Abstract:Background: Primary immune thrombocytopenia (ITP) is an acquired autoimmune hemorrhagic disorder, characterized by immune-mediated platelet destruction and impaired megakaryocyte maturation. Although impaired T cells have been implicated to participate in the pathogenesis of ITP, another immune cell signified as M2 macrophages has not been investigated properly in ITP patients. This study aimed to investigate the role of M2 macrophage subsets in primary immune thrombocytopenia (ITP). Methods: Peripheral blood mononuclear cells (PBMC) from newly diagnosed ITP patients and healthy controls (HC) were isolated. M2-like macrophages (CD68+CD163+) and M2 macrophages (CX3CR1+CD163+) were measured by flow cytometry. The correlation between CD68+CD163+ cells and CX3CR1+CD163+ cells was also analyzed. The CX3CR1+cells were sorted by magnetic bead, CD68+CD163+ in PBMC of ITP patients and healthy controls were then isolated, and the proportion of m2-like macrophages before and after the sorting was analyzed. The PPAR gamma and arg-1 levels of mRNAs and proteins of CX3CR1+ M2 macrophages were examined by Real-time PCR and Western Blot, respectively. Results: CX3CR1+CD163+M2 macrophages were positively correlated with CD68+ CD163+ M2-like macrophages in ITP patients (r = 0.54, p < 0.01). After magnetic bead separation, the proportion of CD68+CD163+ cells in CX3CR1+ cells was significantly increased (p = 0.02). Compared with HC, both the mRNA and protein levels of arg-1 of CX3CR1+ M2 macrophages were significantly increased in patients with ITP. The expression level of PPAR gamma protein was significantly increased in ITP than that of HC. However no statistical difference was detected at mRNA expression level, although it was numerically higher in ITP patients than in HC ( p = 0.19). Conclusion: The peripheral CX3CR1+ M2 macrophage exercises similar phenotypes and functions of M2 macrophage. The remarkably increased expression of arg-1 at both transcription and protein levels and PPAR gamma at protein level of CX3CR1+M2 macrophages in ITP patients suggests potential immunomodulatory functions of these macrophage subsets during ITP pathogenesis. However, no significant change at mRNA level of PPAR gamma indicating that the increased PPAR gamma protein level might be caused by other mechanisms, such as after transcription abnormalities, which warrants further investigation. Role of myeloid-derived suppressor cells in mouse pre-sensitized cardiac transplant model.

Abstract:Harness of sensitized transplantation remains a clinical challenge particularly in parallel with prolonged cold ischemia time (PCI)-mediated injury. Our present study was to test the role of myeloid-derived suppressor cells (MDSCs) in mouse pre-sensitized transplantation. Our findings revealed that CD11b+Gr1(low) MDSC was shown to have strong suppressive activity. MDSCs subsets from the tolerated mice exhibited higher suppressive capacities compared with counterparts from naive (untreated) mice. Depletion of Tregs could not affect splenic CD11b+Gr1(-low) MDSC frequency, but increase peripheral and intragraft CD11b+Gr1(-low) frequency. Intriguingly, boost of Tregs remarkably caused an increase of CD11b+Gr1(-low) frequency in the graft, peripheral blood, and spleen. Furthermore, peripheral CD11b+Gr1(-low) cells were massively accumulated at the early stage when allogeneic immune response was enhanced. Taken together, MDSCs could prevent grafts from PCI-mediated injury independent on Tregs in the pre-sensitized transplant recipients. Utilization of MDSC subset particularly CD11b+Gr1(-low) might provide a novel insight into improving graft outcome under such clinical scenarios.

Abstract:e22094 Background: The immune therapy of human cancer is a promising therapeutic approach. Myeloid-derived suppressor cells (MDSCs) have been confirmed as an indispensable component of immune system and associated with tumor induced immune suppression. Thus, MDSCs play an important role in immune therapy. Although a lot of research work has been done, currently there exists limited data about MDSCs status in cancer patients. The aim of this study was to determine whether MDSCs levels in peripheral blood were different in healthy volunteers, benign breast tumor patients and breast cancer patients, and to investigate the relation between MDSCs and several characteristics in breast cancer patients. Methods: Peripheral blood cells (PBC) were collected from 36 normal healthy volunteers, 52 patients with breast cancer and 62 patients with benign breast tumor. Peripheral blood samples of breast cancer patients were obtained before treatment. Circulating MDSCs (identified as Lin-/Lo HLA-DR-CD33+) were measured by flow cytometry. Statistical methods included two sample t tests and one way ANOVA. Results: The percentage of MDSCs was higher in breast cancer than benign breast tumor patients and healthy volunteers (2.017% vs 1.578% vs1.594%; p=0.0002), and the stage III/IV breast cancer patients had more MDSCs accumulation than those in stage I/II (2.92% vs 2.155%; P=0.0806). The levels of MDSCs in peripheral blood showed a significant correlation with HER-2/neu expression in breast cancer patients, but had no correlation with incidence age, ER status, PR status and P53 expression respectively. The percentage of MDSCs was lower in HER-2/neu over expression breast cancer patients than HER-2/neu negative patients (1.715% vs 2.882%, GP=0.0012). Conclusions: MDSCs seemed to have positive correlation with clinical stage and negative correlation with HER-2/neu expression in breast cancer patients . Further research will contribute to the better understanding of the connection between MDSCs and breast cancer, to develop better approaches to breast cancer therapy.

Abstract:Immune thrombocytopenia (ITP) is an autoimmune disorder, in which megakaryocyte dysfunction caused by an autoimmune reaction can lead to thrombocytopenia, although the underlying mechanisms remain unclear. Here, we performed single-cell transcriptome profiling of bone marrow CD34(+) hematopoietic stem and progenitor cells (HSPCs) to determine defects in megakaryopoiesis in ITP. Gene expression, cell-cell interactions, and transcriptional regulatory networks varied in HSPCs of ITP, particularly in immune cell progenitors. Differentially expressed gene (DEG) analysis indicated that there was an impaired megakaryopoiesis of ITP. Flow cytometry confirmed that the number of CD9(+) and HES1(+) cells from Lin(-)CD34(+)CD45RA(-) HSPCs decreased in ITP. Liquid culture assays demonstrated that CD9(+)Lin(-)CD34(+)CD45RA(-) HSPCs tended to differentiate into megakaryocytes; however, this tendency was not observed in ITP patients and more erythrocytes were produced. The percentage of megakaryocytes differentiated from CD9(+)Lin(-)CD34(+)CD45RA(-) HSPCs was 3-fold higher than that of the CD9(-) counterparts from healthy controls (HCs), whereas, in ITP patients, the percentage decreased to only 1/4th of that in the HCs and was comparable to that from the CD9(-) HSPCs. Additionally, when co-cultured with pre-B cells from ITP patients, the differentiation of CD9(+)Lin(-)CD34(+)CD45RA(-) HSPCs toward the megakaryopoietic lineage was impaired. Further analysis revealed that megakaryocytic progenitors (MkP) can be divided into seven subclusters with different gene expression patterns and functions. The ITP-associated DEGs were MkP subtype-specific, with most DEGs concentrated in the subcluster possessing dual functions of immunomodulation and platelet generation. This study comprehensively dissects defective hematopoiesis and provides novel insights regarding the pathogenesis of ITP.

Abstract:Myelodysplastic syndrome (MDS) is a heterogeneous group of clonal hematopoietic neoplasms. The progression of malignancy is closely associated with immune regulation. Macrophages are indispensable tissue components and have been proposed to play a role in the pathophysiology of hematopoietic malignancies. However, the specific role of macrophages in the development of MDS remains unclear. Here, we investigated the characteristics and phenotypic evolution of macrophages from patients with MDS. Macrophages from patients with MDS expressed CD68, CD86 and CD163. Furthermore, MDS macrophages exhibited more M2-related characteristics. Moreover, a number of phenotype-associated genes in MDS macrophages exhibited diverse responses to iron overload or iron chelation upon stimulation by ferric chloride or deferoxamine (DFO, an iron chelator). Ferric chloride polarized MDS macrophages to exhibit more M1-related characteristics, a phenomenon that could be partially reversed by DFO. Therefore, this study reveals the characteristics and phenotypic evolution of MDS macrophages and broadens the knowledge of macrophage plasticity in hematopoietic malignancies.

Abstract:Primary immune thrombocytopenia is a kind of autoimmune disease,characterized by decreased platelets and an increased risk of bleeding.Besides destruction of platelet by platelet specific antibodies produced by auto-reactive B cells and the imbalance of Th1/Th2 cells,the study of regulatory T lymphocytes is getting more and more attention.Regulatory T lymphocytes are a subset of CD4+T lymphocytes that can suppress immune response.However,recent studies have discovered that a sublineage of CD8+regulatory T lymphocytes is also associated with the pathogenesis and treatment of ITP.This review focuses on the role of abnormalities in the number and function of regulatory T lymphocytes in the pathogenesis of ITP.

Abstract:Abstract Abstract 597 The acquisition of genetic abnormalities that lead to ineffective hematopoiesis is a characteristic of MDS. This event is mediated in part by an interaction of inflammatory intermediates with the bone marrow (BM) microenvironment; however, the mechanism by which chromosomal damage occurs to the stem/progenitor compartment (HSC/HPC) is unknown. We have identified a unique population of myeloid derived suppressor cells (MDSC) bearing a LIN-HLA-DR-CD33+ surface phenotype. These MDSC elicit hematopoietic suppression through the elaboration of nitric oxide, arginase, and inflammatory cytokines. This study found that MDSCs accumulated in excess in the BM of patients with lower risk MDS (mean, 32.32% ± 13.3; n=12) compared to BM from healthy age matched controls (mean 2.1%±0.9; n=8) and non-MDS cancer patients (n=8 mean 2.06%±1.5 p<0.0001). Fluorescence in situ hybridization (FISH) was performed to determine whether MDSCs represent a distinct cell population from the abnormal MDS clone. MDSCs from BM of 5 patients having chromosomal abnormality were separated by FACS sorting based on LIN-HLA-DR-CD33+ phenotype and the presence of chromosomal abnormalities was determined in this population and compared to non-MDSCs. Monosomy 7 and deletion of 5q chromosomal abnormalities resided within the non-MDSC hematopoietic compartment. This indicates that MDSCs in low risk MDS patients may represent a unique cell population from the HPCs with clonal potential. Furthermore, the key cytokines involved in MDSC suppressive function, TGFβ, VEGF and IL-10, were higher in MDSCs isolated from MDS patients compared to controls. Using four-color immunostaining, we discovered that MDSCs are capable of direct cytotoxicity against autologous erythroid precursors (CD71+ and CD235a+), as evidenced by increased polarized granule mobilization toward the site of cellular contact. Moreover, it was observed that the accumulation of MDSC in the BM from MDS patients has an impact on hematopoietic differentiation. In the presence of MDSCs, the formation of BFU-E (burst forming unit erythroid) was significantly suppressed in all patients tested (n=6). In contrast depletion of the MDSC by FACS sorting reconstituted the formation of BFU-E (21± 4.2 in MDSC depleted bone marrow verse 0.8± 0.6 in unsorted BM cells, respectively). These data strongly suggest that the presence of MDSCs in the BM microenvironment of MDS patients contribute to suppression of HPC development. Based on these findings, a novel form of adaptive immunotherapy based on the induction of MDSC maturation can be envisioned. DAP12 (DNAX-activating protein of 12kDa) is an adaptor protein that mediates signaling of dentritic cell and monocyte maturation. In order to determine if DAP12 signaling induces MDSC maturation and reversal of suppressive function, a genetically modified, constitutively activated form was introduced into BM mononuclear cells (BM-MNC) using both recombinant adenoviral and lentiviral gene transduction. Infection of BM-MNC from MDS patients with constitutively active DAP12 increased expression of maturation surface markers CD14, CD15 and HLA-DR and increased BFU-E colony formation (31± 2.1 verse control 7±0.7) after 14 days. These results suggest that active signaling through DAP12 has potential therapeutic implications in MDS by driving maturation of immature myeloid cells reversing the inflammatory changes that contribute to malignant transformation. In conclusion, our previously unknown findings have begun to elucidate whether the presence of MDSCs in the BM microenvironment of MDS patients contributes to the pathogenesis of MDS by providing a suppressive microenvironment and repress BM maturation associated with impaired HSC/HPC development. Additionally, DAP12 may be a potential therapeutic pathway in which to stimulate MDSC maturation, reversing the suppressive effects on HSC/HPC development. Clinical and Molecular Characteristics of Megakaryocytes in Myelodysplastic Syndrome

Abstract:Objective Myelodysplastic syndrome (MDS) is a malignant clonal disorder of hematopoietic stem cells which is characterized by morphologic dysplasia. However, the pathological characteristics of megakaryocytes (MKs) in MDS patients with gene mutation are not well established. Methods Bone marrow MK specimens from 104 patients with primary MDS were evaluated, and all patients were distributed into two groups according to gene mutation associated with functional MKs. The morphologic and cellular characteristics of MKs and platelets were recorded and compared. Results The more frequently mutated genes in MDS patients were TUBB1 (11.54%), VWF (8.65%), NBEAL2 (5.77%), and the most common point mutation was TUBB1 p.(R307H) and p.(Q43P). Patients with MK mutation showed a decrease in adenosine diphosphate-induced platelet aggregation, high proportion of CD34 + CD61 + MKs (10.00 vs. 4.00%, p = 0.012), and short overall survival (33.15 vs. 40.50 months, p = 0.013). Further, patients with a higher percent of CD34 + CD61 + MKs (≧20.00%) had lower platelet counts (36.00 × 10 9 /L vs. 88.50 × 10 9 /L, p = 0.015) and more profound emperipolesis ( p = 0.001). By analyzing RNA-sequencing of MKs, differentially expressed mRNA was involved in physiological processes including platelet function and platelet activation, especially for MDS patients with high percent of CD34 + CD61 + MKs. The high levels of expression of CD62P, CXCL10, and S100A9 mRNA, shown by RNA sequencing, were validated by PCR assay. Conclusion High proportion of CD34 + CD61 + MKs was a poor prognostic factor in MDS patients with MK mutation. CD62P, CXCL10, and S100A9 may be the potential targets to evaluate the molecular link between gene defects and platelet function.

Myeloid-derived suppressor cells (MDSC) are a heterogeneous population of cells of myeloid origin and include immature macrophages, dendritic cells (DC) and other myeloid cells. In mice are phenotypically characterized as CD11b+Gr-1+ cells, while in human they have an immature phenotype, including lineage negative (Lin-), CD14-, HLA-DR-, CD15+, CD34+, CD11b+, CD33+, and CD13+ cells. MDSC reduce activated T-cell number and inhibit their function through different mechanisms including: L-arginine metabolism, nitric oxide (NO), up-regulation of reactive oxygen species (ROS), and secretion of immunosuppressive cytokines. MDSC also promote tumor-dependent angiogenesis as well as tumor metastasis. Their accumulation has been described in patients affected by some solid tumors but information on haematological neoplasms are lacking. Our study investigated by flow cytometry the presence of MSDC in the peripheral blood of patients affected by Hodgkin Lymphoma (HL). Methods We studied 14 patients with HL at diagnosis and 10 age-matched healthy controls (HC). Peripheral blood mononuclear cells were stained with the following monoclonal antibodies:CD11b, CD13, CD14, CD34, CD45, for 20 minutes at room temperature. After lysing red cells, cells were analyzed by flow cytometry. Results we observed a increased number of MDSC (CD11b+,CD13+,CD34+,CD14-, CD45+) in the peripheral blood of patients with HL compared to HC (13,37 ± 17,77 ×109/l vs 1,45± 0,98 ×109/l, p=0,0007). We also found that patients with advanced-stage Hodgkin disease (III and IV) have higher number of MDSC, compared to patients stage I and II (p= 0,04). Conclusion These data suggest a role for myeloid-derived suppressor cells in promoting tumor cell proliferation in hodgkin lymphoma. Disclosures: No relevant conflicts of interest to declare.

Abstract:Background. Monocytic myeloid-derived suppressor cells (M-MDSCs) characterized with the phenotype of CD14+HLA-DRlow/- have attracted a lot of attention in the field of human tumor immunology. However, little is known about the roles of M-MDSCs in acute myeloid leukemia (AML) as opposed to their multiple roles in solid tumors. Methods. We examined the frequencies of M-MDSCs identified for CD14+HLA-DRlow/- by flow cytometry in the peripheral circulating blood of 109 newly diagnosed adult patients with AML and 30 healthy controls (HC). Then, we, respectively, validated the clinic significance of circulating M-MDSCs on the relevance of spectral features for diagnostic stratification, induction therapy response, treatment effect maintenance, and long-term survival in AML. Results. Circulating M-MDSC frequencies of AML were significantly higher than those of HC both in CD14+ monocytes ( 46.22 % ± 2.95 % vs. 1.07 % ± 0.17 % , p < 0.01 ) and peripheral blood mononuclear cells (PBMCs) ( 4.21 % ± 0.80 % vs. 0.17 % ± 0.03 % , p < 0.01 ). Elevated circulating M-MDSCs in patients with AML were significantly associated with low complete remission (CR) rate, high relapse/refractory rate, and poor long-term survival, but had no correlation with common clinic risks and cytogenetic molecular risk categories. Conclusions. It was demonstrated that circulating M-MDSCs are elevated and associated with poor prognosis in AML, suggesting M-MDSCs might be a prognostic indicator for AML.

[Number and Function of Myeloid-Derived Suppressor Cells in Patients with Adult Primary Immune Thrombocytopenia]

Abstract:Objective: To analyze the number of myeloid-derived suppressor cells(MDSC) and the level of prostaglandin E2(PGE2) in the bone marrow of adult ITP patients, and to explore their possible mechanisms involved in the pathogenesis of this disease. Methods: Twenty-five patients of newly diagnosed ITP, 25 patients of complete remission group and 15 patients of control group were selected. The number of MDSC in the bone marrow between 3 groups was detect by flow cytometry (FCM). The serum level of prostaglandin E2 (PGE2) in 3 groups was determined by enzyme linked immunosorbent assay (ELISA). The relative expression of IFN-γ mRNA in bone marrow mononuclear cells was measured by real time fluorescence quantitative polymerase chain reaction (RT-qPCR) in each groups. Results: The number of MDSC in the complete remission group was significantly higher than that in the control group (P<0.05); the number of MDSC in the newly diagnosed group was higher than that in the control group; the number of MDSC in the complete remission group was higher than that in the newly diagnosed group. The serum level of PGE2 in bone marrow of ITP patients in the newly diagnosed group was higher than that of the control group（P<0.05）. The serum level of PGE2 in the bone marrow of ITP patients of the complete remission group was higher than that of the control group (P<0.05）. The level of PGE2 in bone marrow serum of ITP patients of the newly diagnosed group was lower than that in the complete remission group(P<0.05）. The relative expression level of IFN-gamma in bone marrow mononuclear cells of the ITP patients in newly diagnosed group was higher than that in the control group and the complete remission group（P<0.001）. The relative quantification (RQ) of IFN-γ in bone marrow mononuclear cells was 2.60 between the newly diagnosed group and the complete remission group. Conclusion: When adult ITP disease is remitted, the number of MDSC rises and correlates with the therapeutic response and PGE2 level in the bone marrow.

Abstract:Abstract Primary immune thrombocytopenia (ITP) is an autoantibody-mediated hemorrhagic disorder in which B cells play an essential role. Previous studies have focused on peripheral blood (PB), but B cells in bone marrow (BM) have not been well characterized. We aimed to explore the profile of B-cell subsets and their cytokine environments in the BM of patients with ITP to further clarify the pathogenesis of the disease. B-cell subpopulations and their cytokine/chemokine receptors were detected by using flow cytometry. Plasma concentrations of cytokines/chemokines were measured by using enzyme-linked immunosorbent assay. Messenger RNA levels of B cell–related transcription factors were determined by using quantitative polymerase chain reaction. Regulatory B cell (Breg) function was assessed by quantifying their inhibitory effects on monocytes and T cells in vitro. Decreased proportions of total B cells, naive B cells, and defective Bregs were observed in patients with ITP compared with healthy controls (HCs), whereas an elevated frequency of long-lived plasma cells was found in BM of autoantibody-positive patients. No statistical difference was observed in plasmablasts or in short-lived plasma cells between patients with ITP and HCs. The immunosuppressive capacity of BM Bregs from patients with ITP was considerably weaker than HCs. An in vivo study using an active ITP murine model revealed that Breg transfusion could significantly alleviate thrombocytopenia. Moreover, overactivation of CXCL13-CXCR5 and BAFF/APRIL systems were found in ITP patient BM. Taken together, B-cell subsets in BM were skewed toward a proinflammatory profile in patients with ITP, suggesting the involvement of dysregulated BM B cells in the development of the disease.

Abstract:Background: The tumor microenvironment is essential for tumor survival, growth and progression. There are only a few studies on the tumor microenvironment in cutaneous CD30-positive lymphoproliferative disorders. Methods: We assessed the composition of the tumor microenvironment using immunohistochemistry studies in skin biopsies from cases diagnosed with lymphomatoid papulosis (LyP: 18 specimens), primary cutaneous anaplastic large-cell lymphoma (PC-ALCL: 8 specimens), and reactive diseases harboring CD30-positive cells (18 specimens). Results: The predominant cells present in LyP and PC-ALCL were CD163+ M2 macrophages (44.7%, 35%), followed by CD8+ tumor infiltrating lymphocytes (11%, 15%), FOXP3+ T-regulatory cells (9%, 4.5%) and programmed cell death 1(PD-1) + lymphocytes (2.2%, 6.8%). In contrast, CD30-positive reactive inflammatory and infectious disorders were characterized by higher numbers of CD123+ plasmacytoid dendritic cells (6.3%) when compared to LyP (1%), and PC-ALCL (1.1%). Conclusions: Key differences exist between the microenvironment of CD30-positive lymphoproliferative disorders and reactive conditions harboring CD30-positive lymphocytes. The high number of tumor associated macrophages, and the close vicinity of these immune cells to the CD30-positive tumor cells might suggest that tumor associated macrophages have direct influence on tumorigenesis in LyP and ALCL. Therefore, modulation of M2 macrophages may represent a new therapeutic strategy in cutaneous CD30-positive lymphoproliferative disorders.

Abstract:Primary immune thrombocytopenia (ITP) is an acquired autoimmune bleeding disorder. Monocytes and macrophages are the major cells involved in autoantibody-mediated platelet clearance in ITP. In the present study, we found increased percentages of peripheral blood proinflammatory CD16(+) monocytes and elevated frequencies of splenic tumor necrosis factor-alpha (TNF-alpha)-expressing macrophages in ITP patients compared with healthy controls. Concurrently, we observed elevated TNF-alpha secretion in plasma as well as higher TNF-alpha mRNA expression in total peripheral blood mononuclear cells and CD14(+) monocytes of ITP patients. Of note, in vitro TNF-alpha blockade with neutralizing antibody remarkably reduced polarization to M1 macrophages by inhibiting the nuclear factor kappa B (NF-kappa B) signaling pathway. Moreover, TNF-alpha blockade dampened macrophage phagocytosis and T cell stimulatory capacity. Finally, in passive and active murine models of ITP, anti-TNF-alpha therapy reduced the number of nonclassical monocytes and M1 macrophages, ameliorated the retention of platelets in spleen and liver, and increased the platelet count of ITP mice. Taken together, TNF-alpha blockade decreased the number and function of proinflammatory subsets of monocytes and macrophages by inhibiting the NF-kappa B signaling pathway, leading to remarkable attenuation of antibody-mediated platelet destruction. Thus, TNF-alpha blockade may be a promising therapeutic strategy for the management of ITP.

Abnormally activated CD8+ T cells are important in its pathogenesis and majority of the patients having this abnormity are diagnosed as aplastic anemia (AA). Abstract:Besides their role as potent antigen-presenting cells, myeloid dendritic cells (MDCs), but not plasmacytoid dendritic cells (PDCs), have been reported to have cytotoxic or cytostatic activity on some tumor cells. In this article, we analyzed the tumoristatic potential of a distinct peripheral blood monocyte-derived MDC subset which co-expressed PDC-specific marker CD123. CD123(+) MDCs represented a subset of small-sized DCs and accounted for 45-60% of peripheral blood monocytes cultured with granulocyte-macrophage colony-stimulating factor and interleukine-4 (IL-4) for 7 d. They exhibited more significant antiproliferative activity toward hematological tumor cell lines of Jurkat, HL60, and myelodysplastic syndromes over-leukemia than CD123(-) MDCs even at a low effecter/target ratio. Pretreatment of MDC and their supernatant with TRAIL-R2:Fc significantly reduced the tumoristatic effect of CD123(+) MDCs but not of CD123(-) MDCs and their supernatant. CD123(+) MDCs expressed higher level of cytoplasmic TNF-alpha-related apoptosis-inducing ligand (TRAIL) than CD123(-) MDCs, whereas both expressed very little surface and soluble TRAIL. These results reveal that CD123(+) cells represented a predominant subset of MDCs generated from peripheral blood monocytes in vitro, characterized by their potential tumoristic activity partially via cytoplasmic TRAIL.
