Abstract:Polycyclic aromatic compounds are important constituents of pharmaceuticals and other materials. We have developed a series of Rh-catalyzed tandem carbonylative benzannulations for the synthesis of tri-, tetra-, and pentacyclic heterocycles from different types of aryl propargylic alcohols. These tandem reactions provide efficient access to highly substituted carbazoles, furocarbazoles, pyrrolocarbazoles, thiophenocarbazoles, and indolocarbazoles. While tricyclic heterocycles could be derived from vinyl aryl propargylic alcohols, tetra- and pentacyclic heterocycles were synthesized from diaryl propargylic alcohols. The tandem carbonylative benzannulation is initiated by a π-acidic rhodium(I) catalyst-mediated nucleophilic addition to alkyne to generate a key metal-carbene intermediate, which is then trapped by carbon monoxide to form a ketene species for 6π electrocyclization. Overall, three bonds and two rings are formed in all of these tandem carbonylative benzannulation reactions.

Abstract:A Cp*Rh-III-catalyzed annulative carbooxygenation of challenging 1,1-disubstituted alkenes triggered by C-H activation of N-aryloxyacetamides has been established, which affords 2,3-dihydrobenzofuran derivatives with a quaternary carbon center in good to excellent yields under mild redox-neutral conditions. An amide group on the alkenes is essential for the process, and may inhibit the beta-H elimination from C(sp3)-Rh species by saturating the rhodium center through coordination. Furthermore, mechanistic insights obtained from control experiments suggest a mechanism involving a Rh-III-Rh-V-Rh-III catalytic cycle.

Abstract:A novel protocol of rhodium-catalyzed tandem C-H annulation of aldehyde and aniline with diazonaphthalen-1(2H)-one via transient directing group strategy and sequential nucleophilic substitution has been demonstrated in moderate to excellent yields. This reaction proceeds smoothly via intramolecular cyclization to form 6H-benzo[c]chromen-6-ol and is followed by nucleophilic substitution of para-carbon of aniline to construct 6-aryl-6H-benzo[c]chromene derivatives. A series of 6-aryl-6H-benzo[c]chromenes were synthesized in one-pot via the tandem C-H annulation.

Abstract:Rh (III)-catalyzed C-H activation of benzoylacetonitriles in coupling with diazo compounds was developed to synthesize diversified substituted benzo[ de]chromenes via a formal (4 + 2) cycloaddition with a diazo compound and subsequent tandem (4 + 2) cycloaddition with another diazo compound. Intriguingly, synthesis of substituted benzo[ de]chromenes and their decarboxylation products could be realized by controlling the reaction conditions. These reactions have a broad range of substrates, moderate to good yields, and high regioselectivity.

Abstract:Benzofused nitrogen heterocycles are prevalent as key core structural motifs in functional molecules. Major syntheses of benzofused nitrogen heterocycles focus on construction of the heterocyclic ring starting from (poly)substituted benzene derivatives. Given that polysubstituted benzene derivatives are not always easily available, the flexibility of these methods may be problematic. Abstract:Ring expansion reactions have been proven to be efficient tools for building medium-sized rings with broad applications in synthetic and medicinal chemistry. However, the strategy involving pericyclic or pseudopericyclic pathways via sulfur ylide-initiated [1,4]-sigmatropic rearrangement remains an under-exploited area. Herein, we disclose an interesting ring expansion of thiochromenes and aromatic thiophenes via rhodium catalysis, which enables the straightforward assembly of polyaromatic oxathionines and oxathiocines with diverse functionalities. The mechanistic investigation via DFT calculations revealed this ring expansion transformation to be a [1,4]-sigmatropic rearrangement via a metal-free sulfur-based ylide.

Abstract:An intermolecular efficient dimerization of diynes via Rh-catalyzed cycloaddition is presented. This protocol allows for the expedient synthesis of structurally diverse benzene derivatives, which retain an alkyne unit for post-functionalization to realize molecules with potential medicinal value and extended molecular complexity. The transformation is carried out in toluene and employs Rh(COD)2OTf3 as the metal source. Diversely decorated diynes tethered by nitrogen atom and ester were compatible with these conditions and furnished substituted benzenes with excellent selectivity.

Abstract:Dual activation of both C-I and vicinal C-H bonds of diaryliodonium salts allowing for diarylation is a subject of rapid construction of π-extended frameworks. Here, we report palladium-catalyzed cascade of C8-arylation/intramolecular Friedel-Crafts acylation of α-naphthoic acids in the synthesis of benzanthrone derivatives. The step-economical protocol tolerates various substrates, which resulted in a potential molecular library for developing functional polycyclic scaffolds. The approach relies on the synergistic action of strong acid with palladium catalysts to form two bonds in a one-pot procedure.

Abstract:A novel rhodium-catalyzed C–C bond olefination of ring-fused benzocyclobutenols with ethyl acrylate is described. A variety of versatile olefinated products were efficiently generated at room temperature and then readily converted into many useful polycyclic compounds by one-step processes. Abstract:Transition-metal-catalyzed directed C-H functionalization has emerged as a powerful and straightforward tool to construct C-C bonds and C-N bonds. Among these processes, the intramolecular annulative alkene hydroarylation reaction has received much attention because this intramolecular annulation can produce more complex and high value-added structural motifs found in numerous natural products and bioactive molecules. Despite remarkable progress, these annulative protocols developed to date remain limited to hydroarylation and functionalization of one side of alkenes, thus largely limiting the structural diversity and complexity. Herein, we developed a rhodium(iii)-catalyzed tandem annulative arylation/amidation reaction of aromatic tethered alkenes to deliver a variety of 2,3-dihydro-3-benzofuranmethanamine derivatives bearing an all-carbon quaternary stereo center by employing 3-substituted 1,4,2-dioxazol-5-ones as an amidating reagent to capture the transient C(sp(3))-Rh intermediate. Notably, by simply changing the directing group, a second, unsymmetrical ortho C-H amidation/annulation can be achieved to provide tricyclic dihydrofuro[3,2-f]quinazolinones in good yields.

Abstract:Synthetically important scaffolds, fused tricyclic frameworks containing a 2,3-cyclo[b]dihydrofuran unit, play a crucial role in drug discovery. In this study, we demonstrate that rhodium(II)/N-fluorobenzenesulfonimide can catalyze the in situ generation of highly reactive alkene intermediates from commonly accessible alkanes, which undergo intermolecular [3+2] tandem cyclization with the simultaneously generated beta-dicarbonyl radical to synthesize a series of fused tricyclic frameworks containing a 2,3-cyclo[b]dihydrofuran unit with a quaternary carbon center.

Abstract:Cp*Rh(III)-catalyzed chelation-assisted direct C-H bond functionalization of 1-(2-pyridyl)-2-pyridones with internal alkynes that can be controlled to give three different products in good yields has been realized. Depending on the reaction conditions, solvents and additives, the reaction pathway can be switched between alkenylation, alkenylation/directing group migration and rollover annulation. These reaction manifolds allow divergent access to a variety of valuable C6-alkenylated 1-(2-pyridyl)-2-pyridones, (Z)-6-(1,2-diaryl-2-(pyridin-2-yl)vinyl)pyridin-2(1H)-ones and 10H-pyrido[1,2-a][1,8]naphthyridin-10-ones from the same starting materials. These protocols exhibit excellent regio- and stereoselectivity, broad substrate scope, and good tolerance of functional groups. A combination of experimental and computational approaches have been employed to uncover the key mechanistic features of these reactions.
