Abstract:OBJECTIVE:Human embryonic stem (hES) cells are considered to be a valuable resource for research in regenerative medicine, drug screening, and developmental studies. However, hES cells are usually established and maintained on mouse embryonic fibroblast feeder layers, and the risk of animal origin contamination from feeder layer generally excludes the clinical use of these hES cells. The main emphasis over the last several years has been in finding defined serum-and feeder layer-free system for derivation and culture of hES cells to enable the clinical use of hES cell for cell transplantation.

Abstract:Mouse embryonic stem cells (mESCs) have played a key role in the newly emerging fields of stem cell research. The traditional derivation and culture of mESCs have been based on the use of mouse embryonic fibroblasts (MEFs) treated with exogenous leukemia inhibitory factor (LIF). However, the rapid senescence of MEFs, coupled with the high cost of LIF, has significantly hampered the widespread use of mESCs in stem cell research. Thus, we present a novel exogenous LIF-free culture system for general mESCs applications, comprising fibroblast-like cells derived from the rabbit spleen (RSFs). We demonstrated that mESCs cultured on RSFs (mESCs-RSFs) maintained all mESC features after prolonged LIF-free culture, including alkaline phosphatase, cell surface markers (SSEA-1), molecular markers (OCT-4, NANOG, TERT, REX-1), karyotype, and pluripotency. The high expression level of both LIF and WNT3A in the RSFs may account for their ability to maintain mESCs without exogenous LIF. Moreover, this exogenous LIF-free culture system was verified to be of microbiological quality through analysis with electron transmission microscopy.
Study on Culture of Human Embryo Blastocyst and Its Related Factors

Abstract:Objectives To investigate the culture of human embryo blastocyst in vitro and its related factors. Methods The 54 IVF surplus embryos of day 3 from 18 volunteers were transferred to different mediums (IVC-Three medium,blastocyst medium or HTF medium) containing 10%SSS for advanced culture.The growth of embryos was observed until the embryo degeneration or 7 days after fertilization.Results The total blastulation rates of 54 embryos were 29.6%.The positive correlation between the cell number of day 3 embryo and blastulation rate was significant(r=0.99,P0.01).And the higher blastulation rate was positively related to embryo quality score (34.9% for 1～2.5 degree versus 9.1% for 3～4degree).The blastulation rate of IVC-Three medium was significantly higher than that of blastocyst medium and HTF(χ21=5.77,χ22=3.95,P0.05).Conclusion Advanced culture of surplus embryo will reach some blastulation.The cell number of day 3 embryo and quality score have some indication to blastulation.IVC-Three blastocyst medium may have potential clinical use for embryo blastocyst culture.

Abstract:Human embryonic stem(hES) cells are pluripotent cells derived from the inner cell mass(ICM) cells of preimplantation blastocysts with the potential to self-renew and differentiate.As hES cells can be induced to differentiate into numerous cell types of all three germ layers under in vitro and in vivo conditions,they are potentially valuable for the basic research and clinical application,including researches on development of early human embryo,screening drugs and toxins,cell transplantation,gene therapy,etc.Many hES cell lines have been established under different conditions in the world.The firstly established hES cell lines were cultured on mouse embryonic fibroblast(MEF) cells with medium containing many undefined animal compo-nents such as fetal bovine serum,which may cause cross-transfection with animal pathogen and mycoplasm.In recent years,scientists had made great efforts to optimize the culture conditions for hES cells and achieved considerable progresses forward.Today,hES cells are derived and cultured under defined serum-free and feeder-free conditions without xenogeneic proteins,which to some extent solved the problems mentioned above.In this review,We will discuss the new progresses and unsolved issues on optimizing culture condi-tions for hES derivation and maintaining its undifferentiated state,mainly about feeder cell layer,feeder-free culture system,matrix and cytokines.

Abstract:Human embryonic stem cells(hESC) are self-renew unlimitedly and pluripotent,they have an ability to differentiate to multiple cell types representing all primitive embryonic germ layers: ectoderm,mesoderm and endoderm under appropriate conditions.These properties imply great potential in regenerative medicine(cell therapy),drug screening,and developmental studies.However,the medical application of hESCs is hampered by many animal products in the culture medium and/or the complexity of conditions required for hESC growth.Therefore,it is important to establish a defined medium without animal products.This review summarizes the progress on the culture of hESC for clinical use and studies.

Abstract:Objective: The embryonic stem cell(ES) culture system(feeder layer+RH-CM medium) was used in this experiment to coculture mouse early stage embryos in order to study whether this system could ensure the development of the embryos and increase the rate of the embryo development.Methods: Five hundred and twenty-one embryos were divided into 10 groups according to the different feeder layers(mouse embryonic fibroblast,MEF;SIM mouse embryo-derived thioquanine and ouabain(resistant) cells,STO; oviduct epithelial cell,OEC) and conditioned medium.The effect of different culture conditions on the rate of embryo development in vitro was compared.Results: The ES cell culture system was better than the conventional culture system for the embryo development(P<0.05).When embryos were cultured in ES culture system,STO feeder achieved a(higher) rate of embryo development than MEF,while both STO and MEF were significantly better than OEC for the embryo development(P<0.05).Conclusions: The results suggest that this system can apparently increases the development rate of the mouse early stage embryos.Using ES cell culture system to coculture early stage embryos,STO is a better feeder than MEF for the embryo development.

Abstract:The experiment was designed to study the influencing factors of isolation and culture of KM mouse embryonic stem cell(ESC).In this experiment,conditioned medium was made by cadiocyte from 3～5 d rat for the first time,effect of four culture system and different feeder layers on isolation and culture of ESC were studied.The result showed that culture medium adding LIF or RH-CM had a significant difference in adherent ratio,ICM formation ratio,primary ESC formation and inhibiting differentiation comparing with the culture medium without LIF or RH-CM,feeder layer made by MEF had a better effect of isolation and culture of ESC than feeder layer made by mouse sertoli cells.

Abstract:【Objective】 By comparing the undifferentiated and pluripotent marks of hESC in four different culture systems to evaluate and promote more optimal hESC culture environment.【Methods】 The four culture systems consist of xeno-free human foreskin fibroblasts(XF-HFF) combined with chemically-defined medium(CDM)(XF-HFF/CDM group),XF-HFF combined with 200 mL/L HS medium(XF-HFF/HS group),HS-matrix coated on plates without feeder layers combined with CDM(HS-matrix/CDM group),and xeno-contained human foreskin fibroblasts(XC-HFF) combined with 200 mL/L KSR medium(XC-HFF/KSR group).The culture efficiencies were evaluated by morphology,cell growth curve,flow cytometry and quantitative reverse transcription-polymerase chain reaction(qRT-PCR) among these culture systems.Further more,we characterized the stemness of hESC after long-term culture in XF-HFF/CDM culture system.【Results】 The rates of characteristic surface undifferentiated markers(SSEA4,Tra1-60) and proliferate factors(hTERT,Oct3/4) expression of hESC cultured in XF-HFF/CDM were significantly higher than which in XF-HFF/HS and in HS-matrix/CDM(P < 0.01),but were comparable to which in XC-HFF/KSR.After long-term cultured in XF-HFF/CDM,hESC still expressed surface undifferentiated markers of SSEA3,SSEA4 and pluripotent markers of hTERT,Oct3/4,and maintained pluripotency and karyotype stability,over 40 passages.【Conclusion】 The new xeno-free culture conditions demonstrated similar culture efficiency to KSR medium with HFF,and were more effective than CDM with HS-matrix,or HS medium with HFF.

Abstract:We have developed a combined micro-channel and micro-well system for easy cell loading, culture and post-culture operation on a chip. To demonstrate the reliability of the system, on chip cell culture and differentiation were performed with different types of substrates made of culture dish, glass cover slide and polydimethylsiloaxe (PDMS). As expected, mouse embryo fibroblasts (MEF) showed different adhesion and growth rate on different substrates. When embryonic stem (ES) cells were co-cultured with MEFs, the formation of ES colonies is efficient on both glass and Petri dish, although PDMS could also be used. Finally, ES cell differentiation with neuron growth factors was performed on different substrates, showing clear advantages of using culture Petri dish over both glass and PDMS.

Abstract:Background: Elimination of all animal components during derivation and long-term culture of human embryonic stem cells (hESCs) is necessary for future applications of hESCs in clinical cell therapy.Methods: In this study, we established the culture system of xeno-free human foreskin fibroblast feeders (XF-HFF) in combination with chemically defined medium (CDM). XF-HFF/CDM was compared with several conventional culture systems. The hESCs cultured in different media were further characterized through karyotype analysis, pluripotency gene expression, and cell differentiation ability.Results: The hESCs in the XF-HFF/CDM maintained their characteristics including typical morphology and stable karyotype. In addition, hESCs were characterized by fluorescent immunostaining of pluripotent markers and teratoma formation in vivo. RT-PCR analysis shown that the stem cell markers OCT3/4, hTERT, SOX2, and Nanog were present in the cell line hESC-1 grown on XF-HFF/CDM. Furthermore, the results of cell growth and expression of bFGF, Oct-4, and hTERT indicated that XF-HFF/CDM had better performance than human serum-matrix/CDM and XF-HFF/human serum.Conclusion: The comparison of different xeno-free culture conditions will facilitate clarifying the key features of self-renewal, pluripotency, and derivation and will shed light on clinic applications of hESCs.
Optimization of separation methods and culture system of chicken embryonic stem cells in vitro

Abstract:BACKGROUND:Embryonic stem cells are undifferentiated permanent cell line derived from inner cell mass cells and primordial germ cells of animal's early embryos.Chicken embryonic stem cells are derived from the blastodermal of a X-stage embryo.OBJECTIVE:To optim the separation method and in vitro cultural system of chicken embryonic stem cells.METHODS:The X-stage chicken embryos were isolated by using a small square of ?lter paper with a hole punched in the center,and the blastodermal cells were isolated by using the hair loop.STO cells were used to make feeder layer;at the same time,BRL-CM and cytokine were also used for chicken embryonic stem cells in vitro cultural system.RESULTS AND CONCLUSION:The filter paper loop and the hair loop could obtain complete the blastoderm,and the successful percentage was 75%-85%.The colony formation rate was about 50%.After culture in the BRL-CM + feeder layer + cytokine culture system,the passage of CES cells is the seventh generation;BRL-CM + feeder layer + cytokines,cultured chicken embryonic stem cells could passage to the 25th generation.Isolated chicken embryonic stem cells were in an undifferentiated state detected by alkaline phosphatase staining and SSEA-1 staining.The findings indicate that this experiment not only optimized the isolation method of chicken embryonic stem cells to obtain complete and pure embryos,but also further improved the in vitro culture system of chicken embryonic stem cells.

Abstract:Background: The importance of embryonic stem cells (ESCs) in chickens is undeniable, as they can be applied across various fields, including animal modeling, developmental biology, cell fate research, drug screening, toxicity testing, and gene function studies. However, a widely applicable culture system for chicken ESCs has yet to be developed. Objectives: This study aimed to investigate the effects of different culture systems on the derivation and maintenance of chicken ESCs, with a focus on optimizing the selected culture conditions. Methods: To achieve this, we tested the effectiveness of various species-specific ESC media in the derivation and culture of chicken PGCs, while incorporating different small molecule compounds to optimize the process. The pluripotency and differentiation potential of the resulting ESC-like cells were also evaluated. Results: The combination of PD0325901, SB431542, and LIF (R2i+LIF system) was found to be effective in generating chicken ESC-like clones. Further experiments showed that enhancing the R2i+LIF system with cytokines such as SCF and FGF2 significantly extended the culture period and increased the passage number of chicken ESC-like cells. These ESC-like cells were characterized through positive alkaline phosphatase staining and the expression of pluripotency markers POUV, NANOG, and SOX2. Additionally, differentiation assays confirmed their ability to form the three germ layers. Conclusions: The newly developed culture system provides suitable conditions for the short-term culture of chicken ESCs. However, further optimization is required to establish a system that can sustain long-term maintenance.

Abstract:The ideal medium for human pluripotent stem cells (hPSCs) culture should be feeder-free, xeno-free, and completely defined. The present study aims to establish a new feeder-free and xeno-free system for culturing hPSCs. The system consists of the matrix, which was prepared from human umbilical cord blood serum (UCBS) and used to coat the culture plates, and the xeno-free medium, which was conventional serum-free hES medium supplemented with high concentrations of bFGF and Fibronectin. Compared with matrigel and mouse embryonic fibroblasts (MEFs), the UCBS matrix was proved to be equally suitable for the growth of hPSCs. After a series of experiments with different media and cytokins, the UCBS matrix was found worked the best with the basic medium (BM) supplemented with 20 ng/mL bFGF, 10 ug/mL fibronectin and Y-27632 for culture of hES cells. The hPSCs maintained normal karyotype, high proliferation rate and self-renewal ability after continuous culture more than 10 passages in this feeder-free and xeno-free system. Furthermore, a new human embryonic stem (hES) cell line was derived from discarded day 3 embryos in this newly developed culture system. In conclusion, this feeder-free and xeno-free system could not only be used to the culture hPSCs, but could also be used to derive new hES cell lines.

Abstract:The several factors were studied such as the producing methods of embryos, the development stages of embryos and the culture media in order to provide some academic and practical data for the preparation of mouse embryonic stem (ES) cells. Fibroblasts were acquired from 13.5 d embryos of Kunming mouse. The feeder layer was made by fibroblasts of passage treated with mitomycin C. 3.5-day embryos were cultured on the feeder layer for 4-5 days. Inner cell mass (ICM) was separated from the embryonic cells, which was in a good growth status, obvious eminence and without morphological differentiation. The first generation ICM was separated into small pieces of cell mass and then was cultured on the feeder layer in ES culture medium. The passages were carried out according to the appearance of new ES colonies, which grew well and had no differentiation. ICM and different generation of ES cell were identified by morphological observation and alkaline phosphatase staining. The results were showed as following: development ability of embryos from natural mating was better than from superovulation, blastulas had more developmental potential than morulas in the establishment of ES cell lines. DMEM with both low sucrose and high sucrose could be used to culture ES cell, and the medium supplemented with 15% serum was better in culture of ES cell than 10% and 20% group. Bovine Embryonic Stem Cells Culture on Mixed Feeder Layers with Mouse and Bovine Embryonic Fibroblasts in Vitro

Abstract:【Objective】The aim of this study was to establish a suitable culture environment for bovine embryo stem cell.【Method】Bovine and mouse fibroblast cells were treated for 2.0-3.0 h with mitomycin C,respectively.Mixed feeder layer was that cells which were mixed according to 1﹕0,1﹕1,1﹕2,2﹕1,0﹕1 were cultured in the center plate coated by gelatin at 2-2.5×104 per well.Growth states of bovine embryonic stem cells were observed in different feeder layers and undifferentiated phenotypes were detected,including expression of alkaline phosphatase,and presence of OCT-4,SOX2,NANOG and cell marker OCT-4,SSEA-1.Without feeder layer,bovine embryonic stem cell differentiation was observed in vitro.【Result】Comparison of bovine embryonic stem cell growth state on different feeder layers,the clonal morphology on the mixed feeder layer was significantly better than that on the mouse embryonic fibroblast feeder layer or bovine embryonic fibroblast feeder layer.When mouse embryonic fibroblasts and bovine embryonic fibroblasts were mixed at a ratio of 1﹕1 bovine embryonic stem cells accumulated significantly and cloning edge was clear,obvious and full.Expression of alkaline phosphatase,antigen of OCT-4 and SSEA-1 were strongly positive.Specific bands of OCT-4,SOX2 and NANOG mRNA appeared,respectively.Bovine embryonic stem cells on mixed feeder layer was able to form embryoid bodies.【Conclusion】Compared with conventional feeder layers prepared by mouse embryonic fibroblasts or bovine embryonic fibroblasts,the mixed cells feeder layer mixed at a ratio of 1﹕1 may be better suitable for bovine embryonic stem cell culture in vitro and obtain better bovine rembryonic stem cell clonal morphology.
Optimization of Culture Condition of Human Bone Marrow Stromal Cells in Terms of Purification, Proliferation, and Pluripotency.

Abstract:Human bone marrow stromal cells (hBMSCs) possess multilineage differentiation potential and play an important role in modern tissue engineering. However, the development of culture media to maintain hBMSCs in an undifferentiated, self-renewing state during their robust proliferation remains a challenge. We developed and tested modified growth medium [medium 1: epidermal growth factor (EGF), platelet-derived growth factor (PDGF), low glucose, 2% fetal calf serum (FCS)] on hBMSCs by comparing primary cell isolation, multipassage expansion, culture morphology, proliferation, and cellular phenotype, and performing an expression analysis of intrinsic-regulated genes to other two media. Cell morphology, proliferation, and phenotype varied among the media, while cells cultured in medium 1 displayed small, spindle-shaped morphology with the highest rate of growth capacities and the expected phenotype. RT-PCR analysis showed that medium 1 displayed the lowest expression levels of osteogenic genes, chondrogenic genes (osteonectin, runt-related transcription factor 2, cartilage oligo matrix protein, and SOX9), and adipogenic genes (lipoprotein lipase). The expression of another adipogenic gene, peroxisome proliferator-activator receptor-γ2, was higher in medium 1 but did not reach significance. In addition, hBMSCs expanded in medium 1 showed the highest expression ratio of self-renewing-related genes Krüppel-like factor 2 (KLF2) and KLF5. In conclusion, medium 1 allows for better expansion and pluripotency maintenance of hBMSCs and serves as a preferred alternative to traditional serum-containing media for research applications and future clinical use.

Abstract:The research general situation in native and abroad on the mammalian embryonic stem cell (ES) and embryonic germ cells (EG) were reviewed in the article. Especially for the native research development of the ES/EG cells on the feeder making, derive differentiation, germline chimera, fibroblast cell's cropreservation were discussed. The common character and the origin of the ES cells and EG cells were discussed. EG have superiority of more culture materials, longer existing time, no difference of genus than ES.

This study reports a feeder layer-free and serum-free system for culture of rES cells. rES cells could be cultured through at least 22 passages on laminin in medium supplemented with serum replacement (SR), basic fibroblast growth factor (bFGF) and transforming growth factor beta 1 (TGF beta 1), and maintained stable proliferation rates and normal karyotypes, while displaying all the embryonic stem cell characteristics including morphology, alkaline phosphatase (AKP), Oct-4, cell surface markers SSEA-3, SSEA-4, TRA-1-60 and TRA-1-81, and formed cystic embryoid bodies in vitro. In addition, the studies showed that TGF beta 1, bFGF and laminin are necessary for maintaining the undifferentiated growth of rES cells in long-term culture. Moreover, withdrawal of TGF beta 1 increased the differentiation rate by decreasing the expression of integrins. Therefore, this system would provide a well-defined culture system for rES cells, and would facilitate research into self-renewal and differentiation mechanisms of rES cells.
