Abstract:Switchgrass is an important bioenergy crop valued for its biomass yield and abiotic tolerance. Alkali stress is a major abiotic stress that significantly impedes plant growth and yield due to high salinity and pH; however, the response mechanism of switchgrass to alkali stress remains limited. Here, we characterized PvARL1, an ARF-like gene, which was up-regulated in both the shoot and root tissues under alkali stress conditions. Overexpression of PvARL1 not only improved alkali tolerance but also promoted biomass yield with more tiller and higher plant height in switchgrass. Moreover, PvARL1 overexpression lines displayed higher capacities in the maintenance of water content and photosynthetic stability compared with the controls under alkali treatments. A significant reduction in the ratio of electrolyte leakage, MDA content, and reactive oxygen species (ROS) showed that PvARL1 plays a positive role in protecting cell membrane integrity. In addition, PvARL1 also negatively affected the K+ efflux or uptake in roots to alleviate ion toxicity under alkali treatments. Overall, our results suggest that PvARL1 functions as a positive regulator in plant growth as well as in the plant response to alkali stress, which could be used to improve switchgrass biomass yield and alkali tolerance genetically.

Abstract:In plants, Δ1-pyrroline- 5-carboxylate synthase (P5CS) is the rate-limiting enzyme in proline biosynthesis. In this study, we introduced the LpP5CS (Lolium perenne L.) gene into switchgrass by Agrobacterium-mediated transformation. The transgenic lines (TG) were classified into two groups based on their phenotypes and proline levels. The group I lines (TG4 and TG6) had relatively high proline levels and improved biomass yield. The group II lines (TG1 and TG2) showed low proline levels, severely delayed flowering, stunted growth and reduced biomass yield. Additionally, we used RNA-seq analysis to detect the most significant molecular changes, and we analyzed differentially expressed genes, such as flowering-related and CYP450 family genes. Moreover, the biomass yield, physiological parameters, and expression levels of reactive oxygen species scavenger-related genes under salt stress all indicated that the group I plants exhibited significantly increased salt tolerance compared with that of the control plants, in contrast to the group II plants. Thus, genetic improvement of switchgrass by overexpressing LpP5CS to increase proline levels is feasible for increasing plant stress tolerance.

Abstract:Understanding the regulation of proline metabolism necessitates the suppression of two Δ1-pyrroline-5-carboxylate synthetase enzyme (P5CS) genes performed in switchgrass (Panicum virgatum L.). The results reveal that overexpressing PvP5CS1 and PvP5CS2 increased salt tolerance. Additionally, transcript levels of spermidine (Spd) and spermine (Spm) synthesis and metabolism related genes were upregulated in PvP5CS OE-transgenic plants and downregulated in the PvP5CS RNAi transformants. According to salt stress assay and the measurement of transcript levels of Polyamines (PAs) metabolism-related genes, P5CS enzyme may not only be the key regulator of proline biosynthesis in switchgrass, but it may also indirectly affect the entire subset of pathway for ornithine to proline or to putrescine (Put). Furthermore, application of proline prompted expression levels of Spd and Spm synthesis and metabolism-related genes in both PvP5CS-RNAi and WT plants, but transcript levels were even lower in PvP5CS-RNAi compared to WT plants under salt stress condition. These results suggested that exogenous proline could accelerate polyamines metabolisms under salt stress. Nevertheless, the enzymes involved in this process and the potential functions remain poorly understood. Thus, the aim of this study is to reveal how proline functions with PAs metabolism under salt stress in switchgrass.
Whether the ADP-ribosyltransferase Activity of Ta-sro1, a Noncanonical PARP Protein, Contributes to Its Function in Salinity-Stress Tolerance?

Abstract:AbstractADP-ribosylation mediated by ADP-ribosyltransferases (ARTs) is an intricate modification that regulates diverse cellular processes including DNA repair, chromatin remodeling and gene transcription responding to stresses. In addition to the canonical poly(ADP-ribose) polymerases (PARPs), plant specific SRO (Similar to RCD One) family also contain the catalytic core of the PARP domain. However, whether the PARP domains in SROs execute the ART function is still under debate. In 2014, we reported a wheat SRO, Ta-sro1, had the ADP-ribosyltransferase activity and enhanced wheat seedling growth and abiotic stress resistance, however, a recent work by Vogt et al. showed that Ta-sro1 without ADP-ribosyltransferase activity. Based on the recent progress on PARPs and SROs in relation to ADP-ribosyltransferase activity, along with our former and recent evolving results, we argued that Ta-sro1 is a non-canonical ADP-ribosyltransferase with the enzymatic activity. Although we have revealed the novel mechanism of Ta-sro1 regulate redox homeostasis and enhance salinity stress tolerance through interacting with TaSIP1, it is of interest to further clarify whether and how the enzymatic activity of Ta-sro1 responsible for the salinity tolerance of wheat. Our study raises some interesting points and caveats that helpful for understanding the research progresses and debates about the enzymatic activity of SROs.
Small GTPase PvARFR2 Interacts with Cytosolic ABA Receptor Kinase3 to Enhance Alkali Tolerance in Switchgrass

Abstract:Soil alkalization has become a serious problem that limits plant growth through osmotic stress, ionic imbalance, and oxidative stress. Understanding how plants resist alkali stress has practical implications for alkaline-land utilization. In this study, we identified a small GTPase, PvARFR2 (ADP ribosylation factors related 2), that positively regulates alkali tolerance in switchgrass (Panicum virgatum) and uncovered its potential mode of action. Overexpressing PvARFR2 in switchgrass and Arabidopsis (Arabidopsis thaliana) conferred transformant tolerance to alkali stress, demonstrated by alleviated leaf wilting, less oxidative injury, and a lower Na+/K+ ratio under alkali conditions. Conversely, switchgrass PvARFR2-RNAi and its homolog mutant atgb1 in Arabidopsis displayed alkali sensitives. Transcriptome sequencing analysis showed that cytosolic abscisic acid (ABA) receptor kinase PvCARK3 transcript levels were higher in PvARFR2 overexpression lines compared to the controls and were strongly induced by alkali treatment in shoots and roots. Phenotyping analysis revealed that PvCARK3-OE x atgb1 lines were sensitive to alkali similar to the Arabidopsis atgb1 mutant, indicating that PvARFR2/AtGB1 functions in the same pathway as PvCARK3 under alkaline stress conditions. Application of ABA on PvARFR2-OE and PvCARK3-OE switchgrass transformants resulted in ABA sensitivity. Moreover, we determined that PvARFR2 physically interacts with PvCARK3 in vitro and in vivo. Our results indicate that a small GTPase, PvARFR2, positively responds to alkali stress by interacting with the cytosolic ABA receptor kinase PvCARK3, connecting the alkaline stress response to ABA signaling. A small GTPase interacts with a cytosolic abscisic acid receptor kinase to enhance alkali tolerance in switchgrass via the abscisic acid signaling pathway.

Abstract:以耐盐性较强的大豆( Glycine max L . )品种铁丰8号为试验材料，克隆到1个A亚族bZIP类转录因子基因，命名为 GmAREB ( Glycine max ABA responsive element binding protein)。该基因由1 317个核苷酸组成，编码439个氨基酸残基，包括4个保守的磷酸化位点区域(C1、C2、C3和C4)、1个核定位信号区(KVVE)和1个bZIP转录因子保守域。聚类分析显示GmAREB蛋白与拟南芥ABF2、水稻OsTRAB1具有较高的同源性，并存在较近的亲缘关系。采用凝胶阻滞实验方法证明GmAREB蛋白与ABRE顺式元件具有体外结合特异性。功能分析结果表明, 在干旱胁迫条件下, GmAREB 转基因拟南芥的存活率(50%)显著高于野生型(5%)；气孔统计分析显示转基因植株的气孔开度(0.8 μm)明显比对照开度小(2.6 μm)。氧化胁迫结果显示 GmAREB 转基因拟南芥在甲基紫精溶液中叶绿素含量比野生型高(7.3 mg g –1 FW)。转基因拟南芥RT-PCR分析表明， GmAREB 基因过表达能够增强下游胁迫相关基因 ABI1 、 ABI2 的表达，抑制气孔开闭相关基因 KAT1 、 KAT2 的表达。综上所述， GmAREB 基因过表达有效调控了转基因拟南芥下游靶基因表达，加速了气孔关闭，减少了水分蒸发和叶绿素降解，从而提高了转基因拟南芥对干旱、氧化胁迫耐性。
PvNAC1 Increases Biomass and Enhances Salt Tolerance by Decreasing Na+ Accumulation and Promoting ROS Scavenging in Switchgrass (panicum Virgatum L.).

Abstract:Switchgrass (Panicum virgatum L.) is a bioenergy crop; thus, it is important to improve biomass to effectively produce bioethanol, particularly under adverse stress conditions. NAC transcription factors are involved in the abiotic stress response. PvNAC1 was isolated in the nucleus of switchgrass, with its C-terminal region containing a transcriptional activation domain. PvNAC1 expression was induced by dehydration, salt, H2O2, and abscisic acid treatments. Overexpressing (OE) PvNAC1 improved growth performance, leading to significantly taller and heavier (dry weight) plants. Moreover, cellulose content was significantly higher in OE plants, indicating that PvNAC1 plays an important role regulating growth and bioethanol production. PvNAC1 RNA interference (RNAi) switchgrass plants exhibited reduced dry weight and cellulose content. OE PvNAC1 enhanced tolerance to salt stress, through higher reactive oxygen species scavenging ability and less Na+ and more K+ accumulation in roots and shoots. RNAi plants were more sensitive to salt stress. The quantitative polymerase chain reaction results revealed that some stress responsive genes, three antioxidant enzymatic genes, and an ion homeostasis-related gene were upregulated in OE plants and downregulated in RNAi plants. These results show that PvNAC1 functions as a transcriptional activator in response to salt stress and growth.
Enhanced Growth Performance and Salinity Tolerance in Transgenic Switchgrass via Overexpressing Vacuolar Na (K)/H Antiporter Gene ().

Abstract:Switchgrass (Panicum virgaturn L.) has been increasingly recognized as one of the most valuable perennial bioenergy crop. To improve its biomass production, especially under salt stress, we isolated a putative vacuolar Na+ (K+)/H+ antiporter gene from switchgrass and designated as PvNHX1 Subcellular localization revealed that this protein was localized mainly on the vacuole membrane. The PvNHX1 was found to be expressed throughout the entire growth period of switchgrass, exhibited preferentially expressed in the leaf tissue, and highly induced by salt stress. Transgenic switchgrass overexpressing PvNHX1 showed obvious advantages with respect to plant height and leaf development compared to the wild-type (WT) and transgenic control (EV, expressing the empty vector only) plants, suggesting PvNHX1 may serve as a promoter in switchgrass growth and development. Moreover, transgenic switchgrass were more tolerant than control plants with better growth-related phenotypes (higher shoot height, larger stem diameter, longer leaf length, and width) and physiological capacities (increased proline accumulation, reduced malondialdehyde production, preserved cell membrane integrity, etc.) under high salinity stress. Furthermore, the genes related to cell growth, flowering, and potassium transporters in transgenic switchgrass exhibited a different expression profiles when compared to the control plants, indicating a pivotal function of PvNHX1 in cell expansion and K+ homeostasis. Taken together, PvNHX1 is essential for normal plant growth and development, and play an important role in the response to salt stress by improving K+ accumulation. Our data provide a valuable foundation for further researches on the molecular mechanism and physiological roles of NHXs in plants.

Abstract:KEY MESSAGE:Overexpression of OsDST-SRDX chimeric gene in switchgrass promotes plant growth and improves the salt tolerance of transgenic switchgrass by improving its antioxidative ability. Switchgrass (Panicum virgatum L.) is a forage and model feedstock plant. To avoid competing with crops in arable land utilization, improving salt tolerance of switchgrass is required to use marginal saline land for switchgrass production. To improve salt tolerance of switchgrass, a chimeric DROUGHT AND SALT TOLERANCE (DST) gene OsDST-SRDX was constructed using the Chimeric REpressor gene-Silencing Technology (CRES-T), and introduced into switchgrass genome by Agrobacterium-mediated transformation. Compared to wild-type (WT) plants, OsDST-SRDX transgenic (TG) switchgrass plants showed wider leaves and thicker stems. They performed better under salt stress, had higher relative leaf water content, lower electrolyte leakage and lower malondialdehyde (MDA) content, and accumulated less Na+ and more K+ than WT controls. The transgenic plants had also higher activities of antioxidant enzymes associated with suppressed expressing of genes in H2O2 homeostasis, including glutathione S-transferase (GST2, GST6), cytochrome P450, peroxidase 24 precursor, and induced expressing of CAT and SOD under salt stress to eliminate excess H2O2. Our results indicate that overexpression of the chimeric gene OsDST-SRDX improves salt tolerance of switchgrass, a C4 biofuel crop.

Improved drought tolerance and higher water use efficiency (WUE) are important for its successful field establishment and production, especially on marginal lands. Aquaporins are key channels and regulators for water transportation and maintenance of cellular water status. In this study, the functional role of an aquaporin gene,PvPIP2;9, in switchgrass was studied. Expression ofPvPIP2;9was regulated by diurnal oscillation and osmotic stress. Constitutive over-expressingPvPIP2;9in switchgrass significantly improved its leaf length, plant height, aboveground biomass and biomass protein contents under both drought- and non-stressed conditions, and also resulted in higher cellulose content in drought-stressed transgenic than wild-type plants. Under 21 days of drought treatment, transgenic plants showed less electrolyte leakage rates, but higher relative water contents, photochemical efficiencies, chlorophyll contents and WUE, indicating that PvPIP2;9 positively regulated plant drought tolerance. Moreover, expression patterns of all 14 switchgrassPIP2subfamily genes were checked during the drought treatment, and the result showed that over-expressingPvPIP2;9also affected transcript levels of most otherPIP2genes. Together, this study showed that improved biomass yield, drought tolerance and higher WUE can be achieved by manipulating the expression ofPvPIP2;9and also suggestedPIP2subfamily genes were transcriptionally regulated in a coordinated manner, which can be used for potential targets for crop drought resistance breeding efforts.

Proline improves switchgrass growth and development by reduced lignin biosynthesis

Abstract:Transgenic switchgrass overexpressing Lolium perenne L. delta1-pyrroline 5-carboxylate synthase (LpP5CS) in group I (TG4 and TG6 line) and group II (TG1 and TG2 line) had significant P5CS and ProDH enzyme activities, with group I plants (TG4 and TG6) having higher P5CS and lower ProDH enzyme activity, while group II plants had higher ProDH and lower P5CS enzyme activity. We found group II transgenic plants showed stunted growth, and the changed proline content in overexpressing transgenic plants may influence the growth and development in switchgrass. RNA-seq analysis showed that KEGG enrichment included phenylpropanoid biosynthesis pathway among group I, group II and WT plants, and the expression levels of genes related to lignin biosynthesis were significantly up-regulated in group II. We also found that lignin content in group II transgenic plants was higher than that in group I and WT plants, suggesting that increased lignin content may suppress switchgrass growth and development. This study uncover that proline can appropriately reduce lignin biosynthesis to improve switchgrass growth and development. Therefore, appropriate reduction in lignin content and increase in biomass are important for bioenergy crop to lower processing costs for biomass fermentation-derived fuels.
PvMYB106 Increases Switchgrass Biomass Via Activating Photosystem II Subunit Protein PvPsbP and Carbonic Anhydrase PvCA Expression

Abstract:Switchgrass (Panicum virgatum) is a model herbaceous bioenergy crop performing C4 photosynthesis as an alternative to fossil-based feedstocks for industrial applications. Improving photosynthetic capacity is one of the potential strategies to increase switchgrass yield. Through genome-wide bioinformatic analyses of MYB genes in switchgrass, we identified several MYBs that might play regulatory roles in plant photosynthesis, including PvMYB106. Here, we show that PvMYB106 is localized in the nucleus and cytoplasm, has transcriptional activation activity, and its expression pattern is in agreement with photosynthetic rhythm. Overexpressing PvMYB106 in switchgrass promotes the biomass accumulation to a maximum of 82% as compared to the control plants, we further show that the increased biomass in switchgrass is due to enhanced photosynthetic capacity as manifested by higher Pn (net photosynthetic rate). Comparative transcriptomic analyses and dual-luciferase experiment confirm that PvMYB106 mediates photosynthetic process through up-regulating expression levels of photosynthetic genes including genes encoding subunit of photosystem II protein (PvPsbP) and carbonic anhydrase (PvCA). Taken together, our study demonstrates that PvMYB106 is a new elicitor for switchgrass photosynthesis enhancement, and provides a subset of potential downstream candidate genes for genetic improvements of biofuel crops.
Characterization of the M6a Gene Family in Switchgrass and Functional Analysis of PvALKBH10 During Flowering

Abstract:N6-methyladenosine (m6A), a nucleotide modification that is frequently seen in RNA, plays a crucial role in plant growth, development and stress resistance. However, the m6A regulatory machinery in switchgrass (Panicum virgatum L.), a model plant for cellulose-to-ethanol conversion, remains largely unknown. In this study, we identified 57 candidate genes involved in m6A-regulation in the switchgrass genome, and analyzed their chromosomal distribution, evolutionary relationships, and functions. Notably, we observed distinct gene expression patterns under salt and drought stress, with salt stress inducing writer and eraser genes, alongside drought stress predominantly affecting reader genes. Additionally, we knocked out PvALKBH10, an m6A demethylase gene, via CRISPR/Cas9 and found its potential function in controlling flowering time. Inoculation with Azorhizobium caulinodans ORS571 enhances plant growth and salt tolerance of switchgrass (Panicum virgatum L.) seedlings

Abstract:Background: Switchgrass (Panicum virgatum L.) is an important biofuel crop that may contribute to replacing petroleum fuels. However, slow seedling growth and soil salinization affect the growth and development of switchgrass. An increasing number of studies have shown that beneficial microorganisms promote plant growth and increase tolerance to salinity stress. However, the feasibility of inoculating switchgrass with Azorhizobium caulinodans ORS571 to enhance the growth and salt tolerance of its seedlings is unclear. Our previous study showed that A. caulinodans ORS571 could colonize wheat (Triticum aestivum L.) and thereby promote its growth and development and regulate the gene expression levels of microRNAs (miRNAs). Results: In this study, we systematically studied the impact of A. caulinodans ORS571 on switchgrass growth and development and the response to salinity stress; we also studied the underlying mechanisms during these biological processes. Inoculation with A. caulinodans ORS571 significantly alleviated the effect of salt stress on seedling growth. Under normal conditions, A. caulinodans ORS571 significantly increased fresh plant weight, chlorophyll a content, protein content, and peroxidase (POD) activity in switchgrass seedlings. Under salt stress, the fresh weight, dry weight, shoot and root lengths, and chlorophyll contents were all significantly increased, and some of these parameters even recovered to normal levels after inoculation with A. caulinodans ORS571. Soluble sugar and protein contents and POD and superoxide dismutase (SOD) activities were also significantly increased, contrary to the results for proline. Additionally, A. caulinodans ORS571 may alleviate salt stress by regulating miRNAs. Twelve selected miRNAs were all upregulated to different degrees under salt stress in switchgrass seedlings. However, the levels of miR169, miR171, miR319, miR393, miR535, and miR854 were decreased significantly after inoculation with A. caulinodans ORS571 under salt stress, in contrast to the expression level of miR399. Conclusion: This study revealed that A. caulinodans ORS571 increased the salt tolerance of switchgrass seedlings by increasing their water content, photosynthetic efficiency, osmotic pressure maintenance, and reactive oxygen species (ROS) scavenging abilities and regulating miRNA expression. This work provides a new, creative idea for improving the salt tolerance of switchgrass seedlings.
Downregulation of miR156-Targeted PvSPL6 in Switchgrass Delays Flowering and Increases Biomass Yield

Abstract:MiR156/ SQUAMOSA PROMOTER BINDING-LIKE s ( SPL s) module is the key regulatory hub of juvenile-to-adult phase transition as a critical flowering regulator. In this study, a miR156-targeted PvSPL6 was identified and characterized in switchgrass ( Panicum virgatum L.), a dual-purpose fodder and biofuel crop. Overexpression of PvSPL6 in switchgrass promoted flowering and reduced internode length, internode number, and plant height, whereas downregulation of PvSPL6 delayed flowering and increased internode length, internode number, and plant height. Protein subcellular localization analysis revealed that PvSPL6 localizes to both the plasma membrane and nucleus. We produced transgenic switchgrass plants that overexpressed a PvSPL6 - GFP fusion gene, and callus were induced from inflorescences of selected PvSPL6-GFP OE transgenic lines. We found that the PvSPL6-GFP fusion protein accumulated mainly in the nucleus in callus and was present in both the plasma membrane and nucleus in regenerating callus. However, during subsequent development, the signal of the PvSPL6-GFP fusion protein was detected only in the nucleus in the roots and leaves of plantlets. In addition, PvSPL6 protein was rapidly transported from the nucleus to the plasma membrane after exogenous GA 3 application, and returned from the plasma membrane to nucleus after treated with the GA 3 inhibitor (paclobutrazol). Taken together, our results demonstrate that PvSPL6 is not only an important target that can be used to develop improved cultivars of forage and biofuel crops that show delayed flowering and high biomass yields, but also has the potential to regulate plant regeneration in response to GA 3 .

Abstract:Arylalkylamine N-acetyltransferase (AANAT) catalyses the acetylation of serotonin, a rate-limiting process in melatonin biosynthesis. To obtain better insight into the underlying mechanism of AANAT's actions in switch-grass growth, flowering and defence, we performed integrated morphological, physiological and omics analyses between overexpressed oAANAT transgenic lines in wild-type and transgenic control (expressing only the empty vector) plants. We showed that oAANAT played pivotal roles in modulating plant growth through its regulation of cell elongation, and regulating flowering through photoperiod and GA pathways. In relation to photosynthesis, oAANAT promoted photosynthetic efficiency primarily through regulating leaf anatomical structures, stomatal development and chlorophyll metabolism. Moreover, oAANAT overexpression can trigger a number of defence responses or strategies, including antioxidant enzymatic properties, non-enzymatic capacity, significantly activated phenylpropanoid biosynthesis, and adaptive morphological characteristics. This study unveils the possible molecular mechanisms underlying oAANAT dependent melatonin functions in switchgrass, providing an important starting point for further analyses.

Abstract:Improving plant biomass yield and/or feedstock quality for highly efficient lignocellulose conversion has been the main research focus in genetic modification of switchgrass (Panicum virgatum L.), a dedicated model plant for biofuel production. Here, we proved that overexpression of miR396 (OE‐miR396) leads to reduced plant height and lignin content mainly by reducing G‐lignin monomer content. We identified nineteen PvGRFs in switchgrass and proved thirteen of them were cleaved by miR396. MiR396‐targeted PvGRF1, PvGRF9 and PvGRF3 showed significantly higher expression in stem. By separately overexpressing rPvGRF1, 3, and 9, in which synonymous mutations abolished the miR396 target sites, and suppression of PvGRF1/3/9 activity via PvGRF1/3/9‐SRDX overexpression in switchgrass, we confirmed PvGRF1 and PvGRF9 played positive roles in improving plant height and G‐lignin content. Overexpression of PvGRF9 was sufficient to complement the defective phenotype of OE‐miR396 plants. MiR396‐PvGRF9 modulates these traits partly by interfering GA and auxin biosynthesis and signaling transduction and cell wall lignin, glucose and xylan biosynthesis pathways. Moreover, by enzymatic hydrolysis analyses, we found that overexpression of rPvGRF9 significantly enhanced per plant sugar yield. Our results suggest that PvGRF9 can be utilized as a candidate molecular tool in modifying plant biomass yield and feedstock quality.

Abstract:BackgroundIntracellular Na+ (K+)/H+ antiporters (NHXs) have pivotal functions in regulating plant growth, development, and resistance to a range of stresses. To gain insight into the molecular events underlying their actions in switchgrass (Panicum virgatum L.), we analyzed transcriptomic changes between PvNHX1-overexpression transgenic lines and wild-type (WT) plants using RNA sequencing (RNA-seq) technology.ResultsThe comparison of transcriptomic data from the WT and transgenic plants revealed a large number of differentially expressed genes (DEGs) in the latter. Gene ontology (GO) and KEGG pathway analyses showed that these DEGs were associated with a wide range of functions, and participated in many biological processes. For example, we found that PvNHX1 had an important role in plant growth through its regulation of photosynthetic activity and cell expansion. In addition, PvNHX1 regulated K+ homeostasis, cell expansion and pollen development, indicating that it has unique and specific roles in flower development. We also found that transgenic switchgrass exhibited a higher level of transcription of defense-related genes, especially those involved in disease resistance.ConclusionWe showed that PvNHX1 had an important role in plant growth and development through its regulation of photosynthetic activity, cell expansion, K+ homeostasis, and pollen development. Additionally, PvNHX1 overexpression activated a complex signal transduction network in response to various biotic and abiotic stresses. In relation to plant growth, development, and defense responses, PvNHX1 also had a vital regulatory role in the formation of a series of plant hormones and transcription factors (TFs). The reliability of the RNA-seq data was confirmed by quantitative real-time PCR. Our data provide a valuable foundation for further research into the molecular mechanisms and physiological roles of NHXs in plants.
