Cancer cell membrane-coated up-conversion nanoparticles for homologous-targeting multimodality imaging

Abstract:1026 Objectives: At present, triple-negative breast cancer (TNBC) has no specific molecular target. Cell-derived nanoparticles have been gaining increasingly attention due to their ability to mimic the properties of their source cells. Herein, we design a new platform of cancer cell membrane-coated up-conversion nanoparticles (CCm-UCNP) for homologous-targeting multi-modal imaging, including up-conversion luminescence imaging (UCL), magnetic resonance imaging (MRI) and positron emission tomography (PET) tumor imaging, for specific diagnosis and differential diagnosis of TNBC. Methods: Differential centrifugation was used to gain pure cancer cell membranes of MDA-MB-231 cells (CCm231), and coating up-conversion nanoparticles (UCNPs) by physical extrusion through a 400 nm porous polycarbonate membrane. The characterization was measured, and the protein expression before and after cancer cell membrane-coated was confirmed. Inserting 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-(polyethylene glycol)2000-Cy5 (DSPE-PEG-Cy5) into CCm231 to label nanoparticles, and homologous-targeting ability and immune evasion ability were observed through incubating Cy5-DSPE-PEG-CCm231-UCNPs with different kinds of breast cancer cells and Raw264.7 cells (a kind of mice macrophage) by confocal laser scanning microscope. CCK-8 assay was used to evaluate the cytotoxicity. CCm231-UCNPs and UCNPs were injected intravenously into MDA-MB-231 and MCF-7 tumor bearing mice for UCL and MRI respectively. 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[azido(polyethylene glycol)-2000] (DSPE-PEG-N3) was inserted into CCm231to react with DBCO by click chemistry for radiolabeling 18F used for PET imaging. N3-DSPE-PEG-CCm231-UCNPs were injected intravenously into MDA-MB-231 and MCF-7 tumor bearing mice of 24 h pre-positioning respectively, then DBCO-L-NETA-Al18F were injected, followed by serial PET imaging and ex vivo biodistribution study. For evaluating systematic toxicity, BALB/c-nu mice (n = 6) received injection of the same concentration of CCm231-UCNPs, UCNPs and CCm231, PBS were used as control. Results: Core-shell structure of CCm231-UCNPs of ~ 25 nm in size, with the similar protein profile before and after membrane-coated were successfully synthesized. The fluorescence signal of MDA-MB-231 group was extremely higher than those of other cell groups. Raw264.7 had nearly no uptake of Cy5-DSPE-PEG-CCm231-UCNPs, which exhibited immune evasion ability. No obvious cytotoxicity of CCm231-UCNPs had been observed. In the image of UCL and MRI, CCm231-UCNPs to MDA-MB-231 tumor group showed highest tumor site uptake, and the best imaging time was 24 h post-injection. Lower fluorescence signal of liver was observed compared to the group of UCNPs. As for PET imaging, N3-DSPE-PEG-CCm231-UCNPs to MDA-MB-231 tumor group showed highest tumor site uptake (6.39±1.12, 4.97±0.52, 4.70±0.9 and 4.05±0.5 %ID/g at 0.5, 1, 2 and 4h respectively) compared with those in MCF-7 tumor group (1.48±0.18, 1.16±0.10, 1.28±0.09 and 1.20±0.14 %ID/g at 0.5, 1, 2 and 4h respectively). Also, the liver and spleen were not visible. Neither death nor significant difference in body weight among four groups were observed after 30 days. Blood routine, hepatic and renal functions tests, and histology analysis suggested good biocompatibility of CCm231-UCNPs in vivo. Conclusions: We have successfully constructed CCm231-UCNPs, and the immune evasion ability and homologous targeting ability were confirmed by in vitro experiments and in vivo multi-modal imaging, including UCL, MRI and PET. No significant cytotoxicity and biological toxicity of these biomimetic nanoparticles were detected. Our study provides a new method for specific diagnosis and differential diagnosis of TNBC.
