Abstract:The application of small molecules as synthetic blocks is a field of great meaning and full of challenges, especially the use of inert ether molecules, such as THF as starting materials. A versatile and easily handled cross-dehydrogenative coupling between ethers and quinaldine is reported here. Compared to the developed reaction, Selectfluor is used as a mild oxidant, and a variety of functional groups are tolerated. Nitrogen protection, anhydrous systems, and external base are not needed, making this method a convenient route to functional heterocycles.

Abstract:A general and highly efficient method for the electrochemical C(sp2)-H amination of aldehyde hydrazones with azoles has been developed. This reaction proceeds under exogenous metal-, catalyst-, and oxidant-free conditions to provide aminated hydrazone derivatives in good to excellent yields. This strategy applies to both aromatic and aliphatic aldehyde hydrazones and tolerates a broad range of functional groups.

Abstract:A visible-light-induced highly efficient C(sp3)-H amination of ethers with amides and azoles has been presented under mild conditions via a nitrogen- and carbon-centered radical coupling process. This protocol successfully utilizes 2,3-dichloro-5,6-dicyano-p-benzoquinone (DDQ) and tert-butyl nitrite (TBN) as cocatalysts to deliver the aminated products of ethers under aerobic conditions. Notably, the developed reaction features the corresponding products in good yields (up to 93%) with a wide substrate scope. The mechanistic study indicates that C-N bond formation proceeds via a direct radical cross-coupling process. Preliminary biological activity analysis indicates that the resulting products have good and selective inhibitory activity on osteosarcoma (OS) cell lines and are promising for use as hits for drug discovery.

Abstract:Here, we report the copper-catalyzed C2 selective cross-dehydrogenative C–N bond formation of azines with azoles. This straightforward method enables us to address the key limitation of prior N–O activation strategy in C2 amination of azines. The wide substrate scope, high functional group tolerance, and ease of operation of the present method are expected to promote its potential application in synthetic chemistry.

Abstract:A Cu-catalyzed tandem synthesis of azole-fused pyrimido[1,2-c]quinazolines and imidazo[1,2-c]quinazolines has been developed. The reaction is based on a C-N cross-coupling/C-H functionalization reaction of 2-(2-bromophenyl)-1,4,5,6-tetrahydropyrimidines with azoles. Abstract:Catalyst- and chemical oxidant-free electrochemical azole C-H aminations were accomplished via cross-dehydrogenative C-H/N-H functionalization. The catalyst-free electrochemical C-H amination proved feasible on azoles with high levels of efficacy and selectivity, avoiding the use of stoichiometric oxidants under ambient conditions. Likewise, the C(sp3 )-H nitrogenation proved viable under otherwise identical conditions. The dehydrogenative C-H amination featured ample scope, including cyclic and acyclic aliphatic amines as well as anilines, and employed sustainable electricity as the sole oxidant.

Abstract:β-Azolyl propanoic acid derivatives are frequently found in biologically active molecules and pharmaceuticals. Here, we report the oxidative heteroarylation of unactivated C(sp3)-H bonds with azole C(sp2)-H bonds via copper or nickel catalysis with the aid of removable bidentate auxiliary, which provides a rapid pathway to β-azolyl propanoic acid derivatives. A variety of azoles such as oxazole, benzoxazole, thiazole, benzothiazoles, benzimidazole, purine, and even [1,2,4]triazolo[1,5-a]pyrimidine could be engaged in this protocol.

Abstract:A copper-promoted intramolecular C–H oxidative amination reaction between secondary amine (N–H) and C(sp3)–H at the benzylic position of azaarenes or α-position of ketones for the synthesis of aziridine derivatives has been developed. Moreover, a practical annulation of electron-deficient vinylarenes with an unprotected primary alkyl amine by a Yb(OTf)3–CuI relay system has also been reported. The reactions were carried out with oxygen as the sole oxidant to give the N-alkyl- and N-arylaziridines in good yields.

Abstract:Copper-catalyzed aerobic oxidative C-H/N-H coupling between simple ketones and diamines was developed toward the synthesis of a variety of pyrazines. Various substituted ketones were compatible for this transformation. Preliminary mechanistic investigations indicated that radical species were involved. X-ray absorption fine structure experiments elucidated that the Cu(II) species 5 coordinated by two N atoms at a distance of 2.04 Å and two O atoms at a shorter distance of 1.98 Å was a reactive one for this aerobic oxidative coupling reaction. Density functional theory calculations suggested that the intramolecular coupling of cationic radicals was favorable in this transformation.

Abstract:A palladium-catalyzed oxidative amination protocol of electron-deficient olefins by aza-Wacker-type reaction with N-alkylsulfonamides was developed. The presence of the stoichiometric amount of methanesulfonic acid was crucial for the success of this transformation. The reactions were conducted in green solvent under mild conditions and scalable with excellent E-type stereoselectivity. In addition, a Pd(II)/Pd(0) catalytic cycle with the existence of a very strong oxidant (Selectfluor) was proposed.

Abstract:The coupling of azoles and azine N‐oxides with the Fischer‐type copper carbene species bearing a α‐siloxyl group which can be in situ generated from acylsilanes catalytically under photoirradiation and redox‐neutral conditions, is realized. This reaction represents the first example of Cu‐catalyzed coupling reaction of electron‐rich Fischer carbene species with hard nucleophiles. KIE studies, kinetic analysis, and in situ NMR experiments implied a bimetallic relay process. The transition metal‐catalyzed direct coupling reactions involving electron‐rich Fischer carbene species are largely underdeveloped and remain a big challenge. Here, a direct coupling reaction of azoles and azine N‐oxides is reported with Fischer copper carbene species bearing an α‐siloxy group i, which can be in situ generated from acylsilanes catalytically under photoirradiation and redox‐neutral conditions. This coupling reaction between electron‐rich α‐siloxy Fischer Cu‐carbene species with hard carbanion nucleophiles may undergo a bimetallic relay process, which is confirmed by the kinetic analysis and in situ NMR analysis. This reaction features mild conditions and remarkable heterocycle compatibility. Notably, this protocol tolerates a series of azole or azine N‐oxide derivatives, including benzoxazole, benzothiazole, benzoimidazole, benzoisoxazole, oxazole, oxadiazole, triazolo[4,3‐a]pyridine, purine, caffeine, pyridine N‐oxide, quinoline N‐oxide, pyrazine N‐oxide, pyridazine N‐oxide, etc. The synthetic value of this approach is demonstrated by the efficient synthesis of a histamine h4 receptor ligand and a marketed drug carbinoxamine.

Abstract:A highly efficient nickel/silver co-catalyzed C-H amination at the C5 position of 8-aminoquinolines with azodicarboxylates at room temperature is reported. The reaction undergoes a self-redox process without the necessity of external oxidant. It proceeded under simple and mild conditions without any additional ligand, base or oxidant and provided the desired products in good to excellent yields. This method also possessed the merits of good functional group compatibility and air and moisture tolerance. It provides an efficient strategy for the synthesis of useful quinoline derivatives.

Abstract:A highly efficient method for the selective formal C–H/C–H cross-coupling of azoles and (hetero)arenes was established through arylsulfonium intermediates under transition-metal catalysis, which produced a variety of 2-(hetero)aryl azoles in good to excellent yields.
