Abstract:Modifications at polypeptide side chains have been found to affect conformational and properties,which is a vital strategy to prepare functional polypeptides.This contribution describes the synthesis of homo and co-polypeptides containing unsaturated bonds and thioether moieties,poly(S-allylcysteine)(PAC).The pendant vinyl groups provide an active binding site which are able to be further modified with cysteine by thiol-ene click reaction.The sulfoxide from thioether oxidation bestows the polypeptide water solubility and enzymatic catalytic sites.The self-assembly micelles of PEG-b-PAC are ruptured along with oxidation to release the Nile red payloads.Moreover,poly(S-allyl-cysteine sulfoxide),aka.polyalliin,is the oxidation product of PAC which undergoes side chain cleavage catalyzed by alliinase enzyme.The polypeptides are biocompati-ble as confirmed by cell viability assays and may find applications in drug delivery,biosensing and nanoreactor.

Abstract:Stimuli-responsive poly(alpha-amino acids) (P alpha AAs) are protein mimics that can alter their conformational or other physicochemical properties in response to external triggers. We report here the synthesis and characterization of novel photo and thermal responsive P alpha AAs by the ring-opening polymerization of alpha-amino acid N-carboxyanhydrides (NCAs). Specifically, a series of NCA monomers bearing azobenzene (Azo) and oligoethylene glycol (OEG) of different lengths (OEG(m)-AzoNCA, m = 2, 4, 6) are synthesized in a modular route and polymerized in a controlled manner using hexamethyldisilazane (HMDS) as the initiator. The afforded P(OEG(m)-Azo)(n)s show interesting aggregation behaviors in both organic and aqueous solutions. Moreover, P(OEG(m)-Azo)(n)s exhibit a reversible photo-responsiveness accompanied by the conformational switch of the peptidic backbone, and an irreversible thermal-responsiveness. This new biodegradable and stimuli-responsive system holds considerable promise for applications such as on-demand delivery, liquid crystalline, and hierarchical self-assembly.

Abstract:ABSTRACTA series of side‐chain‐functionalized α‐helical polypeptides, i.e., poly(γ‐4‐(3‐chloropropoxycarbonyl)benzyl‐L‐glutamate) (6) have been prepared from n‐butylamine initiated ring‐opening polymerization (ROP) of γ‐4‐(3‐chloropropoxycarbonyl)benzyl‐L‐glutamic acid‐based N‐carboxyanhydride. Polypeptides bearing oligo‐ethylene‐glycol (OEG) groups or 1‐butylimidazolium salts were prepared from 6 via copper‐mediated [2+3] alkyne‐azide 1,3‐dipolar cycloaddition or nuleophilic substitution, respectively. CD and FTIR analysis revealed that the polymers adopt α‐helical conformations both in solution and the solid state. Polymers bearing OEG (m = 3) side‐chains showed reversible LCST‐type phase transition behaviors in water while polymers bearing 1‐butylimidazolium and I− counter‐anions exhibited reversible UCST‐type transitions in water. Variable‐temperature UV‐vis analysis revealed that the phase transition temperatures (Tpts) were dependent on the main‐chain length and polymeric concentration. © 2015 Wiley Periodicals, Inc. J. Abstract:Well-defined azobenzene-containing side-chain liquid crystalline diblock copolymers composed of poly[6-(4-methoxy-azobenzene-4'-oxy) hexyl methacrylate] (PMMAZO) and poly(gamma-benzyl-L-glutamate) (PBLG) were synthesized by click reaction from alkyne- and azide-functionalized homopolymers. The alkyne-terminated PMMAZO homopolymers were synthesized by copper-mediated atom transfer radical polymerization with a bromine-containing alkyne bifunctional initiator, and the azido-terminated PBLG homopolymers were synthesized by ring-opening polymerization of gamma-benzyl-L-glutamate-N-carboxyanhydride in DMF at room temperature using an amine-containing azide initiator. The thermotropic phase behavior of PMMAZO-b-PBLG diblock copolymers in bulk were investigated using differential scanning calorimetry and polarized light microscopy. The PMMAZO-b-PBLG diblock copolymers exhibited a smectic phase and a nematic phase when the weight fraction of PMMAZO block was more than 50%. Photoisomerization behavior of PMMAZO-b-PBLG diblock copolymers and the corresponding PMMAZO homopolymers in solid film and in solution were investigated using UVvis. In solution, transcis isomerization of diblock copolymers was slower than that of the corresponding PMMAZO homopolymers. Abstract:Supramolecular polymers are able to change their structure, morphology and function in response to external stimuli. However, controlling the independence of stimuli-responses in these systems is challenging. Herein, we exploit halogen bonding (XB) as a reversible network element to regulate the photoresponsive and adaptive behavior of supramolecular polymers. To this end, we have designed a system comprising an amphiphilic XB acceptor with the ability to self-assemble in aqueous media (OPE-Py) and a molecule with a dual photoresponsive and XB donor function [(E)-Azo-I]. OPE-Py self-assembles in aqueous media into supramolecular polymers, which transform into nanoparticle assemblies upon co-assembly with (E)-Azo-I. Interestingly, a third type of assembly (2D sheets) is obtained if OPE-Py is treated with (E)-Azo-I and exposed to photoirradiation. At ambient conditions, both nanoparticles and 2D sheets remain invariant over time. However, heating dissociates the XB interactions present in both assemblies, resulting in their transformation to the original fiber-like morphology of OPE-Py. Thus, breaking the communication between self-assembly and the stimuli-responses upon heating restores the original state of the system, drawing parallels to feedback loops in programming language. This work broadens the still limited scope of XB in solution assemblies and paves the way for multifunctional adaptive supramolecular systems.

Theoretical Studies of A Novel Photo-Switchable Self-Organized Peptide System

Abstract:Synthetic organic functional nanomaterials have been the subject of intense study lately due to their potential use in many fields. As a result, theoretical studies of such materials have also received extensive attention. In this paper, seven models of a novel photo-switchable self-organized peptide system were optimized using the semi-empirical molecular orbital method AM1, and single point calculations of three of them were investigated by means of the density functional B3LYP/3-21G* method. The geometries, energetics and frontier orbital interactions of these photo-switchable peptide subsystems were calculated and analyzed. Our results provide insight into the formation, self-assembly and photo-isomerization of the system. Weak interactions between two polypeptide rings, especially hydrogen-bonding interactions, are crucial for stabilizing the conformation and self-assembly. Remarkably, oligomers of the E form show almost no cooperative effect, strongly supporting the notion that it is possible to break the intermolecular hydrogen bonds to enable E --> Z isomerization reactions. Moreover, the polypeptide rings strengthen the conjugation effect on azobenzene subunits and lead to a reduction in the frontier molecular orbital energy differences. The self-assembling process of the E form also reinforces frontier orbital interactions and favors E --> Z isomerization.
Synthesis and photoresponsive properties of two liquid crystalline polymers bearing branched azobenzene-containing side chains

Abstract:We synthesized two photoresponsive liquid crystalline polymers bearing branched azobenzene-containing side-chains. The side-chain liquid crystal polymers (SCLCPs) were designed to contain two and three azobenzene moieties in each monomeric unit, which were attached to the backbones via a linkage of biphenylene and flexible spacers. The SCLCPs were synthesized by radical polymerization of corresponding methacrylate-based monomers. The polymers were characterized by FT-IR, H-1 NMR, UV-vis, GPC and small angle X-ray scattering (SAXS). Both polymers formed the smectic-A mesophase in the LC states, whilst the polymer containing three azobenzene-containing branches in each repeat unit showed a high order mesophase in the lower temperature range. The polymers exhibited trans-cis photoisomerization and photoinduced orientation under light irradiation at suitable wavelengths.

Abstract:The synthesis of a series of new photoresponsive side-chain liquid crystalline polymethacrylates with amide group-substituted azobenzene (azo) mesogens and different length of flexible spacers and terminal tails via conventional free radical polymerization is described. The resulting azo polymers proved to have high thermal stability and good solubility in common organic solvents (e.g., tetrahydrofuran and chloroform). Differential scanning calorimetry, polarizing optical microscopy, and small angle X-ray scattering studies confirmed the presence of obvious enantiotropic smectic C liquid crystalline phases (with a bilayer lamellar structure) for all these polymers. The introduction of an amide group into the azo mesogen led to the formation of strong hydrogen bonding among the side chains of the polymers (as revealed by variable temperature FT-IR), which played a decisive role in forming and stabilizing the liquid crystalline mesophases of the polymers. In addition, the length of the flexible spacers and terminal tails also significantly influenced their phase transition behaviors. Furthermore, the photoresponsivity of the polymer solutions was verified and the effects of the molecular structures of the polymers on their photoresponsive properties were also studied.

Abstract:Azobenzene‐containing polymers (azopolymers) are a kind of fascinating stimuli‐responsive materials with broad and versatile applications. In this work, a series of syndiotactic C1 type azopolymers of Pm‐Azo‐Cn with side‐chain azobenzene mesogens of varied length alkoxy tails (n = 1, 4, 8, 10) and different length alkyl spacers (m = 6, 10) have been prepared via Rh‐catalyzed carbene polymerization. The thermal properties and ordered assembly structures of thus synthesized side chain liquid crystalline polymers (SCLCPs) have been systematically investigated with differential scanning calorimetry (DSC), polarized optical microscopy (POM) and variable‐temperature small/wide‐angle X‐ray scattering (SAXS/WAXS) analyses. P10‐Azo‐C1 and P10‐Azo‐C4 with shorter alkoxy tails exhibited hierarchical structures SmB/Colob and transformed into SmA/Colob at a higher temperature, while P10‐Azo‐C8 and P10‐Azo‐C10 with longer alkoxy tails only displayed side group dominated layered SmB phase and transformed into SmA phase at higher temperatures. For P6‐Azo‐C4 with a shorter spacer only showed a less ordered SmA phase owing to interference by partly coupling between the side chain azobenzene mesogens and the helical backbone. More importantly, the series high densely substituted syndiotactic C1 azopolymer thin films, exhibited evidently and smoothly reversible photoresponsive properties, which demonstrated promising photoresponsive device applications.

Abstract:ABSTRACTIn this study, we report the synthesis, characterization, and photo‐responsive properties of a new Y‐shaped amphiphilic azo triblock copolymer composed of two isotropic polyethylene glycol (PEG) blocks and an azobenzene liquid crystalline block. The azo block, with two ending groups suitable for the azo coupling reaction, is polymerized by atom transfer radical polymerization with a synthesized initiator containing two functional terminal groups. The macromolecular diazonium salts are prepared by the diazotization of PEG terminated with an amino group. The triblock copolymer is obtained by the azo coupling reaction between the azo block and macromolecular diazonium salts in DMF under mild condition. The intermediates and the obtained triblock copolymer are characterized by 1H NMR, FT‐IR, GPC, POM, DSC, TEM, and UV‐vis. The photoinduced isomerization behavior of the azo copolymer is investigated by UV‐vis. With the addition of water into the solution of the triblock copolymer, spherical aggregates with an average diameter of about 400 nm can be easily obtained. Abstract:A series of novel azo polyelectrolytes have been synthesized based on an extremely reactive precursor polymer, poly(acryloyl chloride) (PAC), prepared from acryloyl chloride by radical polymerization. The precursor polymer was post-functionalized by the Schotten-Baumann reaction of PAC and several aromatic azo reactants containing hydroxyl end groups. The degrees of functionalization were controlled by selecting suitable feed ratios between the azo reactants and poly(acryloyl chloride) and the unreacted acyl chloride groups were hydrolyzed to obtain ionizable carboxyl groups. The products were characterized by elemental analysis, FT-IR, H-1 NMR and UV-vis spectroscopy. Irradiated by 365 nm UV light, azo polyelectrolytes PPAPE, PEAPE and PCAPE showed a significant photochromic effect. The contact angles of water on the surfaces of spin-coated films of PPAPE and PEAPE decreased evidently upon UV irradiation. The extent of the photoinduced contact angle changes depends on the type of the azo chromophores and the degree of functionalization. Self-assembled multilayers of the azo polyelectrolytes were fabricated by a layer-by-layer adsorption method. A significant photochromic effect from cis-trans isomerization of the azo chromophores was observed for the multilayers. The photoinduced contact angle changes of water on the self-assembled multilayers were also observed.

Abstract:The synthesis of a series of novel photoresponsive side-chain liquid crystalline polymers (LCPs) bearing an N-hydroxysuccinimide carboxylate-substituted azobenzene mesogen and their use in generating LCP networks are described. The obtained polymers and their monomers were characterized with H-1 NMR, UV-vis, TGA, DSC, POM and SAXS techniques. The length of the flexible spacers in the polymers and their monomers show significant influence on their properties such as the phase transition behaviors and the monomer reactivity. The polymerization reactivity of the monomers increases with an increase in the spacer length and only those monomers and polymers with a flexible spacer >= (CH2)(6) show liquid crystallinity with their mesophases being smectic A. In addition, the polymers have proven to be easily cross-linkable with a difunctional primary amine under mild conditions both in solution and in film, demonstrating that they are very useful precursors for photoresponsive LCP networks. Furthermore, the photoresponsivity of the polymer solution in tetrahydrofuran and the crosslinked polymer film was also confirmed by their occurrence of the UV and visible light-induced trans/cis photoisomerization.

Abstract:Polystyrenes (PSs) that provide photochemical stability are commonly used as the chromophore macromolecular carriers. Herein, PS-based alternating copolymers, i.e., alt-P(D-SiH/Ip), alt-P(D-2SiH/Ip) and alt-P(D-SiH/St), are synthesized through anionically copolymerized Si-H functionalized 1,1-diphenylethylene (DPE-SiH/2SiH) with isoprene (Ip)/styrene (St). Anionically polymerized (4-vinylphenyl)-dimethyl-silane is used as the uniform hydrosilylated PS, i.e., Uni.-P(St-SiH). A series of photo-responsive azobenzene side-chain liquid crystalline PSs combine the above PS-based main chains and the mesogenic moiety composed of azobenzene (Azo) and cholesterol (Chol) groups, i.e., Azo-Chol (M), via the hydrosilylation. The polarized optical microscopic (POM) morphologies and thermal properties of Azo-Chol (M) and Azo-Chol-PSs are studied. As desired, they produce the cholesteric phase of wide temperature range due to the PSs and Chol-groups. Moreover, the photo-responsive behavior irradiated under UV-light of different wavelengths are quantitatively evaluated by time-dependent UV-Vis spectroscopy. Azo-Chol-PSs undergo rapid and reversible trans-cis isomerization under the alternating irradiation of UV and visible light. The trans-to -cis isomerization efficiency under 365 nm UV-light is over that under 290-320 nm UV-light, while over 80% of cis-to-trans efficiency can be realized under 400-500 nm Vis-light irradiation This work would provide guidelines for the effective design of photo-responsive anisotropic materials.
Manipulating the helix-coil transition profile of synthetic polypeptides by leveraging side-chain molecular interactions

Abstract:Polypeptides with trigger-responsive helix-coil transition behaviours are interesting biomaterials due to their helix-specific assemblies and biomedical performances. Based on the pH-sensitive, conformationally switchable triazole polypeptides, we reported the manipulation of the helix-coil transition profile, which was determined by the combined molecular interactions of triazole and other side-chain functionalities. Specifically, the introduction of side-chain hydrophobic moieties or hydrogen bonding acceptors neutralized the helix-disrupting effect of side-chain triazoles, which altered the pH-responsive conformational transition profile of the polypeptides. These results inspired us to design new triazole polypeptides bearing dimethylamino side chains, which exhibited interesting helix-coil-helix transition behaviours as the pH decreased.

Abstract:Polypeptide hybrid triblock copolymer, poly(L-glutamic acid)-b-poly(propylene oxide)-b-poly (L-glutamic acid) (PLGA-b-PPO-b-PLGA), was synthesized by the ring-opening polymerization of benzyl-L-glutamic N-carboxyanhydride (BLG-NCA) using poly(propylene glycol) bis(2-aminopropyl ether) as initiator, followed by the subsequent deprotection step. The obtained double hydrophilic triblock copolymer exhibits “schizophrenic” micellization behavior in aqueous solution upon dually playing with solution pH and temperature. The multi-responsive micellization behavior of this polypeptide hybrid triblock copolymer has been thoroughly investigated by 1H NMR, laser light scattering (LLS), temperature-dependent optical transmittance, and circular dichroism spectroscopy (CD).

Abstract:Stimuli-responsive polymeric materials have attracted significant attention in a variety of high-value-added and industrial applications during the past decade. Among various stimuli, light is of particular interest as a stimulus because of its unique advantages, such as precisely spatiotemporal control, mild conditions, ease of use, and tunability. In recent years, a lot of effort towards the synthesis of a biocompatible and biodegradable polypeptide has resulted in many examples of photo-responsive nanoparticles. Depending on the specific photochemistry, those polypeptide derived nano-assemblies are capable of crosslinking, disassembling, or morphing into other shapes upon light irradiation. In this mini-review, we aim to assess the current state of photo-responsive polypeptide based nanomaterials. Firstly, those ‘smart’ nanomaterials will be categorized by their photo-triggered events (i.e., crosslinking, degradation, and isomerization), which are inherently governed by photo-sensitive functionalities, including O-nitrobenzyl, coumarin, azobenzene, cinnamyl, and spiropyran. In addition, the properties and applications of those polypeptide nanomaterials will be highlighted as well. Finally, the current challenges and future directions of this subject will be evaluated.

Abstract:Novel polycatenar liquid crystals containing two 1,3,4-thiadiazole rings interconnected by an azobenzene central linkage have been synthesized and investigated by polarizing microscopy, DSC, X-ray scattering, SEM, UV-vis spectroscopy and photoluminescence measurements. These compounds can self-assemble into SmC, Col(hex)/p6mm and Cub(I)/Pm (3) over barn liquid crystalline phases in the bulk states and form multistimuli responsive organogels in organic solvents. They have reversible photoresponsive properties in solution, liquid crystalline states and gel states. They also show fluorescence emission with large Stokes shift in solution and binding selectivity to Cu2+ among a series of cations in CH3CN-CH2Cl2 solution.

Abstract:The synthesis of novel (co‐)aspartates bearing ortho‐fluorinated azobenzenes as photochromic group in the sidechain is presented. The influence of photo‐induced E‐Z‐isomerization on the secondary structure is studied revealing helix‐to‐helix and helix‐to‐coil transitions. Polyaspartates forming LLC phases in organic solvents allow studying the effect of photoswitching on macroscopic behavior with distinct anisotropic environments becoming accessible by irradiation with visible light. Many polypeptides form stable, helical secondary structures enabling the formation of lyotropic liquid crystalline (LLC) phases. Contrary to the well‐studied polyglutamate, their counterparts based on polyaspartates exhibit a much lower helix inversion barrier. Therefore, the helix sense is not solely dictated by the chirality of the amino acid used, but additionally by the nature and conformation of the polymer sidechain. In this work, polymers responsive to irradiation with visible light are designed achieving conformational transitions from helix‐to‐coil and helix‐to‐helix. The synthesis and the application as LLC mesogens of several (co‐)polyaspartates bearing ortho‐fluorinated azobenzene (FAB) as a photochromic group are presented. Many of the obtained polymers undergo changes in their secondary structure upon E‐Z‐isomerization of the FAB‐containing sidechain. Of special interest are copolymers that exhibit photo‐responsive helix inversion without loss of their helical secondary structure. These copolymers form stable LLC phases in helicogenic solvents, where the effect of photo‐switching on the macroscopic behavior is studied by NMR spectroscopy. Especially, the irradiation of the different LLC phases of the helix inversion polymers displays a change in the LLC order experienced by the solvent. These peculiar properties are promising for future applications as photo‐responsive alignment media for structure elucidation in NMR.
