Abstract:A self-assembly of an active tumor-cell-targeted photothermal agent, functionalized with a cyclic Arg-Gly-Asp peptide and ibuprofen, for killing cancer cells and eliciting anti-inflammatory responses was developed. In vitro and in vivo experiments also demonstrated that the photothermal material exhibited low cytotoxicity, good biological compatibility and excellent tumor inhibitory effects.

Abstract:Phototherapy has emerged as a promising way to treat cancer with high selectivity and non-invasiveness. With a vision towards solving the safety problems of current inorganic materials, tremendous efforts have been dedicated to search for much safer and more efficient organic nanoagents for cancer phototherapy. Herein, we design and synthesize a new porphyrin compound (TPAPor) by covalent conjugation of porphyrin and triphenylamine. The typical donor-acceptor-donor (D-A-D) structure of TPAPor facilitates the red-shift of absorption to the nearinfrared (NIR) region. Subsequently, the amphiphilic organic molecule is self-assembled into nanoparticles (TPAPor NPs) with homogeneous size (about 80 nm), enhanced NIR absorption and excellent biocompatibility. TPAPor NPs display low singlet oxygen (O-1(2)) generation and excellent photothermal conversion efficiency (55.7%) under irradiation of a single NIR laser (690 nm). Moreover, TPAPor NPs could ablate the HeLa cells by NIR laser-induced ROS and heat generation, with the half maximal inhibitory concentrations (IC50) about 18.3 mu g/mL. This study indicates TPAPor NPs are promising nanoagents for cancer therapy through photothermal/photodynamic combination manner.
Polypyrrole-Coated Mesoporous TiO2 Nanocomposites Simultaneously Loading DOX and Aspirin Prodrugs for a Synergistic Theranostic and Anti-Inflammatory Effect

Abstract:Although a number of therapeutic strategies have been applied in cancer therapy, treatment for cancer metastasis is challenging due to unsatisfactory cure rate and easy cancer recurrence. In our work, nanocomposites (NCs) based on polypyrrole-coated mesoporous TiO2 with a suitable size are prepared through a modified soft-templating strategy, which integrates double prodrugs (doxorubicin (DOX) prodrug and aspirin prodrug) with superior drug loading capacity. Under external stimulation of near-infrared (NIR) and ultrasound (US), the prepared nanocomposites have an excellent photothermal conversion efficiency (over 50.8%) and a satisfactory sonodynamic therapeutic effect, and simultaneous prodrug activation and drug release occur rapidly under external stimulation. Through intravenous injection, the tumor area can be clearly seen through thermal imaging, benefiting from the enhanced permeability and retention (EPR) effect. Through synergistic therapy, cancer cell toxicity and the tumor inhibition effect are significantly enhanced. Moreover, downregulated inflammatory factors also reduce the risk of cancer recurrence. In general, the designed NCs provide a potential alternative for synergistic therapy as well as downregulation of inflammatory cytokines.

[Construct validity of the Information and Decisions in Asthma questionnaire].

Abstract:OBJECTIVE To establish the construct validity of a new scale based on the Information and Decisions in Asthma (IDEA) questionnaire, which was designed to detect an asthmatic's desire to receive information, active seeking out of information and ability to make decisions about his or her disease. METHODOLOGY The IDEA questionnaire containing 31 items in three subscales: desire for information (DI), seeking out information (SI) and decision making (DM) was answered by 120 asthmatic adults (54 men and 66 women; 86 intrinsic and 44 extrinsic) in stable condition and with varying levels of severity of disease. After interviews to take down medical histories (age of onset, years of evolution, visits to the emergency room within the past year), educational level and economic status, all patients completed a battery of tests assessing the following: quality of life (QL), alexithymia (TAS-20), health opinions (HOS), state-trait anxiety (STAI-E/R), asthma symptoms control (ASC), health locus control (HLC), somatosensory amplification (SAS) and personality (NEO-PI). RESULTS Although most patients expressed interest in obtaining information, their level of active seeking and decision making were much lower. DI and SI were associated with higher educational levels and economic status, whereas DM correlated with the presence of atopy, age (young patients), sex (women) and few visits to the emergency room. Multiple regression analysis of psychological variables showed that DI and SI were accounted for mainly by the absence of alexithymia, whereas DM depended on an individual's behavioral involvement with his or her disease. CONCLUSION The global analysis of these results indicate that the IDEA questionnaire has satisfactory construct validity, given that its dimensions are associated with sociodemographic and psychological characteristics that can be expected given its conceptual content.
Injectable nano-composite hydrogels based on hyaluronic acid-chitosan derivatives for simultaneous photothermal-chemo therapy of cancer with anti-inflammatory capacity

Abstract:Nowadays, the photothermal therapy (PTT) has received widespread attention and research by rapidly killing tumors with local high temperature. However, due to the irregular edges of tumor and the blurred boundary between normal and necrotic tissues, the desirable treatment cannot be achieved by the single PTT, and excessive heat will cause serious inflammation in local tissues. Herein, an injectable composite hydrogel is prepared by the oxidized hyaluronic acid (OHA) and hydroxypropyl chitosan (HPCS) via the imine bonds, which is employed as the delivery substrate for functional substances. In the gel medium, the mesoporous polydopamine (MPDA) nanoparticles are incorporated as the high efficiency photothermal agent and a reservoir of DOX, which can achieve the good photothermal conversion performance and pulsed drug release. Besides, the addition of the curcumin-cyclodextrin host-guest inclusion complex (CUR@NH<sub>2</sub>-CD) in the composite hydrogel could reduce the inflammation caused by PTT. The composite hydrogel shows favorable the Hepa1-6 tumor inhibition in vivo by virtue of the comprehensive effect of the admired photothermal efficacy of MPDA, chemotherapy of DOX and anti-inflammatory of CUR. It can be predicted that the composite hydrogel has a broad prospect in the field of comprehensive therapy for tumor.

A Photothermal-Hypoxia Sequentially Activatable Phase-Change Nanoagent for Mitochondria-Targeting Tumor Synergistic Therapy.

Abstract:To enhance the specificity and efficiency of anti-tumor therapies, we have designed a multifunctional nanoparticle platform for photochemotherapy using fluorescence (FL) and photoacoustic (PA) imaging guidance. Nanoparticles (NPs) composed of a eutectic mixture of natural fatty acids that undergo a solid-liquid phase transition at 39 °C were used to encapsulate materials for the rapid and uniform release of the hypoxia-activated prodrug tirapazamine (TPZ) and the photosensitizer IR780, which targets the mitochondria of tumor cells and can be used to induce hypoxic cell death via photodynamic therapy and photothermal therapy. In vitro, the NPs containing TPZ and IR7890 exhibited appreciable cell uptake and triggered drug release when irradiated with a NIR laser. In vivo, photochemotherapy of the NPs achieved the best anti-tumor efficacy under PA and FL imaging guidance and monitoring. By combining IR780 mitochondria-targeting phototherapy with TPZ, we observed improved anti-tumor effectiveness and this has the potential to reduce the side effects of traditional chemotherapy. Herein, we demonstrate a new intracellular photochemotherapy nanosystem that co-encapsulates photosensitizers and hypoxia-activated drugs to enhance the overall anti-tumor effect precisely and efficiently.

Abstract:Design of new nanoagents that intrinsically have both diagnostic imaging and therapeutic capabilities is highly desirable for personalized medicine. In this work, a novel nanotheranostic agent is fabricated based on polydopamine (PDA)-functionalized Co-P nanocomposites (Co-P@PDA) for magnetic resonance imaging (MRI)-guided combined photothermal therapy and chemotherapy. The ultrahigh relaxivity of 224.61 mm(-1) s(-1) can enable Co-P@PDA to be applied as an excellent contrast agent for MRI in vitro and in vivo, providing essential and comprehensive information for tumor clinical diagnosis. Moreover, Co-P@PDA exhibit excellent photothermal performance owing to the strong near-infrared (NIR) absorbance of both Co-P nanocomposite and PDA. Highly effective ablation of tumors is achieved in a murine tumor model because the NIR laser not only induces photothermal effects but also triggers the chemotherapeutic drug on-demand release, which endows the Co-P@PDA with high curative effects but little toxicity and few side effects. These findings demonstrate that Co-P@PDA are promising agents for highly effective and precise antitumor treatment and warrant exploration as novel theranostic nanoagents with good potential for future clinical translation.

Abstract:A rational combination of different therapeutic modalities within one single nanostructure is promising to enhance the therapeutic response, especially to achieve a synergistic therapeutic efficacy for tumor treatment. Herein, a near-infrared (NIR) photothermally activated nanoagent, which could achieve a spatially controllable codelivery of different curative molecules and a temporally controlled responsive release, was designed to perform effective genechemo- photothermal therapy of malignant tumors. The nanoagent consisted of a gold nanorod (AuNR) functionalized with mPEG, DNA, and small interfering RNA (siRNA). With the aid of aptamer AS1411-mediated recognition and endocytosis, the nanoagents were selectively delivered into cancer cells; subsequently, the photothermal conversion of AuNRs happened while under NIR irradiation, which successfully achieved an effective photothermal therapy, induced dehybridization of DNA duplexes, and simultaneously released doxorubicin (DOX) and siRNA. Then, the released siRNAY silenced the expression of the multidrug resistance associated protein 1 (MRP1), the primary cause of the undesirable expelling of DOX in PC-3 cells, yielding a remarkable improvement in the efficiency of gene-chemo therapy. All of the results of in vitro and in vivo studies revealed that our prepared nanoagents exhibited an excellent performance in synergistic gene-chemo-photothermal therapy and successfully inhibited tumor growth. This work provides an interesting concept of a nanoscale therapeutic agent in achieving a dramatically enhanced therapeutic ability for tumor ablation, which benefits from the spatiotemporally controllable properties and the synergistic combination of chemotherapy, gene therapy, and photothermal therapy in one single nanoagent.
Mitochondria-targeting polydopamine-coated nanodrugs for effective photothermal- and chemo-synergistic therapies against lung cancer

Abstract:Abstract Targeting mitochondria via nano platform emerged as an attractive anti-tumor pathway due to the central regulation role in cellar apoptosis and drug resistance. Here, a mitochondria-targeting nanoparticle (TOS-PDA-PEG-TPP) was designed to precisely deliver polydopamine (PDA) as the photothermal agent and alpha-tocopherol succinate (α-TOS) as the chemotherapeutic drug to the mitochondria of the tumor cells, which inhibits the tumor growth through chemo- and photothermal- synergistic therapies. TOS-PDA-PEG-TPP was constructed by coating PDA on the surface of TOS NPs self-assembled by α-TOS, followed by grafting PEG and triphenylphosphonium (TPP) on their surface to prolong the blood circulation time and target delivery of TOS and PDA to the mitochondria of tumor cells. In vitro studies showed that TOS-PDA-PEG-TPP could be efficiently internalized by tumor cells and accumulated at mitochondria, resulting in cellular apoptosis and synergistic inhibition of tumor cell proliferation. In vivo studies demonstrated that TOS-PDA-PEG-TPP could be efficiently localized at tumor sites and significantly restrain the tumor growth under NIR irradiation without apparent toxicity or deleterious effects. Conclusively, the combination strategy adopted for functional nanodrugs construction aimed at target-delivering therapeutic agents with different action mechanisms to the same intracellular organelles can be extended to other nanodrugs-dependent therapeutic systems.

Abstract:Nanomedicine has attracted growing attention due to its designability and functionality, as well as its excellent pharmacokinetics with limited side effects, and recently, combined therapies have become desirable as they can obtain enhanced therapeutic efficacy by using nanomedicine. Herein, we have reported a functional drug delivery system with a dual response to temperature and reactive oxygen species to efficiently eliminate pancreatic cancer cells in a combined therapy strategy. Functional micelles with camptothecin (CPT) in the core and indocyanine green (ICG) on the surface could effectively accumulate in tumor sites through the EPR effect. The ROS in the tumor microenvironment trigger the conversion of an amino-based copolymer to a carboxy based copolymer, releasing the loaded ICG to reduce the size of the micelles with high penetration in tumor tissue. On the one hand, under 808 nm light irradiation, the micelles will produce the heat to kill tumor cells via photothermal therapy. On the other hand, the generated heat could further trigger the transition of a copolymer from a hydrophobic to a hydrophilic state, releasing the loaded CPT into the deep tumor cells to achieve chemotherapy. The in vitro and in vivo experiments revealed that this combined therapy could combat pancreatic cancer cells with an enhanced therapeutic effect.

Abstract:Due to the high incidence and mortality rates of cancer, it is necessary to seek more targeted and effective treatment methods. Traditional cancer treatment methods have problems such as incomplete treatment and significant side effects. In recent years, photothermal therapy (PTT) has been considered one of the most promising treatment methods in the biomedical field. The treatment principle of PTT is to use materials with high photothermal conversion efficiency to absorb near-infrared light (NIR) and generate heat to ablate tumors. PTT has the advantages of low toxicity, non-invasiveness, strong specificity, and high controllability. However, the self-protective mechanism related to heat shock proteins (HSPs) gives cancer cells thermal tolerance. To minimize the thermal tolerance of cancer cells and improve the efficacy of PTT, a nano-assembly composed of Pluronic F-127 encapsulating the HSP inhibitor APO and the high photothermal conversion efficiency photosensitizer Cy7-TCF has been developed. The nano-assembly can accumulate at the tumor site and generate heat under near-infrared light irradiation. At the same time, it inhibits the expression of heat shock proteins, achieving the effect of mild photothermal therapy and reducing thermal damage to surrounding tissues.

Abstract:Photothermal therapy, in which light is converted into heat and triggers local hyperthermia to ablate tumors, presents an inherently specific and noninvasive treatment for tumor tissues. In this area, the development of efficient photothermal agents (PTAs) has always been a central topic. Although many efforts have been made on the investigation of novel molecular architectures and photothermal materials over the past decades, PTAs can cause severe damage to normal tissues because of the poor tumor aggregate ability and high irradiation density. Recently, dual-targeted photothermal agents (DTPTAs) provide an attractive strategy to overcome these problems and enhance cancer therapy. DTPTAs are functionalized with two classes of targeting units, including tumor environment targeting sites, tumor targeting sites and organelle targeting sites. In this perspective, typical targeted ligands and representative examples of photothermal therapeutic agents with dual-targeted properties are systematically summarized and recent advances using DTPTAs in tumor therapy are highlighted.
Self-assembling combretastatin A4 incorporated protamine/nanodiamond hybrids for combined anti-angiogenesis and mild photothermal therapy in liver cancer

Abstract:Tumor angiogenesis has been identified as an important factor in the development and progression of tumors, and anti-angiogenesis therapy has been recognized as an effective tumor therapy pattern. The unique characteristics of nanodiamonds (NDs) have been explored for photothermal therapy (PTT) against cancer, while the efficiency of mild PTT mediated by bare NDs was limited. The combination of different therapies into a single nanoplatform has shown great potential for synergistic cancer treatment. In this investigation, we integrated hydrophobic antiangiogenesis agent combretastatin A4 (CA4) into the protamine sulfate (PS) functionalized NDs hybrids (NDs@PS) with a noncovalent self-assembling method (CA4-NDs@PS) for potential combined anti-angiogenesis and mild PTT in liver cancer. The resulted CA4-NDs@PS NDs exhibited high drug loading ability, good dispersibility and colloidal stability. The near-infrared (NIR) laser irradiation could trigger the release of CA4 from CA4-NDs@PS NDs and elevate the temperature of CA4-NDs@PS NDs aqueous solution. In vitro results illustrated that CA4-NDs@PS coupled with laser irradiation could remarkably enhance HepG-2 cells killing efficiency, leading to an enhanced photocytotoxicity. Furthermore, in vivo experiments revealed that CA4-NDs@PS exhibited a highly synergistic anticancer efficacy with NIR laser irradiation in HepG-2 tumor-bearing mice. Altogether, our present study fabricated a novel NDs@PS-based nanoplatform for combined anti-tumor angiogenesis and mild PTT against liver cancer.

Abstract:Image-guided combined chemo-thermal therapy assists in optimizing treatment time, enhancing therapeutic efficiency, and circumventing side effects. In the present study, we developed a chemo-photothermal theranostic platform based on polydopamine (PDA)-coated gold nanorods (GNRs). The PDA coating was thin; however, it significantly suppressed the cytotoxicity of the cetyltrimethylammonium bromide template and allowed high cisplatin loading efficiency, arginine-glycine-aspartic acid (RGD) peptide (c(RGDyC)) conjugation, and chelator-free iodine-125 labeling (RGD-125IPt-PDA@GNRs). While loaded cisplatin was released in a pH-sensitive manner, labeled 125I was outstandingly stable under biological conditions. RGD-125IPt-PDA@GNRs had a high specificity for αvβ3 integrin, and consequently, they could selectively accumulate in tumors, as revealed by single photon emission computed tomography/CT imaging, and in target tumor angiogenic vessels, as shown by high-resolution photoacoustic imaging. As RGD-125IPt-PDA@GNRs targets tumor angiogenesis, it is a highly potent tumor therapy. Combined chemo-photothermal therapy with probes could thoroughly ablate tumors and inhibit tumor relapse via a synergistic antitumor effect. Our studies demonstrated that RGD-125IPt-PDA@GNRs is a robust platform for image-guided, chemo-thermal tumor therapy with outstanding synergistic tumor killing and relapse inhibition effects.

Abstract:Photothermal therapy (PTT) is an emerging field, and the photothermal agents could convert visible or near-infrared (NIR) radiation into heat to kill tumor cells. However, the low photothermal conversion efficiency of photothermal agents and their limited antitumor activities delay the development of these agents into monotherapies for cancer. Herein, we have fabricated an ultrasmall polyvinylpyrrolidone (PVP)-Fe-Cu-Ni-S (PVP-NP) nano-agent via a simple hot injection method with an excellent photothermal conversion efficiency (~96%). Photothermal therapy with this nano-agent effectively inhibits tumor growth without apparent toxic side effects. Mechanistically, our results demonstrated that, after NIR irradiation, PVP-NPs can induce ROS/singlet oxygen generation, decrease mitochondrial membrane potential, and release extracellular Fe2+ and consume glutathione, triggering autophagy and ferroptosis of cancer cells. Moreover, PVP-NPs exhibit excellent contrast enhancement according to magnetic resonance imaging (MRI) analysis. In summary, PVP-NPs have a high photothermal conversion efficiency and can be applied for MRI-guided synergistic photothermal/photodynamic/chemo-dynamic cancer therapy, solving the bottleneck of existing phototherapeutic agents.

Synergistic Chemo-thermal Therapy of Cancer by DNA-Templated Silver Nanoclusters and Polydopamine Nanoparticles

Abstract:Herein, we develop a novel and effective combination nanoplatform for cancer theranostics. Folic acid (FA) is first modified on the photothermal agent of polydopamine (PDA), which possesses excellent near-infrared (NIR) absorbance and thermal conversion features. Temperature-sensitive silver nanoclusters (AgNCs) are then synthesized on the DNA template that also loads the anticancer drug doxorubicin (Dox). After accumulation in cancer cells, PDA generates cytotoxic heat upon excitation of NIR light for photothermal therapy. On the other hand, the temperature increment is able to destroy the template of AgNCs, leading to the fluorescence variation and controlled release of Dox for chemotherapy. The combined nanosystem exhibits outstanding fluorescence tracing, NIR photothermal transduction, as well as chemo drug delivery capabilities. Both in vitro and in vivo results demonstrate excellent tumor growth suppression phenomena and no apparent adverse effects. This research provides a powerful targeted nanoplatform for cancer theranostics, which may have great potential value for future clinical applications.
A Photoactivatable Self-Assembled Nanoagonist for Synergistic Therapy against Pancreatic Ductal Adenocarcinoma

Abstract:Immunotherapy has revolutionized the cancer treatment paradigm, yet efficient immunotherapeutic responses against immune-cold/desert cancers remain challenging. Herein, we report that photoactivatable nanoagonists yield a potent antitumor synergy of photoimmunotherapy against pancreatic ductal adenocarcinoma (PDAC). The nanoagonist was fabricated by assembling an amphiphilic boron dipyrromethene-derived polymer conjugated with a Toll-like receptor agonist via a photocleavable linker and stimulator of interferon genes agonist. The nanoagonist enables light-induced generation of reactive oxygen species and on-demand release of the agonists to yield synergistic photoimmunotherapy. The produced tumor antigens promote dendritic cell maturation, which is further amplified by these agonists for eliciting adaptive immunity, accompanied by apparently abscopal and long-term memory effects. The nanoagonist further alleviates the fibrosis of tumor stroma and the immunosuppressive microenvironment, leading to the deep infiltrations of clinically used therapeutics and immune cells to yield preferable combinational treatments against PDAC models. These results provide valuable insights into activatable nanoparticles for cancer therapy against immune-desert cancers.
