Abstract:In this paper, we constructed a label-free ratiometric fluorescence nanoprobe for ascorbic acid (AA) on the basis of the change in dual-emission signals by the specific reaction between cobalt oxyhydroxide (CoOOH) and AA. CoOOH nanoflakes were used as the recognition unit while polymer dots (PFO dots) and gold nanoclusters (AuNCs) served as luminescent units of the nanoprobe. After electrostatic assembly of PFO dots and AuNCs on the surface of CoOOH nanoflakes, the energy transfer between the PFO dots and CoOOH could occur and thereby quench the fluorescence of the PFO dots. When AA was present, CoOOH nanoflakes were reduced to Co2+, resulting in a recovery of the fluorescence of the PFO dots. In the meantime, the generated Co2+ could combine with the surface ligands of AuNCs, accompanied by a reduction in the luminescence intensity of the AuNCs. Therefore, the fluorescence intensity ratio of PFO dots to AuNCs would be related to the concentration of AA. The nanoprobe enabled highly sensitive analysis of AA with a detection limit of 1.9 μM and showed great performance in human serum samples and living cells. This report may not only endow a CoOOH-based nanoprobe with improved ability but also offer a novel strategy for the construction of ratiometric nanoprobes for AA and have potential applications in research into and clinical diagnosis of AA-related diseases.
Catalytic Formation of Silver Nanoparticles by Bovine Serum Albumin Protected-Silver Nanoclusters and Its Application for Colorimetric Detection of Ascorbic Acid

Abstract:We established a simple spectrophotometric and colorimetric method for detection of ascorbic acid based on the growth of silver nanoparticles in bovine serum albumin protected-silver nanoclusters (BSA–AgNCs) and Ag+ mixture. Due to the catalysis of BSA–AgNCs, ascorbic acid could reduce Ag+ to silver nanoparticles (NPs) at room temperature. The color of the mixture changed from colorless to yellow and a strong absorption band near 420nm could be found in their absorption spectra owing to localized surface plasmon resonance (LSPR) of produced silver NPs. The absorbance changes at 420nm had a good relationship with ascorbic acid concentration. Thus, we proposed a spectrophotometric and colorimetric method to determine ascorbic acid in concentration range from 2.0 to 50.0μM, with the corresponding limits of determination (3σ) of 0.16μM.
A Nanoclay-Based Magnetic/fluorometric Bimodal Strategy for Ascorbic Acid Detection

Abstract:Ascorbic acid (AA) plays an essential role in biochemical processes, as a scavenger for preventing oxidative damage. Although various optical measurement strategies for AA detection have been developed, there is an increasing demand for a multi-modal sensor to obtain detailed information. Herein, we propose a facile ion-exchange/coordination strategy for the preparation of a magnetic resonance (MR)/fluorescence nanohybrid by using laponite (Lap) as a matrix and Eu(DPA)(3)/Gd(DPA)(3) (DPA = Pyridine-2, 6-dicarboxylic acid) as a bimodal signal source. This nanohybrid was observed to have a higher MR response than Gd DPA, because Gd DPA was coordinated with Lap to affect proton relaxation parameters, while excellent Eu DPA fluorescence remained. Moreover, potential applications of the nanohybrid had been extended to transparent films and MR/fluorescent inks. Interestingly, the MR/fluorescence of this nanohybrid was simultaneously suppressed by AA. We found that this decrease was attributed to a synergistic effect of acidity and redox. The MR/fluorescence bimodal detection of Eu(DPA)(3)/Gd(DPA)(3)@Lap makes it as a potential biological probe for clinical applications and commercialization because of its simple preparation procedure, excellent MR/fluorescence properties, high sensitivity, and high selectivity. A Novel Electrochemical Aptasensor Based on Polyaniline and Gold Nanoparticles for Ultrasensitive and Selective Detection of Ascorbic Acid.

Abstract:Ascorbic acid (AA) is involved in many physiological activities of the body and plays an important role in maintaining and promoting human health. It is also present in many natural and artificial foods. Therefore, the development of highly sensitive and accurate AA sensors is highly desirable for human health monitoring, as well as other commercial application fields. Herein, an ultrasensitive and selective electrochemical sensor based on an aptamer was developed for the determination of AA for the first time. The aptasensor was fabricated by modifying a composite made of polyaniline (PANI) and gold nanoparticles (AuNPs) on a glassy carbon electrode. The morphologies and electrochemical properties of the resulting electrodes were characterized by various analytical methods. The results indicated relatively good electrical conduction properties of PANI for accelerated electron transfer. The modification with AuNPs provided signal amplification, suitable for applications as novel platforms for the sensitive sensing of AA. Under optimized conditions, the proposed aptasensor displayed a wide linear response toward the detection of AA from 1.0 to 1.0 x 10(5) ng L-1 coupled with a low detection limit of 0.10 ng L-1. The sensor also exhibited excellent selectivity and high stability, with at least 2000-fold higher sensitivity than similar previously reported methods. Importantly, the aptasensor exhibited promising properties for the determination of AA in real fruits, vegetables, and infant milk powder, thereby showing potential for food analysis.

Abstract:In this work, a ratiometric fluorescent nanoplatform for the detection of ascorbic acid (AA) was constructed based on the Ag2S quantum dots (QDs) and multifunctional hydroxyl cobalt oxide nanoflakes (CoOOH NFs). Ag2S QDs can be assembled on the surface of CoOOH NFs by electrostatic adsorption, resulting in the quenching of the NIR fluorescence emission of Ag2S QDs at 680 nm effectively through the inner filter effect (IFE). o-Phenylenediamine (OPD), a common substrate of oxidase-like (OXD) mimic, is rapidly oxidized into the fluorescent product of 2,3-diaminophenazine (DAP) with the appearance of an emission peak at 575 nm under the catalysis of CoOOH NFs. After AA was added, the fluorescence emission of DAP declined because of the decline in the OXD-like activity of CoOOH NFs due to the transformation of Co2+. Simultaneously, Ag2S QDs were released, accompanied by the recovery of red fluorescence. These two fluorescent signals can be excited at the same excitation wavelength, simplifying the detection procedure. Using F575/F680 as the readout, the quantification of AA can be realized with the linear range and detection limit of 0.2 μM-20 mM and 0.014 μM, respectively. The ratiometric fluorescence sensor can be effectively used to determine the content of AA in real samples such as juice and serum. This work integrates the in-situ formation of the fluorescent species via the catalysis of the nanozyme and the redox reaction to destroy the CoOOH NFs nanozyme as well as the two dimensional nanoflake induced turn-off-on strategy for Ag2S QDs, which provides a specific strategy for the selective detection of AA and may offer a reliable approach for the construction of other biosensing platforms.

Abstract:Fluorescence detection is a commonly used analytical method with the advantages of fast response, good selectivity and low destructiveness. However, fluorescence detection, a single-mode detection method, has some limitations, such as background interference that affects the accuracy of the fluorescence signal, lack of visualization of the detection results, and low sensitivity for detecting low-concentration samples. In order to overcome the shortcomings of fluorescence single-mode detection, we used the dual-mode method of fluorescence and colorimetry to detect ascorbic acid. The dual-mode detection of AA by fluorescence and colorimetry in the probe system enhances the specificity and accuracy of the detection. This bimodal detection method solved the problem of low detection sensitivity in the low concentration range of the analytes to be tested, and was linear in the lower (0-50 {\mu}M) and higher (50-350 {\mu}M) concentration ranges, respectively, and had a lower detection limit (0.034 {\mu}M). This glutathione-based gold cluster assay is characterized by simplicity, rapidity and accuracy, and provides a new way for the quantitative analysis of ascorbic acid. In addition, the method was validated during the determination of AA in beverages, which has the advantages of high sensitivity and fast response time.

Fluorescent Sensing of Ascorbic Acid Based on Iodine Induced Oxidative Etching and Aggregation of Lysozyme-Templated Silver Nanoclusters

Abstract:In this work, we developed a sensitive and highly selective fluorescent approach for the detection of ascorbic acid (AA) by taking advantage of the oxidative etching effect of iodine (I-2) on the lysozymestabilized silver nanoclusters (dLys-AgNCs) with fluorescence quenching. I-2 could be produced from the redox reaction between iodate (IO3-) and AA, and thus the fluorescence intensity of dLys-AgNCs was turned off significantly in the coexistence of IO3- and AA. The fluorescence quenching of dLys-AgNCs had a good linear relationship with AA concentration, which allowed the detection of AA in the range from 0.05 to 45.0 mmol L-1 with a detection limit of 20 nmol L-1. The quenching mechanism was elucidated by Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS), transmission electron microscopy (TEM), zeta potential, and dynamic light scattering (DLS) measurements, confirming that the fluorescence quenching of the dLys-AgNCs was attributed to the oxidative etching of the in situ generated I-2, inducing aggregation of the dLys-AgNCs probe by forming Ag@ AgI nanocomposite. The dLys-AgNCs probe exhibited excellent selectivity for AA sensing over several common reducing agents tested. Moreover, this approach was extended to the detection of AA in orange juice and urine with recovery rates in the range of 96.0% (RSD: 4.11) to 100.9% (RSD: 3.28) and 94.5% (RSD: 6.40) to 99.2% (RSD: 5.36), respectively. Abstract:By using Fe 7 S 8 nanosheets (NSs) as catalyst, O -phenylenediamine (OPD) as chromogenic substrate, H 2 O 2 as oxidant, and ascorbic acid (AA) as regulator, a dual ratio colorimetric and fluorescence sensing system for determining AA was constructed. The inherent peroxidase like activity of Fe 7 S 8 NSs enables it to catalyze the oxidation of OPD in the existence of H 2 O 2 , resulting in the formation of yellow-colored 2, 3-diaminophenazine (DAP) with a characteristic absorption peak at 450 nm and a fluorescence emission peak at 560 nm. However, the presence of AA could suppress the oxidation of OPD and induce the generation of quinoxaline derivative (QXD) 3-(dihydroxyethyl) furo [3,4-b] quinoxaline-1-one with absorption and fluorescence emission peaks located at 345 and 435 nm, respectively. Thus, employing the absorption of QXD at 345 nm and emission at 435 nm as the reporting signals, and the absorption of DAP at 450 nm and emission at 560 nm as the reference signals, a ratio colorimetric ( A 345 / A 450 ) and ratio fluorescence ( F 435 / F 560 ) dual signal detection method for AA was developed. Under the optimized conditions, the value of A 345 / A 450 was found to be linear with AA concentration in the ranges of 1.0–40 μM and 40–100 μM, and the value of F 435 / F 560 was strongly correlated with AA concentration in the ranges of 1.0–30 μM and 30–100 μM. When applied for the assay of AA in human serum samples, an acceptable recovery ranging from 93.1 to 106.6 % was obtained, confirming the potential of the sensing system in practical applications.

Abstract:Gold nanorods (GNRs) with suitable aspect ratio were synthesized with a template technique and then dispersed in a saturated sodium citrate solution by ultrasonication to form a GNR suspension. A GNR-modified electrode was fabricated using the GNR suspension. The oxidation of dopamine at the GNR/GC electrode exhibited surprisingly high electrocatalytic activity and adsorption-controlled characteristics. Square-wave voltammetry was used to detect dopamine. At the GNR/GC electrode, the linear concentration range of DA is from 1×10−8M to 1×10−7M and the detection limit (s/n=3) is as low as 5.5×10−9M. The current sensitivity is 3.280μA/μM, and 1000-fold ascorbic acid (AA) cannot interfere with the determination of DA. All these performances are greatly superior to those of the bare GC electrode.

Abstract:We report on an amperometric sensor for ascorbic acid (AA) that is based on highly dense gold-silver nanotubes in a chitosan film on a glassy carbon electrode. The nanotubes were synthesized by a poly(vinyl pyrrolidone)-mediated polyol method employing a replacement reaction with silver nanowires as templates, and were characterized by scanning electron microscopy, energy-dispersive X-ray spectroscopy, and X-ray diffraction. Under the optimal conditions, the sensor exhibits good electrocatalytic activity towards the oxidation of AA, and this enables the determination of AA in the 5 μM to 2 mM concentration range, with a detection limit at 2 μM (at an S/N of 3). The response time is 2 s. The sensor displays good reproducibility, selectivity, sensitivity, and long-term stability.

Abstract:<p>A simple, fast, one-step method is described for ratiometric colorimetric detection of ascorbic acid (AA) based on 4-mercaptophenylboronic acid (4-MPBA) and silver nanoparticles (AgNPs). In the presence of AA, the aggregation process of AgNPs can be triggered and accelerated, due to the interaction between 4-MPBA and the <em>cis</em> 1,2-diol moieties of AA through a reversible esterification, resulting in a distinguish color change from yellow to brownish red. The AA concentration-dependent color change can be quantified by the bare eyes or a UV–Vis spectrometer. The whole AA detection procedure was able to complete in 10 min with an excellent linear relationship (1–500 μM) and a low limit of detection (0.70 μM). The proposed approach was also used to accurately evaluate AA levels in orange juice. In our perception, this assay is rapid, sensitive, and specific, which demonstrates its potential on constructing biochemical and clinical sensors.</p>

Abstract:In this study, a new method for the detection of ascorbic acid (AA) was proposed. It was based on the protective effect of AA on silver triangular nanoplates (Ag TNPs) against Cl- induced etching reactions. Cl- can attack the corners of Ag TNPs and etch them, causing a morphological shift from triangular nanoplates to nanodiscs. As a result, the solution changes color from blue to yellow. However, in the presence of AA, the corners of Ag TNPs can be protected from Cl- etching, and the blue color of the solution remains unchanged. Using this effect, a selective sensor was designed to detect AA in the range of 0-40.00 & mu;M with a detection limit of 2.17 & mu;M. As the concentration of AA varies in this range, color changes from yellow to blue can be easily observed, so the designed sensor can be used for colorimetric detection. This method can be used to analyze fruit juice samples.
A Non-Oxidation Reduction Strategy for Highly Selective Detection of Ascorbic Acid by “On-off-on” Photoluminescence Switches

Abstract:Great progress was made to detect ascorbic acid (AA) based on the photoluminescent nanoprobes/oxidant complexes by using the reducibility of AA. However, some reducing substances including cysteine and gluta-thione disturbed the accuracy of AA assay. Herein, a non-oxidation reduction regulation strategy was fabricated to construct a novel "on-off-on" photoluminescence model for highly selective determination of AA. Initially, copper nanoclusters capsulated into zeolitic imidazolate framework-8 (CuNCs@ZIF-8) was achieved. And then inorganic phosphate ions quenched the emission of CuNCs@ZIF-8 owing to the decomposition of ZIF-8 shell, rendering the signal in "turn-off" state. The linear range for phosphate ions was 1.0-60 mu M with LOD of 0.35 mu M. Interestingly, upon introducing AA into CuNCs@ZIF-8/phosphate, the emission of CuNCs@ZIF-8 recovered possibly due to the electronegativity improvement of metal core and surface rigidified through hydrogen bond interaction, and the system signal was "turned-on". The LOD of AA was as low as 51 nM in a good linear range from 0.1 to 40 mu M. Furthermore, CuNCs@ZIF-8 were used for AA detecting in tablets, beverages, and fresh fruits with the satisfactory recoveries from 96% to 104%. More importantly, a paper-based analytical device containing CuNCs@ZIF-8/phosphate were evaluated for the visual sensing of AA in food safety.

Abstract:The low activity of nanozymes poses a significant scientific challenge, limiting their application. Developing noble metal-based nanozymes with high activity is considered as an effective strategy to address this issue. In this study, core–shell Au@PdNi nanozyme was synthesized by co-precipitating Pd 2+ and Ni 2+ on Au cores. The oxidase (OXD)-like property of Au@PdNi was thoroughly investigated, revealing a specific activity reaching 194.2 U mg −1 . The high OXD-like activity was attributed to several factors elucidated through experimental analysis and Density Functional Theory (DFT) calculations. Firstly, the core–shell structure of Au@PdNi provides a substantial surface area for catalytic reactions. Secondly, the presence of nickel enhances the current density of the material, augmenting its catalytic activity. Lastly, the electronic interplay between the Au core and the PdNi shell layer improves the binding and reaction kinetics with the target substrate. The Au@PdNi nanozyme demonstrates remarkable performances in colorimetric and electrochemical detection of ascorbic acid (AA), exhibiting high sensitivity and selectivity. Moreover, a portable electrochemical sensor was fabricated for the determination of AA in spiked fruit samples, demonstrating satisfactory recovery rates in real sample analysis.

Abstract:The development of a specific reaction of nanomaterials and reactive species is of fundamental importance for the determination of biomolecules. Here we report a novel nanoprobe for detection and imaging of ascorbic acid (AA) in living cells and in vivo based on the specific reaction of cobalt oxyhydroxide (CoOOH) and AA. Persistent luminescence nanoparticles (PLNPs) were used as the luminescence unit, and CoOOH nanoflakes served as the quencher. When CoOOH was modified on the surface of the PLNPs, the luminescence of the PLNPs was efficiently quenched by the CoOOH. In the presence of AA, CoOOH was reduced to Co(2+) and the luminescence of PLNPs was restored. The nanoprobe showed high selectivity and an instantaneous response. The luminescence property permits detection and imaging without external excitation, which could effectively avoid background noise and scattering of light from biological matrixes produced by in situ excitation. The current strategy provides an effective platform for monitoring and imaging reactive species in living cells and in vivo.
Fe 3 O 4 @Au-metal organic framework nanozyme with peroxidase-like activity and its application for colorimetric ascorbic acid detection.

Abstract:A free radical scavenging system based on FeO@Au/MOF-ABTS˙ has formed the basis of a novel method for the highly sensitive and specific spectrophotometric determination of ascorbic acid (AA). The FeO@Au/MOF nanozyme with magnetic separation properties was effectively prepared and evaluated using an environmentally friendly technique. Nanomaterials have the advantages of superparamagnetism, biocompatibility, chemical stability, and enhanced synergistic peroxidase-like activity, which can be utilized in catalysis to oxidise the peroxidase substrate 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) into a green-colored product in the presence of HO. AA as an antioxidant has scavenging effects on ABTS radicals and can reduce green ABTS˙ to uncolored ABTS, contributing to a substantial reduction in green color. Based on such a premise, a highly selective and sensitive chromogenic sensing method depending on the peroxidase-like activity of the nanocomposites was developed in order to achieve the efficient detection of AA in real samples. Under optimum conditions, the proposed technique had a detection range of 0.001-0.1 mmol L, a limit of detection of 0.098 μmol L, and a detection time of only 30 seconds. The newly proposed colorimetric analysis method devoid of enzymes has broad application potential in the areas of quality control and quality and safety detection.
Microfluidic chemistry assisted synthesis of cobalt quantum dot embedded nitrogen doped carbon with oxidase-like properties toward ascorbic acid detection

Abstract:Ascorbic acid (AA) is a powerful antioxidant in food safety and disease treatment. It is of great significance to develop a low-cost, high-stability, and easy-to-operate colorimetric method for quantitative detection of AA in food or human body. Although various nanozymes have been developed for the colorimetric detection of AA, the size regulation of the catalytic center of nanozymes remains a challenge. In this work, we propose a combined strategy of flow chemistry synthesis and pyrolysis to realize the controllable adjustment of the catalytic center size of nanozymes. Zinc-cobalt zeolitic imidazole frameworks (ZnCo-ZIFs) with different sizes are synthesized by flow chemistry. Nitrogen-doped carbon materials with different Co catalytic centers (80 nm-10 nm) are then obtained by pyrolysis of ZnCo-ZIFs precursors. Among them, cobalt quantum dot embedded nitrogen-doped carbon (Co QDs/N-C) exhibits excellent oxidase activity, with Vmax and Km of 4.19 × 10-7 M s-1 and 0.12 mM. Therefore, a simple, low-cost, and stable colorimetric method for the detection of AA is established with a good linear relationship (3-500 μM) and low detection limit (0.40 μM). This work has certain guiding significance for the size regulation of catalytic center of nanozyme, and the detection method has broad application prospects in biochemical sensing field.

Abstract:In this work, we developed a simple and selective luminescence sensing system for detecting ascorbic acid (AA) based on NaGdF4:Yb,Er@NaYF4 upconversion nanoparticles (UCNPs) and oxidase-like CoOOH nanoflakes. When p-phenylenediamine (PPD) and oxidase-like CoOOH nanoflakes were added to the UCNPs solutions, PPD, as a typical substrate of oxidase, could be oxidized to PPDox. The luminescence intensity of UCNPs was quenched because PPDox could play a role as an energy acceptor. When AA was added to the above solution mixture, the CoOOH nanoflakes were destroyed, and then, the luminescence intensity was recovered. Under the best experimental conditions, the linear range for detection of AA was 0.8 to 280 μM, and the detection limit was 0.25 μM. Furthermore, the system could be applied to real sample analysis with satisfactory results.
