Abstract:This paper reports on an inkjet printing method for the fabrication of lateral flow immunochromatographic devices made from a single piece of filter paper by patterning microfluidic channels and dispensing immunosensing inks, requiring only a single printing apparatus. This "paperfluidic" immunosensing device allows for a less time-consuming and more low-cost fabrication compared with the conventional immunochromatographic strips requiring multiple pads, plastic or nylon backing, and a plastic case. A sandwich immunoreaction was performed on the patterned immunosensing paper device, and the sensitivity of the device was optimized with an IgG model analyte. Inkjet-printed antibodies on the test line and the control line were immobilized by physical adsorption, resulting in a very simple fabrication method applicable for pure cellulose surfaces. The color intensity in the test line and the control line was determined both by naked eye and by means of a color scanner in combination with a simple computer program. With the resulting paperfluidic immunosensing device, human IgG concentrations at least down to 10 μg/l could be detected within 20 min. Additionally, in order to demonstrate the feasibility of a total multianalyte sensing system, a combined immuno-chemical sensing device was also fabricated by patterning an additional microfluidic channel for a chemical assay onto the same paper substrate. This low-cost multianalyte paperfluidic sensing device thus demonstrates the feasibility of simple, portable, and disposable tools for pathogen detection in the field of medical, environmental, and food analyses, possibly resulting in useful devices in remote settings and less-industrialized countries.

Abstract:Fluorescence immunoassay technology is widely recognized as the most sensitive detection technology, now widely used in clinical testing. The research of quantitative analyzer for fluorescence immune chromatography strips is of great significance. In this thesis, a quantitative analyzer for fluorescence immune chromatography strips has realized, based on photoelectric detection principle and immunochromatographic. The system uses LPC1788 and MSP430 as processors. Using some algorithms such as low-pass filtering, least square method and area method to analyze the detected waveform and eliminate noise with high accuracy. In our experiment, the minimum detection concentration is at several mIU/ml levels, the relative standard deviation is less than 2% and it has good linear response characteristics in a concentration range from 0.5125 to 952mIU/ml while the correlation coefficient R2 is 0.981. The instrument has a good prospect.

Abstract:We report a novel chemiluminescence diagnosis system for high-throughput human IgA detection by inkjet nanoinjection on a multicapillary glass plate. As proof-of-concept, microhole-based polydimethylsiloxane (PDMS) sheets were aligned on a multicapillary glass plate to form a microwell array as microreactors for enzyme-linked immunosorbent assay (ELISA). The multicapillary glass plate was utilized as a switch that controlled the holding/passing of the solution. Further, anti-IgA-labeled polystyrene (PS) microbeads was assembled into the microwell array, and an inkjet nanoinjection was specially used to distribute the sample and reagent solution for chemiluminescence ELISA, enabling high-throughput detection of human IgA. As a result, the performance of human IgA tests revealed a wider range for the calibration curve and a lower limit of detection (LOD) of 0.1 ng mL(-1) than the ELISA by a standard 96-well plate. The analysis time and reagent consumption were significantly decreased. The IgA concentrations in saliva samples were determined after 10000-fold dilution by the developed ELISA system showing comparable results by conventional immune assay with 96-wells. Thus, we believe that the inkjet nanoinjection for high-throughput chemiluminescence immunoassay on a multicapillary glass plate will be promising in disease diagnosis.

Abstract:A portable heterogeneous immunoassay system is presented in this paper. It consists of a poly(dimethylsiloxane) (PDMS) based microfluidic chip as the immunoreactor, a miniaturized programmable high voltage sequencer as the power supply and the flow controller, and a laser-optical fiber fluorescence detection module as the signal reader. The operation of this immunoassay system is automatic. The sequential reagent dispensing and washing processes are controlled by the programmable sequencer. The reagent consumption was only 12 microL and the assay time was only 26 min. The detection limit for Escherichia coli O157:H7 bacterial antigen with this miniaturized system was 0.3 ng/muL, lower than that obtained using fluorescence microscope in previous studies.

Abstract:The paper describes a new online quantitative electrophoretically mediated microanalysis (EMMA) for use in immunoassays based on the unique drop-by-drop introduction of a sample by means of an inkjet for capillary electrophoresis (CE). Plugs of a fluorescein-labeled antibody (Anti-humanIgG-DyL550) and human IgG were alternately injected into a capillary using the inkjet, followed by the merging of the plugs and the subsequent immune reaction. The antigen-antibody complex that was formed in the merged zone was then separated by CE. As a proof-of-concept, the method was used to determine human IgG. As a result, both the consumption of the reaction solution and the analysis time were significantly reduced. The method showed a wide linear range (10-2000 ng mL(-1), R-2 = 0.9912) of calibration and the detection limit (5 ng mL(-1)) was substantially lower than that by for conventional methods. Abstract:In this article, a kind of microfluidic method based on MEMS technology combined with gold immunochromatographic assay (GICA) is developed and discussed. Compared to the traditional GICA, this method supplies us convenient, multi-channel, in-parallel, low cost and similar efficiency approach in the fields of alpha-fetopro-tei (AFP)detection. Firstly, we improved the adhesion between the model material SU-8 and Silicon wafer, optimized approaches of the fabrication of the SU-8 model systematically, and fabricate the PDMS micro fluid chip with good reproduction successfully. Secondly, Surface modification and antibody immobilization methods with the GICA on the PDMS micro fluid analysis chip are studied, we choose the PDMS material and transfer GICA to the PDMS micro fluid chip successfully after researching the antibody immobilization efficiency of different materials utilized in fabrication of the micro fluid chip. In order to improve the reaction efficiency of the immobilized antibody, we studied the characteristics of micro fluid without the gas drive, and the fluid velocity control in our design; we also design structure of grove to strengthen the ability of immobilizing the antibody. The stimulation of the structure shows that it achieves great improvement and experiments prove the design is feasible.

Abstract:A fluorescence detection system for a microfluidic device using an organic light-emitting diode (OLED) as the excitation light source and a charge-coupled device (CCD) as the photo detector was developed. The OLED was fabricated on a glass plate by photolithography and a vacuum deposition technique. The OLED produced a green luminescence with a peak emission at 512 nm and a half bandwidth of 55 nm. The maximum external quantum efficiency of the OLED was 7.2%. The emission intensity of the OLED at 10 mA/cm(2) was 13 μW (1.7 mW/cm(2)). The fluorescence detection system consisted of the OLED device, two band-pass filters, a five microchannel poly(dimethylsiloxane) (PDMS) microfluidic device and a linear CCD. The fluorescence detection system was successfully used in a flow-based enzyme-linked immunosorbent assay on a PDMS microfluidic device for the rapid determination of immunoglobulin A (IgA), a marker for human stress. The detection limit (S/N=3) for IgA was 16.5 ng/mL, and the sensitivity was sufficient for evaluating stress. Compared with the conventional 96-well microtiter plate assay, the analysis time and the amounts of reagent and sample solutions could all be reduced.

Abstract:In the work, a novel fluorescence point-of-care testing (POCT) system was developed for sample-to-answer detection of immunoglobulin E (IgE), which is the first time that a cloth material has been applied to the fluorescence detection of proteins. The system included an lateral/vertical flow-based microfluidic (LVFM) model, POCT detector, and smartphone-based data processing module. In the LVFM model, the non-woven cloth and screen-printing methods were applied to break the restrictions of traditional lateral flow strips on nitrocellulose membrane material and fabrication method. The 3D-printed POCT detector was used to supply a dark environment, with advantages of small volume, lightweight, and being easy-to-use. The smartphone-based data processing module was applied for image acquisition, image analysis and data display. After the fluorescent image was taken, the image analysis was carried out by the self-developed APP. By using the proposed POCT system, on the optimized conditions, the desired IgE detection was obtained, with a linear dynamic detection range of 10–1000 ng mL −1 , and detection limit of 8.5 ng mL −1 . In addition, the presented system showed good selectivity, repeatability and stability, and had the potential to be used in the field of detection of allergic diseases.

Abstract:A novel 3D microfluidic paper-based immunodevice, integrated with blood plasma separation from whole blood samples, automation of rinse steps, and multiplexed CL detections, was developed for the first time based on the principle of origami (denoted as origami-based device). This 3D origami-based device, comprised of one test pad surrounded by four folding tabs, could be patterned and fabricated by wax-printing on paper in bulk. In this work, a sandwich-type chemiluminescence (CL) immunoassay was introduced into this 3D origami-based immunodevice, which could separate the operational procedures into several steps including (i) folding pads above/below and (ii) addition of reagent/buffer under a specific sequence. The CL behavior, blood plasma separation, washing protocol, and incubation time were investigated in this work. The developed 3D origami-based CL immunodevice, combined with a typical luminuol-H(2)O(2) CL system and catalyzed by Ag nanoparticles, showed excellent analytical performance for the simultaneous detection of four tumor markers. The whole blood samples were assayed and the results obtained were in agreement with the reference values from the parallel single-analyte test. This paper-based microfluidic origami CL detection system provides a new strategy for a low-cost, sensitive, simultaneous multiplex immunoassay and point-of-care diagnostics.

Abstract:A novel immunosensor of human immune globulin (IgG) was fabricated based on the fluorescence transfer between luminescent semiconductor quantum dots (QDs) and gold nanoparticles (AuNPs). AuNPs and CdSe/ZnS QDs were respectively labeled with immune reaction pair:IgG and goat anti-human immunoglobulin (GIgG), by optimizing the conditions including pH value and protein amount. In the assembled QD-GIgG-IgG-AuNP fluorescence resonance energy transfer (FRET) immunocomplex system, the presence of AuNP-IgG directly reduced the fluorescence intensity of the GIgG conjugated QDs. As a result, the concentration of AuNP-IgG had a linear relationship with the fluorescence decrease in a range of 0-1.57 microg/mL. Furthermore, the mechanism of the QDs' fluorescence decay has also been discussed and attributed to the light-induced photobleaching. This novel sensing method achieves quantitative detection of trace proteins, suggesting the potential of biomolecule-AuNPs conjugation based analytical methods in further application.

Abstract:Quick & easy: A simple and positive-readout fluorescent immunosensor was developed based on a quencher composed of a 3 nm gold nanoparticle (GNP) covalently linked to a Fab fragment, which proved to be stable and specific. The method allows for direct and sensitive detection of target antigens in homogeneous solutions in one step without any separation steps. It has been successfully applied to rapidly quantify the amount of secretory immunoglobulin A (sIgA) in human saliva samples.

Abstract:Fluorescence-based microarrays are promising diagnostic tools due to their high throughput, small sample volume requirements, and multiplexing capabilities. However, their low fluorescence output has limited their implementation for in vitro diagnostics applications in point-of-care (POC) settings. Here, by integration of a sandwich immunoassay microarray within a plasmonic nanogap cavity, we demonstrate strongly enhanced fluorescence which is critical for readout by inexpensive POC detectors. The immunoassay consists of inkjet-printed antibodies on a polymer brush which is grown on a gold film. Colloidally synthesized silver nanocubes are placed on top and interact with the underlying gold film creating high local electromagnetic field enhancements. By varying the thickness of the brush from 5 to 20 nm, up to a 151-fold increase in fluorescence and 14-fold improvement in the limit-of-detection is observed for the cardiac biomarker B-type natriuretic peptide (BNP) compared to the unenhanced assay, paving the way for a new generation of POC clinical diagnostics.

Abstract:Microfluidic paper-based sensors as a new type of microsample detection technology are widely used in medical diagnosis, environmental monitoring, and food safety testing. Inkjet printing has the advantages of simplicity, speed, flexibility, high resolution, low cost, and efficient mass production and has become one of the most cutting-edge technologies in the manufacture of paper-based sensors. In this work, a fully inkjet printing preparation method was proposed for paper-based sensors, which can achieve high-precision, multichannel, and visual fluorescence detection. Three kinds of fluorescent carbon dots (CDs; r-CDs, b-CDs, and y-CDs) were fabricated into inkjet ink by adding a suitable ratio of solvent, PEG, and surfactant FS3100 to control its viscosity, surface tension, and other influencing factors, obtaining the best-visualized fluorescence response on paper. To optimize the full inkjet printing process of the paper-based sensor, we studied the influence of factors such as the hydrophobic material AKD formula, postprocessing conditions, and the structure of the hydrophilic and hydrophobic channels on the paper-based detection accuracy, and it was found that proper AKD concentration, curing time, and temperature can make AKD fully react with paper-based surface groups and produce more hydrophobic groups on the surface and inside of the filter paper, which can form paper-based microfluidic sensors with clear boundaries and fast transmission speed at low cost and high efficiency. The fabricated sensor is used for the fluorometric determination of vitamin C (AA), NO2-, and sunset yellow (SY) at the same time, and the limits of visual detection by eyes are 6 mmol/L (NO2-), 60 μmol/L (SY), and 40 mmol/L (AA). The mechanism of inkjet printing is investigated in detail, which is simple, reliable, and easy to realize mass production and can realize highly sensitive, on-site, and visual detection for food additives.

Abstract:The robust identification and quantification of various biomarkers is of utmost significance in clinical diagnostics and precision medicine. Fluorescent immunoassays are widely used and considered as a gold standard for biomarker detection due to their high specificity and accuracy. However, current commercial immunoassay tests suffer from limited detection sensitivity and complicated, labor-intensive operation procedures, making them impractical for point-of-care diagnosis, particularly in resource-limited regions. Recently, microfluidic immunoassay devices integrated with plasmonic nanostructures have emerged as a powerful tool for sensitive detection of biomarkers, addressing specific issues, such as integration schemes, easy operation, multiplexed detection, and sensitivity enhancement. In this paper, we provide a discussion on the recent advances in the plasmonic nanostructures integrated with microfluidic devices for fluorescent immunoassays. We shed light on the nanofabrication strategies and various fluidic designs for rapid, sensitive, and highly efficient sensing of antigens. Finally, we share our perspectives on the potential directions of these integrated devices for practical applications.

Abstract:The spread of the FluA virus poses significant public health concerns worldwide. Fluorescent lateral flow immunoassay (LFIA) test strips have emerged as vital tools for the early detection and monitoring of influenza infections. However, existing quantitative virus-detection methods, particularly those utilizing smartphone-based sensing platforms, encounter accessibility challenges in resource-limited areas and among the elderly population. Despite their advantages in speed and portability, these platforms often lack user-friendliness for these demographics, impeding their widespread utilization. To address these challenges, this study proposes leveraging the optical pick-up unit (OPU) sourced from commercial optical drives as a readily available fluorescence excitation module for the quantitative detection of antibodies labeled with quantum-dot fluorescent microspheres. Additionally, we utilize miniaturized and high-performance optical components and 3D-printed parts, along with a customized control system, to develop an affordable point-of-care testing (POCT) device. Within the system, a stepping motor scans the test strip from the T-line to the C-line, enabling the calculation of the fluorescence-intensity ratio between the two lines. This simple yet effective design facilitates rapid and straightforward field or at-home testing for FluA. The proposed prototype platform demonstrates promising performance, achieving a limit of detection (LOD) of 2.91 ng/mL, a total detection time of no more than 15 min, and dimensions of 151 mm × 11.2 mm × 10.8 mm3. We believe that the proposed approach holds great potential for improving access to an accurate influenza diagnosis.

Abstract:A lab-on-a-CD immunoassay system was designed, fabricated, and tested. The actuator for the centrifugal platform consists of a flyball governor and a group of spring plungers. The microfluidic structures were fabricated using replica processes with poly-dimethylsiloxane (PDMS) as structural material. For better control, the valves in the microfluidic system were designed to be mechanical-pinch ones and actuated. The burst frequencies of the micro-valves can be tuned independently by adjusting the heights and prestresses of corresponding spring plungers. For the research work reported in this paper, four sequential valves and one inward pump were integrated on a centrifugal microfluidic system to develop an automated immunoassay system. Mouse IgG was used as a model analyte to test the feasibility of using the proposed device for immunoassay applications. Sample solution of goat anti-mouse IgG conjugated with magnetic nanoparticles was used for magnetic trapping mechanism to eliminate the blocking process. Rabbit anti-mouse IgG labeled with fluorescent molecules was used as the signal reporter. The entire microfluidic assay process, including beads trapping, immunoassay reaction, washing and pumping was completed in a centrifugal platform. The detection sensitivity may also be improved by using higher quality monoclonal antibodies. This new immunoassay platform offers some significant advantages over those reported in the field: simple fabrication, robust control, easy integration, and lower cost. The system may also be modified for detection of multiple targets. The technology has great potential for applications in complex fluid handling needed in biological, chemical and medical application in Lab-on-a-CD systems. (C) 2017 Elsevier B.V. Abstract:Abstract Smart fluid manipulation with automatically controlled paper valves will enable automated and multi-step immunoassays on paper-based microfluidic devices. In this work, we present an integrated paper-based microfluidic platform with shape-memory polymer (SMP)-actuated fluid valves capable of automated colorimetric enzyme-linked immunosorbent assays (ELISAs). A single-layer microfluidic paper-based analytical device (μPAD) was designed to store all the reagents on the chip, and sequentially transfer reagents to a paper test zone following a specific ELISA protocol through automatic fluidic flow control by the multiple SMP-actuated valves. The actuation of a paper valve was based on the thermally responsive, duel-state shape transformation of a SMP sheet attached to the root of a paper cantilever beam for driving a hydrophilic paper bridge to connect and disconnect two paper channels. A portable colorimetric reader was developed to control the on-chip valve operations, quantify the colorimetric signal output, display the assay result, and wirelessly transmit the data to a smart phone for the application of telemedicine. Reliable operations of the paper valve and the entire μPAD were demonstrated with success rates of 97% and 93%, respectively. A detection mechanism for valve malfunction was designed and confirmed effective to identify any mal-operation of individual valves, thus rendering our platform reliable in real assays. For device calibration, we conducted direct ELISAs of rabbit IgG in phosphate-buffered saline (PBS), and achieved a low limit of detection (LOD) of 27 pM (comparable to that of standard and paper-based ELISAs). In order to demonstrate the clinical application of our multi-step immunoassay platform, we also conducted sandwich ELISAs to quantify the protein level of an inflammatory cytokine, namely tumor necrosis factor (TNF)-α, in surgically injured laryngeal tissues of rats. Abstract:In this paper, a new microfluidic array device has been fabricated with screen printing technology. In contrast to traditional microfabrication processes, our method is simple, inexpensive and also suitable for mass production. The device is used for sandwich-type electrochemical immunoassay, in which probes are covalently attached to the electrode surface via electropolymerized polypyrrole propylic acid (PPA) film. This novel microfluidic system enables the whole array preparation and detection processes, including the probe immobilization, sample injection, enzyme incubation and electrochemical detection, to be conducted in the sealed microchannels. For a demonstration, mouse IgG is selected as the target analyte and its detection is realized by sandwich ELISA with goat anti-mouse IgG, rat anti-mouse IgG (conjugated to alkaline phosphatase) and p-aminophenyl phosphate (PAPP) as the primary antibody, second antibody, and enzyme substrate, respectively. A detection limit of 10 ng mL(-1) (67 pM) is achieved with a dynamic range of 100 ng mL(-1)-10 microg mL(-1). In addition, anti-goat IgG is also immobilized as an alternative probe to test mouse IgG in the solution, in order to demonstrate the multiplexing capability as well as the specificity of the device. As expected, the electrochemical responses are much lower than that using anti-mouse IgG as the probe, indicating good selectivity of the immunoassay device. These results indicate a great promise toward the development of miniaturized, low-cost protein biochips for clinical, forensics, environmental, and pharmaceutical applications.

Abstract:The feasibility of performing the enzyme-amplified sandwich immunoassay with protein G capillary affinity chromatography and laser induced fluorescence (LIF) detection was investigated using the determination of human immunoglobulin G (hIgG) as a model system. The incubated mixture of samples containing hIgG and alkaline phosphatase (ALP) conjugated goat anti-human IgG F-(ab')2 fragment was loaded onto the capillary column packed with recombinant protein G bound perfusive support in neutral pH. After nonretained compounds were eluted, the fluorogenic ALP substrate, fluorescein diphosphate (FDP), was loaded onto the capillary column followed by stop-flow incubation.Finally, the product of ALP-mediated hydrolysis of FDP, fluorescein, was swept out of the capillary column and detected with a LIF detector using the 488 nm line of an argon ion laser as the excitation source. Chromatographic conditions were optimized. The calibration curve for hIgG was linear over the range of 0.5-50 pmol/L (r(2)=0.999) with the limit of detection of 0.2 pmol/L.
Microfluidic Systems Integrated With Two-Dimensional Surface Plasmon Resonance Phase Imaging Systems For Microarray Immunoassay

Abstract:This study reports a microfluidic chip integrated with an arrayed immumoassay for surface plasmon resonance (SPR) phase imaging of specific bio-samples. The SPR phase imaging system uses a surface-sensitive optical technique to detect two-dimensional (2D) spatial phase variation caused by rabbit immunoglobulin G (IgG) adsorbed on an anti-rabbit IgG film. The microfluidic chip was fabricated by using micro-electromechanical-systems (MEMS) technology on glass and polydimethylsiloxane (PDMS) substrates to facilitate well-controlled and reproducible sample delivery and detection. Since SPR detection is very sensitive to temperature variation, a micromachine-based. temperature control module comprising micro-heaters and temperature sensors was used to maintain a uniform temperature distribution inside the arrayed detection area with a variation of less than 0.3 degrees C. A self-assembled monolayer (SAM) technique was used to pattern the surface chemistry on a gold layer to immobilize anti-rabbit IgG on the modified substrates. The microfluidic chip is capable of transporting a precise amount of IgG solution by using micropumps/valves to the arrayed detection area such that highly sensitive, highly specific bio-sensing can be achieved. The developed microfluidic chips, which employed SPR phase imaging for immunoassay analysis, could successfully detect the interaction of anti-rabbit IgG and IgG. The interactions between immobilized anti-rabbit IgG and IgG with various concentrations have been measured. The detection limit is experimentally found to be 1 x 10(-4) mg/ml (0.67 nM). The specificity of the arrayed immunoassay was also explored. Experimental data show that only the rabbit IgG can be detected and the porcine IgG cannot be adsorbed. The developed microfluidic system is promising for various applications including medical diagnostics, microarray detection and observing protein-protein interactions. (c) 2007 Elsevier B.V.
