In this paper, hierarchical carbon nanofibers (CNFs)/MnCo2O4.5 nanofibers as efficient oxidase mimics are reported. The products are synthesized by an electrospinning technique and an electrochemcial deposition process in which the CNFs are used as the working electrode where MnCo2O4.5 nanosheets deposit on. The resulting binary metal oxide-based nanocomposites exhibit a good oxidase-like activity toward the oxidations of 3,3', 5,5' tetramethylbenzi-dine (TMB), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium (ABTS) salt and o-phenylenediamine (OPD) without exogenous addition of H2O2. The system of CNFs/MnCo2O4.5-TMB can be used as a candidate to detect sulfite and ascorbic acid via a colorimetric method with a high sensitivity. This work provides the efficient utilization and potential applications of binary metal oxide-based nanocomposites with oxidase activities in biosensors and other biotechnologies.

Co, N-doped Carbon Dot Nanozymes with Acid Ph-Independence and Substrate Selectivity for Biosensing and Bioimaging

Abstract:Nanozymes are more stable, economical, and easier to produce than natural enzymes; however, their low activity in non-acidic environments and lack of substrate selectivity severely limit the applications of nanozymes. In this study, we developed Co- and N-doped carbon dot nanozymes (CoNCDs) that specifically exhibited peroxidase-like activity toward o-phenylenediamine in a non-acidic environment. Based on the peroxidase-like activity and fluorescence property of CoNCDs in a neutral environment, a one-pot multi-mode sensing platform (colorimetric and ratiometric fluorescence imaging) was developed to detect cholesterol and xanthine in solution and human serum samples. This platform was simple to operate, and the CoNCDs demonstrated a reduced detection time and an improved analytical performance compared to most of the reported nanozymes. Moreover, the improved peroxidase activity at neutral pH, good biocompatibility, and photostability of CoNCDs facilitated their use for monitoring endogenous H2O2 levels in vivo. This study not only provides a novel approach for developing an acid pH-independent and substrate selectivity nanozyme but also demonstrates the application of ratiometric fluorescence imaging using the developed CoNCDs both in vitro and in vivo.

Abstract:Artificial nanozymes are designed for pursuing the functions of splendid catalytic efficiency and prominent selectivity of natural enzymes, meanwhile obtaining higher stability than that of natural enzymes. This emerging technology shows widespread application in the crossing field between nanotechnology and biomedicine. In this work, we employed a universal approach to fabricate a Co@N-CNTs hybrid nanocomposite as an oxidase mimic, in which fine Co nanoparticles were wrapped in N-doped carbon nanotubes, stacking on a hollow dodecahedron carbon skeleton. The synergistic effects of nanostructure engineering, N-doping and carbon coating, as well as the derived interfacial effect contribute to the glorious oxidase-like activity, stability and reusability. It can catalytically oxidize the colorless substrate 3,3',5,5'-tetramethylbenzidine (TMB) to a blue oxidation product (ox-TMB). As a result, a colorimetric technique with excellent selectivity and sensitivity for detecting ascorbic acid (AA) with naked eyes was established, in view of specific inhibitory effects towards oxidation of TMB. Under optimal detection conditions, this method exhibits a good linearity ranging from 0.1 to 160 μM with a low limit of detection (LOD) of 0.076 μM. For practical applications, Co@N-CNTs hybrid catalyst as a mimic oxidase was used for the determination of AA in human serum, which yielded satisfactory results. This work may serve as a new research thought to guide the design of high-performance nanozymes and establish a sensing platform for the detection of AA.
Colorimetric Detection of Ascorbic Acid and Alkaline Phosphatase Activity Based on the Novel Oxidase Mimetic of Fe–Co Bimetallic Alloy Encapsulated Porous Carbon Nanocages

Abstract:A novel catalyst of FeCo nanoparticles (FeCo NPs) incorporated porous nanocages (FeCo NPs@PNC) was first synthesized by encapsulating of FeCo alloy into ZIF-8 and further carbonation of the composite. The FeCo NPs@PNC displays enhanced intrinsic oxidase-like activity compared to the individual FeCo NPs and porous nanocages (PNC). The FeCo NPs@PNC can catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to oxidized TMB (oxTMB) without H2O2, producing a blue color with a maximum absorption peak at 652 nm. The catalytic mechanism was investigated and it found that the intermediate (O-2(center dot-)) produced from the catalytic process in the system of TMB-O-2-FeCo NPs@PNC can accelerate the oxidation of TMB to oxTMB. However, ascorbic acid (AA) can reduce the oxTMB and result in a conspicuous blue color fading. Therefore, a novel colorimetric platform was constructed to quantify AA with the linear range of 0.5-28 mu M and detection limit of 0.38 mu M (at 3 sigma/m). Owing to the alkaline phosphatase (ALP) can catalyze the hydrolysis of AA 2-phosphate (AAP) into AA, ALP can also be quantified by the above method. And the linear range for ALP is 0.6-10 U L-1 and the limit of detection is 0.49 U L-1. The FeCo NPs@PNC also shows excellent stability and reproducibility. This study provides a new alternative oxidase mimetic on the basis of easily obtained metal-organic frameworks derivatives to replace the expensive natural enzymes and noble metal based nanoenzymes, which will show great potential in biological assays.

Abstract:Rational design of high-performance nanozyme is of great significance for sensing applications. Here, N-doped carbon nanocages containing Co-N-x active sites (CoNx-NC) were fabricated by simple acid etching of Co@NC, which is the product of Co-Co Prussian blue analogues carbonized in an N-2 atmosphere. Both optimized Co@NC-650 and CoNx-NC-650 exhibit catalase- and oxidase-like properties. They rapidly decompose H2O2 into O-2 and oxidize colorless 3,3',5,5'-tetramethylbenzidine (TMB) into a blue product. This enables them with no peroxidase-like activity. The oxidase-like activity of CoNx-NC-650 is 3.9 times that of unetched Co@NC-650. The acid treatment dissolves Co nanoparticles generated during carbonization, forming extra mesoporous structures and increasing the exposure of more Co-N-x active species. The Michaelis-Menten constant for CoNx-NC-650 with a TMB substrate is 0.186 mM, which is 3.2 times lower than that of oxidase-mimicking CeO2 nanoparticles, suggesting a higher affinity to TMB. Given its excellent oxidase-like activity, the CoNx-NC-650 was used to sensitively and selectively determine acetylcholinesterase (AChE) activity based on thiocholine inhibition of the TMB color reaction. Thiocholine is produced via hydrolysis of acetylthiocholine catalyzed by AChE. The colorimetric biosensor has a linear response to AChE over 0.6-800 mU L-1 concentration range and a detection limit (3s) of 0.2 mU L-1. The assay was successfully applied to determine AChE activity in biological samples.

Nanostructured Cobalt Phosphates As Excellent Biomimetic Enzymes to Sensitively Detect Superoxide Anions Released from Living Cells

Abstract:Monitoring superoxide anion radicals in living cells has been attracting much academic and industrial interest due to the dual roles of the radicals. Herein, we synthesized a novel nanostructured cobalt phosphate nanorods (Co3(PO4)2 NRs) with tunable pore structure using a simple and effective micro-emulsion method and explored their potential utilization in the electrochemical sensing of superoxide anions. As an analytical and sensing platform, the nanoscale biomimetic enzymes Co3(PO4)2 NRs exhibited excellent selectivity and sensitivity towards superoxide anion (O2•-) with a low detection limit (2.25nM), wide linear range (5.76-5396nM), and long-term stability. Further, the nanoscale biomimetic enzyme could be efficiently applied in situ to electrochemically detect O2•- released from human malignant melanoma cells and normal keratinocyte, showing excellent real time quantitative detection capability. This material open up exciting opportunities for implementing biomimetic enzymes in nanoscale transition metal phosphates and designing enzyme-free biosensors with much higher sensitivity and durability in health and disease analysis than those of natural one.

Abstract:Mixed transition-metal oxides (MTMOs) have attracted much research interest because of their promising applications in artificial enzymes. In this work, uniform hollow MnCo2O4 nanofibers have been fabricated via an electrospinning technique followed by a calcination process, which can be used as efficient oxidase mimics. In detail, the as-prepared hollow MnCo2O4 nanofibers display excellent oxidase-like activity toward the oxidation of 3,3', 5,5'-tetramethylbenzidine (TMB) superior to their corresponding single-metal oxides without the addition of H2O2. Based on the high catalytic activity of hollow MnCo2O4 nanofibers and their inhibition effect by specific substances, the system of hollow MnCo2O4 nanofibers-TMB has potential to be applied in assays to detect sulfite and L-cysteine using a colorimetric approach with high sensitivity and selectivity. This work implies a broad application prospect of MTMOs in bio-sensors, environmental protection, food safety and medical science.
High Peroxidase-Like Activity of Metallic Cobalt Nanoparticles Encapsulated in Metal–organic Frameworks Derived Carbon for Biosensing

Abstract:Encapsulating metal nanoparticles (NPs) into carbon materials is a promising strategy for the fabrication of nanozymes with high activity. Here, for the first time, the proof-of-concept is demonstrated by fabricating CoNPs embedded in NH2-MIL-88(Fe) MOFs-derived magnetic carbon (MC) through in situ reduction of cobalt precursors by NaBH4. The obtained CoNPs/MC was characterized by TEM, SEM, XPS, XRD, Raman spectrum, and FT-IR. The CoNPs/MC exhibits much higher peroxidase-like activity than pure CoNPs and magnetic carbon. The high peroxidase-like activity of the CoNPs/MC is attributed to the decomposition of H2O2 to generate center dot OH and O-2(center dot-)-radicals, resulting in TMB color reaction. Also, CoNPs/MC is robust in a wide range of pH and temperature, indicating a promising peroxidase-like candidate. On this basis, a colorimetric assay was developed by combining glucose oxidase with CoNPs/MC for glucose biosensing. The linear range for glucose detection is from 0.25 to 30 mu M with a lower detection limit of 156 nM. The CoNPs/MC-based colorimetric method has been successfully used to assay the content of glucose in real biological samples such as human serums. Our work reveals the high potential of encapsulating metal NPs into carbon materials for the fabrication of nanozymes with stable and high activity. (C) 2017 Elsevier B.V. All rights reserved.

Abstract:Mn(II) ion has emerged as a cofactor in heme-copper oxidase to confer O 2 reduction to H 2 O, which shares the similar mechanism with oxidase mimicking reaction, so it is fascinating and interesting to explore the oxidase mimetic activity of Mn(II) compound. Here, MnO nanoparticles supported on porous carbon (MnO NPs/PC) was controllably prepared via in-situ conversion of MnO 2 nanobelts supported on porous carbon (MnO 2 NBs/PC). The oxidase mimetic activity of MnO NPs/PC was investigated using 3,3′,5,5′-tetramethyl-benzidine (TMB) as chromogenic substrate. MnO NPs/PC shows excellent oxidase mimetic activity with a low kinetic parameter K m of 0.0214 mM and a high V max of 7.21 × 10 −7 M/s. Due to the inhibition effect on TMB color reaction, dopamine (DA) can be detected using MnO NPs/PC-TMB chromogenic system, even in human serum samples. This work not only provides a controllable synthetic method for small-sized MnO NPs, but also throws the light on MnO oxidase mimetic activity for the first time.

Abstract:A novel nanozyme of bimetallic (Ni/Co) metal-organic framework (Ni/Co-MOF) was synthesized using a simultaneous precipitation and acid etching method with a zeolitic imidazolate framework ZIF-67 as the template. The as-synthesized Ni/Co-MOF catalyst presented a three-dimensional hollow nanocage structure and exhibited excellent intrinsic oxidase-like activity. It was demonstrated that Ni/Co-MOF could directly catalyze the oxidation of 3, 3′, 5, 5′-tetramethylbenzidine (TMB) to produce a blue product (oxidized TMB, oxTMB) in the absence of H2O2. The mechanisms and kinetics of this nanozyme activity were investigated, and it was determined that the catalytic activity of Ni/Co-MOF was closely related to temperature and solution pH. Owing to its strong reducibility, ascorbic acid (AA) could reduce oxTMB, and the blue color of the reaction mixture faded over time. Therefore, a novel colorimetric platform was constructed to detect AA based on the oxidase-like activity of Ni/Co-MOF. Under optimal conditions, the absorbance of ox-TMB at 652 nm decreased linearly over the 0.015-50 μM AA range with a detection limit of 0.004 μM.

Co₃O₄/Au Hybrid Nanostructures as Efficient Peroxidase Mimics for Colorimetric Biosensing

Abstract:Nanomaterials with enzyme-like characteristics (nanozymes) have emerged as potential replacements for natural enzymes due to their potential to overcome several critical limitations of natural enzymes, including low stability as well as high costs in preparation and purification. Herein, we have developed hybrid nanostructures that incorporate cobalt oxide nanoparticles (Co₃O₄ NPs) and gold nanoclusters (AuNCs) through electrostatic attraction induced by simple incubation in an aqueous buffer for 2 hours. Owing to the synergistic effect of Co₃O₄ NPs and AuNCs, the constructed Co₃O₄/Au hybrid nanostructures yielded highly enhanced peroxidase-like activity and enabled rapid catalytic oxidation of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB), producing a blue colored solution depending on the amount of H₂O₂. Moreover, we observed catalytic activity of the Co₃O₄/Au hybrid over a broad pH range, especially at physiologically relevant pH in the range of 5.0-7.4, which is advantageous for applications in biological systems. Using the hybrid as peroxidase mimic, we successfully determined the level of target H₂O₂ or glucose by coupling with glucose oxidase with excellent specificity and sensitivity. Based on this study, we expect that Co₃O₄/Au hybrid nanostructures can serve as potent peroxidase mimics for the detection of clinically important target molecules.

Abstract:Research about constructing multi-dimensional carbon nanomaterials is of great significance in electrocatalytic and sensing fields in order to integrate structural merits of each individual unit. Also, nanomaterials with enzyme-like activities are prospective candidates for artificial enzyme design and electrochemical application. Herein, we fabricate Co, N co-doped hierarchical hybrid (/NC) nanozyme, which integrates of both N-doped carbon nanotubes (NCNTs) and N-doped carbon sheets (NC). The three-dimensional (3D) porous carbon composite is prepared by thermal treatment of metal-organic framework (MOF) which was synthesized by growing of ZIF-67 on ZIF-L at room temperature. The obtained nanomaterial not only possesses an improved oxidase-like activity that can catalyze 3,3′,5,5′-tetramethylbenzidine (TMB) in the absence of hydrogen peroxide (H2O2), but also constructs a signal amplification platform towards dopamine (DA) due to the synergistic catalysis of Co species and N-doped porous carbon architecture. The electrocatalytic performance for DA detection shows a broad linear range from 30 nM to 710 μM and a detection limit of 9 nM. The /NC/GCE is employed to practically detect DA in human serum and artificial cerebrospinal fluid (aCSF) samples with satisfactory results. The present work exhibits a great promising in colorimetric and electrochemical sensing fields and presents a new sight for the fabrication of MOF-derived nanostructured nanozyme.

Abstract:Compared to natural enzymes, nanomaterial-based artificial enzymes have attracted immense attention because of their high stability and cost-effectiveness. In this study, cobalt nitride (Co4N) as a noble-metal-free artificial enzyme exhibiting highly intrinsic peroxidase-like activity and good stability was reported. Kinetic studies revealed that the resultant Co4N nanowires (NWs) exhibited a stronger affinity for 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2 than HRP. Compared to Co3O4 NWs, Co4N NWs exhibited highly improved catalytic activities, with H2O2 exhibiting an apparent Km approximately 2 orders of magnitude less than that of Co3O4. In particular, the peroxidase-like activity of Co4N was maintained well over a wide range of temperatures and ionic strength. A Co4N-based method was further developed for the detection of glucose with good sensitivity and reliability. Because of advantages such as easy storage, cost-effectiveness, high sensitivity, and outstanding stability, Co4N NWs demonstrate the potential for replacing noble-metal-based peroxidase mimetics in a wide range of promising applications.
Cobalt-doped MoS2 nanocomposite with NADH oxidase mimetic activity and its application in colorimetric biosensing of NADH

Abstract:NADH oxidase (NOX) is an oxidoreductase that catalyzes the oxidation of NADH to NAD+ with the participation of oxygen. NOX can artificially regulate metabolic processes as an important regulatory enzyme for NAD+ regeneration. In this study, cobalt-doped molybdenum sulfide (Co-MoS2) nanocomposite was prepared by hydrothermal synthesis, which exhibited NOX mimics. The catalytic mechanism of NOX mimics was studied. The NOX-like activity of Co-MoS2 is optimal at pH 4.5 and in the temperature range of 20 − 45℃. Further, its catalytic activity maintained almost the same after 5 cycles of repetitive addition of NADH, suggesting that Co-MoS2 nanozyme possessed significant catalytic persistence. The produced H2O2 catalyzed by Co-MoS2 can be detected by horseradish peroxidase (HRP) and 3,3',5,5'-tetramethylbenzidine at pH4.5. Thus, interestingly, a colorimetric NADH sensor was proposed on the basis of the enzymatic cascade reaction of NADH oxidase-like conjugated with HRP. This as-established method had wide detection range (0.5 − 800 μM) with the detection limit of 0.25 μM NADH, capable of detecting NADH in real sample.

Abstract:In contrast to peroxidase-mimicking nanozymes, oxidase mimics directly employ molecular oxygen (O2) as oxidant to generate reactive oxygen species (ROS). Generally, transition metal-based nanozymes exhibit poor oxidase-like activity. We herein constructed a colorimetric sensor based on Co-based oxidase mimic, where the Co nanoparticles were encapsulated in hollow N-doped carbon nanospheres (Co-N/C). Co nanoparticles confined catalyst effectively anchored the active part, which greatly influenced the catalytic activity of the oxidase-mimic nanozymes. Due to the antioxidative property of L-Cysteine (L-Cys) and the specific complexation between L-Cys and Hg2+, the rationally designed Co-N/C could be utilized to detect L-Cys and Hg2+. The proposed sensing platform exhibited wide linear ranges of 1-30 mu M for L-Cys and 0.6-30 mu M for Hg2+, respectively. The excellent selectivity of the constructed sensor showed the reliability and practicality for the assay of L-Cys and Hg2+ in fetal bovine serum. The spatial confinement can prevent the aggregation of Co nanoparticles, boosting the stability and catalytic performance in colorimetric sensing.

Abstract:Single-atom catalysts are becoming a hot research topic owing to their unique characteristics of maximum specific activity and atomic utilization. Herein, atomically dispersed Co on a N-doped carbon matrix with an enzyme-like M-N-x structure has been developed as a bifunctional biosensor to detect hydrogen peroxide (H2O2) and dopamine (DA). It features 100% atomic utilization, high electro-chemical activity and selectivity, and strong stability in various pH environments. The abundance of Co-N-x sites can be increased via regulating calcination temperature, and as a result, the sensing performance is significantly improved. This platform could selectively catalyze the oxidation of DA and the reduction of H2O2 at different holding potentials. The optimal Co-N-C-800 affords high sensitivity (H2O2: 943.9 mu A mM(-1) cm(-2) and DA: 979.6 mu A mM(-1) cm(-2)), low detection limit (H2O2: 0.13 mu M and DA: 0.04 mu M), high selectivity, and robust stability. At the same time, H2O2 and DA released by PC12 can be detected, which proved the feasibility of the enzyme-like single-atomic materials in electrochemical biosensing.
