of the spinneret lumen at the press part by large shear stress. This is similar to the case of the spinneret of Bombyx mori silkworm, indicating that the structural change in the silk fibroin of S.c. ricini silkworm occurs exclusively at the silk press part due to large shear stresses. Molecular dynamics (MD) calculations were then performed to study the structural change that occurs in the crystalline region of S.c. ricini silk fibroin under shear stress. Namely, using the peptide AGGAGG(A)12GGAGAG as a model of the crystalline part of the silk fibroin under different shear stresses in the presence of water molecules and followed by molecular mechanics (MM) calculation after removal of water molecules. The simulation indicates that the Ala residues in the model peptides adopt a predominantly β-sheet structure under shear stresses of above 1.0GPa.

Abstract:Silk and elastin are exemplary protein materials that exhibit exceptional material properties. Silk is uniquely strong, surpassing engineering materials such as Kevlar and steel, while elastin has exquisite flexibility and can reversibly fold into a more structured form at high temperatures when many other proteins would unfold and denature. This phenomenon in elastin is termed the inverse temperature transition. It is a reversible, controllable process that motivates applications in drug delivery, shape change materials, and biomimetic devices. Silk-elastinlike protein polymers (SELPs), which combine repeating B. mori silk and elastin blocks, have been introduced as biologically inspired materials that combine the distinctive properties of the component parts to achieve strong and extensible, tunable biomaterials. Abstract:Silk fibers are one of the most attractive natural materials that exhibit enchanting luster and superior mechanical properties. Exploring the unique hierarchical architecture structures of natural silk fibers is the basis to understanding these unique properties. Here, we report observations of 3.1 +/- 0.8 nm silk nanofibrils and 3.7 +/- 0.9 A silk molecule chains exfoliated from natural silkworm silk fibers. Interestingly, the individual nanofibrils and protein chains show periodic diameters fluctuating along their axes. We further find that the thicker regions are relatively softer and the thinner regions are stiffer, and these can be assigned to alternatively distributed alpha-helix and beta-sheet domains, respectively. Based on these observations, we proposed a refined structure model of natural silk, which ranges from the molecular level to the fiber scale. These findings provide new opportunities to understand and exploit the unique structure-property relationships found in natural silk fibers, including inspiring the design of new artificial materials.

Abstract:Silk features exceptional mechanical properties such as high tensile strength and great extensibility, making it one of the toughest materials known. The exceptional strength of silkworm and spider silks, exceeding that of steel, arises from β-sheet nanocrystals that universally consist of highly conserved poly-(Gly-Ala) and poly-Ala domains. This is counterintuitive because the key molecular interactions in β-sheet nanocrystals are hydrogen bonds, one of the weakest chemical bonds known. Here we report a series of large-scale molecular dynamics simulations, revealing that β-sheet nanocrystals confined to a few nanometres achieve higher stiffness, strength and mechanical toughness than larger nanocrystals. We illustrate that through nanoconfinement, a combination of uniform shear deformation that makes most efficient use of hydrogen bonds and the emergence of dissipative molecular stick–slip deformation leads to significantly enhanced mechanical properties. Our findings explain how size effects can be exploited to create bioinspired materials with superior mechanical properties in spite of relying on mechanically inferior, weak hydrogen bonds.

Abstract:A comprehensive review on the five levels of hierarchical structures of silk materials and the correlation with macroscopic properties/performance of the silk materials, that is, the toughness, strain-stiffening, etc., is presented. It follows that the crystalline binding force turns out to be very important in the stabilization of silk materials, while the β-crystallite networks or nanofibrils and the interactions among helical nanofibrils are two of the most essential structural elements, which to a large extent determine the macroscopic performance of various forms of silk materials. In this context, the characteristic structural factors such as the orientation, size, and density of β-crystallites are very crucial. It is revealed that the formation of these structural elements is mainly controlled by the intermolecular nucleation of β-crystallites. Consequently, the rational design and reconstruction of silk materials can be implemented by controlling the molecular nucleation via applying sheering force and seeding (i.e., with carbon nanotubes). In general, the knowledge of the correlation between hierarchical structures and performance provides an understanding of the structural reasons behind the fascinating behaviors of silk materials.

A Nanofibril Network Model of Biological Silks

Abstract:Biological silks produced by, for examples, spiders and silkworms exhibit exceptional mechanical properties, including high strength, toughness, and extensibility, which arise from their optimized hierarchical structures. Understanding the relationship between their structures and properties is crucial for the design of bioinspired materials. However, it remains a challenge to recapitulate their macroscopic mechanical behaviors of biological silks with a micromechanical model that integrates the physical mechanisms of deformation and failure, such as molecular unfolding and nanofibrous remodeling. In this paper, we propose a nanofibril network (NFN) model to analyze the deformation, damage, and fracture processes of biological silks. The NFN model reveals how molecular unfolding affects the deformation and fracture of biological silks at the nanofibrous scale. It is observed that molecular unfolding consecutively occurs, evolving into band or ellip-tical morphologies with increasing tensile strain. Intermolecular hydrogen bonds tend to stabilize the expansion of unfolding zones. The NFN model also unveils how the instability of molecular unfolding is suppressed by nanofibrous architecture and how intermolecular hydrogen bonds interact with intramolecular unfolding to toughen the biological silk. This work provides a theoretical tool for studying and designing biomimetic nanofibrous materials.

Abstract:Natural silk fibers achieve remarkable toughness by exploiting hierarchical structure organizations over many length scales. These hierarchical architectures are often decorated with defects that usually lead to catastrophic failure of artificial polymer materials. Silk fibers are self-tolerant of intrinsic/artificial defects to give surprising fracture toughness, but how they fulfill this function and the underlying mechanism remain elusive. Here, we unveil this puzzle by experimentally and theoretically investigating the tensile and facture behaviors of Antheraea pernyi (A. pernyi) silk monofilaments containing an artificial notch of controlled depth. The introduced transverse notch can be blunted effectively by ahead developed longitudinal crack through fibrillar separation. The notched silk fibers show ductile failure with tensile behavior dominated by the magnitude of longitudinal crack, which depends not only on the notch size but also on the interface interaction between nanofibrils. We propose that the highly oriented, disentangled nanofibrillar structure of A. pernyi silk offers it distinct tensile properties, path of crack propagation, and fracture mode in comparison with highly oriented synthetic fibers.

Abstract:Silk protein builds up one of the strongest fibers superior to most synthetic and natural polymers. However, the strengthening mechanisms of the silk proteins remain largely elusive because of their complex nanocomposite structures. Here, we report an unusual behavior of this kind of material that is distinctively different from those of metals and other polymers. We find that there are multiple interface microcracks nucleating and stacking under the shear loading, dividing the interchain interface into small segments, by which the silk protein can achieve a high strength even with the ultralong chains. This is a new strategy of microstructure design of soft matter that could avoid the "larger is weaker" fate due to the increase of the chain length. This novel mechanism is crucial for building strong polymer materials with long chain molecules and at the same time retaining their complex functional and structural properties.

Abstract:Natural materials, such as silk, nacre, and bone, possess superior mechanical properties which are derived from their unique hierarchical structures. Individual carbon nanotubes (CNTs) are considered as one of the strongest materials. However, macroscopic CNT fibers usually have breaking strength far below that of individual CNTs. In this work, by mimicking the structure of natural silk fibers, strong and stiff CNT fibers are prepared by infiltrating silk fibroin (SF) into CNT fibers. There are abundant hydrogen bonds in SF, contributing to the enhanced interactions between neighboring CNTs. Glycerol is selected to promote the formation of β‐sheet conformation in SF, leading to further enhanced strength and modulus. Remarkably, the SF infiltrated CNT fibers show breaking strength of 1023 MPa, toughness of 10.3 MJ m−3, and Young's modulus of 81.3 GPa, which are 250%, 132%, and 442% of the pristine CNT fibers. The structure of the SF and the interactions between CNTs and SF are studied via Fourier transformed infrared spectroscopy and molecular dynamics simulation. Mimicking the hierarchical structures of natural silk fibers and enhance the interfacial load transfer by infiltrating SF are effective for reinforcing CNT fibers, which may be useful in the design and preparation of other structural materials.

Abstract:Ductile and damage-tolerant fibers (DDTFs) are desired in aerospace engineering, mechanical engineering, and biomedical engineering because of their ability to prevent the catastrophic sudden structural/mechanical failure. However, in practice, design and fabrication of DDTFs remain a major challenge due to finite fiber size and limited processing techniques. In this regard, animal silks can provide inspirations. They are hierarchically structured protein fibers with an elegant trade-off of mechanical strength, extensibility and damage tolerance, making them one of the toughest materials known. In this article, we confirmed that nanofibril organization could improve the ductility and damage-tolerance of silk fibers through restricted fibril shearing, controlled slippage and cleavage. Inspired by these strategies, we further established a rational strategy to produce polyamide DDTFs by combining electrospinning and yarn-spinning techniques. The resultant polymeric DDTFs show a silk-like fracture resistance behavior, indicating potential applications in smart textile, biomedicine, and mechanical engineering.Graphic Abstract

Abstract:The heat‐induced self‐assembly of silk fibroin (SF) is studied by combing fluorescence assessment, infrared nanospectroscopy, wide‐angle X‐ray scattering, and Derjaguin−Muller−Toporov coupled with atomic force microscopy. Several fundamental issues regarding the formation, structure, and mechanical performance of silk nanofibrils (SNFs) under heat‐induced self‐assembly are discussed. Accordingly, SF in aqueous solution is rod‐like in shape and not micellar. The formation of SNFs occurs through nucleation‐dependent aggregation, but the assembly period is variable and irregular. SF shows inherent fractal growth, and this trend is critical for the short‐term assembly. The long‐term assembly of SF, however, mainly involves an elongation growth process. SNFs produced by different methods, such as ethanol treatment and heat incubation, have similar secondary structure and mechanical properties. These investigations improve the in‐depth understanding of fundamental issues related to self‐assembly of SNFs, and thus provide inspiration and guidance in designing of silk nanomaterials.

Biomimicking the structure of silk fibers via cellulose nanocrystal as β-sheet crystallite

Abstract:Inspired by the ordered beta-sheet crystalline structure and molecular orientation of natural silkworm silk, biomimetic silk fibers with refined crystalline structure were creatively produced through incorporating cellulose nanocrystals (CNCs) into silk fibroin (SF) matrix to mimic the beta-sheet crystallites in natural silk via wet-spinning assembly technology. The influence of CNCs and post-draw on the structural characteristics, and thermal and mechanical properties of regenerated cellulose nanocrystal/silk fibroin (CNC/SF) fibers were comparatively studied with those of degummed silk. The resultant CNC/SF fibers exhibited a uniform, circular shape as well as dense morphology. The CNCs were uniformly dispersed into SF matrix and aligned along the fiber axis, which were beneficial to the formation of more ordered structure through intermolecular hydrogen bonding interactions. The crystallinity and overall molecular orientation of CNC/SF fibers were increased with increases of draw ratio and CNC contents, although lower than those of degummed silk. DSC and TGA analysis revealed that thermal decomposition behavior of CNC/SF fibers presented different features depending on the CNC contents, whereas maximum decomposition temperature of CNC/SF fibers improved sharply at first and then decreased slightly with the increase of CNC contents. Moreover, the tan delta peak temperatures of SF were increased with the addition of CNC, indicating restrained SF molecular mobility in the vicinity of the CNC surface. These results indicated that the CNC could substantially enhance the mechanical properties of SF and this enhancement could be attributed to the strong hydrogen bonding interactions as well as CNC-induced crystallization and orientation. Meanwhile, Young's modulus, tensile strength and breaking strain of the CNC/SF fibers with 5 wt% CNC were significantly increased to 28.8 +/- 2.6 GPa, 728.5 +/- 36.4 MPa and 23.1 +/- 1.6% respectively, higher than those of degummed silk. Thus it was demonstrated that biomimicking the natural silk crystalline structure using CNC as beta-sheet crystallite is a promising strategy for production of artificial silk fiber with improved strength without compromising the toughness.

Abstract:Spider dragline silk is distinguished through the highest toughness of all natural as well as artificial fiber materials. To unravel the toughness’s molecular foundation and to enable manufacturing biomimetic analogues, we investigated the morphological and functional structure of recombinant fibers, which exhibit toughness similar to that of the natural template, on the molecular scale by means of vibrational spectroscopy and on the mesoscale by X-ray scattering. Whereas the former was used to identify protein secondary structures and their alignment in the natural as well as artificial silks, the latter revealed nanometer-sized crystallites on the higher structural level. Furthermore, a spectral red shift of a crystal-specific absorption band demonstrated that macroscopically applied stress is directly transferred to the molecular scale, where it is finally dissipated. Concerning this feature, both the natural as well as the biomimetic fibers are almost indistinguishable, giving rise to the toughness of both fiber materials.

Abstract:Animal silks are built from pure protein components and their mechanical performance, such as strength and toughness, often exceed most engineered materials. The secret to this success is their unique nanoarchitectures that are formed through the hierarchical self-assembly of silk proteins. This natural material fabrication process in sharp contrast to the production of artificial silk materials, which usually are directly constructed as bulk structures from silk fibroin (SF) molecular. In recent years, with the aim of understanding and building better silk materials, a variety of fabrication strategies have been designed to control nanostructures of silks or to create functional materials from silk nanoscale building blocks. These emerging fabrication strategies offer an opportunity to tailor the structure of SF at the nanoscale and provide a promising route to produce structurally and functionally optimized silk nanomaterials. Here, we review the critical roles of silk nanoarchitectures on property and function of natural silk fibers, outline the strategies of utilization of these silk nanobuilding blocks, and we provide a critical summary of state of the art in the field to create silk nanoarchitectures and to generate silk-based nanocomponents. Further, such insights suggest templates to consider for other materials systems.

Abstract:Abstract Silk is a unique, remarkably strong biomaterial made of simple protein building blocks. To date, no synthetic method has come close to reproducing the properties of natural silk, due to the complexity and insufficient understanding of the mechanism of the silk fiber formation. Here, we use a combination of bulk analytical techniques and nanoscale analytical methods, including nano-infrared spectroscopy coupled with atomic force microscopy, to probe the structural characteristics directly, transitions, and evolution of the associated mechanical properties of silk protein species corresponding to the supramolecular phase states inside the silkworm’s silk gland. We found that the key step in silk-fiber production is the formation of nanoscale compartments that guide the structural transition of proteins from their native fold into crystalline β -sheets. Remarkably, this process is reversible. Such reversibility enables the remodeling of the final mechanical characteristics of silk materials. These results open a new route for tailoring silk processing for a wide range of new material formats by controlling the structural transitions and self-assembly of the silk protein’s supramolecular phases.

Optimal Protein Sequence Design Mitigates Mechanical Failure in Silk β-Sheet Nanocrystals

Abstract:The excellent mechanical strength and toughness of spider silk are well characterized experimentally and understood atomistically using computational simulations. However, little attention has been focused on understanding whether the amino acid sequence of β-sheet nanocrystals, which is the key to rendering strength to silk fiber, is optimally chosen to mitigate molecular-scale failure mechanisms. To investigate this, we modeled β-sheet nanocrystals of various representative small/polar/hydrophobic amino acid repeats for determining the sequence motif having superior nanomechanical tensile strength and toughness. The constant velocity pulling of the central β-strand in the nanocrystal, using steered molecular dynamics, showed that homopolymers of small amino acid (alanine/alanine-glycine) sequence motifs, occurring in natural silk fibroin, have better nanomechanical properties than other modeled structures. Further, we analyzed the hydrogen bond (HB) and β-strand pull dynamics of modeled nanocrystals to understand the variation in their rupture mechanisms and explore sequence-dependent mitigating factors contributing to their superior mechanical properties. Surprisingly, the enhanced side-chain interactions in homopoly-polar/hydrophobic amino acid models are unable to augment backbone HB cooperativity to increase mechanical strength. Our analyses suggest that nanocrystals of pristine silk sequences most likely achieve superior mechanical strength by optimizing side-chain interaction, packing, and main-chain HB interactions. Thus, this study suggests that the nanocrystal β-sheet sequence plays a crucial role in determining the nanomechanical properties of silk, and the evolutionary process has optimized it in natural silk. This study provides insight into the molecular design principle of silk with implications in the genetically modified artificial synthesis of silk-like biomaterials.

Abstract:There is an emerging interest in natural silkworm silks as alternative reinforcement for engineering composites. Here, we summarize the research on two common silkworm silks and silk fibre reinforced plastics (SFRPs) from the authors over the past few years in the context of related research. Silk fibres from silkworms display good strength and toughness under ambient and cryogenic conditions owing to their elastic-plastic deformation mechanism. In particular, the wild Antheraea pernyi (A. pernyi) silk also displays micro- and nano-fibrillation as an important mechanism for toughness and impact resistance. For SFRP composites, we found: (i) it is critical to achieve silk fibre volume fraction to above 50% for an optimal reinforcement and toughening effect; (ii) the tougher A. pernyi silks present a better reinforcement and toughening agent than B. mori silks; (iii) impact and toughness properties are advantageous properties of SFRPs; (iv) hybridization of natural silk with other fibres can further improve the mechanical performance and economics of SFRPs for engineering applications; and (v) the lightweight structure designs can improve the service efficiency of SFRPs for energy absorption. The understanding on the comprehensive mechanical properties and the toughening mechanisms of silks and silk fibre-reinforced polymer composites (SFRPs) could provide key insights into material design and applications.
Bioinspired Tough and Strong Fibers with Hierarchical Core-Shell Structure

Abstract:Strong and tough bio-based fibers are attractive for both fundamental research and practical applications. In this work, strong and tough hierarchical core-shell fibers with cellulose nanofibrils (CNFs) in the core and regenerated silk fibroins (RSFs) in the shell are designed and prepared, mimicking natural spider silks. CNF/RSF core-shell fibers with precisely controlled morphology are continuously wet-spun using a co-axial microfluidic device. Highly-dense non-covalent interactions are introduced between negatively-charged CNFs in the core and positively-charged RSFs in the shell, diminishing the core/shell interface and forming an integral hierarchical fiber. Meanwhile, shearing by microfluidic channels and post-stretching induce a better ordering of CNFs in the core and RSFs in the shell, while ordered CNFs and RSFs are more densely packed, thus facilitating the formation of non-covalent interactions within the fiber matrix. Therefore, CNF/RSF core-shell fibers demonstrate excellent mechanical performances; especially after post-stretching, their tensile strength, tensile strain, Young's modulus, and toughness are up to 635 MPa, 22.4%, 24.0 GPa, and 110 MJ m(-3), respectively. In addition, their mechanical properties are barely compromised even at -40 and 60 degrees C. Static load and dynamic impact tests suggest that CNF/RSF core-shell fibers are strong and tough, making them suitable for advanced structural materials.

Abstract:Silk protein fibres produced by silkworms and spiders are renowned for their unparalleled mechanical strength and extensibility arising from their high-β-sheet crystal contents as natural materials. Investigation of β-sheet-oriented conformational transitions in silk proteins at the nanoscale remains a challenge using conventional imaging techniques given their limitations in chemical sensitivity or limited spatial resolution. Here, we report on electron-regulated nanoscale polymorphic transitions in silk proteins revealed by near-field infrared imaging and nano-spectroscopy at resolutions approaching the molecular level. The ability to locally probe nanoscale protein structural transitions combined with nanometre-precision electron-beam lithography offers us the capability to finely control the structure of silk proteins in two and three dimensions. Our work paves the way for unlocking essential nanoscopic protein structures and critical conditions for electron-induced conformational transitions, offering new rules to design protein-based nanoarchitectures.
