Abstract:Skin-interfaced electrochemical sensing devices are widely developed for biochemical sensing at molecular levels. However, the sensing electrodes fabricated with photolithography or printing technique are hard to achieve high stretchability without special designs like serpentine shape. Also, most of them require wired connections with external electrochemical workstations for data acquisition or on-board batteries to power the circuits, which largely restrain the flexibility, simplification, and miniaturization of the devices. Here, a battery-free, wireless, and epidermal electrochemical system is presented for in situ biochemical sensing. The sensing component of the system is a stretchable electrode array all-printed with soft polymer and conductive silver nanowires. The electrodes show stable electrical properties within strain range of 0-50%, without serpentine designs. The electronic component of the system is a fully integrated flexible circuit board with near field communication module, which enables wireless energy harvesting and data transmission with smartphones. The system demonstrates good performance in real-time on-body sweat analysis for simultaneous quantitative detections of glucose, hydrogen, sodium, and potassium. This battery-free and wireless epidermal electrochemical system provides a simplified, miniaturized, and flexible solution for a wide range of biochemical platforms, including wearable and implanted bioelectronics.

Abstract:Capabilities in health monitoring enabled by capture and quantitative chemical analysis of sweat could complement, or potentially obviate the need for, approaches based on sporadic assessment of blood samples. Established sweat monitoring technologies use simple fabric swatches and are limited to basic analysis in controlled laboratory or hospital settings. We present a collection of materials and device designs for soft, flexible, and stretchable microfluidic systems, including embodiments that integrate wireless communication electronics, which can intimately and robustly bond to the surface of the skin without chemical and mechanical irritation. This integration defines access points for a small set of sweat glands such that perspiration spontaneously initiates routing of sweat through a microfluidic network and set of reservoirs. Embedded chemical analyses respond in colorimetric fashion to markers such as chloride and hydronium ions, glucose, and lactate. Wireless interfaces to digital image capture hardware serve as a means for quantitation. Human studies demonstrated the functionality of this microfluidic device during fitness cycling in a controlled environment and during long-distance bicycle racing in arid, outdoor conditions. The results include quantitative values for sweat rate, total sweat loss, pH, and concentration of chloride and lactate.

Abstract:Skin-interfaced wearable systems with integrated colorimetric assays, microfluidic channels, and electrochemical sensors offer powerful capabilities for noninvasive, real-time sweat analysis. This Perspective details recent progress in the development and translation of novel wearable sensors for personalized assessment of sweat dynamics and biomarkers, with precise sampling and real-time analysis. Sensor accuracy, system ruggedness, and large-scale deployment in remote environments represent key opportunity areas, enabling broad deployment in the context of field studies, clinical trials, and recent commercialization. On-body measurements in these contexts show good agreement compared to conventional laboratory-based sweat analysis approaches. These device demonstrations highlight the utility of biochemical sensing platforms for personalized assessment of performance, wellness, and health across a broad range of applications.This article has not yet been cited by other publications.

Abstract:A wearable sweat sensor, which could continuously monitor biomolecules related to the human physiological state, is emerging as a promising piece of health surveillance equipment. However, current sensors cannot simultaneously achieve a detection performance that equates to that of traditional sensors and satisfactory mechanical strength. Herein, a wearable sweat sensor with excellent detection performance and mechanical stability is designed and fabricated. Based on the integration of laser-induced graphene electrodes and a screen printing technique, this wearable sweat sensor could realize both the separate and simultaneous detection of uric acid (UA), tyrosine (Tyr), and ascorbic acid (AA) with high sensitivity. Good UA sensing performance in artificial sweat could be maintained even after 20 000 bends. In addition, the sensor can operate well in the wearing state or in a complex bovine whole blood sample. For the detection of human sweat, the changes in UA concentration after a purine-rich meal are continuously monitored and the results are in accordance with the corresponding serum UA detection results tested with a commercial serum UA meter. These results suggest its application potential in health monitoring for both gout patients and healthy humans.

Abstract:Wearable biosensors are attracting great interest thanks to their high potential for providing clinical-diagnostic information in real time, exploiting non-invasive sampling of biofluids. In this context, sweat has been demonstrated to contain physiologically relevant biomarkers, even if it has not been exhaustively exploited till now. This biofluid has started to gain attention thanks to the applications offered by wearable biosensors, as it is easily collectable and can be used for continuous monitoring of some parameters. Several studies have reported electrochemical and optical biosensing strategies integrated with flexible, biocompatible, and innovative materials as platforms for biospecific recognition reactions. Furthermore, sampling systems as well as the transport of fluids by microfluidics have been implemented into portable and compact biosensors to improve the wearability of the overall analytical device. In this review, we report and discuss recent pioneering works about the development of sweat sensing technologies, focusing on opportunities and open issues that can be decisive for their applications in routine-personalized healthcare practices.

Abstract:Wearable non-invasive biosensors for the continuous monitoring of metabolites in sweat can detect a few analytes at sufficiently high concentrations, typically during vigorous exercise so as to generate sufficient quantity of the biofluid. Here we report the design and performance of a wearable electrochemical biosensor for the continuous analysis, in sweat during physical exercise and at rest, of trace levels of multiple metabolites and nutrients, including all essential amino acids and vitamins. The biosensor consists of graphene electrodes that can be repeatedly regenerated in situ, functionalized with metabolite-specific antibody-like molecularly imprinted polymers and redox-active reporter nanoparticles, and integrated with modules for iontophoresis-based sweat induction, microfluidic sweat sampling, signal processing and calibration, and wireless communication. In volunteers, the biosensor enabled the real-time monitoring of the intake of amino acids and their levels during physical exercise, as well as the assessment of the risk of metabolic syndrome (by correlating amino acid levels in serum and sweat). The monitoring of metabolites for the early identification of abnormal health conditions could facilitate applications in precision nutrition.

Abstract:The urgent need for real-time and noninvasive monitoring of health-associated biochemical parameters has motivated the development of wearable sweat sensors. Existing electrochemical sensors show promise in real-time analysis of various chemical biomarkers. These sensors often rely on labels and redox probes to generate and amplify the signals for the detection and quantification of analytes with limited sensitivity. In this study, we introduce a molecularly imprinted polymer (MIP)-based biochemical sensor to quantify a molecular biomarker in sweat using electrochemical impedance spectroscopy, which eliminates the need for labels or redox probes. The molecularly imprinted biosensor can achieve sensitive and specific detection of cortisol at concentrations as low as 1 pM, 1000-fold lower than previously reported MIP cortisol sensors. We integrated multimodal electrochemical sensors with an iontophoresis sweat extraction module and paper microfluidics for real-time sweat analysis. Several parameters can be simultaneously quantified, including sweat volume, secretion rate, sodium ion, and cortisol concentration. Paper microfluidic modules not only quantify sweat volume and secretion rate but also facilitate continuous sweat analysis without user intervention. While we focus on cortisol sensing as a proof-of-concept, the molecularly imprinted wearable sensors can be extended to real-time detection of other biochemicals, such as protein biomarkers and therapeutic drugs.

Abstract:The absolute configurations of three new enantiomerically pure ferrocenylphosphole compounds, namely (2S,4S,S(Fc))-4-methoxymethyl-2-[2-(9-thioxo-9lambda5-phosphafluoren-9-yl)ferrocenyl]-1,3-dioxane, [Fe(C5H5)(C23H22O3PS)], (III), (S(Fc))-[2-(9-thioxo-9lambda5-phosphafluoren-9-yl)ferrocenyl]methanol, [Fe(C5H5)(C18H14OPS)], (V), and (S(Fc))-diphenyl[2-(9-thioxo-9lambda5-phosphafluoren-9-yl]ferrocenylmethyl]phosphine, [Fe(C5H5)(C30H23P2)], (VIII), have been unambiguously established. All three ligands contain a planar chiral ferrocene group, bearing a dibenzophosphole and either a dioxane, a methanol or a diphenylphosphinomethane group on the same cyclopentadienyl. In compound (V), the occurrence of O-H...S and C-H...S hydrogen bonds results in the formation of a two-dimensional network parallel to (001). The geometry of the ferrocene frameworks agrees with related reported structures.

Abstract:The significant impact of stress on health necessitates accurate assessment methods, where traditional questionnaires lack reliability and objectivity. Current advancements like wearables with electrocardiogram (ECG) and galvanic skin response (GSR) sensors face accuracy and artifact challenges. Molecular biosensors detecting cortisol, a critical stress hormone, present a promising solution. However, existing cortisol assays, requiring saliva, urine, or blood, are complex, expensive, and unsuitable for continuous monitoring. Our study introduces a passive, molecularly imprinted polymer-radio-frequency (MIP-RF) wearable sensing system for real-time, non-invasive sweat cortisol assessment. This system is wireless, flexible, battery-free, reusable, environmentally stable, and designed for long-term monitoring, using an inductance-capacitance transducer. The transducer translates cortisol concentrations into resonant frequency shifts with high sensitivity (∼ 160 kHz/(log (μM))) across a physiological range of 0.025–1 μM. Integrated with near-field communication (NFC) for wireless and battery-free operation, and three-dimensional (3D)-printed microfluidic channel for in-situ sweat collection, it enables daily activity cortisol level tracking. Validation of cortisol circadian rhythm through morning and evening measurements demonstrates its effectiveness in tracking and monitoring sweat cortisol levels. A 28-day stability test and the use of cost-effective 3D nanomaterials printing enhance its economic viability and reusability. This innovation paves the way for a new era in realistic, on-demand health monitoring outside the laboratory, leveraging wearable technology for molecular stress biomarker detection.

Abstract:Stretchable sweat sensors are promising technology that can acquire biomolecular insights for health and fitness monitoring by intimate integration with the body. However, current sensors often require microfabricated microfluidic channels to control sweat flow during lab-on-body analysis, which makes effective and affordable sweat sampling a significant practical challenge. Here, we present stretchable and sweat-wicking patches that utilize bioinspired smart wettable membranes for the on-demand manipulation of sweat flow. In a scalable process, the membrane is created by stacking hydrophobic elastomer nanofibers onto soft microfoams with predefined two-dimensional superhydrophobic and superhydrophilic patterns. The engineered heterogeneous wettability distribution allows these porous membranes to achieve enhanced extraction and selective collection of sweat in embedded assays. Despite the simplified architecture, the color reactions between sweat and chemical indicators are inhibited from directly contacting the skin to achieve a largely improved operation safety. The sensing patches can simultaneously quantify pH, urea, and calcium in sweat through digital colorimetric analysis with smartphone images. The construction with all compliant materials renders these patches soft and stretchy to achieve conformal attachment to the skin. Successfully analyzing sweat compositions after physical exercises illustrates the practical suitability of these skin-attachable sensors for health tracking and point-of-care diagnosis.

Abstract:Real-time monitoring of biological markers in sweat is a valuable tool for health assessment. In this study, we have developed an innovative wearable biosensor for precise analysis of glucose in sweat during physical activities. The sensor is based on a single-atom catalyst of platinum (Pt) uniformly dispersed on tricobalt tetroxide (Co 3 O 4 ) nanorods and reduced graphene oxide (rGO), featuring a unique three-dimensional nanostructure and excellent glucose electrocatalytic performance with a wide detection range of 1-800 μM. Additionally, density functional theory calculations have revealed the synergetic role of Pt active sites in the Pt single-atom catalyst (Co 3 O 4 /rGO/Pt) in glucose adsorption and electron transfer, thereby enhancing sensor performance. To enable application in wearable devices, we designed an S-shaped microfluidic chip and a point-of-care testing (POCT) device, both of which were validated for effectiveness through actual use by volunteers. This research provides valuable insights and innovative approaches for analyzing sweat glucose using wearable devices, contributing to the advancement of personalized healthcare.

Abstract:Bioreceptor‐based wearable electrochemical sensors have opened new avenues for on‐body and continuous molecular detections, enabling personalized and preventive healthcare. This review summarises recent innovations and applications of those wearable biosensing systems in detecting a variety of biochemical markers, such as small molecules, hormones, drugs, and macromolecules, in biofluids including interstitial fluid, sweat, wound exudate, saliva, and tears. Recent advancements in wearable electrochemical biosensors have opened new avenues for on‐body and continuous detection of biomarkers, enabling personalized, real‐time, and preventive healthcare. While glucose monitoring has set a precedent for wearable biosensors, the field is rapidly expanding to include a wider range of analytes crucial for disease diagnosis, treatment, and management. In this review, recent key innovations are examined in the design and manufacturing underpinning these biosensing platforms including biorecognition elements, signal transduction methods, electrode and substrate materials, and fabrication techniques. The applications of these biosensors are then highlighted in detecting a variety of biochemical markers, such as small molecules, hormones, drugs, and macromolecules, in biofluids including interstitial fluid, sweat, wound exudate, saliva, and tears. Additionally, the review also covers recent advances in wearable electrochemical biosensing platforms, such as multi‐sensory integration, closed‐loop control, and power supply. Furthermore, the challenges associated with critical issues are discussed, such as biocompatibility, biofouling, and sensor degradation, and the opportunities in materials science, nanotechnology, and artificial intelligence to overcome these limitations.

Abstract:Sweat, as a biofluid with the potential for noninvasive collection, provides profound insights into human health conditions, because it contains various chemicals and information to be utilized for the monitoring of well-being, stress levels, exercise, and nutrition. Recently, wearable sweat sensors have been developed as a promising substitute to conventional laboratory sweat detection methods. Such sensors are promising to realize low-cost, real-time, in situ sweat measurements, and provide great opportunities for health status evaluation analysis based on personalized big data. This review first presents an overview of wearable sweat sensors from the perspective of basic components, including materials and structures for specific sensing applications and modalities. Current strategies and specific methods of the fabrication of wearable power management are also summarized. Finally, current challenges and future directions of wearable sweat sensors are discussed.

Abstract:In the field of wearable sensing, small and precise sensors can greatly reduce the burden on the wearer and improve the sense of experience, which is the future direction of sensing development. Herein, we introduce an ultra-small wearable biosensor system that integrates an MS02 chip for real-time and highly accurate sweat detection. The whole system mainly includes flexible electrodes and a printed circle board (PCB). The size of the PCB is only 1.5 cm × 0.8 cm, which greatly minimizes the size of the sweat system and improves wearing comfort. Notably, this miniaturized system is comparable to a commercial electrochemical workstation, ensuring the reliability and accuracy of real-time analysis. The core processing MS02 chip, with a dimension of 1.2 mm × 1.1 mm, is used to perform electrochemical signal processing. By performing electrochemical characterization and measurements of the ultra-small wearable biosensor system, on-body monitoring of four biomarkers (glucose, lactate, Na+, and K+) in sweat of human volunteers has been successfully achieved. With the help of this electrochemical sensor system, mass of biochemical data from perspiration can be acquired to better understand the body's response to daily activities, which will facilitate the early prediction of abnormal physiological changes in the future.

An Integrated Wearable Sweat Sensing Patch for Passive Continuous Analysis of Stress Biomarkers at Rest

Abstract:The wearable sweat sensing patch fabricated integrates a microfluidic chip, an electrochemical sensing patch, and an on‐site signal processing circuitry capable of simultaneously detecting cortisol, magnesium ions, and pH in sweat. The detection results can be wirelessly transmitted to smartphones in real time to assess a person's mental health. Real‐time monitoring of mental stress biomarkers in sweat provides the possibility to evaluate mental status in a precise manner. In general, wearable sweat sensors suffer from inconvenient sweat collection, low levels of diagnostic biomarkers in sweat, sophisticated signal processing, and challenges with data visualization. To overcome these challenges, herein an integrated wearable sweat‐sensing patch for continuous analysis of stress biomarkers (cortisol, Mg2+, and pH) at rest is demonstrated. The sweat sensing patch comprised a microfluidic chip, a highly sensitive sensing platform, an on‐site signal processing circuitry (SPCs), and a smartphone installed with a home‐developed display software. The sweat collection at rest is realized using a microfluidic chip without perspiration assistance. A ternary composite electrode is designed to obtain good conductivity, high surface area, and massive reactive sites, thereby yielding excellent electrochemical performances and high sensitivity to trace stress biomarkers. The on‐site SPC has the function of signal transduction, conditioning, processing, and wireless transmission. The detection results can be displayed on a smartphone through the software. This work represents a significant scientific and technological advancement toward indexing mental stress status and can be used as an innovative tool for psychological diagnosis.

Abstract:Sweat analysis has advanced from diagnosing cystic fibrosis and testing for illicit drugs to noninvasive monitoring of health biomarkers. This article introduces the rapid development of wearable and flexible sweat sensors, highlighting key milestones and various sensing strategies for real-time monitoring of analytes. We discuss challenges such as developing high-performance nanomaterial-based biosensors, ensuring continuous sweat production and sampling, achieving high sweat/blood correlation, and biocompatibility. The potential of machine learning to enhance these sensors for personalized healthcare is presented, enabling real-time tracking and prediction of physiological changes and disease onset. Leveraging advancements in flexible electronics, nanomaterials, biosensing, and data analytics, wearable sweat biosensors promise to revolutionize disease management, prevention, and prediction, promoting healthier lifestyles and transforming medical practices globally.
