Abstract:The three-component reaction between an isoquinoline, a dialkyl acetylenedicarboxylate and 2,2,2-trifluoro-1-phenylethanone involving a 1,4-dipolar cycloaddition cascade approach is developed using a continuous-flow microreactor. The reaction conditions (solvent, flow rate and temperature) are optimized leading to the synthesis 2-(trifluoromethyl)-2H-[1,3]oxazino[2,3-a]isoquinoline derivatives in 79-83% isolated yields, whilst eliminating the formation of 11bH-pyrido[2,1-a]isoquinoline-12,3,4-tetracarboxylate by-products. The continuous-flow method is also applied to similar reactions using aromatic aldehydes as the carbonyl component.

Abstract:A novel palladium-catalyzed [4 + 1] cycloaddition to give spirotetrahydrofuran oxindoles has been developed, in which diphenyl 2-oxoindolin-3-yl phosphates were used as the both electrophilic and nucleophilic C1 synthons at C-3 of the oxindole unit and 2-methylidenetrimethylene carbonate was used as the 1,4-dipole. The cycloannulation was performed at room temperature and provided the corresponding spirotetrahydrofuran oxindoles in good to excellent yields. The present method affords a new strategy for the construction of spirooxindole derivatives with unique three-dimensional structures.

Abstract:Asymmetric synthesis of spiro[indoline‐3,4‐pyrrolo [3,4‐b]pyridines] derivatives was first developed through organocatalytic cascade Knoevenagel/Michael/cyclization reaction using a quinidine ‐derived squaramide. Under the optimized conditions, the three‐component reaction of isatins, cyanoacetates, and 3‐aminomaleimides yield the desired heterocycle‐fused spirooxindoles in good yields (78–91%) with 53‐99% enantiomer excess (ee). Notably, this reaction enabled a broad substrate scope under mild conditions, and provided a convenient method for enantioselective construction of diverse spirooxindoles combined with dihydropyridine and maleimide skeleton, which brought great potential to build new bioactive chemical entities.

Abstract:An organocatalytic and highly diastereo- and enantioselective reaction of 3-aminooxindoles with 2-enoylpyridines for the synthesis of chiral spiro[pyrrolidin-3,2'-oxindole] derivatives has been achieved. With the cinchonidine-based thiourea catalyst, the asymmetric Michael/cyclization reaction sequence of 3-aminooxindoles with 2-enoylpyridines followed by the dehydration and deprotection with concentrated hydrochloric acid provided a wide range of chiral 3',4'-dihydrospiro[indoline-3,2'-pyrrol]-2-ones, bearing two adjacent tri- and tetrasubstituted stereocenters, in moderate to good yields with overall excellent stereoselectivities. The synthetic application of this methodology was presented by the scale-up experiment and transformation of product 5a into the spiro-oxindole-based pyrrolidine 6. Furthermore, a plausible transition state for this cascade reaction sequence was also proposed. And this work will provide a new way to access chiral spiro[pyrrolidin-3,2'-oxindole] derivatives.

Abstract:A novel electrochemical multicomponent cascade reaction of indole‐tethered alkenes with CF3SO2Na, n‐Bu4NI, and H2O has been developed, which proceeds via oxidation of CF3SO2Na, coupling with alkene, spirocyclization, oxidation of sulfinate, iodide substitution, and water coupling enabling the rapid assembly of iodomethyl spiropyrrolidinyl‐oxindoles in good yields. Comprehensive Summary A novel electrochemical multicomponent cascade reaction of indole‐tethered alkenes with CF3SO2Na and n‐Bu4NI has been developed, which enables the rapid assembly of spiropyrrolidinyl‐oxindoles in good yields. The experimental results and DFT calculations suggest that this reaction proceeds through the oxidation of CF3SO2Na, radical coupling with alkene, spirocyclization, oxidation of sulfinate, iodide substitution, and water coupling. This strategy features mild reaction conditions, easy‐to‐handle reactants, and good chemical yields. This finding not only enriches the research contents of indole‐tethered alkenes but also provides a green strategy for the construction of spiropyrrolidinyl‐oxindoles compared with the existing methodologies.

Abstract:We report a visible‐light‐mediated, catalyst‐free multicomponent reaction for the efficient synthesis of spirooxindole‐linked fused pyrans. The reaction involves isatin, malononitrile or its derivatives, and various cyclic 1,3‐dicarbonyl compounds such as 4‐hydroxy‐1‐methyl‐2(1H)‐quinolone, 4‐hydroxycoumarin, 4‐hydroxythiocoumarin, or barbituric acid derivatives, yielding the corresponding spirooxindole‐linked fused pyrans. Key features of this method include the use of a water–ethanol solvent mixture, visible light as the energy source, and catalyst‐free conditions. A green and efficient method for the preparation of spirooxindole‐linked fused pyrans has been developed utilizing a catalyst‐free visible‐light‐driven one‐pot multicomponent reaction strategy. The reaction was performed in a water and ethanol mixture, using the combination of isatin, malononitrile, and cyclic 1,3‐dicarbonyls such as 4‐hydroxy‐1‐methyl‐2(1H)‐quinolone, 4‐hydroxycoumarin, 4‐hydroxythiocoumarin, or barbituric acid derivatives. This catalyst‐free, environmentally friendly, visible‐light‐driven, and room temperature method is a valuable addition to the synthetic toolbox for constructing complex heterocyclic scaffolds under one‐pot conditions. It holds promise for future applications in medicinal chemistry and drug discovery.

Abstract:A cost-effective, highly regioselective and metal-free version for the synthesis of indolizine derivatives by means of Tf2O-mediated cascade reaction of pyridyl-enaminones and thiophenols/thioalcohols under mild reaction conditions has been reported. Diverse electron-rich indolizine derivatives could be obtained in up to 94% yield via the selective 1,4-addition of vinyl iminium triflate tandem cyclization/aromatization, which allowed the simultaneous construction of C-N and C-S/and one example of C-Se bonds.

Abstract:Various multifunctional chromone-spiroindolinone-cyclopentene derivatives were synthesized through a phosphine-catalyzed (3+2) annulation of Morita-Baylis-Hillman (MBH) carbonates with oxindole-chromones in generally high yields with good to excellent diastereoselectivities. The synthetic method has several advantages such as mild reaction conditions, wide substrate scope, high yields and good stereoselectivity. Abstract:An interesting asymmetric organocatalytic cascade reaction of 3-hydroxyoxindoles with coumarins was accomplished by chiral bisguanidinium hemisalt. A series of enantioenriched spirobenzofuran indolinones were afforded in high yields with high diastereo- and enantioselectivities. The reaction was disclosed to involve in several steps: intermolecular Michael reaction, transesterification, retro-Michael reaction, substitution and decarboxylation, then an intramolecular Michael reaction to yield the product, in which the final step is disclosed to be stereo-determining step. In addition, it provided a facile route for the late-stage modification of several drug molecules via installation of spirobenzofuran indolinone scaffold.

Abstract:The development of green and efficient methods for construction of azepinoindole skeletons remains highly desirable yet challenging. Described herein is the EtOH-mediated cascade C(sp3)‒H alkylation reactions of 4-dialkylamino-indole-3-carbaldehydes for green and divergent synthesis of spirocyclic azepino[4,3,2-cd]indole derivatives. This protocol proceeded through an in situ assembly of pre-aromatics/aromatization-driven [1,6]-hydride transfer/cyclization sequence, which exhibited many advantages such as green bio-sourced EtOH as the reaction medium, metal-free and redox-neutral conditions, high step-/atom-economy, water as waste, high yields, excellent diastereoselectivites (up to ˃20:1 dr), wide substrate scope, and diverse transformations.

Abstract:An efficient cascade cyclization strategy was developed to synthesize aminobenzofuran spiroindanone and spirobarbituric acid derivatives utilizing 2-bromo-1,3-indandione, 5-bromo-1,3-dimethylbarbituric acid, and ortho-hydroxy α-aminosulfones as substrates. Under the optimized reaction conditions, the corresponding products were obtained with high efficiency, exceeding 95% and 85% yields for the respective derivatives. This protocol demonstrates exceptional substrate versatility and robust scalability up to the Gram scale, establishing a stable platform for the synthesis of 3-aminobenzofuran derivative. The successful synthesis paves the way for further biological evaluations with potential implications in scientific research.
