Abstract:Migrasomes are recently discovered cellular organelles that form as large vesicle-like structures on retraction fibres of migrating cells. While the process of migrasome formation has been described before, the molecular mechanism underlying migrasome biogenesis remains unclear. Here, we propose that the mechanism of migrasome formation consists of the assembly of tetraspanin- and cholesterol-enriched membrane microdomains into micron-scale macrodomains, which swell into migrasomes. The major finding underlying the mechanism is that tetraspanins and cholesterol are necessary and sufficient for migrasome formation. We demonstrate the necessity of tetraspanins and cholesterol via live-cell experiments, and their sufficiency by generating migrasome-like structures in reconstituted membrane systems. We substantiate the mechanism by a theoretical model proposing that the key factor driving migrasome formation is the elevated membrane stiffness of the tetraspanin- and cholesterol-enriched macrodomains. Finally, the theoretical model was quantitatively validated by experimental demonstration of the membrane-stiffening effect of tetraspanin 4 and cholesterol.

Abstract:Migrasomes are recently discovered signalling organelles, enriched with tetraspanin proteins (TSPAN)[1][1]. They form by local swelling of retraction fibers, the cylindrical protrusions of cell membranes that form as a result of cell migration along external substrates. Migrasomes can grow up to several micrometers in diameter[2][2], and allow cells to release contents such as chemokines at specific locations, hence, transmitting signals to surrounding cells through the relevant chemokine receptors. Recently, evidence emerged showing that migrasomes play essential roles in fundamental cellular processes such transfer of mRNA and proteins[3][3], organ morphogenesis[4][4], and mitochondria quality control[5][5]. Thus, understanding the mechanism of migrasome biogenesis is of outstanding importance. Previously, it was established that the molecules crucial for migrasome formation are tetraspanin proteins and cholesterol forming macrodomains in the migrasome membrane, while the physical forces driving local swelling of the retraction fibers originate from membrane tension and bending rigidity[1][1]. Abstract:The migrasome is a newly discovered organelle of migrating cells. Migrasomes play diverse physiological roles including mitochondrial quality control, lateral transfer of material between cells, and delivery of signaling molecules to spatially defined locations. The formation of migrasomes is dependent on tetraspanins, a group of membrane proteins containing four transmembrane domains, which form membrane microdomains named tetraspanin-enriched microdomains (TEMs). In this review, we will discuss the mechanisms for migrasome biogenesis, with a focus on the role of TEMs and the organizing principles underlying the formation of TEMs.
