Effect of Dietary Xylan on Immune Response, Tight Junction Protein Expression and Bacterial Community in the Intestine of Juvenile Turbot (scophthalmus Maximus L.)

Abstract:The present study investigated the effects of dietary xylan on intestinal barrier function and bacteria community of turbot (Scophthalmus maximus L.). A 12-week feeding trial was conducted using three isonitrogenous and isolipidic diets containing 0%, 1.25% and 5% xylan, respectively. Dietary xylan at 1.25% significantly increased the gene expression of transforming growth factor β and tight junction protein Tricellulin, and decreased the expression of pro-inflammatory cytokine il-1β. Dietary xylan at 5% significantly increased the expression of il-1β and muc-2, and decreased the gene expression of tight junction protein Claudin-3 and Claudin-like. Sequencing of intestinal bacterial 16s rRNA V4 region revealed separate clustering of bacterial community in turbot fed different diets, but the overall bacterial community structure was not significantly affected by diet. LEfSe and MetaStat analysis indicated that dietary xylan at both 1.25% and 5% altered the composition and abundance of intestinal bacteria at different taxonomic levels. In the group with 1.25% xylan, the abundance of Proteobacteria, especially the protease-producing bacteria Halomonas, showed a significant increase. Meanwhile, the abundance of Bacteroides and Bifidobacterium significantly decreased. However, dietary xylan at 5% resulted in significantly up-regulated abundance of Clostridium, Escherichia coli and Prevotella copri associated with intestinal disease. Collectively, this study suggested xylan at 1.25% had prebiotic effect in enhancing the intestinal health of juvenile turbot, whereas 5% xylan exerted adverse effects. Attention should be paid on the dose of xylan when plant protein is used in fish feed.
