Abstract:As an important chemical component of grapes, viniferins are essential dimers of natural product resveratrol with good antioxidant activity. In this study, we have examined the antioxidant activity of two viniferin molecules (δ-viniferin and ε-viniferin) with •OH/•OOH radicals. Density functional theory (DFT) calculations at the M062X/6-311++G(d,p)-SMD//M062X/6-311G(d,p) level of theory in the gas phase, water, and benzene used to examine two crucial radical scavenging pathways: hydrogen atom transfer (HAT) and the radical adduct formation (RAF). Then, the radical scavenging activities of the two molecules are compared kinetically and thermodynamically. The Gibbs free energy of activation (ΔG≠) and rate constant (kTST) are calculated using the conventional transition state theory (TST), and the results reveal that HAT and RAF processes compete in different solvents. Furthermore, molecular electrostatic potential (MEP) and frontier molecular orbital (FMO) of both the compounds are computed to elucidate their antioxidant activity. The results show that the RAF site appears at the >C7=C8< double bond position of δ- and ε-viniferins, and the most favorable HAT positions of δ-viniferin and ε-viniferin are 12′-OH and 12-OH, respectively; in addition, ε-viniferin is a more potent radical scavenging agent than δ-viniferin. These findings provide useful insights on the antioxidant activity of viniferins and different radical scavenging mechanisms of antioxidants, which can be beneficial for improving the practical utilization of resveratrol dimer and other analogs.

Abstract:<p>Viniferin, the dimer of resveratrol, is an important natural product class with anti-oxidant activities. δ-viniferin and ε-viniferin are two different isomers of viniferin. In this context, a systematic theoretical study by density functional theory (DFT) calculations is carried out on these two isomers' radical scavenging activity, which focuses on the thermodynamically preferred mechanism, anti-oxidant sites, and anti-oxidant activity sequence of δ-viniferin and ε-viniferin. Three radical scavenging mechanisms named hydrogen atom transfer (HAT), single electron transfer followed by proton transfer (SET-PT) and sequential proton loss electron transfer (SPLET) were discussed. The anti-oxidative capacities of δ-viniferin and ε-viniferin in the gas phase and solvents were elucidated by comparing the thermodynamic parameters such as bond dissociation enthalpy (BDE), ionization potential (IP), proton dissociation enthalpy (PDE), proton affinity (PA), and electron transfer enthalpy (ETE). The computational results showed that δ-viniferin is a more potent anti-oxidant than ε-viniferin. The obtained results may have some important implications for better understanding the relationship between structure and anti-oxidant activity, designing more active anti-oxidants, and providing theoretical guidance for developing and utilizing resveratrol derivatives.</p>

Abstract:The structural and antioxidant activity properties of three flavonols kaempferol, galangin and morin have been investigated at density functional level of theory with the aim of verifying experimental findings. The potentialities of antioxidant activity are highly related to their capabilities to scavenge free radicals. Two potential working mechanisms of the hydrogen-atom transfer and single-electron transfer are reported by which antioxidants can play their role. Two parameters of the O-H bond dissociation enthalpy (BDE) and ionization potential (IP) in the presence of water medium are computed to estimate the antioxidant capacities. Results indicate that the order of antioxidant efficacies predicted theoretically in this work is in agreement with that reported by experimental results of oxygen radical-scavenging capacity (ORAC) assay. This demonstrates the importance of the hydrogen-atom and single-electron transfer mechanisms to explain their capacities to scavenge peroxyl radical.
Evaluation of resveratrol derivatives as potential antioxidants and identification of a reaction product of resveratrol and 2, 2-diphenyl-1-picryhydrazyl radical

Abstract:Resveratrol (3,4‘,5-trihydroxy-trans-stilbene), an antioxidant from grapes, and five other polyhydroxystilbenes were synthesized. Their antioxidative properties were evaluated in two model systems [pure lipid oxidation using the Rancimat method and 2,2-diphenyl-1-picryhydrazyl (DPPH) free radical scavenging model.] 3,3‘,4,5‘-Tetrahydroxystilbene, 3,3‘,4,5,5‘-pentahydroxystilbene, and 3,4,4‘,5-tetrahydroxystilbene were found to be more active than resveratrol in both models. A dimer of resveratrol was identified as the major radical reaction product when resveratrol was reacted with DPPH radicals. Insights into the Inhibitory Mechanism of Viniferifuran on Xanthine Oxidase by Multiple Spectroscopic Techniques and Molecular Docking

Abstract:Viniferifuran was investigated for its potential to inhibit the activity of xanthine oxidase (XO), a key enzyme catalyzing xanthine to uric acid. An enzyme kinetics analysis showed that viniferifuran possessed a strong inhibition on XO in a typical anti-competitive manner with an IC50 value of 12.32 μM (IC50 for the first-line clinical drug allopurinol: 29.72 μM). FT-IR and CD data analyses showed that viniferifuran could induce a conformational change of XO with a decrease in the α-helix and increases in the β-sheet, β-turn, and random coil structures. A molecular docking analysis revealed that viniferifuran bound to the amino acid residues located within the activity cavity of XO by a strong hydrophobic interaction (for Ser1214, Val1011, Phe914, Phe1009, Leu1014, and Phe649) and hydrogen bonding (for Asn768, Ser876, and Tyr735). These findings suggested that viniferifuran might be a promising XO inhibitor with a favorable mechanism of action.

Abstract:Aim:In order to improve the solubility and stability of resveratrol,nanoethosome was prepared to encapsulate resveratrol to study its anti-oxidation effect.Methods:Nanoethosomes loaded resveratrol were prepared by ethanol injection method.The anti-oxidative effects of resveratrol nanoethosomes in vitro were evaluated by DPPH free radical scavenging assay,ABTS free radical scavenging assay and ferrous reduction method (FRAP) respectively.Results:The results demonstrated that resveratrol nanoethosomes had higher clearance rate than free resveratrol on DPPH and ABTS.The FeSO4equivalent of resveratrol nanoethosomes were also higher than free resveratrol.Conclusion:These results indicate that nanoethosomes can enhance the antioxidant capacity of resveratrol in vitro.
Theoretical Insight into the Antioxidative Activity of Isoflavonoid: the Effect of the C2=C3 Double Bond.

Abstract:Isoflavonoids are one of the most important groups of naturally occurring antioxidants. Their structural features are important for evaluating their antioxidative activity. In this work, density functional theory (DFT) methods were applied to investigate the influence of the C2=C3 double bond on the antioxidative activity of isoflavonoids based on three currently accepted radical scavenging mechanisms from the viewpoint of thermodynamics. The C2=C3 double bond can make the compounds more flat, which would extend the conjugated system in the molecule and make the isoflavonoids higher antioxidant activity. The C2=C3 double bond would not alter the strongest antioxidative hydroxyl group of the isoflavonoids. In the gas, benzene and CHCl3 phases, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids by lowering the bond dissociation enthalpies of the hydroxyl groups in the B ring that are the strongest antioxidative sites for the hydrogen atom transfer (HAT) mechanism. In polar phases, a similar result is obtained by weakening the proton affinity of 7-OH that is the strongest antioxidative hydroxyl group in the sequential proton loss electron transfer (SPLET), mechanism. Thus, the C2=C3 double bond will enhance the antioxidative activity of isoflavonoids irrespective of the studied phases.

Abstract:Viniferin is the generic term of oligomers of resveratrol, which acts as phytoalexin in Leguminosae, Polygonaceae, Vitaceae, Ranunculaceae, Dipterocarpaceae and other plants. Viniferin plays important physiological roles in protecting against UV damage and resisting bacterial fungal and viral infection in plants. Nevertheless, these oligomers have shown various pharmacological activities including antioxidative activities, anti-pathogenic, anti-inflammatory and anti-tumor activities. This paper review the recent advances in research of viniferins microbes to show their key pharmaceutical activities for pharmaceutic references.

Abstract:Resveratrol monomer (Res) and its oligomers are considered as nutritional components distributed in edible plants. Three naturally occurring resveratrol dimers, namely parthenocissin A (Par), quadrangularin A (Qua) and pallidol (Pal), were synthesized and evaluated for their ability to scavenge reactive oxygen species (ROS) and to activate the transcription factor Nrf2, which regulates cellular antioxidant systems. In vitro studies with different ROS and radical assay models showed that all the three dimers are strong DPPH quenchers and selective singlet oxygen ((1)O2) scavengers (IC50=4.90, 1.05 and 5.50 μM, respectively). However, they were ineffective against hydroxyl radical (OH) or superoxide anion (O2(-)). Exposing the dimers to an antioxidant response element (ARE) reporter cell line revealed that only pallidol was able to activate Nrf2 at 30 μM, while parthenocissin A and quadrangularin A had no significant effect on Nrf2. Our data demonstrates the distinct difference between reservatrol monomer and its dimers in activating the Nrf2/ARE signalling pathway.

Abstract:The molecular structure and radical scavenging activity of three novel antioxidants from Lespedeza Virgata, lespedezavirgatol, lespedezavirgatal, and lespedezacoumestan, have been studied using density functional theory with the B3LYP and BhandHLYP methods. The optimized geometries of neutral, radical cation, radical and anion forms were obtained at the B3LYP/6–31G(d) level, in which it was found that all the most stable conformations contain intramolecular hydrogen bonds. The same results were obtained from the MP2 method. The homolytic O—H bond dissociation enthalpy and the adiabatic ionization potential of neutral and anion forms for the three new antioxidants and adiabatic electron affinity and H-atom affinity for hydroxyl radical, superoxide anion radical, and hydrogen peroxide radical were determined both in gas phase and in aqueous solution using IEF-PCM and CPCM model with UAHF or Bondi cavity. The antioxidant activities and reactive oxygen species scavenging mechanisms were then discussed, and the results obtained from different methods are consistent. Furthermore, the antioxidant activities are consistent with the experimental findings of the compounds under investigation.
Synthesis and anti-oxidative activities of resveratrol analogs

Abstract:Objective: To synthesize the resveratrol analogs with more potent anti-oxidative activities. Methods: The A ring of resveratrol was substituted by TMP ring or 4-aminoquinoline ring. The DPPH radical scavenging abilities of the target molecules 9, 12 and intermediate 8, 11 were tested. Results and Conclusion: Two novel resveratrol analogues were synthesized, and their structures were confirmed by MS and 1H-NMR. Compoud 8 possesses more potent DPPH radical scavenging ability than resveratrol at low concentration and holds potent anti-oxidative activity.

Abstract:Viniferin is a resveratrol derivative. Resveratrol is the most prominent stilbenoid synthesized by plants as a defense mechanism in response to microbial attack, toxins, infections or UV radiation. Different forms of viniferin exist, including alpha-viniferin (α-viniferin), beta-viniferin (β-viniferin), delta-viniferin (δ-viniferin), epsilon-viniferin (ε-viniferin), gamma-viniferin (γ-viniferin), R-viniferin (vitisin A), and R2-viniferin (vitisin B). All of these forms exhibit a range of important biological activities and, therefore, have several possible applications in clinical research and future drug development. In this review, we present a comprehensive literature search on the chemistry and biosynthesis of and the diverse studies conducted on viniferin, especially with regards to its anti-inflammatory, antipsoriasis, antidiabetic, antiplasmodic, anticancer, anti-angiogenic, antioxidant, anti-melanogenic, neurodegenerative effects, antiviral, antimicrobial, antifungal, antidiarrhea, anti-obesity and anthelminthic activities. In addition to highlighting its important chemical and biological activities, coherent and environmentally acceptable methods for establishing vinferin on a large scale are highlighted to allow the development of further research that can help to exploit its properties and develop new phyto-pharmaceuticals. Overall, viniferin and its derivatives have the potential to be the most effective nutritional supplement and supplementary medication, especially as a therapeutic approach. More researchers will be aware of viniferin as a pharmaceutical drug as a consequence of this review, and they will be encouraged to investigate viniferin and its derivatives as pharmaceutical drugs to prevent future health catastrophes caused by a variety of serious illnesses.

New Theoretical Perspective for Easy Evaluation of the Antioxidant Properties of Typical Flavonoids

Abstract:To intensively investigate the antioxidant property of flavonoids and their different phenolic hydroxyl groups, we selected six types of typical flavonoids and carried out experimental analysis and quantum chemical calculations. By calculating the Mulliken charges, frontier molecular orbitals, molecular dipole moments, and interaction model hydrogen bond lengths of the compounds, the theoretical analysis results were compared with the free radical scavenging assays. Results show that the three flavonoids with different numbers of hydroxyl groups on the B ring are ranked in antioxidant activity as myricetin > quercetin > kaempferol. The glycoside linked in the C ring has a great effect on bioavailability, and the antioxidant activity is ranked as quercetin > isoquercetin > rutin. Besides, the results of the novel interaction model calculations show that quercetin with an meta-hydroxy group on the B ring structure has stronger antioxidant capacity than morin with an ortho-hydroxy group. The theoretical evaluation method established in this work is expected to provide new ideas and references for the evaluation of antioxidant properties of natural active substances from the electronic structure perspective.
The Antioxidant Effect of Imine Resveratrol Analogues

Abstract:Twenty five Imine resveratrol analogues (IRAs) were synthesized, replacing the CC bond in resveratrol with CN bond, as well as substitution modifications on aromatic rings. Radical scavenging activities against DPPH, along with singlet oxygen quenching capacities were evaluated, and further confirmed using density functional theory calculations (DFT). It was found that IRAs bearing ortho-OH on B ring have better radical scavenging activities against DPPH than resveratrol, these compounds were also found to be effective 1O2 quenchers. Theoretical studies on the reaction mechanism of these compounds with 1O2 suggest that the 1,3-addition to a double bond with a –OH group with the formation of allylic hydroperoxide is the most probable reaction route.
Isolation and Purification of Two Antioxidant Isomers of Resveratrol Dimer from the Wine Grape by Counter-Current Chromatography

Abstract:Resveratrol dimers belong to a group of compounds called stilbenes, which along with proanthocyanidins, anthocyanins, catechins, and flavonols are natural phenolic compounds found in grapes and red wine. Stilbenes have a variety of structural isomers, all of which exhibit various biological properties. Counter-current chromatography with a two-phase solvent system composed of n-hexane/ethyl acetate/methanol/water (2:5:4:5, v/v/v/v) was applied to isolate and purify stilbene from the stems of wine grape. Two isomers of resveratrol dimers trans-ε-viniferin and trans-δ-viniferin were obtained from the crude sample in a one-step separation, with purities of 93.2 and 97.5%, respectively, as determined by high-performance liquid chromatography. The structures of these two compounds were identified by (1) H and (13) C NMR spectroscopy. In addition, their antioxidant activities were assessed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. The antioxidant activities of trans-δ-viniferin were higher than that of trans-ε-viniferin in this model. This work demonstrated that counter-current chromatography is a powerful and effective method for the isolation and purification of polyphenols from wine grape. Additionally, the DPPH radical assay showed that the isolated component trans-δ-viniferin exhibited stronger antioxidant activities than trans-ε-viniferin and a little bit weaker than vitamin E at the same concentration.
Radical Scavenging Activity of Myricetin

Abstract:Density functional theory (DFT) calculations have been performed to explore the molecular structure, electronic structure, and O-H bond dissociation enthalpy of myricetin. Possible antioxidation mechanisms between lipid peroxide radical CH3OO center dot and myricetin have been discussed. DFT calculations at the M06-2X/6-31++G(d, p) level indicated that the 4 '-OH group of myricetin is the most active site on the basis of the stability of dehydrogenated myricetin radicals, O-H bond dissociation enthalpy, and hydrogen abstraction activation barrier, as well as kinetic data for myricetin determined at different temperatures. The relatively high activity of the 4 '-OH site can be ascribed to weak hydrogen- bonding interactions between the oxygen radical of the reactive OH group and the adjacent OH group in the B- ring, which is retained upon going from free myricetin to reactant complex to product according to atoms in molecule (AIM) analysis. The hydrogen- bond helps to stabilize the electronic deficiency generated on the oxygen radical during the hydrogen abstraction reaction. All calculations are in agreement with the structure- activity relationship previously established for myricetin by considering its antioxidant activity. Present calculations provide theoretical basis for the designing new antioxidants.

Abstract:Anthocyanidins are an important class of plant pigment, in the present work the antioxidative properties of anthocyanidins have been explored by density functional theory calculations, three main antioxidative mechanisms, which include H atom transfer (HAT), single electron transfer (SET) and sequential proton loss electron transfer (SPLET), have been investigated at B3LYP/6-311G(d,p) level of theory. The O–H bond dissociation enthalpies (BDEs), ionization potentials (IPs), electron affinities (EAs), proton affinities (PAs) and electron transfer enthalpies (ETEs) are investigated in gas phase and aqueous solution. Results show 3-OH and 4′-OH posses lower BDE as compared to other OH groups, the substituents in B-ring influence 4′-OH BDE, but exhibit negligible influence on other OH BDEs. Among all investigated anthocyanidins, pelargonidin has the highest IP and EA in gas phase, substituent in ortho position of 4′-OH leads to remarkable decrease in IP and EA. OH substituent in B-ring almost show no influence on PAs and ETEs of 3-, 5-, and 7-phenolate anions, but OCH3 substituent influence them significantly. For PAs and ETEs of 4′-phenolate anion, two types of substituents both exhibit remarkable influence. Base on the simplest anthocyanidins, pelargonidin, molecular simplification has been performed to explore the necessary pharmacophores responsible for the antioxidative activity of anthocyanidins.

Abstract:The antioxidant capacities of the selected flavonoids quercetin, luteolin and taxifolin have been investigated at density functional level of theory with the aim of verifying the cellular antioxidant activity (CAA) values representative of experimental findings. The selected flavonoids were believed to act through the H-atom transfer mechanism. Their potentiality of hydrogen abstraction was evaluated by computing the OH bond dissociation enthalpy (BDE) in gas-phase and in dimethylsulfoxide solution. Results indicate that the order of antioxidant efficacies calculated in this work is in agreement with that reported by experimental results of CAA. Time-dependent density functional theory (TDDFT) calculations were also performed both in gas-phase and in dimethylsulfoxide to reproduce the electronic UV–vis spectra of the selected flavonoids.

Abstract:Density functional theory (DFT) calculations have been used to explore the molecular structure, electronic structure and O-H bond dissociation enthalpy of quercetin, one of flavonoid compounds. The relationships between the properties and the active site of quercetin are analyzed. Possible reaction mechanisms between center dot OH, center dot OOH and O-2(-center dot) with quercetin involved in the antioxidation activity of quercetin have been discussed. The DFT calculations under the level of B3LYP/6-31 + G(d) show that the 4'-OH of quercetin is the most active site on the basis of stability of dehydrogenated quercetin radicals, O-H bond dissociation enthalpy as well as activation energies of hydrogen abstraction from quercetin. This relatively high activity can be ascribed to weak hydrogen-bond interactions between two nearby hydroxides in quercetin molecule. The theoretical investigations on the antioxidation mechanism of quercetin are useful for the design and synthesis of novel antioxidants with enhanced activity.
