Lnc-LFAR1 Regulates Osteoarthritic Chondrocytes Proliferation and Apoptosis Through NF-κB Signaling Pathway

Abstract:Osteoarthritis (OA) is a common disease in elderly patients and the drugs currently used to treat OA are mostly palliative. Inflammatory factors are one of the critical factors in the development of OA, but the exact mechanism has not been elucidated. LncRNA is involved in the progression of several diseases. Lnc-LFAR1 was confirmed to be involved in physiological and pathological activities. However, the role and mechanism of lnc-LFAR1 in OA remains poorly understood. The bone and joint cartilage tissues of OA patients and healthy control were collected. The expression of lnc-LFAR1 was detected by real-time PCR. The chondrocytes of OA patients were isolated and divided into 3 groups, control group, lnc-LFAR1 siRNA group, and lnc-LFAR1 group followed by analysis of cell proliferation by MTT assay, cell apoptosis by flow cytometry, caspase 3 activity by Caspase 3 activity detection kit, NF-kappa B expression by Western blot as well as the levels of inflammatory factor tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) by ELISA. The expression of lnc-LFAR1 was significantly increased in OA patients (P < 0.05). Lnc-LFAR1 siRNA transfection significantly down-regulated lnc-LFAR1 expression, promoted cell proliferation, inhibited cell apoptosis, attenuated Caspase 3 activity, reduced NF-kappa B expression, as well as TNF-alpha and IL-6 secretions (P < 0.05). pcDNA lnc-LFAR1 plasmid transfection significantly upregulated lnc-LFAR1 expression, suppressed cell proliferation, facilitated cell apoptosis, enhanced Caspase 3 activity, elevated NF-kappa B expression, and increased TNF-alpha and IL-6 secretions (P < 0.05). Lnc-LFAR1 expression is increased in OA patients. Lnc-LFAR1 downregulation inhibited articular chondrocyte apoptosis, promoted articular chondrocyte proliferation, and restrained joint inflammation possibly by suppressing NF-kappa B signaling pathway.
