Abstract:A drug's aqueous solubility is defined as the ability to dissolve in a particular solvent, and it is currently a major hurdle in bringing new drug molecules to the market. According to some estimates, up to 40% of commercialized products and 70–90% of drug candidates in the development stage are poorly soluble, which results in low bioavailability, diminished therapeutic effects, and dosage escalation. Because of this, solubility must be taken into consideration when developing and fabricating pharmaceutical products. To date, a number of approaches have been investigated to address the problem of poor solubility. This review article attempts to summarize several conventional methods utilized to increase the solubility of poorly soluble drugs. These methods include the principles of physical and chemical approaches such as particle size reduction, solid dispersion, supercritical fluid technology, cryogenic technology, inclusion complex formation techniques, and floating granules. It includes structural modification (i.e., prodrug, salt formation, co-crystallization, use of co-solvents, hydrotrophy, polymorphs, amorphous solid dispersions, and pH variation). Various nanotechnological approaches such as liposomes, nanoparticles, dendrimers, micelles, metal organic frameworks, nanogels, nanoemulsions, nanosuspension, carbon nanotubes, and so forth have also been widely investigated for solubility enhancement. All these approaches have brought forward the enhancement of the bioavailability of orally administered drugs by improving the solubility of poorly water-soluble drugs. However, the solubility issues have not been completely resolved, owing to several challenges associated with current approaches, such as reproducibility in large scale production. Considering that there is no universal approach for solving solubility issues, more research is needed to simplify the existing technologies, which could increase the number of commercially available products employing these techniques.

Abstract:The low water solubility of pharmacoactive molecules limits their pharmacological potential, but the solubility parameter cannot compromise, and so different approaches are employed to enhance their bioavailability. Pharmaceutically active molecules with low solubility convey a higher risk of failure for drug innovation and development. Pharmacokinetics, pharmacodynamics, and several other parameters, such as drug distribution, protein binding and absorption, are majorly affected by their solubility. Among all pharmaceutical dosage forms, oral dosage forms cover more than 50%, and the drug molecule should be water-soluble. For good therapeutic activity by the drug molecule on the target site, solubility and bioavailability are crucial factors. The pharmaceutical industry's screening programs identified that around 40% of new chemical entities (NCEs) face various difficulties at the formulation and development stages. These pharmaceuticals demonstrate less solubility and bioavailability. Enhancement of the bioavailability and solubility of drugs is a significant challenge in the area of pharmaceutical formulations. According to the Classification of Biopharmaceutics, Class II and IV drugs (APIs) exhibit poor solubility, lower bioavailability, and less dissolution. Various technologies are discussed in this article to improve the solubility of poorly water-soluble drugs, for example, the complexation of active molecules, the utilization of emulsion formation, micelles, microemulsions, cosolvents, polymeric micelle preparation, particle size reduction technologies, pharmaceutical salts, prodrugs, the solid-state alternation technique, soft gel technology, drug nanocrystals, solid dispersion methods, crystal engineering techniques and nanomorph technology. This review mainly describes several other advanced methodologies for solubility and bioavailability enhancement, such as crystal engineering, micronization, solid dispersions, nano sizing, the use of cyclodextrins, solid lipid nanoparticles, colloidal drug delivery systems and drug conjugates, referring to a number of appropriate research reports.

Abstract:Background: In the development of drug delivery systems, drugs' solubility remains the most challenging constraint. Many newly synthesized chemical compounds are available, but they involve low solubility and poor permeability restrictions. Among various drug delivery systems, the utilization of solid dispersion technologies has become more focused due to their promising benefits. Objective: This technology has attracted extensive attention for dissolution rate improvement along with substantial bioavailability enhancement of poorly water-soluble drug candidates. Methods: Many approaches have been employed for preparing solid dispersions, such as the melting method, hot melt extrusion, solvent evaporation process, fusion and kneading method, spray drying technique, co-grinding and freeze drying, supercritical fluid technology, etc. Result: A wide variety of hydrophilic and hydrophobic materials are available as carriers, which are employed in the formulation of solid dispersions. Depending on the carrier characteristics, immediate- release solid dispersions and/or controlled-release solid dispersions can be formulated. Multiple hydrophilic materials have been explored for heightening dissolution features with enhanced bioavailability of poorly water-soluble drug molecules. The availability of commercially available products further validates the utility of solid dispersion technology in drug delivery systems. Conclusion: In the current manuscript, an attempt has been made to highlight the comprehensive development techniques, characterization techniques, recent solid dispersion technologies, clinical trial studies, and patented technology, along with studies heightening the dissolution behavior of numerous poorly aqueous soluble drugs. Abstract:High solubility in water and physiological fluids is an indispensable requirement for the pharmacological efficacy of an active pharmaceutical ingredient. Indeed, it is well established that pharmaceutical substances exhibiting limited solubility in water are inclined towards diminished and inconsistent absorption following oral administration, consequently resulting in variability in therapeutic outcomes. The current advancements in combinatorial chemistry and pharmaceutical design have facilitated the creation of drug candidates characterized by increased lipophilicity, elevated molecular size, and reduced aqueous solubility. Undoubtedly, the issue of poorly water-soluble medications has been progressively escalating over recent years. Indeed, 40% of the top 200 oral medications marketed in the United States, 33% of drugs listed in the US pharmacopoeia, 75% of compounds under development and 90% of new chemical entities are insufficiently water-soluble compounds. In order to address this obstacle, formulation scientists employ a variety of approaches, encompassing both physical and chemical methods such as prodrug synthesis, salt formation, solid dispersions formation, hydrotropic substances utilization, solubilizing agents incorporation, cosolvent addition, polymorphism exploration, cocrystal creation, cyclodextrins complexation, lipid formulations, particle size reduction and nanoformulation techniques. Despite the utilization of these diverse approaches, the primary reason for the failure in new drug development persists as the poor aqueous solubility of pharmaceutical compounds. This paper, therefore, delves into the foundational principles that underpin the implementation of various formulation strategies, along with a discussion on the respective advantages and drawbacks associated with each approach. Additionally, a discourse is provided regarding methodological frameworks for making informed decisions on selecting an appropriate formulation strategy to effectively tackle the key challenges posed during the development of a poorly water-soluble drug candidate.

Abstract:This article is intended to combine recent literature on solid dispersion technology for solubility enhancement with special emphasis on mechanism responsible for the same by solid dispersion, various preparation methods, and evaluation parameters. Solubility behavior is the most challenging aspect for various new chemical entities as 60% of the new potential products possess solubility problems. This is the biggest reason for new drug molecules not reaching to the market or not reaches to full potential. There are various techniques to enhance the drug solubility such as particle size reduction, nanosuspension, use of surfactants, salt formation, solid dispersion, etc. From this article it may be concluded that solid dispersion is an important approach for improvement of bioavailability of poor water-soluble drugs.

Abstract:The emerging trends in the combinatorial chemistry and drug design have led to the development of drug candidates with greater lipophilicity, high molecular weight and poor water solubility. Majority of the failures in new drug development have been attributed to poor water solubility of the drug. Issues associated with poor solubility can lead to low bioavailability resulting in suboptimal drug delivery. About 40% of drugs with market approval and nearly 90% of molecules in the discovery pipeline are poorly water-soluble. With the advent of various insoluble drug delivery technologies, the challenge to formulate poorly water soluble drugs could be achieved. Numerous drugs associated with poor solubility and low bioavailabilities have been formulated into successful drug products. Several marketed drugs were reformulated to improve efficacy, safety and patient compliance. In order to gain marketing exclusivity and patent protection for such products, revitalization of poorly soluble drugs using insoluble drug delivery technologies have been successfully adopted by many pharmaceutical companies. This review covers the recent advances in the field of insoluble drug delivery and business prospects.

Several elements influence the absorption of oral medications, including their solubility, permeability across mucosal membranes, and stability within the gastrointestinal (GI) environment. Research has delved into comprehending physicochemical, biochemical, metabolic, and biological obstacles that impact the bioavailability of a drug. To improve oral drug absorption, several pharmaceutical technologies and delivery methods have been studied, including cyclodextrins, micelles, nanocarriers, and lipid-based carriers. This review examines both traditional and innovative drug delivery methods, as well as the physiological and pharmacological barriers influencing medication bioavailability when taken orally. Abstract:Poorly water-soluble drugs are frequently encountered and present a most challengeable difficulty in pharma-ceutical development. Poor solubility of drugs can lead to suboptimal bioavailability and therapeutic efficiency. Increasing efforts have been contributed to improve the solubility of poorly water-soluble drugs for better pharmacokinetics and pharmacodynamics. Among various solubility enhancement technologies, protein-based strategy to address poorly water-soluble drugs issues has special interests for natural advantages including versatile interactions between proteins and hydrophobic drugs, biocompatibility, biodegradation, and metabo-lization of proteins. The protein-drug formulations could be formed by covalent conjugations or noncovalent interactions to facilitate solubility of poorly water-soluble drugs. This review is to summarize the advances using proteins including plant proteins, mammalian proteins, and recombinant proteins, to enhance water solubility of poorly water-soluble drugs.

Abstract:Low water solubility is the main hindrance in the growth of pharmaceutical industry. The main intention of present review is to depict the conventional and novel strategies to overcome the solubility-related problems of Biopharmaceutical Classification System Class-II drugs. More than 100 articles published in the last 5 years were reviewed to have a look at the strategies used for solubility enhancement. pH modification, salt forms, amorphous forms, surfactant solubilization, cosolvency, solid dispersions, inclusion complexation, polymeric micelles, crystals, size reduction, nanonization, proliposomes, liposomes, solid lipid nanoparticles, microemulsions, and self-emulsifying drug delivery systems are the various techniques to yield better bioavailability of poorly soluble drugs. Abstract:Introduction: Most drugs used in therapy have low water-solubility, a factor that could reduce their dissolution rate and oral bioavailability, representing a challenge in pharmaceutical development. Nanonization of drugs is the reduction of particles to nanoscale, increasing the surface area and consequently the saturation solubility and dissolution rate and resulting in higher bioavailability. Areas covered: This review provides an overview of the consequences of the poor water-solubility and the main strategies applied to increase the solubility of poorly water-soluble drugs. The relationship between the biopharmaceutical classification system and the solubilization process of the drug is also considered. Finally, it includes how drug nanoparticles and nanocarriers, especially lipid-based nanosystems, can overcome these challenges and which of these approaches are already available on the market. Expert opinion: Due to the growing importance of nanomedicines, especially for applications in poorly water-soluble drugs, it is important to clearly establish the specifications and quality criteria for nanonized drugs to ensure the quality and safety of nanoparticles.

Abstract:Abstract Background: Fenofibrate (FF) is a BCS class II compound whose poor solubility poses challenges in drug delivery and bioavailability. Solid self-micro emulsifying drug delivery systems (S-SMEDDS) have emerged as a promising solution to address these issues. These systems are aimed at enhancing the solubility and dissolution rates of poorly soluble drugs, such as FF, by formulating them into solid dosage forms. Methods: FF solubility was investigated in various oils, surfactants, and co-surfactants to identify the most suitable components for formulating S-SMEDDS. The preparation of S-SMEDDS was carefully evaluated according to parameters including drug content, morphological characteristics, and structural features. Two methods, freeze-drying, and spray-drying, were compared for their efficacy in producing S-SMEDDS. Additionally, in vitro dissolution studies were conducted to assess the dissolution rates of FF-loaded S-SMEDDS tablets compared with conventional tablets. Results: Among the oils tested, oleic oil achieved the highest FF solubility, whereas Tween 80 and Transcutol HP were identified as the optimal surfactant and co-surfactant, respectively. The preparation method significantly influenced the properties of S-SMEDDS. Freeze-drying outperformed the other methods by enhancing dissolution rates, primarily through increased surface area. Moreover, the solid-state characteristics of S-SMEDDS were dependent on the polymer concentration and processing method. In vitro dissolution studies demonstrated that FF-loaded S-SMEDDS tablets exhibited faster drug release than conventional tablets, owing to the inclusion of the super disintegrating agent CCS and the S-SMEDDS component. Freeze-drying was superior to spray-drying in enhancing dissolution, albeit with potentially higher production costs. Conclusions: The study highlights the potential of S-SMEDDS to overcome the solubility and bioavailability challenges associated with FF. Freeze-drying emerged as the preferred method for producing S-SMEDDS, because of its superior dissolution enhancement capabilities, despite potentially higher production costs, whereas spray-dried S-SMEDDS offers economic and environmental benefits, but may achieve lower dissolution rates. Overall, our findings underscore the importance of formulation strategy in enhancing the efficacy of poorly soluble drugs such as FF.

Abstract:The solid dispersions with poloxamer 188 (P188) and solid solutions with polyvinylpyrrolidone K30 (PVPK30) were evaluated and compared in an effort to improve aqueous solubility and bioavailability of a model hydrophobic drug. All preparations were characterized by differential scanning calorimetry, powder X-ray diffraction, intrinsic dissolution rates, and contact angle measurements. Accelerated stability studies also were conducted to determine the effects of aging on the stability of various formulations. The selected solid dispersion and solid solution formulations were further evaluated in beagle dogs for in vivo testing. Solid dispersions were characterized to show that the drug retains its crystallinity and forms a two-phase system. Solid solutions were characterized to be an amorphous monophasic system with transition of crystalline drug to amorphous state. The evaluation of the intrinsic dissolution rates of various preparations indicated that the solid solutions have higher initial dissolution rates compared with solid dispersions. However, after storage at accelerated conditions, the dissolution rates of solid solutions were lower due to partial reversion to crystalline form. The drug in solid dispersion showed better bioavailability in comparison to solid solution. Therefore, considering physical stability and in vivo study results, the solid dispersion was the most suitable choice to improve dissolution rates and hence the bioavailability of the poorly water soluble drug.

Abstract:Water solubility has been identified as a critical parameter and the main responsible by affecting poor performance of oral drug delivery. Poorly soluble drugs can originate unsatisfactory ADME properties leading to low oral bioavailability, insufficient chemical stability, low half-life, fast pre-systemic metabolism and difficulties in formulation. In this context, the prodrug design is an alternative in order to improve physicochemical, biopharmaceutical and pharmacokinetic properties such as permeability, solubility, bioavailability, chemical stability and metabolism of molecules presenting poor drug-like properties. In this article we highlight the importance of the prodrug design in the early stages of drug discovery and development process, in an attempt to diminish the attrition rate and end up falling into the valley of death. Selected examples of this strategy are provided in this review and they are classified by some basic functional groups that are amenable to the prodrug approach with the aim of increasing aqueous solubility of poorly water-soluble compounds. Over the past decade, the number of approved prodrugs is considerable among all drugs launched in the market, emphasizing the importance of this tool on drug design. It is reported that 10% of all marketed drug worldwide can be classified as prodrugs. Furthermore, prodrugs designed to be more water soluble launched in the past decade are summarized in a table to have a closer look and finally state that the prodrug design is an amenable approach to increase water solubility.

Abstract:Despite numerous research efforts, drug delivery through the oral route remains a major challenge to formulation scientists. The oral delivery of drugs poses a significant challenge because more than 40% of new chemical entities are practically insoluble in water. Low aqueous solubility is the main problem encountered during the formulation development of new actives and for generic development. A complexation approach has been widely investigated to address this issue, which subsequently improves the bioavailability of these drugs. This review discusses the various types of complexes such as metal complex (drug-metal ion), organic molecules (drug-caffeine or drug-hydrophilic polymer), inclusion complex (drug-cyclodextrin), and pharmacosomes (drug-phospholipids) that improves the aqueous solubility, dissolution, and permeability of the drug along with the numerous case studies reported in the literature. Besides improving solubility, drug-complexation provides versatile functions like improving stability, reducing the toxicity of drugs, increasing or decreasing the dissolution rate, and enhancing bioavailability and biodistribution. Apart, various methods to predict the stoichiometric ratio of reactants and the stability of the developed complex are discussed.

Enhanced Bioavailability of the Poorly Water-Soluble Drug Fenofibrate by Using Liposomes Containing a Bile Salt

Abstract:The main purpose of this study was to evaluate oral bioavailability of the poorly water-soluble drug fenofibrate when liposomes containing a bile salt were used as oral drug delivery systems. Liposomes composed of soybean phosphotidylcholine (SPC) and sodium deoxycholate (SDC) were prepared by a dry-film dispersing method coupled with sonication and homogenization. Several properties of the liposomes, including particle size, entrapment efficiency and membrane fluidity, were extensively characterized. In vitro release experiments indicated that no more than 20% of total fenofibrate was released from SPC/cholesterol (CL) and SPC/SDC liposomes at 2 h, in contrast with near complete release for micronized fenofibrate capsules. Strikingly, in vivo measurements of pharmacokinetics and bioavailability demonstrated higher rates of fenofibrate absorption from both SPC/SDC and SPC/CL liposomes than micronized fenofibrate. The bioavailability of SPC/SDC and SPC/CL liposomes was 5.13- and 3.28-fold higher, respectively, than that of the micronized fenofibrate. The disparity between oral bioavailability and in vitro release for liposomes strongly suggests alternative absorption mechanisms rather than enhanced release. Importantly, SPC/SDC liposomes exhibited a 1.57-fold increase in bioavailability relative to SPC/CL liposomes, indicating that liposomes containing bile salts may be used to enhance oral bioavailability of poorly water-soluble drugs.
