Abstract:"Dragon's blood" is the name given to a deep red resin obtained from a variety of plant sources. The resin extracted from stems of Dracaena cochinchinensis is one such source of "dragon's blood". It has a reputation for facilitating blood circulation and dispersing blood stasis. In traditional Chinese medicine, this resinous medicine is commonly prescribed to invigorate blood circulation for the treatment of traumatic injuries, blood stasis and pain. Modern pharmacological studies have found that this resinous medicine has anti-bacterial, anti-spasmodic, anti-inflammatory, analgesic, anti-diabetic, and anti-tumor activities, while it is also known to enhance immune function, promote skin repair, stop bleeding and enhance blood circulation. Various compounds have been isolated from the plant, including loureirin A, loureirin B, loureirin C, cochinchinenin, socotrin-4'-ol, 4',7-dihydroxyflavan, 4-methylcholest-7-ene-3-ol, ethylparaben, resveratrol, and hydroxyphenol. The present review summarizes current knowledge concerning the botany, phytochemistry, pharmacological effects, toxicology studies and clinical applications of this resinous medicine as derived from D. cochinchinenesis.

Dragon's Blood from Dracaena Worldwide: Species, Traditional Uses, Phytochemistry and Pharmacology

Abstract:Dragon's blood (DB) refers mainly to the crimson resin of many Dracaena spp. DB has been used by different traditional medicine systems worldwide, including Arabic medicine, African medicine, traditional Chinese medicine, Thai medicine, etc. DB are mainly used to heal wounds, kill pain, stop bleeding, and cure various diseases such as diarrhea, dysentery and ulcers for over 1000 years. 11 Dracaena spp. and 3 subspecies are reported to be able to produce red resin. However, the resources are extremely deficient. Several Dracaena spp. are in threatened status. Over 300 compounds have been isolated from Dracaena spp., mainly including flavonoids, steroids, and phenolics. DB exhibits anti-inflammatory, analgesic, antithrombotic, anti-oxidant, antimicrobial, antidiabetic, and anticancer properties, which explain its wound healing effects, preventive effects on cardiovascular and cerebrovascular diseases, dual-directional regulation of blood flow, neuroprotection and radioprotective effects. No apparent side effects or toxicity have been reported. DB are restricted from being exploited due to limited resources and unclear resin formation mechanism. It is necessary to expand the cultivation of Dracaena spp. and fully understand the mechanism underlying the resin formation process to develop an effective induction method for the sustainable utilization of DB.
Simultaneous Quantification of Five Major Constituents in Stems of Dracaena Plants and Related Medicinal Preparations from China and Vietnam by Hplc-Dad

Abstract:A high-performance liquid chromatographic (HPLC) method was developed for the simultaneous quantification of five major bioactive constituents in the stems of resiniferous Dracaena plants from China and Vietnam, as well as those in the related traditional Chinese medicinal preparations. A diode array detector with the wavelength of 330 nm was used to monitor resveratrol, 7,4'-dihydroxyflavone and pterostilbene, while loureirin A and loureirin B were monitored at 280 nm. The five constituents were separated on an Agela SB C(18) column by gradient elution using 0.008% (v/v) formic acid solution (A) and acetonitrile (B) as the mobile phase. The validation of the method included recovery, linearity, accuracy and precision (intra- and inter-day variation). The range of recoveries of this method was 98.1-104.9%, with all the constituents showing good linearity (r(2) > 0.9999). The accuracy and precision were satisfactory, with the overall intra- and inter-day variation being less than 4%. The present method has been successfully applied for the determination of all five constituents in 21 related herbal samples including 10 D. cochinchinensis stem samples, seven D. cambodiana stem samples and four purchased medicinal preparations. The contents of these constituents were analyzed using principal component analysis, which can efficiently identify raw herb of Dracaena from different sources. The study may be considered helpful to the quality control of Dracena plants and its medicinal preparations.

Abstract:Many plants such as Dracaena cinnabari have been used in traditional medicine since ancient times due to their including bioactive compounds such as phenolic compounds, tocopherols, and organic acids. The aim of present study is to determine the presence and diversity of bioactive components such as phenolic, organic acids and tocopherols of Dracaena cinnabari resin extracts, and to reveal which solvents can be effective in the extraction of these components and to what extent. Methanol, ethanol, chloroform, water, and vinegar were used in extraction of Dracaena cinnabari resin. The widest diversity was obtained with vinegar for organic acids, methanol for tocopherols, and methanol solvent for phenolic compounds. Fourteen phenolic compounds, four tocopherols and ten organic acids were detected in Dracaena cinnabari resin extracts, the most abundant phenolic was apigenin, while the most tocopherol was ?-tocopherol. Dracaena cinnabari resin extracts appear to be rich in apigenin, kaempferol, luteolin and rutin flavonoids.

Abstract:Object To study the chemical constituents of Dracaena cochinchinensis (Lour.) S. C. Chen. Methods The constituents were isolated on silica gel chromatography, preparative TLC, and spectral data. Results Six compounds were isolated and identified from the resin of D. cochinchinensis as: 1, 2, 4, 5-tetrachloro-3, 6-dimethoxybenzene (Ⅰ), cholest-4α-methyl-7-en-3β-ol (Ⅱ), cholest-4α-methyl-7-en-3-one (Ⅲ), hexacosane (Ⅳ), cholest-7-en-3β-ol (Ⅴ), cholest-7-en-3-one (Ⅵ). Conclusion Compounds Ⅳ-Ⅵ were isolated from D. cochinchinensis for the first time.
Antifungal and wound healing promotive compounds from the resins of Dracaena cochinchinensis

Abstract:Five new compounds (xuejieins A–E), including three new phenolic glycosides (1, 2, and 5) and two new flavonoids (10 and 11), together with six known compounds were isolated from the resins of Dracaena cochinchinensis (Chinese dragon's blood). The structures of the new compounds were confirmed by extensive spectroscopic methods and electronic circular dichroism (ECD) data analysis. Especially, the absolute configurations of the sugar moieties in compounds 1, 2, and 5 were clarified by GC analysis after acid hydrolysis. All isolated compounds have been tested for antifungal and wound healing promoting activities, The results showed that compound 9 shows significant antifungal activities against Botrytis cinerea, Magnaporthe grisea, Penicillium digitatum, and Sclerotinia sclerotiorum. In addition, compound 4 could significantly stimulate human keratinocytes (HaCAT) proliferation, mobility, and human umbilical vein vascular endothelial cells (HUVECs) tube formation at 40 μM.

Abstract:Phenolic compounds are the most important group of secondary metabolites widely distributed in plants,playing important roles in diverse physiological processes such as plant quality,coloring,flavor and stress resistance.The natural antioxidant activities of phenolic compounds become a hot spot in terms of research and utilization at present.In this review,the structural characterization,distribution,composition and diverse functions of phenolic compounds in plants and human bodies were summarized,and the preparation methods and progress in phenolic compounds biosynthesis genes and their regulation modes were concluded as well in order to elucidate the physiological,nutritional and antioxidant functions of different phenolic compounds in plants and human.

Abstract:OBJECTIVE:To study the anatomy of Dracaena cochinchinensis systematically, and find out the distribution and detect the constituents of its resin, in order to provide substantial foundation for the formation mechanism of its red resin.METHOD:The microscopic structures of D. cochinchinensis were systematically observed by using color micrographics, including stem with and without resin, roots, barks and leaves. The HPLC fingerprints of the stem with and without resin were compared.RESULT:Characteristics of the tangentical longitudinal section of stem with resin and surface view of leaves were elucidated. Besides xylem vessels and fibers of the stem, it was found that the red resin also exists in the cortex parenchyma cells of the stem and the medulla and xylem of the root. According to the HPLC fingerprint analysis result of the stems with and without resin, a number of flavones and stilbenoids were detected in the stem in which resin appeared after it wounded.CONCLUSION:No secretory tissue to secrete resin was found in D. cochinchinensis, further study is needed to elucidate the formation mechanism of its resin.

Abstract:A schematic diagram of the mature stem was created based on the structural characteristics of the different developmental stages of stems of Dracaena cambodiana Pierre ex Gagnep. The main substance histological location of the stems was also studied. Dracaena cambodiana Pierre ex Gagnep is an important plant resource for producing dragon's blood and one of most popular ornamental trees in China. For a better understanding of the physiological function of the stem, the structural characteristics and main substance histological location of the stems of D. cambodiana were studied. The structural characteristics of the different developmental stages of stems of D. cambodiana were observed and described detailly. And then a schematic diagram of the mature stem was created. Histochemical staining showed that two kinds of polysaccharides distributed in parenchymal cells. Saponins distributed mainly in ground tissue and phenolic compounds distributed mainly in the thick cell walls. An abundant of calcium oxalate raphide bundles were identified in cortex and primary tissue. Finally, the role of the above results in the taxonomy of Dracaena species and in their strong adaptability was discussed.

Abstract:Two new dihydrochalcones, 4-hydroxy-2,4'-dimethoxydihydrochalcone (1) and 3,4'-dihydroxy-2,4,6-trimethoxydihydrochalcone (2), and a new homoisoflavane, 7,3'-dihydroxy-8,4'-dimethoxyhomoisoflavane (3), along with 12 known compounds (4-15), were isolated from the red resin of Dracaena cochinchinensis (Chinese dragon's blood). Their structures were assigned by a variety of spectroscopic techniques. Diversity of cleavage pathways were proposed for dihydrochalcones and homoisoflavanes based on the mass spectroscopic behaviors of those identified compounds using hybrid ion trap-time of flight mass spectrometry. All the compounds were evaluated for their inhibitory effects on nitric oxide production in lipopolysaccharide-induced RAW 264.7 macrophages, and compound 9 exhibited mild inhibition of NO production in this assay with IC50 value of 50.3 mu M. Identification and Characterization of Chalcone Isomerase Genes Involved in Flavonoid Production in Dracaena cambodiana

Abstract:Dragon’s blood is a traditional medicine in which flavonoids are the main bioactive compounds; however, the underlying formation mechanism of dragon’s blood remains largely poorly understood. Chalcone isomerase (CHI) is the key enzyme in the flavonoid biosynthesis pathway. However, CHI family genes are not well understood in Dracaena cambodiana Pierre ex Gagnep, an important source plant of dragon’s blood. In this study, 11 CHI family genes were identified from D. cambodiana , and they were classified into three types. Evolutionary and transcriptional profiling analysis revealed that DcCHI1 and DcCHI4 might be involved in flavonoid production. Both DcCHI1 and DcCHI4 displayed low expression levels in stem under normal growth conditions and were induced by methyl jasmonate (MeJA), 6-benzyl aminopurine (6-BA, synthetic cytokinin), ultraviolet-B (UV-B), and wounding. The recombinant proteins DcCHI1 and DcCHI4 were expressed in Escherichia coli and purified by His-Bind resin chromatography. Enzyme activity assay indicated that DcCHI1 catalyzed the formation of naringenin from naringenin chalcone, while DcCHI4 lacked this catalytic activity. Overexpression of DcCHI1 or DcCHI4 enhanced the flavonoid production in D. cambodiana and tobacco. These findings implied that DcCHI1 and DcCHI4 play important roles in flavonoid production. Thus, our study will not only contribute to better understand the function and expression regulation of CHI family genes involved in flavonoid production in D. cambodiana but also lay the foundation for developing the effective inducer of dragon’s blood.

Abstract:Objective: The aim of this study was to comprehensively analyze the differences in Chinese dragon's blood (CDB), specifically Dracaena cochinchinensis and Dracaena cambodiana, from different geographical origins. Methods: Metabolomic analysis of CDB was performed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). A reliable ultrahigh-performance liquid chromatography method with a photodiode array detector (UHPLC-PDA) was developed and applied for the quantitative analysis of 12 phenolic compounds in 51 batches of samples. Results: A total of 1394 metabolites were detected, of which 467 were identified as differentially accumulated metabolites. Multivariate analysis revealed that both origin and species had an effect on the composition of CDB, with greater variation between species. 19 phenolic compounds were selected as quality markers to distinguish D. cochinchinensis (Hdsp) from D. cambodiana (Hdca), and oppositin and spinoflavanone a were identified as quality markers to discriminate D. cochinchinensis samples from Hainan (Hdsp) and Guangxi Provinces (Gdc). Quantitative analysis indicated that four phenolic compounds, including loureirin D, 4H-1-benzopyran-4-one,2,3-dihydro-3,5,7-trihydroxy-3-[(4-methoxyphenyl)methyl]-,(R)-, loureirin B, and pterostilbene, showed significant differences between Gdc and Hdsp. Additionally, five phenolic compounds, namely resveratrol, loureirin D, pinostilbene, 4H-1-benzopyran-4-one,2,3-dihydro-3,5,7-trihydroxy-3-[(4-methoxyphenyl)methyl]-, (R)-, and loureirin B, exhibited significant differences between Hdsp and Hdca. Conclusion: There are significant differences in the quality of CDB from different geographical origins and species, which lays the foundation for the in-depth development and utilization of different sources of CDB.

Abstract:AIM:To study the chemical constituents from the aerial parts of Daphne pedunculata.METHODS:Column chromatography on silica gel and Sephadex LH-20 were used for the isolation and purification of compounds,and physical properties and spectroscopic techniques were used for structural identification.RESULTS:Eight compounds were isolated from the titled plant,and elucidated as daphnetin(1),daphnoretin(2),daphneticin(3),isodaphneticin(4),(-)-pinoresinol(5),daphneolone(6),β-sitosterol(7),and daucostero1(8).CONCLUSION:All compounds were isolated from this plant for the first time.

Abstract:Here we show the therapeutic effects of She-Mei [Duchesnea indica(Andr)Focke],a Chinese herbal medicine for removing toxic heat and eliminating blood stasis.The herb is often used to treat different cancers,along with other Chinese medicines.Some recent studies have found that She-Mei contains phenolic acids and triterpenes,and its crude extracts have a noticeable antioxidant and anticancer activity.This article summarizes a range of studies concerning the chemical composition,pharmacological activities,as well as clinical applications of the herb.It makes a valuable information source for future investigations.

Abstract:Three new spirostan glycosides, dracochinosides A‐C (1‐3), and four known steroidal glycosides (4‐7) were isolated from the aerial parts of Dracaena cochinchinensis (Lour.) S.C.Chen. Their chemical structures were determined by the IR, HR‐ESI‐MS, 1D‐, and 2D‐NMR spectra. Compounds 1‐3, 6 and 7 inhibited nitric oxide production in LPS activated RAW 264.7 cells with IC50 values ranging from 57.5 to 92.8 μM. In addition, all the isolated compounds exhibited at least one of seven tested microbial strains with the MIC values ranging from 0.016 to 0.128 mg/mL. This is the first report of compounds 5‐7 from the genus Dracaena

Abstract:For thousands of years, plant has been widely applied in the medical area and is an important part of human diet. A high content of nutrients could be found in all kinds of plants, and the most outstanding group of nutrients that attracts scientists' attention is the high level of phenolic compounds. Due to the relationship between high phenolic compound content and high antioxidant capacity, plant extracts are expected to become a potential treatment for oxidation stress diseases including diabetes and cancer. However, according to the instability of phenolic compounds to light and oxygen, there are certain difficulties in the extraction of such compounds. But after many years of development, the extraction technology of phenolic compounds has been quite stable, and the only problem is how to obtain high-quality extracts with high efficiency. To further enhance the value of plant extracts, concentration and separation methods are often applied, and when detailed analysis is required, characterization methods including HPLC and LC/GC–MS will be applied to evaluate the number and type of phenolic compounds. A series of antioxidant assays are widely performed in numerous studies to test the antioxidant capacity of the plant extracts, which is also an important basis for evaluating value of extracts. This paper intends to provide a view of a variety of methods used in plants' phenolic compound extraction, separation, and characterization. Furthermore, this review presents the advantages and disadvantages of techniques involved in phenolic compound research and provides selected representative bibliographic examples.

Abstract:Dragon’s blood (Resina Draconis) is the red resin of Dracaena spp, which has a variety of biological activities and pharmacological effects, including anti-thrombotic, anti-inflammatory, anti-bacterial, analgesic, anti-oxidant, anti-tumor, and immunosuppressive. In China, the main source of dragon’s blood is Dranaena Cochinchinensis (Lour.) S.C.Chen. A wide array of studies have speculated that the dragon’s blood derived from Dranaena Cochinchinensis (Lour.) S.C.Chen possesses cardiovascular protective effects. It has been reported that Chinese dragon’s blood can potentially alleviate and treat conditions such as coronary heart disease, myocardial infarction, and myocardial ischemia–reperfusion through its anti-inflammatory and antioxidant properties, which have not been systematically stated in previous reviews. Moreover, the precise underlying pharmacological mechanisms through which the Chinese dragon’s blood exhibits cardioprotective effects are not fully understood. Therefore, this article discusses the pharmacological action and biomolecular mechanism of dragon’s blood from Dranaena Cochinchinensis (Lour.) S.C.Chen and how it prevents and protects against cardiovascular diseases. The review article concludes with prospects for further application of dragon’s blood in respect to cardiovascular diseases.

Abstract:This study reports a chromosome-level genome for Dracaena cambodiana, one of two typical dracaena species in China. This work will help to deepen the understanding of the dracaena species and the conservation and development of plant resources. The final assembly consisted of 54 scaffolds, spanning 1.08 Gb, with a scaffold N50 size of 52.29 Mb, encoded 36860 predicted protein-coding genes. A total of 1,064,810,157 bp of genome sequences were located on 20 chromosomes (2n = 40), accounting for 98.46%. We investigate the possible mechanisms of the longevity of dracaena, the longest-lived plant in the world, which involve DNA repair and post-translational modification. We also performed comparative genomic analysis of the previously assembled genome of Dracaena cochinchinesis with this genome, and found that the two involved interaction of plant−pathogen interaction and ubiquitin mediated proteolysis, which may reveal why Dracaena has environmental adaptability and longevity characteristics.

Abstract:Five new homoisofiavonoid derivatives, including three meta-homoisoflavanes (1-3), one homoisoflavanone (4), and one homoisofiavan (5), along with five known analogues (6-10), were isolated from the red resin of Dracaena cochinchinensis (Chinese dragon's blood). Their structures were elucidated by analysis of spectroscopic data (1D and 2D NMR, IR, and HRESIMS). The absolute configuration of compound 1 was determined by single crystal X-ray crystallographic analysis, and those of 2-5 were established on the basis of experimental and computed ECD data. Compounds 4, 6, and 9 exhibited moderate inhibition of nitric oxide production in lipopolysaccharide-stimulated BV-2 microglial cells with IC50 values of 60.4-75.6 mu M.
