Abstract:In this work, a dicyanoisophorone-based fluorophore, which showed an attractive near-infrared fluorescence as well as a favorable large stokes shift, was used to develop a novel probe (NIR-Probe 1) for effective detection of biothiols. It displayed distinct color changes and a turn-on NIR fluorescence response with a high off/on ratio toward biothiols. We demonstrated that NIR-Probe 1 exhibited fast sensing speed, high selectivity for biothiols and possessed the function of detecting low concentration of biothiols with impressive limit of detection: 12.1 nM for Cys, 14.5 nM for Hcy and 11.9 nM for GSH. Moreover, the utility of NIR-Probe 1 as a bioanalytical molecular tool was demonstrated by fluorescence imaging of biothiols in living cells.(C) 2017 Elsevier B.V. All rights reserved.

Abstract:In this study, a new near-infrared (NIR) fluorescent turn-on probe featuring a large Stokes shift (198 nm) was developed for the detection of biothiols. The probe was based on a dicyanoisophorone derivative serving as the fluorophore and a 2,4-dinitrobenzenesulfonyl (DNBS) group functioning as both a recognition site and a fluorescence quencher. In the absence of biothiols, the fluorescence of the probe was low due to the photoinduced electron transfer (PET) effect between the fluorophore and DNBS. Upon the presence of biothiols, the DNBS group underwent a nucleophilic aromatic substitution reaction with the sulfhydryl group of biothiols, leading to the release of the fluorophore and a notable emission peak at 668 nm. This developed probe exhibited exceptional selectivity and sensitivity to biothiols in solution, with an impressive detection limit of 28 nM for cysteine (Cys), 22 nM for homocysteine (Hcy), and 24 nM for glutathione (GSH). Furthermore, the probe demonstrated its applicability by successfully visualizing both endogenous and exogenous biothiols in living systems.

Abstract:In this work, a novel ratiometric fluorescence nanoprobe based on Au3+-triggered aggregation-induced emission (AIE) behavior of silver nanoclusters (Ag NCs) was designed for the label-free detection of biothiols. The probe was constructed by loading of Au3+ on g-C3N4 nanosheets surface and subsequently aggregating Ag NCs via ion binding. As Au3+ could conversely regulate the emission of g-C3N4 nanosheets and Ag NCs, the remove of Au3+ from nanoprobe by coordination with biothiols would change the emission ratio of nanoprobe that could be used for biothiols detection. The probe provided high sensitivity for glutathione (GSH) determination with the limit of detection as low as 0.8 μM and showed satisfying performance in human serum samples. This report may offer a new sight for the construction of ratiometric probe based on the AIE behavior of Ag NCs and broaden its applications in biosensing.

Abstract:Bio-thiols such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) play a crucial role in physiological processes. Distinguishing between Cys, Hcy, and GSH remains a challenge because of their similar structure and reactivity. Herein, we conceptualized and synthesized a near-infrared (NIR) fluorescent probe TAI based on acridine-conjugated indole salts. This innovative probe demonstrated effectiveness in sensitively distinguishing Cys, Hcy and GSH by NIR dual-channel and time-dependence fluorescence. Probe displays a significant turn-on NIR fluorescent response toward Cys at 654nm under 488nm excitation with Large Strokes shift (about 166nm) and high selectivity, but also shows turn-on NIR fluorescent response for Hcy/GSH at 695nm under 640nm excitation. More interestingly, the probe also exhibited an NIR fluorescence response at 654nm to Hcy and no fluorescence response to GSH when the response time is over 3h. The sensing mechanism was fully confirmed by NMR, HRMS spectra and DFT theoretical calculations. Moreover, successful intracellular imaging experiments in living cells suggest that the probe is suitable for discrimination imaging endogenous and exogenous bio-thiols. This work offers a promising near-infrared fluorescent imaging tool for the differential detection of bio-thiols.

Abstract:Near-infrared (NIR) fluorophores, especially dicyano-based fluorophores and xanthene-based hemicyanines, have been put high expectation in bioimaging application due to their excellent optical properties. However, they suffer from inherent shortages such as short excitation/emission wavelength (less than 700 nm) or small Stokes shift (20-50 nm). Herein, we constructed a novel NIR dicyano-based fluorophore (DCO-HBTN). To our knowledge, it is the first reported dicyano-based fluorophore of which the excitation/emission wavelength is more than 650 nm and Stokes shift is more than 100 nm. To demonstrate the feasibility of our efforts, we developed two NIR fluorescent probes (Probe-Cys and Probe-H2S) based on the fluorophore, Probe-Cys displayed good selective and highly sensitive (LOD = 0.28 mu M) recognition of Cys over Hcy and GSH, which was used to visualize endogenous Cys in tumor tissue. Probe-H2S exhibited an. excellent specific and sensitive (LOD = 0.11 mu M) response to H2S, which was applied in monitoring H2S releasing from the prodrug in vitro and in vivo.

Abstract:Two probes, structurally symmetric CBFB and asymmetric CBFM, constructed by a D–A–D (donor–acceptor–donor) type curcuminoid as the fluorophore and the DNBS (2,4-dinitrobenzenesulfonyl) group as the biothiol recognition site were designed and synthesized here. The DNBS group can quench the emission of the fluorophore by the PET (photoinduced electron transfer) process, and in the presence of biothiols, the emission of the probe was switched on as a result of the cleavage of the quencher by a nucleophilic aromatic substitution reaction. Experimental analyses and theoretical calculations revealed that two recognition moieties in the molecule can quench the fluorescence more efficiently, therefore, CBFB showed a much higher SNR (signal to noise ratio) than CBFM in biothiol detection with an emission maximum at 610 nm. This “low background” and “turn-on” fluorescent probe, CBFB, was successfully utilized to map endogenous biothiols in living cells.

Abstract:The well-known small-molecule biothiols have been used to maintain the normal metabolism of peroxy radicals, forming protein structures, resisting cell apoptosis, regulating metabolism, and protecting the homeostasis of cells in the organism. A large amount of research has found that abnormal levels of the above biothiols can cause some adverse diseases, such as changes in hair pigmentation, a slower growth rate, delayed response, excessive sleep and skin diseases. In order to further investigate the exact intracellular molecular mechanism of biothiols, it is imperative to explore effective strategies for real-time biothiol detection in living systems. In this work, a new near-infrared (NIR) emission fluorescence probe (probe 1) for sensitive and selective detection of biothiols was devised by combining dicyanoisophorone derivatives with the dinitrobenzenesulfonyl (DNBS) group. As expected, probe 1 could specifically detect biothiols (Cys, Hcy and GSH) through the dinitrobenzenesulfonyl group to form dye 2, which works as a signaling molecule for sensing biothiols in real samples. Surprisingly, probe 1 showed superior sensing characteristics and low-limit detection towards biothiols (36.0 nM for Cys, 39.0 nM for Hcy and 48.0 nM for GSH) with a large Stokes shift (134 nm). Additionally, the function of probe 1 as a platform for detecting biothiols was confirmed by confocal fluorescence imaging of biothiols in MCF-7 cells and zebrafish. More importantly, the capability of probe 1 in vivo has been further evaluated by imaging the overexpressed biothiols in tumor tissue. It is reasonable to believe that probe 1 can provide a valuable method to explore the relationship between biothiols and the genesis of tumor.

Abstract:Due to the redox properties closely related to numerous physiological and pathological processes, biothiols, including cysteine (Cys), homocysteine (Hcy) and glutathione (GSH), have received considerable attention in biological science. On account of the important physiological roles of these biothiols, it is of profound significance to develop sensitive and selective detection of biothiols to understand their biological profiles. In this work, we reported an efficient fluorescent probe, PHPQ-SH, for detecting biothiols in vitro and vivo, based on the phenothiazine-HPQ skeleton, with DNBS (2,4-dinitrobenzenesulfonate) as the response unit. Probe PHPQ-SH exhibited brilliant sensing performances toward thiols, including a large Stokes shift (138 nm), excellent sensitivity (for GSH, LOD = 18.3 nM), remarkable fluorescence enhancement (163-fold), low cytotoxicity, rapid response (8 min), and extraordinary selectivity. Finally, the probe PHPQ-SH illustrated herein was capable of responding and visualizing biothiols in MCF-7 cells and zebrafish.

Abstract:A new green-emitting fluorescent probe 1 was developed for biothiol detection. The sensing mechanism was considered to be biothiol-induced cleavage of the 2,4-dinitrobenzene- sulfonate group in probe 1 and resulting inhibition of the probe's photoinduced electron transfer (PET) process. Probe 1 exhibited favorable properties such as excellent selectivity, highly sensitive (0.12 mu M), large Stokes shift (117 nm) and a remarkable turn-on fluorescence signal (148-fold). Furthermore, confocal fluorescence imaging indicated that probe 1 was membrane-permeable and suitable for visualization of biothiols in living A549 cells.

Abstract:Biothiols, such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH), play essential roles in many physiological and pathological processes. In this work, three porphyrin derivatives, XQ1, XQ2 and XQ3, were synthesized by combining the porphyrin framework with nitroethylene, bis(methoxycarbonyl) ethylene and dicyanoethylene moieties, respectively. XQ1 was almost nonfluorescent, which could be utilized as a good starting point for developing fluorescence turn-on probes. Thus, XQ1 exhibits fast fluorescence enhancement and high selectivity towards the biothiols based on the Michael addition mechanism. It also exhibits high sensitivity towards the biothiols with the detection limits of 0.65 -1.1 mu M. In addition, XQ1 was successfully applied to cell imaging in living A549 cells for visualizing the biothiols. The results compose a good example of designing porphyrin derivatives for detecting biothiols, with the advantages of dramatic fluorescence enhancement in the NIR wavelength range, which exhibits good cell membrane permeability and practicability in detecting both exogenous and endogenous biothiols. Abstract:Biothiols are the main antioxidants in regulating the redox balance and resisting oxidative stress in various biological processes, but the long detection time of current fluorescent probes hinders their rapid imaging in vitro and in vivo. To reveal the influx of biothiols, we rationally develop an ortho -activation approach to accelerate the reaction between the probe and biothiols, by introducing electron-withdrawing fluorine atom into the ortho -site of the phenolic hydroxyl group in the NIR probe to generate an ortho -inductive effect. The ortho -fluorine helps to increase the chemical reactivity of the molecular structure, resulting in a significantly shorter detection time (within 5 min) as compared to previous reports (> 20 min for acrylates-based probes in aqueous solution). Based on this approach, our near-infrared probe 2F-RBX can sensitively and efficiently detect endogenous biothiols in living HepG2 cells and in vivo . These data suggest that ortho -activation is a simple and flexible approach to construct sensitive fluorescent probes for rapid imaging of biothiols, and perhaps other molecules in future, under biological circumstances.

Abstract:Near-infrared (NIR) fluorescent probes are attractive molecular tools for bioimaging because of their low autofluorescence interference, deep tissue penetration, and minimal damage to sample. However, most previously reported NIR probes exhibit small Stokes shift, typically less than 30 nm, and low fluorescence quantum yield, strictly limited contrast and spatial resolution for bioimaging. Herein, by expanding the π-conjugated system of rhodamine B, while, at the same time, keeping its rigid and planar structure, we reported an efficient NIR dye, HN7, with large stokes shift of 73 nm and fluorescence quantum yield as high as 0.72 in ethanol, values superior to those of such traditional cyanine NIR dyes as Cy5. Using HN7, living cells, tissues and mice were imaged, and the results showed significantly enhanced contrast, improved spatial resolution, and satisfactory tissue imaging depth when compared to Cy5. Moreover, the nonfluorescent spirocyclic structure of rhodamine B is an inherent component of HN7; therefore, our strategy provided a universal platform for the design of efficient NIR turn-on bioimaging probes for various targets. As a proof-of-concept, two different NIR probes, HN7-N2 and HN7-S for NO and Hg2+, respectively, were designed, synthesized, and successfully applied for the imaging of NO and Hg2+ in living cells, tissues and mice, respectively, demonstrating the potential bioimaging applications of the new probes. In sum, this new type of dye may present new avenues for the development of efficient NIR fluorescent probes for contrast-enhanced imaging in biological applications.

Abstract:A new probe based on the squaraine skeleton with a 2,4-dinitrobenzenesulfonyl unit is reported as a fluorescent probe for biothiol. It shows turn-on properties with red emission towards biothiols in acetonitrile-PBS buffer (1 : 9, v/v) solution, and its properties of rapid response, high selectivity and high sensitivity make it a potential probe with real applications. Moreover, the in vitro assays show that the probe can be used in fluorescence imaging for the detection of intracellular biothiol levels.

Abstract:The homeostasis of biothiols is closely related to the health of organisms. In view of the important role of biothiols, a fluorescent probe ( 7HIN-D ) for the detection of intracellular biothiols was developed based on a simple chalcone fluorophore 7HIN with "ESIPT + AIE" characteristics. The probe 7HIN-D was obtained by introducing a biothiols specific DNBS (2,4-dinitrobenzenesulfonyl) unit as a fluorescence quencher to the fluorophore 7HIN . The nucleophilic substitution reaction between biothiols and probe 7HIN-D will release the DNBS unit and the fluorophore 7HIN , which exhibits a "turn on" AIE fluorescence with a large Stokes shift of 113 nm. The probe 7HIN-D displays high sensitivity and good selectivity to biothiols, and the detection limits value of probe 7HIN-D for GSH, Cys and Hcy were 0.384 μmol/L, 0.471 μmol/L and 0.638 μmol/L, respectively. In addition, the probe has been successfully used for fluorescence detection of endogenous biothiols in living cells due to its excellent performance, good biocompatibility and low cytotoxicity.

Abstract:A fluorescein-based sensor bearing nitroolefin moiety for the detection of thiols was prepared. Upon addition of thiols, such as cysteine (Cys), homocysteine (Hcy) and glutathione, the solution containing sensor showed an increase of absorption intensity at 497 nm with a slight blue-shift as well as the enhancement in fluorescence intensity at 520 nm. The fluorescence enhancement and absorption change are attributed to the Michael addition of thiols to the double bond of nitroolefin moiety in sensor. The probe displays a fast response to Cys with the rate constant (k(obs)) of 2.3 min(-1) and the detection limit to Cys was evaluated to be 0.2 mu M. Abstract:Bio-thiols play an important role in living organisms. Both elevated levels and deficiency of bio-thiols will induce some diseases, such as cardiovascular disease and Alzheimer's diseases, skin lesions, and weakness. Therefore, fast and accurate detection of bio-thiols is of great significance. We have synthesized a ratiometric probe that can respond to bio-thiols with the fluorescence intensity at 568 nm (F-5(68)) increasing and taht at 420 nm (F-420) decreasing. The responding process reaches equilibration in 1 min in the present of Cys or Hcy while in about 50 min with GSH. This probe can further detect bio-thiols in human breast cancer cells (MDA-MB-231) by confocal laser scanning microscope. Fluorescent ratio (R-Y/B) of yellow channel with blue channel increase as reacting with bio-thiols in cells. Ratiometric probe SH-1 can realize the fast detecting of bio-thiols with R-Y/B as detection signal both in vitro and in living cells.

Abstract:A new N-butyl-4-amino-1,8-naphthalimide-based colorimetric and ratiometric fluorescent probe for the detection of biothiols (cysteine, homocysteine, and glutathione) was designed and synthesized. The probe exhibited good selectivity and sensitivity toward biothiols over other non-thiol-bearing amino acids and common anions with significant changes in both color (from colorless to yellow) and fluorescence (from blue to yellow-green). The mechanism is based on cleavage of disulfide by thiols followed by an intramolecular cyclization to give the product, N-butyl-4-amino-1,8-naphthalimide, which showed outstanding intramolecular charge transfer (ICT). Therefore, ratiometric fluorescence signal was observed. Furthermore, the probes were successfully applied for visualizing endogenous thiols in living cells.

In this work, we demonstrated a general strategy for the design of fluorescent probes for the detection of various biological related thiols. The preferred strategy we employed was to introduce a 3,5-dinitropyridyl moiety to a selected fluorophore via ether linkage to afford fluorescent probe. Once the ether bond was thiolyzed by biothiol, the hydroxy-functionalized fluorescent dye would be released and fluorescence emission restored. Herein, some novel fluorescent probes based on this strategy have been developed for sensing biothiols in living cells. All these probes presented here showed desirable properties, such as high sensitivity, high selectivity and preferred reaction kinetics. Moreover, the probes have been used for imaging of intracellular biothiols with emission colors ranging from green to near-infrared (NIR).
