In 2017, over 70% of potato plants showing chlorotic yellow patches of rounded shape were observed in the potato fields in Hangzhou, Zhejiang province, China. To determine the causal agent, a small RNA (sRNA) library was constructed using symptomatic leaf tissues of a local retention potato variety collected in a potato field near the Xixi National Wetland Park in Hangzhou. The sRNA library was sequenced using the Illumina HiSeq next-generation sequencing platform (Lianchuan Bio, Hangzhou, Zhejiang, China). After trimming adapter and removing low-quality sequences, a total number of 2,204,676 clean sRNAs with length between 15 and 29 nucleotides (nt) were obtained. These sRNAs were assembled and analyzed as described earlier (Wu et al. 2018). Results revealed 69, 48, and 61 contigs share about 98, 96, and 59 to 82% nt sequence similarities to the genomic sequences of potato virus A (PVA; genus Potyvirus), potato aucuba mosaic virus (PAMV; genus Potexvirus), and potato virus H (PVH; genus Carlavirus), respectively. These results suggested that this sample was coinfected by at least three viruses. Because PAMV has not been reported in China, the full genome of this PAMV isolate (referred as PAMV-HZ) was determined by reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends. Amplified fragments were ligated into pEASY-Blunt vector (TransGen Biotech, Beijing, China) and sequenced with each fragment of at least four clones. Excluding the poly-A tail, the full genome of PAMV-HZ is 7,064 nt in length (GenBank submission no. MH379106), which shares the highest nucleotide acid identity (96.2%) with a Canadian isolate of PAMV (GenBank accession no. NC_003632). Sequence analysis revealed nine and three nucleotide insertions and deletions between HZ-1 and the Canadian isolate, which led to reading-frame shifts between 403 and 411 amino acids (aa) and 1,615 to 1,647 aa in RNA dependent RNA polymerase, 75 to 80 aa and 121 to 139 aa in triple-gene-block protein 1 (TGBp1), and 55 to 72 aa in TGBp3. To test the infectivity of PAMV-HZ, Nicotiana benthamiana and potato cultivar Jinshu-3 seedlings were mechanically inoculated with sap extracted from the symptomatic potato leaves. N. benthamiana plants developed systematic necrosis symptoms at 7 days postinoculation (dpi), whereas Jinshu-3 plants did not show obvious symptom even at 14 dpi. RT-PCR using primers designed based on assembled contigs showed that PAMV and PVA could be successfully detected in the systemic leaves of N. benthamiana and potato, whereas PVH was only detected in the systemic leaves of potato, agreeing with previous observation that N. benthamiana is not a susceptible host of PVH (Li et al. 2013). We subsequently screened 34 potato samples from five cities in Zhejiang (Hangzhou, Quzhou, Huzhou, Jinhua, and Ningbo) by RT-PCR using primers PAMV-6350F and PAMV-7000R. PAMV was only detected on six samples from Hangzhou and Huzhou. These results suggested that PAMV was transmitted to Zhejiang recently through the transportation of potato germplasm or tuber and has not been broadly distributed on potato in Zhejiang yet. To the best our knowledge, this is the first report of PAMV on potato in China. Given the importance of potato, special attention should be paid to the damage that PAMV may cause to the potato industry in China.References:Li, Y., et al. 2013.
