Abstract:Metabolism and biological functions of the nitrogen-rich compound guanidine have long been neglected. The discovery of four classes of guanidine-sensing riboswitches and two pathways for guanidine degradation in bacteria hint at widespread sources of unconjugated guanidine in nature. So far, only three enzymes from a narrow range of bacteria and fungi have been shown to produce guanidine, with the ethylene-forming enzyme (EFE) as the most prominent example. Here, we show that a related class of Fe 2+ - and 2-oxoglutarate-dependent dioxygenases (2-ODD-C23) highly conserved among plants and algae catalyze the hydroxylation of homoarginine at the C6-position. Spontaneous decay of 6-hydroxyhomoarginine yields guanidine and 2-aminoadipate-6-semialdehyde. The latter can be reduced to pipecolate by pyrroline-5-carboxylate reductase but more likely is oxidized to aminoadipate by aldehyde dehydrogenase ALDH7B in vivo . Arabidopsis has three 2-ODD-C23 isoforms, among which Din11 is unusual because it also accepted arginine as substrate, which was not the case for the other 2-ODD-C23 isoforms from Arabidopsis or other plants. In contrast to EFE, none of the three Arabidopsis enzymes produced ethylene. Guanidine contents were typically between 10 and 20 nmol*(g fresh weight) -1 in Arabidopsis but increased to 100 or 300 nmol*(g fresh weight) -1 after homoarginine feeding or treatment with Din11-inducing methyljasmonate, respectively. In 2-ODD-C23 triple mutants, the guanidine content was strongly reduced, whereas it increased in overexpression plants. We discuss the implications of the finding of widespread guanidine-producing enzymes in photosynthetic eukaryotes as a so far underestimated branch of the bio-geochemical nitrogen cycle and propose possible functions of natural guanidine production.

Abstract:Guanidine is a chemically stable nitrogen compound that is excreted in human urine and is widely used in manufacturing of plastics, as a flame retardant and as a component of propellants, and is well known as a protein denaturant in biochemistry1-3. Guanidine occurs widely in nature and is used by several microorganisms as a nitrogen source, but microorganisms growing on guanidine as the only substrate have not yet been identified. Here we show that the complete ammonia oxidizer (comammox) Nitrospira inopinata and probably most other comammox microorganisms can grow on guanidine as the sole source of energy, reductant and nitrogen. Proteomics, enzyme kinetics and the crystal structure of a N. inopinata guanidinase homologue demonstrated that it is a bona fide guanidinase. Incubation experiments with comammox-containing agricultural soil and wastewater treatment plant microbiomes suggested that guanidine serves as substrate for nitrification in the environment. The identification of guanidine as a growth substrate for comammox shows an unexpected niche of these globally important nitrifiers and offers opportunities for their isolation.

Abstract:Nitrogen is an indispensable element for life and the material world. The development of efficient conversion strategies to transform dinitrogen gas into various valuable nitrogen-containing compounds is of great economic and scientific importance. The activation and transformation of dinitrogen molecule is an eternal topic in chemistry, and it is of profound significance to understand nitrogen fixation from the level of condensed matter chemistry. Several related examples have been illustrated here to discuss the effects of condensed matter phenomena in nitrogen fixation chemistry. Some critical scientific problems in the field are discussed from three aspects: nitrogen fixation in homogeneous solution, heterogeneous ammonia synthesis, and coupling multiple energy for N2 / O2 conversion. We hope this review will inspire more chemists to think about the fundamental nature of nitrogen fixation chemistry from the perspective of condensed matter chemistry, offering more ideas to solve the related problems.
Photochemical Tuning of Tricoordinated Nitrogen Deficiency in Carbon Nitride to Create Delocalized Π Electron Clouds for Efficient CO2 Photoreduction

Abstract:Precisely engineering point defects holds promise for the development of state-of-the-art photocatalysts for CO2 conversion. This study demonstrates the controllable creation of nitrogen vacancies (V-Ns) in the centers of heptazine rings of graphitic carbon nitrides (g-C3N4) via a photochemical-assisted nitrogen etching strategy. Spectroscopic analyses and theoretical simulations elucidate the photochemical process to hydrogenate the nitrogen situated at the center of the g-C3N4 heptazine ring and then release an ammonia molecule, accompanied by the photooxidation of the sacrificial agents. The catalyst with an optimal V-Ns concentration achieves a CO generation rate of 35.2 mu mol g(-1) h(-1) with nearly 100% selectivity, comparable to the performance of the reported g-C3N4 materials. The remarkably improved photoactivity is due to the adjustments of the electronic structures and the midgap states of g-C3N4 by the delocalized pi electron cloud created in the 12-membered ring surrounding the V-N, which maximizes the light-harvesting efficiencies and suppresses the recombination of photogenerated electrons and holes. The V-Ns also activates the neighboring catalytic carbon centers to reduce the energy barrier for CO2 reduction. This work provides a good design concept to regulate catalytic activity by engineering point defects.

Abstract:Abstract The element nitrogen and nitrogenous compounds are vital to life. The synthesis of nitrogen-containing compounds using dinitrogen as the nitrogen source, not through ammonia, is of great interest and great value but remains a grand challenge. Herein, we describe a strategy to realize this transformation by combining the heterogeneous approach with the homogeneous methodology. The N2 molecule was first fixed with carbon and LiH through a one-pot heterogeneous process, forming Li2CN2 as an ‘activated’ nitrogen source with high efficiency. Then subsequent homogeneous treatments of Li2CN2 to construct the organic synthon carbodiimide and the RNA/DNA building block pyrimidines were fulfilled. By using 15N2 as the feedstock, their corresponding 15N-labeled carbodiimide and pyrimidines were readily obtained. This homogeneous–heterogeneous synergy strategy will open a new chapter for N2 transformation.

Abstract:Described here is the de novo design and synthesis of a series of 6H-dipyrido[1,2-e:2',1'-i]purin-6-ones (DPs) as a new class of visible-light photoredox catalysts (PCs). The synthesized DP1-5 showed their λ Abs(max) values in 433-477 nm, excited state redox potentials in 1.15-0.69 eV and -1.41 to -1.77 eV (vs. SCE), respectively. As a representative, DP4 enables the productive guanylation of various amines, including 1°, 2°, and 3°-alkyl primary amines, secondary amines, aryl and heteroaryl amines, amino-nitrile, amino acids and peptides as well as propynylamines and α-amino esters giving diversities in biologically important guanidines and cyclic guanidines. The photocatalytic efficacy of DP4 in the guanylation overmatched commonly used Ir and Ru polypyridyl complexes, and some organic PCs. Other salient merits of this method include broad substrate scope and functional group tolerance, gram-scale synthesis, and versatile late-stage derivatizations that led to a derivative 81 exhibiting 60-fold better anticancer activity against Ramos cells with the IC50 of 0.086 μM than that of clinical drug ibrutinib (5.1 μM).

Abstract:Nitrogen-doped graphene oxide (NGO) was synthesized by irradiation of graphene oxide (GO) in NH3 atmosphere. NGO obtained by irradiation of GO for 10 min has high N content of 13.62 at. %. The photoluminescence (PL) properties of NGO were investigated. The results showed that compared with GO, NGO exhibits significant PL enhancement with a high enhancement ratio of approximately 1501.57%. It may attribute to the high content of amino-like N, which can effectively enhance PL of GO because of the amino conjugation effect.

Abstract:Excess and deficient nitrogen (N) inhibit photosynthesis in the leaves of rice plants, but the underlying mechanism is still unclear. N can improve the chlorophyll content and thus affect photon absorption, but the photosynthetic rate does not increase accordingly. To investigate this mechanism, three concentrations of N treatments were applied to two rice varieties, Zhefu802 and Fgl. The results indicated increased chlorophyll content of leaves with an increased N supply. Little discrepancy was detected in Rubisco enzyme activity and Non-photochemical quenching (NPQ) in the high nitrogen (HN) and moderate nitrogen (MN) treatments. The model that photoinhibition occurs in Zhefu802 due to a lack of balance of light absorption and utilization is supported by the higher malondialdehyde (MDA) content, higher H2O2 content, and photoinhibitory quenching (qI) in HN treatment compared with MN treatment. A lower proportion of N in leaf was used to synthesize chlorophyll for Fgl compared with Zhefu802, reducing the likelihood of photoinhibition under HN treatment. In conclusion, HN supply does not allow ideal photosynthetic rate and increases the likelihood of photoinhibition because it does not sustain the balance of light absorption and utilization. Apart from Rubisco enzyme activity, NPQ mainly contributes to the unbalance. These results of this study will provide reference for the effective N management of rice.

Abstract:Ru‐TiO2 is synthesized and employed as an effective catalyst for urea photo‐synthesis through a nitric acid‐mediated pathway combining nitrogen oxidation and subsequent kinetically advantageous nitrate and CO2 co‐reduction. This study presents groundbreaking insights for nitrogen oxidation activation and provides brand‐new ideas for the direct resource utilization of nitrogen. Photocatalytic urea synthesis, utilizing N2 and CO2 as feedstock and sustainable solar energy, represents an environmentally friendly and promising alternative strategy. However, the conventional unidirectional gas phase co‐reduction techniques typically employes pose stringent demands for the design of key catalysts, reaction control, mass transfer, and other aspects, due to the disparity in physical properties, structure, and catalytic kinetics between N2 and CO2. Herein, Ru‐TiO2 is synthesized and employed as an effective catalyst for urea photo‐synthesis through a nitric acid‐mediated pathway combining nitrogen oxidation and subsequent kinetically advantageous nitrate and CO2 co‐reduction. A urea yield of 24.95 μmol h−1 g−1cat. is obtained through an innovative intermittent illumination method, AQE values of 4.7% and 6.3% at 380 and 420 nm, respectively. The remarkable photo‐activity is attributed to its unique oxygen vacancy‐anchored Ru nanostructure (Ru‐O4Ti1), which effectively activates inert N2 molecules to minish the disparity of orbital energy levels, facilitates the formation of crucial *NN(OH) intermediates, and serves as an "electronic pump" to avoid electronegativity effect for facilitating electron transfer from nitrogen to TiO2 support for urea photosynthesis. This study presents groundbreaking insights for nitrogen oxidation activation and provides brand‐new ideas for the direct resource utilization of nitrogen.

Abstract:Graphitic carbon nitride (g-C3N4) is traditionally obtained by thermal polycondensation of nitrogen-rich organic nonionic precursors. Herein, we provide a new and simple strategy for the synthesis of g-C3N4 through direct thermal condensation of single protic guanidine salts with various anions. The thermal condensation process, and the structure and photophysical properties (optical properties, thermal stabilities, morphology and porosity, and photocatalytic activity) of the obtained g-C3N4 materials are investigated intensively through thermogravimetric analysis, powder X-ray diffraction, X-ray photoelectron spectroscopy, UV/Vis diffuse reflectance spectroscopy, nitrogen adsorption isotherms, and photodegradation of RhodamineB. The results reveal that during thermal condensation, the nitrogen-rich basic core in the guanidine salts serves as a carbon and nitrogen source to form the skeleton of g-C3N4, and the counter anions mediate the thermal condensation insitu and modify slightly the structure and properties of g-C3N4, even though they are not incorporated into the final structure. Among all g-C3N4 materials, including the pristine g-C3N4 from melamine, the g-C3N4 derived from guanidine thiocyanate exhibits a high activity toward the photocatalytic degradation of RhodamineB dye because of its relatively high surface area, enhanced absorption, small bandgap, and low photoluminescence intensity.

Abstract:Nitrate products are widely used in manufacturing as crucial raw materials and fertilizers. The traditional nitrate synthesis process involves high energy consumption and emission, thereby opposing the goals of zero-carbon emission and green chemistry. Thus, a sustainable roadmap for nitrate synthesis that uses green energy input, clean N sources, and direct catalytic processes is urgently required (e.g., developing a novel photosynthesis system). Here, we synthesized TiO2-supported atomically dispersed Cu species for N2 photofixation to nitrate in a flow reactor. The optimized photocatalyst yielded a high nitrate photosynthesis rate of 0.93 μmol h−1 and selectivity of ∼90%, which is superior to most of the values reported thus far. Further, experimental results and in-situ investigations revealed that the atomically dispersed Cu sites in the as-designed sample significantly enhanced the separation and transfer efficiency of photogenerated carriers, adsorption and activation of reactants, and the formation of chemisorbed NOx intermediates, thereby realizing the excellent photofixation of N2 to nitrate.

Abstract:Significant process has been made on the development of semi-artificial photosynthesis catering to the production of H 2 – and CO 2 – derived value-added products. But there are little reports on the application of semi-artificial photosynthetic system to nitrogen fixation areas. In this study we report the successful and highly efficient N 2 fixation in one step at room temperature and ambient pressure by constructing new semi-artificial photosynthetic systems based on material-cell hybrids and microbial photo-electrolysis cells (MPECs). Improved TiO 2 /Chlorophyll composite that annealed at 400 °C was selected as electron donator to replenish additional electrons to Nostoc commune Vauch microalgae cell for N 2 fixation. Ubiquinone-0 (2,3-dimethoxy-5-methyl-1,4-benzoquinone, Q 0 ) was selected as electron relay that guarantees efficient transmission of electrons from abiotic material to microalgae cells. Determined by an ammonia assay kit, at constant concentration of microalgae, the constructed material-cell hybrid system photosynthetically synthesized 40.67 μM/h ammonia, while H-shape MPECs synthesized 132.70 μM/h, 19.23 times higher than that produced by pure microalgae. Using 15 N 2 as the reduction gas, the constructed H-shape MPECs photosynthetically synthesized 28.37 μM/h 15 NH 4 + , 2.80 time higher than that of the MPECs without Q 0 as electron relay.

Abstract:Abstract Recent studies have revealed the prevalence and biological significance of guanidine metabolism in nature. However, the metabolic pathways used by microbes to degrade guanidine or mitigate its toxicity have not been widely studied. Here, via comparative proteomics and subsequent experimental validation, we demonstrate that Sll1077, previously annotated as an agmatinase enzyme in the model cyanobacterium Synechocystis sp. PCC 6803, is more likely a guanidinase as it can break down guanidine rather than agmatine into urea and ammonium. The model cyanobacterium Synechococcus elongatus PCC 7942 strain engineered to express the bacterial ethylene-forming enzyme (EFE) exhibits unstable ethylene production due to toxicity and genomic instability induced by accumulation of the EFE-byproduct guanidine. Co-expression of EFE and Sll1077 significantly enhances genomic stability and enables the resulting strain to achieve sustained high-level ethylene production. These findings expand our knowledge of natural guanidine degradation pathways and demonstrate their biotechnological application to support ethylene bioproduction.

Abstract:Nitrogen (N-2) is an essential element for life, but kinetically stable N-2 in the atmosphere needs to be reduced to biologically available forms as a nutrient for organisms. Abiotic nitrogen fixation is critical to the origin of life on the early Earth, which is due to lightning or mineral-based reduction. Here, synchrotron X-ray-induced silver nitrate formation on a silver copper (AgCu) thin-film is reported. Time-resolved X-ray diffraction measurements show that under intense X-ray exposure, initially formed silver oxides (AgOx) are quickly converted to silver nitrate (AgNO3). Interestingly, AgNO3 is first formed in its high-temperature phase with a space group of R3cH, which gradually transforms to the room temperature phase with a space group of Pbca under continuous X-ray irradiation. The result not only provides a new clue about the abiotic nitrogen reduction prior to life but also demonstrates a novel strategy of materials synthesis using synchrotron X-rays.

Abstract:A low-cost, widely available and environmentally benign compound guanidine hydrochloride was applied to prepare g-C3N4 by thermal-induced polymerization at different temperatures. The as-prepared g-C3N4 was thoroughly characterized by X-ray diffraction, Fourier transform infrared spectroscopy, elemental analyses, N-2 adsorption-desorption isotherms, thermal gravimetric analysis and differential scanning calorimetry, transmission electron microscopy, UV-vis diffuse reflectance spectra and photoluminescence spectra. It was noticeable that g-C3N4 prepared from guanidine hydrochloride possessed a much larger surface area than that prepared from melamine. And its photocatalytic activity was evaluated for degrading Rhodamine B dye which demonstrated that the g-C3N4 synthesized from guanidine hydrochloride exhibited a much higher activity than that prepared from melamine. Moreover, the activity can be further enhanced by increasing the condensation temperature of g-C3N4. In addition, the as-prepared g-C3N4 was also stable as demonstrated in the recycling experiments.

Abstract:Nonribosomal peptide synthetases (NRPSs) are modular enzymes that use a thiotemplate mechanism to assemble the peptide backbones of structurally diverse and biologically active natural products in bacteria and fungi. Unlike these canonical multi-modular NRPSs, single-module NRPS-like enzymes, which lack the key condensation (C) domain, are rare in bacteria, and have been largely unexplored to date. Here, we report the discovery of a gene cluster (gup) encoding a NRPS-like megasynthetase through genome mining. Heterologous expression of the gup cluster led to the production of two unprecedented alkaloids, guanipiperazines A and B. The NRPS-like enzyme activates two l-tyrosine molecules, reduces them to the corresponding amino aldehydes, and forms an unstable imine product. The subsequent enzymatic reduction affords piperazine, which can be morphed by a P450 monooxygenase into a highly strained compound through C-O bond formation. Further intermolecular oxidative coupling forming the C-C or C-O bond is catalyzed by another P450 enzyme. This work reveals the huge potential of NRPS-like biosynthetic gene clusters in the discovery of novel natural products.
High nitrogen-driven photosynthesis limitation in shade-demanding species mediated by stomatal conductance through plasma membrane intrinsic proteins

Abstract:Photosynthesis is susceptible to nitrogen (N) surplus. However, the mechanism of reduced photosynthesis by N surplus need to be further verified in the shade-demanding and high-N-sensitive species. Photosynthetic capacity, N allocation, stomatal parameters, the correlation between PIP (encoding plasma membrane intrinsic proteins) expression and total stomatal conductance (g t ) were analyzed in Panax notoginseng grown under the levels of low N (LN), moderate N (MN) and high N (HN). N content per unit leaf area (N area ) was increased with increasing N application. HN resulted in an increase in N allocation to the light capture system ( P L ) and a decrease in N allocation to the carboxylation system ( P C ). Net photosynthetic rate ( P n ), stomatal conductance (g s ), the maximum electron transport rate ( J max ) and the maximum carboxylation rate ( V cmax ) were lower in the LN and HN plants. Under HN condition, the limitation to stomatal conductance (S L ) and limitation to biochemical factors (B L ) were the main factors for reducing photosynthetic efficiency. The stomatal area index (SAI) was more closely correlated with stomatal density (SD) than with stomatal length (SS). g s was partially correlated with SS or SD. The correlation analysis showed that the relative expression of PIP1.3 , PIP1.4 and PIP2.7 is positively correlated with g t /N area . The results obtained suggest that high-N-driven limitation to photosynthesis in the shade-demanding and HN-sensitive medicinal crop, such as P. notoginseng , is mainly derived from the decrease in g s , and partly from the decrease in the carboxylation capacity of RuBP.

Abstract:Ammonia synthesis under mild conditions is of supreme interest. Photocatalytic nitrogen fixation with water at room temperature and atmospheric pressure is an intriguing strategy. However, the efficiency of this method has been far from satisfied for industrialization, mainly due to the sluggish cleavage of the N/N bond. Herein, we report a carbon-tungstic-acid (WO3 center dot H2O) hybrid for the co-optimization of N-2 activation as well as subsequent photoinduced protonation. Efficient ammonia evolution reached 205 mu mol g(-1)h(-1) over this hybrid under simulated sunlight. Nitrogen temperature-programmed desorption revealed the decisive role of carbon in N-2 adsorption. Photoactive WO3 . H2O guaranteed the supply of electrons and protons for subsequent protonation. The universality of carbon modification for enhancing the N-2 reduction was further verified over various photocatalysts, shedding light on future materials design for ideal solar energy utilization.
Enhanced Foliar 15N Enrichment with Increasing Nitrogen Addition Rates: Role of Plant Species and Nitrogen Compounds

Abstract:Determining the abundance of N isotope (delta N-15) in natural environments is a simple but powerful method for providing integrated information on the N cycling dynamics and status in an ecosystem under exogenous N inputs. However, whether the input of different N compounds could differently impact plant growth and their N-15 signatures remains unclear. Here, the response of N-15 signatures and growth of three dominant plants (Leymus chinensis, Carex duriuscula, and Thermopsis lanceolata) to the addition of three N compounds (NH4HCO3, urea, and NH4NO3) at multiple N addition rates were assessed in a meadow steppe in Inner Mongolia. The three plants showed different initial foliar delta N-15 values because of differences in their N acquisition strategies. Particularly, T. lanceolata (N-2-fixing species) showed significantly lower N-15 signatures than L. chinensis (associated with arbuscular mycorrhizal fungi [AMF]) and C. duriuscula (associated with AMF). Moreover, the foliar delta N-15 of all three species increased with increasing N addition rates, with a sharp increase above an N addition rate of similar to 10 g N m(-2) year(-1). Foliar delta N-15 values were significantly higher when NH4HCO3 and urea were added than when NH4NO3 was added, suggesting that adding weakly acidifying N compounds could result in a more open N cycle. Overall, our results imply that assessing the N transformation processes in the context of increasing global N deposition necessitates the consideration of N deposition rates, forms of the deposited N compounds, and N utilization strategies of the co-existing plant species in the ecosystem.
