Sodium iodide facilitated this transformation by generating iodine in situ which was found to accelerate this transformation. The use of a transition metal-free protocol, readily available reagents, and mild reaction conditions allowed this protocol to be easily scaled up.

Abstract:The copper-catalyzed sulfenylation of indoles with aryl iodide and sulfur powder has been investigated both experimentally and theoretically. This protocol provides a direct and facile approach to prepare 3-sulfenylindoles with moderate to excellent yields and good functional-group tolerance. The in-situ IR analysis provided evidence for that NaOAc could promote the synthesis of diphenyl disulfide by the coupling of aryl iodide and sulfur powder. According to DFT calculations, the coupling pathway involving the intermediate N-methyl-3-iodoindole is more favored than the direct coupling by the CH activation of indole. The N-methyl-3-iodoindole was identified as a crucial intermediate in the catalytic cycle.

Abstract:A new method for phosphine-mediated difluoromethylthiolation of indole derivatives and other electronrich aromatics including pyrroles, pyrazolones, indolizine, and 1,3,5-trimethoxybenzene was developed using difluoromethanesulfonyl chloride. The additive n-Bu4NI was demonstrated to facilitate this reaction by generating iodine in situ, which could promote this transformation. The use of a transition-metal- free protocol, readily available reagents, and mild reaction conditions makes this protocol more practical than traditional methods.

Abstract:Direct chloromethylthiolation of the C(sp2)‐H bonds of indole was achieved with the commonly used solvents dimethyl sulfoxide (DMSO) and dichloroethane (DCE). Initially, chlorodimethylsulfonium ion is generated in situ from the reaction of DMSO and DCE on simple heating at 120 °C and is used for electrophilic methylthiolation followed by chlorination, enabling the synthesis of 2‐chloro‐3‐methylthioindoles.

Abstract:3-Sulfanyloxindoles were synthesised by triphenylphosphine-mediated transition-metal-free thiolation of oxindoles using sulfonyl chlorides as sulfenylation reagents. The above reaction was promoted by iodide anions, which was ascribed to the in situ conversion of sulfenyl chlorides into the more reactive sulfenyl iodides. Moreover, the thiolation of 3-aryloxindoles was facilitated by bases. The use of a transition-metal-free protocol, readily available reagents, and mild reaction conditions make this protocol more practical for preparing 3-sulfanyloxindoles than traditional methods.

Abstract:A metal‐free and novel thiolation of indole with sulfinyl amides in the presence of tetrabutylammonium iodide (TBAI), affording 3‐indole thioethers in good to excellent yields. A mild, efficient and highly regioselective thiolation of indoles by sulfinyl amides via direct C−H functionalization is described. Sulfinyl amides could be reduced to thioethers and submitted as the thiolation reagents were developed. This reaction provides a useful method for the synthesis of 3‐indole thioethers and 2,3‐dithioindole derivatives. This mix‐and‐go approach also tolerates broad substrate scope and outstanding functional group compatibility. In addition, this preparation could be easily conducted on a gram scale with high efficiency.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:We report a base-promoted metal-free approach for the synthesis of thieno[2,3-b]indole derivatives. This method combined four C-H σ-bond cleavage reactions of two different kinds of C-H bonds and two C-S σ-bond formation processes. A series of thieno[2,3-b]indoles were obtained starting from 3-benzylindole derivatives with good yields and high regioselectivity, with the elemental sulfur serving as a cheap and readily available sulfur source. Good efficiency could be maintained even when the reaction was performed on a gram scale. A plausible mechanism was proposed on the basis of mechanistic studies.

Abstract:Triphenylphosphine-mediated metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols by CF3SO2Cl and CHF2SO2Cl to synthesize trifluoromethyl disulfides and difluoromethyl disulfides, respectively, was achieved at room temperature. Iodine generated in situ from iodide facilitated this reaction via the formation of iodotriphenylphosphonium iodide which could serve as a reducing agent in this transformation. Readily available reagents and mild reaction conditions without transition-metals allow this protocol to be more practical than traditional methods.

Abstract:An iron‐catalyzed direct C3‐H radical trifluoromethylthiolation of quinoxalin‐2(1H)‐ones has been developed. This protocol use earth‐abundant FeSO4·7H2O as the catalyst, and AgSCF3 as the trifluoromethylthiolating reagent to provide various SCF3‐containing quinoxalin‐2(1H)‐one derivatives in 26‐88% yields. Preliminary mechanistic investigations indicate that the reaction proceeds through a radical pathway.

Abstract:A novel method for the synthesis of 2-substituted 3-trifluoromethylselenoindole derivatives from readily accessible 2-trifluoromethylseleno ethynylanilines via zinc-mediated SeCF3 group migration is achieved. The tandem reaction starts from cyclization/nucleophilic addition, followed by an unprecedented trifluoromethylseleno group 1,2-migration and then electrophilic substitution. A zincoindole complex with Cindolyl/Otosyl chelation is successfully isolated as reactive intermediate and characterized by X-ray diffraction. Control experiments indicate that both of the nature of selenium atom and N-protecting groups play crucial role in the rearrangement reaction.

Abstract:An unprecedented electrochemically regioselective C–H phosphorothiolation and S- to C-[1,4] phosphoryl migration involving indolizines, elemental sulfur, and H-phosphonates in an undivided cell has been developed. This strategy utilizes elemental sulfur as an odorless sulfur source, providing a new avenue for the formation of C(sp 2 )–S–P bonds under sustainable conditions. Various phosphorothiolated and mercapto-phosphono substituted indolizines were obtained via controllable electrochemical phosphorothiolation and a 1,4-S → C phospho-Fries rearrangement. Additionally, this is the first example of successive electrochemical phosphorothiolation and a 1,4-S → C phospho-Fries rearrangement.

Abstract:Indoles as important N-heteroaromatic skeletons are widely used in the fields of pharmaceuticals, agrochemicals and biological sciences, but the N-trifluoromethyl indoles have been rarely explored. So far, the main accesses to N-trifluoromethyl indoles include oxidative desulfurization-fluorination of dithiocarbamates and Fisher indole synthesis via the reaction of N-CF3 hydrazine and ketones. We report herein the efficient straightforward strategies for the synthesis of N-trifluoromethyl indoles through desulfurization-fluorination/cyclization of 2-alkynylaryl isothiocyanates or 2-alkynylanilines under mild conditions. These cascade reactions possess good substrate scope and functional group compatibility, providing a variety of N-trifluoromethyl indoles in moderate to high yileds.
