Abstract:Transition metal ions are served as disinfectant thousand years ago. However, the in vivo antibacterial application of metal ions is strongly restricted due to its high affinity with proteins and lack of appropriate bacterial targeting method. Herein, for the first time, Zn2+-gallic acid nanoflowers (ZGNFs) are synthesized by a facile one-pot method without additional stabilizing agents. ZGNFs are stable in aqueous solution while can be easily decomposed in acidic environments. Besides, ZGNFs can specifically adhere onto Gram-positive bacteria, which is mediated by the interaction of quinone from ZGNFs and amino groups from teichoic acid of Gram-positive bacteria. ZGNFs exhibit high bactericidal effect toward various Gram-positive bacteria in multiple environments, which can be ascribed to the in situ Zn2+ release on bacterial surface. Transcriptome studies reveal that ZGNFs can disorder basic metabolic processes of Methicillin-resistant Staphylococcus aureus (MRSA). Moreover, in a MRSA-induced keratitis model, ZGNFs exhibit long-term retention in the infected corneal site and prominent MRSA elimination efficacy due to the self-targeting ability. This research not only reports an innovative method to prepare metal-polyphenol nanoparticles, but also provides a novel nanoplatform for targeted delivery of Zn2+ in combating Gram-positive bacterial infections.
Green Synthesis of Silver Nanoparticles with High Antimicrobial Activity and Low Cytotoxicity Using Catechol-Conjugated Chitosan

Abstract:In this study, catechol-conjugated chitosan (CSS) was synthesized to prepare silver nanoparticles in aqueous solution as both a reducing and stabilizing agent. The entire reaction process complied with the principles of green chemistry. The stability and physicochemical properties of CSS-coated silver nanoparticles (CSS–Ag NPs) were well characterized by a series of techniques. A narrow diameter distribution and excellent stability were observed in the target CSS–Ag NPs. Determination of the minimum inhibitory concentration (MIC) and the disk diffusion test were applied to evaluate the antibacterial activities with respect to quantity and quality, respectively. The cytotoxic effects on HepG2 cells were also evaluated with a series of assays. The resulting CSS–Ag NPs combined the unique properties of chitosan and silver nanoparticles, showing effective antibacterial activities and low cytotoxicity. The effective toxic concentration of CSS–Ag NPs towards E. coli and S. aureus is too low to damage HepG2 cells. The results demonstrated that CSS–Ag NPs could be a potential candidate for use in biological and pharmaceutical areas to prevent infections caused by microorganisms.
Catechol-Functional Chitosan/Silver Nanoparticle Composite As A Highly Effective Antibacterial Agent with Species-Specific Mechanisms

Abstract:In this study, silver nanoparticles (Ag NPs) coated with catechol-conjugated chitosan (CSS) were prepared using green methods. Interestingly, we uncovered that CSS-coated Ag NPs (CSS-Ag NPs) exhibited a higher toxicity against gram-negative Escherichia coli (E. coli) bacteria than against grampositive Staphylococcus aureus (S. aureus) bacteria. The differences revealed that the CSS-Ag NPs killed gram bacteria with distinct, species-specific mechanisms. The aim of this study is to further investigate these underlying mechanisms through a series of analyses. The ultrastructure and morphology of the bacteria before and after treatment with CSS-Ag NPs were observed. The results demonstrated the CSS-Ag NPs killed gram-positive bacteria through a disorganization of the cell wall and leakage of cytoplasmic content. In contrast, the primary mechanism of action on gram-negative bacteria was a change in membrane permeability, induced by adsorption of CSS-Ag NPs. The species-specific mechanisms are caused by structural differences in the cell walls of gram bacteria. Gram-positive bacteria are protected from CSS-Ag NPs by a thicker cell wall, while gram-negatives are more easily killed due to an interaction between a special outer membrane and the nanoparticles. Our study offers an in-depth understanding of the antibacterial behaviors of CSS-Ag NPs and provides insights into ultimately optimizing the design of Ag NPs for treatment of bacterial infections.
Carboxymethyl Chitosan-Fe3O4 Nanoparticles: Preparation and Adsorption Behavior Toward Zn2+ Ions

Abstract:A novel magnetic nanoadsorbent was prepared by the covalent binding of carboxymethyl chitosan (CMC) onto the surface of Fe3O4 magnetic nanoparticles, which was developed using a coprecipitating method. This nanoadsorbent was characterized by transmission electron microscopy (TEM) and X-ray diffraction patterns (XRD), etc. Moreover, the adsorption performance of the nanoadsorbent toward Zn2+ ions was investigated. The results showed that the mean diameter of the magnetic nanoadsorbent was 18 nm and the amount of CMC was about 5%. The nanoadsorbent showed high efficiency for the removal of Zn2+ ions. The adsorption rate was so rapid that the equilibrium was achieved within 2 min. The isotherm adsorption data obeyed the Langmuir model, with a maximum adsorption capacity of 20.4 mg·g−1 and an adsorption equilibrium constant of 0.0314 L·mg−1. The thermodynamic calculations indicated that the adsorption process was exothermic and that the enthalpy change was −5.68 kJ·mol−1.

Abstract:Glycopolymer has been developed in bactericidal applications, but the traditional synthesis, such as click chemistry, needs poly-step reactions and complicated processes. Herein, incorporating the glyco-units into the polymers by quaternization has been exploited to give a facile strategy by Copper-mediated reversible deactivation radical polymerization (Cu(0)-RDRP) to prepare quaternized poly(ionic liquid)s (PILs) with pendant saccharide. The obtained PILs have the suitable property of killing typical Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus. Simultaneously, they perform the lower hemolytic rate of blood cells, higher selectivity index against bacteria, and higher affinity with Concanavalin A (ConA) than those quaternized by bromohydrin at the same quaternization degree. This result demonstrated that the introduced saccharide units, such as glucose and mannose, could maintain the electrostatic interaction to kill bacteria, perform lectin recognition behavior and interaction with ConA, and relatively harmlessness against red blood cells. Moreover, the sugar-containing PILs became insoluble and still antibacterial after anion exchange reaction. These new findings illuminated that such quaternized antibacterial agents with saccharide units that are easy-to-prepare, low cytotoxic could pave the way for further water disinfectant and pathogen control application.

Abstract:Porous magnetic chitosan microspheres with average diameter about 600 pm were prepared by high voltage electrostatic method through mixing Fe3O4 particles into CS with silicagel as pore-forming agent and then adding them into thermal NaOH solution to get porous structure. This research studied the preparation technology, as well as the structure and morphology of microsphere with SEM, the results of which indicate that holes of po-rous magnetic chitosan microspheres with the size approximately 50 pm were uniformly dispersed in the microsphere when the mass volume fraction of chitosan acetic acid aqueous solution was 5% and m(s):m(cs) was 4: 1. Due to its large amount of hydroxyl groups and amino groups, porous magnetic chitosan microspheres could be widely used to deal with acidic dyestuff to improve environment and separated from dirty water by adding magnetic field.
Delivery of chiral D-cysteine by pH-responsive mesoporous silica nanoparticles for effective bacterial inhibition

Abstract:Efficient and highly controllable antibacterial effects, as well as good biocompatibility, are required for antibacterial materials to overcome multi-drug resistance in bacteria. Herein, mesoporous silica nanomaterials (MSNs) carriers with a particle mean size of 60 nm and pore size of 7.9 nm were prepared, which was followed by loading with D-cysteine (D-Cys) and modified with Polyethyleneimine (PEI) molecules on the outer surface (named as D@MSNs-P). The prepared D@MSNs-P showed a good pH response in the range of 5-7, and the rate of antibacterial agent D-Cys released from nanocarriers was much faster at lower pH (pH 5) than that at higher pH (pH 6-7), which favors the rapid control of the pathogenic bacteria. In a working pH (pH 5), D@MSNs-P exhibited broad-spectrum antibacterial activities against Escherichia coli, Staphylococcus aureus, Salmonella enteritidis, and Listeria monocytogenes with the highest antibacterial efficiency of 99.9%, 99.8%, 98.1%, and 96.2%, respectively, which is much higher than that of pure D-Cys, pure MSNs, D@MSNs, and PEI group. The outstanding antibacterial activity of D@MSNs-P was attributed to the synergistic effect of the unique structure of MSNs and chiral D-Cys molecules. In addition, the prepared D@MSNs-P has no cytotoxicity to HepG2 cells (Human hepatoma cells) at the concentration of 0.4-12.8 mg/mL and even can promote cell proliferation at high concentrations. Our results open a new door for designing the most promising nanomaterials for pH response release and controllable antimicrobial.
Synergy of bioinspired chimeric protein and silver nanoparticles for fabricating “kill-release” antibacterial coating

Abstract:In view of the problem that medical materials are prone to contamination by bacteria that causing health threats, an antibacterial surface was designed by combining mussel-inspired chimeric protein with silver nanoparticles (AgNPs). The mussel-inspired chimeric proteins have ability to adhere to various substrates through a convenient "one-step" method on account of 3,4-dihydroxyphenyl-alanine (DOPA), and serve as fixed templates to generate AgNPs. The prepared AgNPs protein coatings (Ag-MAP-KE) maintained prominent antifouling property with 97.9% E. coli shielding. In addition, the Ag-MAP-KE also exhibited excellent inhibitory performance against gram-negative bacteria E.coli and gram-positive bacteria S.aureus with 19.97 mm and 17.60 mm inhibition zone, and can be stimulated with water to release 99% covered bacteria. Furthermore, the designed funtionals coatings displayed excellent blood and cell compatibility. The cooperation of protein and AgNPs presents a noval"kill-release" strategy for developing antibacterial surfaces.

Efficient Bacterial Capture with Amino Acid Modified Magnetic Nanoparticles

Abstract:Traditional chemical disinfectants are becoming increasingly defective due to the generation of carcinogenic disinfection byproducts and the emergence of antibiotic-resistant bacterial strains. Functionalized magnetic nanoparticles yet have shown great application potentials in water treatment processes especially for bacterial removal. In this study, three types of amino acids (arginine, lysine, and poly-L-lysine) functionalized Fe3O4 nanoparticles (Fe3O4@Arg, Fe3O4@Lys, and Fe3O4@PLL) were prepared through a facile and inexpensive two-step process. The amino acid modified Fe3O4 nanoparticles (Fe3O4@AA) showed rapid and efficient capture and removal properties for both Gram-positive Bacillus subtilis (B. subtilis) and Gram-negative Escherichia coli 15597 (E. coli). For both strains, more than 97% of bacteria (initial concentration of 1.5 x 10(7) CFU mL(-1)) could be captured by all three types of magnetic nanoparticles within 20 min. With E. coli as a model strain, Fe3O4@AA could remove more than 94% of cells from solutions over a broad pH range (from 4 to 10). Solution ionic strength did not affect cell capture efficiency. The co-presence of sulfate and nitrate in solutions did not affect the capture efficiency, whereas, the presence of phosphate and silicate slightly decreased the removal rate. However, around 90% and 80% of cells could be captured by Fe3O4@AA even at 10 mM of silicate and phosphate, respectively. Bacterial capture efficiencies were over 90% and 82% even in the present of 10 mg L-1 of humic acid and alginate, respectively. Moreover, Fe3O4@AA nanoparticles exhibited good reusability, and greater than 90% of E. coli cells could be captured even in the fifth regeneration cycle. Abstract:In this current work, the magnetic composite microspheres (MCM), consisting of Fe3O4 nanoparticles and poly(acrylic acid) (PAA) blended chitosan (CS), were prepared successfully by a simple method, co-precipitation of the compounds in alkaline solution. SEM, FTIR and TG techniques have been applied to investigate the structures of the MCM materials. The vibrating-sample magnetometer (VSM) measurement illustrated a paramagnetic property as well as a fast magnetic response, which indicated the significant separability of the MCM in the aqueous suspensions. Then, the MCM materials were employed as absorbents for removal of copper(II) (Cu(II)) ions from aqueous solutions. The fundamental adsorption behaviors of MCM were studied also. Experimental results revealed that the CS/PAA-MCM had greater adsorption capacity than CS-MCM, and PAA played an important role for the adsorption of Cu(II) ions. Moreover, the adsorption isotherms were all well described by the Langmuir model, while the adsorption kinetics followed the pseudo-second order equation. Furthermore, the adsorbent could be easily regenerated at lower pH and reused almost without any loss of adsorption capacity. On the contrary, the Cu(II) ions loaded CS-MCM and CS/PAA-MCM were stable enough at pH higher than 4.0, and both exhibited efficient phosphate removal with maximal uptakes around 63.0 and 108.0mgPg−1, respectively.

Abstract:The superior photocatalysis of titanium dioxide (TiO2) had received much attention, but the effect of its separation and agglomeration on the water disinfection performance should not be ignored. Herein, lactose-containing glycopolymers grafted onto magnetic TiO2 nanomaterials were prepared through copper (0) mediated controlled radical polymerization and "grafting to" strategy to improve their separation and dispersibility properties. The physicochemical properties of the Fe3O4@TiO2/glycopolymers were systematically characterized by Fourier transform infrared spectroscopy, thermogravimetric analysis, dynamic light scattering, transition electron microscopy, and vibrating sample magnetometer. The result of the survival bacterial ratio under ultraviolet exhibited the well photocatalytic antibacterial activity of Fe3O4@TiO2/glycopolymers towards E. coli, which retained the photocatalytic sterilization performance of TiO2. Among them, Fe3O4@TiO2@poly(LacA) showed an excellent ability to identify and trap E. coli because of the carbohydrate-protein recognition effect. The endocytosis of E. coli on Fe3O4@TiO2@poly(LacA) was further confirmed by the image of transition electron microscopy. Considering the magnetic characteristics of Fe3O4@TiO2/glycopolymers, it is speculated that these antibacterial agents can be easily recovered by the external magnetic field, and thus be a promising candidate for photocatalysis water disinfection.

Abstract:Bacterial infections have evolved as a life-threatening problem afflicting people due to the abuse of antibiotics and emergence of drug-resistant bacteria. Thus developing novel antibacterial materials is an urgent need. Herein, chitosan-MoS2 (CS-MoS2) nanocomposite was synthesized through thiol ligand functionalization and chemical modification to achieve more efficient bactericidal activity. CS-MoS2 exhibited synergistic remarkable bactericidal capability against Gram-positive Staphylococcus aureus (S. aureus) and Gram-negative Escherichia coli (E.coli), where bacterial viabilities were significantly reduced. After treatment with 10 μg/mL of CS-MoS2, 100% and 98.1% of S. aureus and E.coli cells were killed respectively. A mechanism study revealed that the positively charged CS-MoS2 could interact with cell membrane, then cause damage to the membrane and cellular constituents by generation of reactive oxygen species (ROS) and glutathione (GSH) oxidation, finally inhibit bacterial growth. The CS-MoS2 could be an attractive antibacterial agent with improved efficiency and provide more strategies for application of MoS2.
Synthesis of Chitosan/poly (ethylene Glycol)-Modified Magnetic Nanoparticles for Antibiotic Delivery and Their Enhanced Anti-Biofilm Activity in the Presence of Magnetic Field

Abstract:Biocompatible Fe3O4/chitosan (CS)/poly (ethylene glycol) (PEG)/gentamicin (Gent) magnetic nanoparticles, namely Fe3O4@PEG-Gent NPs, have been successfully developed for antibiotic delivery. In which, PEG dicarboxylic acid was used to modify Fe3O4 NPs for good dispersity as well as offer sufficient carboxyl groups as binding sites. And then the free Gent was facilely loaded onto Fe3O4 NPs so as to achieve powerful antibacterial activity via electrostatic interactions. Under acidic condition, the CS and PEG of Fe3O4@PEG-Gent were protonated to introduce the positive charge to NPs surface, thus facilitating the contact with negatively charged bacterial cell membrane. What is more, the stretches of CS chains triggered by acidic pH may prevent the antimicrobial efficiency of Gent from weakening. Compared with the free antibiotic, these nanocomposites presented better antimicrobial efficacy against gram-positive bacteria S. aureus under acidic condition. Intriguingly, the confocal laser scanning macroscopy imaging suggested that the anti-biofilm efficacy of nanocomposites was significantly enhanced in the presence of an external magnetic field. Due to the superparamagnetic performance of Fe3O4 NPs, these nanocomposites were allowed deeper penetration into a mature biofilm of S. aureus by magnetic field, leading to an effective Gent delivery for eradication of biofilm. The ingenious fabrication of the antibiotic delivery system not only efficiently improved the effectiveness and bioavailability of Gent at acidic media, but also provided an innovative platform to treat bacterial biofilms-associated infection by applying extra environmental factors such as magnetic field.

Abstract:Functionalized magnetic nanoparticles have shown great application potentials in water treatment processes especially for bacterial removal. Antibacterial agent, cetyltrimethylammonium bromide (CTAB), was employed to modify Fe3O4 nanoparticles to fabricate bactericidal paramagnetic nanoparticles (Fe3O4@CTAB). The as-prepared Fe3O4@CTAB could effectively capture both Gram-negative Escherichia coli and Gram-positive Bacillus subtilis from water. For both cell types, more than 99% of bacteria with initial concentration of 1.5 × 10(7)CFU/mL could be inactivated by Fe3O4@CTAB (0.5 g/L) within 60 min. Fe3O4@CTAB could remove more than 99% of cells over a wide pH (from 3 to 10) and solution ionic strength range (from 0 to 1000 mM). The copresence of sulfate and nitrate did not affect the bacterial capture efficiencies, whereas, phosphate and silicate slightly decreased the bacterial removal rates. However, more than 91% and 81% of cells could be captured at 10mM of phosphate and silicate, respectively. Over 80% of cells could be removed even in the presence of 10mg/L of humic acid. Moreover, Fe3O4@CTAB exhibited good reusability, and greater than 83% of cells could be captured even in the fifth regeneration cycle. Fe3O4@CTAB prepared in this study have great application potentials for water disinfection.

Abstract:Three magnetic iron oxide/phosphate nanocomposites, Fe3O4/Ba-3(PO4)(2), Fe3O4/Sr-5(PO4)(3)(OH), and Fe3O4/Sr5xBa3x(PO4)(3)(OH) (denominated FB, FS, and FSB, respectively) were prepared by one-pot hydrothermal method. They exhibited a removal efficiency of 96%, 86%, and 91% for Escherichia coli (E. coli) of 5 x 10(8) CFU/mL within 30 min, respectively. After bacterial removal, the recovered nano-composites by a magnet were reused to treat a mixed solution of Pb2+ and malachite green (MG). The loaded E. coli on the surface of the nanocomposites is beneficial to the subsequent removal of positively charged Pb2+ and MG. On the other hand, the MG in the mixture can contribute substantially to the disinfection of the recovered nanocomposites. These nano-composites are sustainable materials for the successive removal of E. coli and mixed cationic pollutants from water. Removal process by these nano-composites possesses a synergistic effect, leading to more effective water treatment.

Abstract:Magnetic particles with active groups on their surfaces can be applied to wastewater purification to remove harmful ions, but the insufficient number of active groups on their surfaces tends to limit their removal rate of ions. Herein, we grafted different generations of dendritic macromolecular polyamidoamine (PAMAM) onto the magnetic chitosan microspheres (MCS) using ethylenediamine tetraacetic acid (EDTA) as core (MCEPs) for the removal of Cu (II). Firstly, 7 μm-sized MCS with good dispersibility was prepared by adjusting conditions such as glacial acetic acid content, emulsifier content and raw material ratio. Secondly, the effect of MCEPs generation, pH and contact time on the adsorption capacity were investigated. The adsorption experiments showed that the MCEPs had a high adsorption capacity, when the adsorbent was the second generation of MCEP (MCEP G2.0), the maximum adsorption capacity can up to 702.9 mg/g and the adsorption process can be completed in 60 min. Moreover, among various isotherms models, MCEP G2.0 was well-fitted by Langmuir adsorption isotherm. Also, adsorption kinetic studies proved that the adsorption mechanism of MCEP G2.0 adsorbent followed the pseudo-second-order kinetic model, demonstrating that the process was chemisorption. The adsorbent has renewability as well, after five adsorption cycles, its adsorption efficiency was still higher than 80%. Therefore, the adsorbent showed a superior adsorption capacity and renewability to remove Cu (II).

Abstract:Novel eco-friendly and green dimethyldiallylammonium chloride (DMDAAC) grafted chitosan/genipin/cellulose hydrogel beads (CCBG-g-PDMDAAC), were fabricated as selective adsorbents for anionic dyes. The physical and chemical structural changes of the prepared hydrogels were evaluated by FTIR, XRD, SEM and TG-DSC analysis. Results showed CCBG-g-PDMDAAC efficiently and selectively adsorb anionic dyes (Reactive Red 195-RR195 and Methyl orange-MO) from mixture of dye solutions. Endowed chitosan-based hydrogels the advantage of acid insolubility and good adsorption. RR195 and MO adsorption process were described better with pseudo-second-order kinetic model and Langmuir isotherm model with a maximum adsorption capacity of 1333.52 and 190.48 mg/g, respectively, indicating that monolayer chemisorption controlled the sorption process. Moreover, the hydrogels exhibited nice reusability and against S. aureus and E. coli. The hydrogels are promising for the potential application in wastewater treatment and antibacterial simultaneously.

Abstract:Pathogen and biofilm contamination in aqueous systems leave millions of people at risk of waterborne diseases. Herein, to address this issue, a green and highly efficient strategy is developed to concurrently trap and kill bacteria, eliminate the debris and the existing biofilm matrix in water environment via magnetic microparticles. The particles (TPFPs) were prepared from the in-situ deposition of Fe3O4 nanoparticles onto the surface of antibacterial functionalized microcrystalline cellulose (MCC) Noticeably, TPFPs can completely inactivate both S. aureus and E. coli once contacting for 30 min by disrupting the bacterial membrane. Meanwhile, the MCC-based magnetic particles retained 100% biocidal efficiency against E. coli (5 * 104 E. coli/mg particles) during ten recycling procedures without any treatment. More importantly, according to the results of trapping behavior and antibiofilm assays, not only bacteria could be captured by the particles (trapping rate was over 85%), but also the residual debris from dead bacteria and fragmented biofilm was together removed based on the special structure and functions of the antibacterial particles (~ 80%), including extremely rough surfaces, surficial positive charge and magneto-responsive property.
