Abstract:A novel transformation for the construction of Csp3-Csp bonds was achieved via a photo-induced copper-catalysed C-C bond cleavage. This approach was applied to prepare a series of highly functionalized alkynyl nitriles using readily available cyclobutanones and terminal alkynes. Mechanistic exploration showed that the in situ generated copper acetylide complex is an effective photosensitive catalyst to promote the C-C bond cleavage of cycloketoxime esters through a radical process.

Abstract:Here we report an efficient copper-catalysed selective C-C bond cleavage/amination of cyclobutanone oxime esters. This reaction protocol is operationally simple and conducted at ambient temperature, allowing access to a wide range of functionalized 4-(arylamino)butanenitriles in moderate to excellent yields. This transformation shows high chemo-selectivity and wide functional-group compatibility and can be easily scaled up to the gram level with a useful yield. A mechanism involving copper-catalysed capture of alkyl radical intermediates by amine nucleophiles is proposed.

Abstract:We have developed a modular, convenient and practical copper-catalyzed platform for the preparation of alkene nitriles from corresponding cycloketoxime esters via radical-mediated C-C bond cleavage. A broad scope of cycloketoxime esters (4-membered to 8-membered) and some complex substrates derived from natural product derivatives performed well, affording corresponding ring opening products in the presence of copper salts. Moreover, the etherification, oxidation and alkynylation of cycloketoxime esters could also be achieved under variable copper-catalyzed conditions.

Abstract:A novel structural reorganization of cycloketoxime esters beyond the traditional Beckmann rearrangement process has been established to build cyano-containing ketones in the presence of photocatalyst. This novel transformation is remarkable with selective CC bond cleavage and an oxidation process enabled by DMSO used as the solvent, oxidant, and oxygen source avoiding acid, base and toxic cyanide salts as the cyano source. Abstract:A metal-free, visible-light-induced oxidative C-C bond cleavage of cycloketones with molecular oxygen is described. Cooperative Brønsted-acid catalysis and photocatalysis enabled selective C-C bond cleavage of cycloketones to generate an array of γ-, δ- and ϵ-keto esters under very mild conditions. Mechanistic studies indicate that singlet molecular oxygen (1 O2 ) is responsible for this transformation.

Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:The first example of intermolecular olefination of cyclobutanone oximes with alkenes via selective C-C bond cleavage leading to the synthesis of nitriles in the presence of a cheap copper catalyst is reported. The procedure is distinguished by mild and safe reaction conditions that avoid ligand, oxidant, base, or toxic cyanide salt. A wide scope of cyclobutanones and olefin coupling components can be used without compromising efficiency and scalability. The alternative visible-light-driven photoredox process for this coupling reaction was also uncovered.

Abstract:The copper-catalyzed cyclization of activated alkenes with cyclobutanone O-acyl oximes under redox-neutral conditions has been reported. This facile protocol provided an efficient approach to a variety of cyanoalkylated oxindoles and dihydroquinolin-2(1H)-ones with a broad substrate scope and excellent functional group tolerance. In this reaction, sequential C-C bond cleavage, radical addition, and cyclization processes were involved, wherein multiple bonds were constructed in a one-pot reaction. Mechanistic studies suggest that the reaction probably proceeded via a radical pathway.

Abstract:The construction of carbon–heteroatom bonds is one of the most active areas of research in organic chemistry because the function of organic molecules is often derived from the presence of heteroatoms. Although considerable advances have recently been achieved in radical-involved catalytic asymmetric C–N bond formation, there has been little progress in the corresponding C–O bond-forming processes. Here, we describe a photoinduced copper-catalyzed cross-coupling of readily available oxime esters and 1,3-dienes to generate diversely substituted allylic esters with high regio- and enantioselectivity (>75 examples; up to 95% ee). The reaction proceeds at room temperature under excitation by purple light-emitting diodes (LEDs) and features the use of a single, earth-abundant copper-based chiral catalyst as both the photoredox catalyst for radical generation and the source of asymmetric induction in C–O coupling. Abstract:Base-promoted C-H cleavage without transition metals opens a practical alternative for the one based on noble metals or radical initiators. The resulting carbanion can pass through radical addition to unsaturated bonds like C-N or C-C triple bonds, in which stoichiometric oxidants are needed. When in situ C-H cleavage meets catalytic carbanion-radical relay, it turns to be challenging but has not been accomplished yet. Here we report the combination of base-promoted benzylic C-H cleavage and copper-catalyzed carbanion-radical redox relay. Catalytic amount of naturally abundant and inexpensive copper salt, such as copper(II) sulfate, is used for anion-radical redox relay without any external oxidant. By avoiding using N-O/N-N homolysis or radical initiators to generate iminyl radicals, this strategy realizes modular synthesis of N-H indoles and analogs from abundant feedstocks, such as toluene and nitrile derivatives, and also enables rapid synthesis of large scale pharmaceuticals.

Abstract:Selective activation and transformation of multiple inert chemical bonds is a challenging yet intriguing direction in synthetic chemistry. We report a copper‐mediated complete cleavage and selective transformation of sp2C—H bond in indoles, three sp3C—H bonds and one C—N bond in a methyl carbon atom in TMEDA, and the C≡N triple bond in CH3CN for cyanation of indoles via tandem carbon and nitrogen atom transfer. Comprehensive Summary The activation of inert chemical bonds is an exciting area of research in chemistry because it enables the direct utilization of readily available starting materials and promotes atom‐ and step‐economic synthesis. Undoubtedly, selectively activating and transforming multiple inert chemical bonds is an even more intriguing and demanding task in synthetic chemistry. However, due to its inherent complexity and extreme challenges, this endeavour is rarely accomplished. We report a copper‐mediated complete cleavage and selective transformation of multiple inert chemical bonds of three easily available feedstocks, i.e., a sp2C—H bond in indoles, three sp3C—H bonds and one C—N bond in a methyl carbon atom in TMEDA, and the C≡N triple bond in CH3CN. This reaction proceeds via tandem carbon and nitrogen atom transfer, and allows for the direct and efficient cyanation of indoles, presenting a simple and direct alternative for synthesizing 3‐cyanoindoles.

Abstract:A regioselective C-C bond cleavage/aminocarbonylation cascade is presented. In recent years, tremendous progress has been made in the alkoxyl radical-mediated C-C bond cleavage of unstrained monocarbocycles. In contrast, the deconstruction and functionalization of bicyclic skeletons has been less developed and has mainly focused on the ring expansion process. Inspired by the aromatization-driven C-C bond cleavage, here we demonstrate a ring-opening/aminocarbonylation cascade under copper catalysis, in which the formation of a stable γ-lactam or succinimide skeleton reverses the selectivity of C-C cleavage. Remarkably, the photo and thermal assistance is not required when the succinimide skeleton is formed during the ring opening process. DFT calculations revealed that this unexpected ring-opening process is thermodynamically and kinetically favourable.

Abstract:Photocatalytic coupling reactions have developed rapidly in the field of organic synthesis. However, highly efficient, broad-spectrum, low-cost, and energy-saving catalytic systems are urgently needed. Herein a desirable alternative combining NaI–PPh3 photoredox catalyst and Cu(I) catalyst was theoretically designed and evaluated from photophysical processes and potential energy surfaces. This metallaphotoredox catalysis could achieve C(sp3)–O/N cross-couplings of alkyl N-hydroxyphthalimide esters with phenols/secondary amines via a radical mechanism merging photoexcited radical decarboxylation and a low-valent CuI–CuII–CuI cycle. More importantly, a series of target reactions can be realized with high yield (≥90%) at room temperature under only irradiation with 10 W blue light-emitting diodes for 4 h without additional precious photocatalysts, which is mild, convenient, and environmentally friendly.
