Abstract:Surface enhanced Raman scattering (SERS) fiber sensors have shown great potential in sensitive biosensing and medical diagnostics. However, current SERS fiber probes are most commonly based on stiff silica fibers, which, unfortunately, are not mechanically compliant with soft biological tissues. In addition, the poor bio-compatibility of silica fibers sets another barrier that hinders their development for biomedical applications. Here, we present, to the best of our knowledge, the first demonstration of soft-polymer-optical-fiber-based SERS (SPOF-SERS) probes with physio-mechanical properties suitable for implantation, and demonstrate their potential applications for in situ detection of bioanalysts. The SPOFs are made from porous hydrogel materials that are soft, elastic, and biocompatible. The three-dimensional porous structures of the hydrogels enable high loading of metal nanoparticles to provide a large amount of SERS "hot spots" for high sensitivity. We tested the SPOF-SERS sensor for detection and discrimination of rhodamine 6G and 4-mercaptopyridine in situ with detection limits of 10(-7) M and 10(-8 )M, respectively. We also demonstrated the capability of SPOF-SERS probes in multiplexing detection. Abstract:The in vivo detection of biomarkers in a liquid environment is very important for the early diagnosis of diseases. Spectroscopy methods are employed in ultraviolet-visible-infrared wavelengths, fluorescence or Raman spectra are detected for clinical diagnose. The dual-mode image can provide more diagnostic information and has been realized in some research work. However, there is still lacking simple and sensitive dual-mode sensors to satisfy the in vivo detecting demands. In this paper, a dual-mode fiber sensor for Surface-enhanced Raman Scattering (SERS) and fluorescence detection is proposed. The sensor is formed by a tapered optical fiber, half of the fiber tip surface is coated with Ag nanoparticles. In the detection of Rhodamine 6G (R6G) aqueous solution, the minimum detectable concentrations in SERS and fluorescence tests are of the same order of magnitude. By combining the Raman spectral features and the fluorescence intensity, the recognition and quantitation of target molecules were obtained reliably. It is the first time, to our knowledge, that the Raman-fluorescence dual-mode detection is realized in one single fiber, which was manufactured with micro-machinery techniques. It is a label-free, general-purpose fiber sensor, which can be applied for liquid biopsy, helping to diagnose and treat diseases in vivo.
Uniaxially Stretched Flexible Surface Plasmon Resonance Film for Versatile Surface Enhanced Raman Scattering Diagnostics.

Abstract:Surface-enhanced Raman scattering (SERS) spectroscopy affords a rapid, highly sensitive, and nondestructive approach for label-free and fingerprint diagnosis of a wide range of chemicals. It is of great significance to develop large-area, uniform, and environmentally friendly SERS substrates for in situ identification of analytes on complex topological surfaces. In this work, we demonstrate a biodegradable flexible SERS film via irreversibly and longitudinally stretching metal deposited biocompatible poly(ε-caprolactone) film. This composite film after stretching shows surprising phenomena: three-dimensional and periodic wave-shaped microribbons array embedded with a high density of nanogaps functioning as hot-spots at an average gap size of 20 nm and nanogrooves array along the stretching direction. The stretched polymer surface plasmon resonance film gives rise to more than 10 times signal enhancement in comparison with that of the unstretched composite film. Furthermore, the SERS signals with high uniformity exhibit good temperature stability. The polymer SPR film with excellent flexibility and transparency can be conformally attached onto arbitrary nonplanar surfaces for in situ detection of various chemicals. Our results pave a new way for next-generation flexible SERS detection means, as well as enabling its huge potentials toward green wearable devices for point-of-care diagnostics.

Abstract:Surface Plasmon Resonance (SPR) is a powerful technique for directly sensing in biological studies, chemical detection and environmental pollution monitoring. In this paper, we present polymer optical fiber application in SPR sensors, including wavelength interrogation SPR sensor and surface enhanced Raman scattering (SERS) probe. Polymer optical fiber (POF) is used to effectively couple the incident light from light source, and collect reflective light into spectrometer. It also makes the sensing system more flexible and reliable. Gold nanorods with 30 nm long axis and average aspect ratios 3:1 are synthesized using seed-mediated growth method. Combining Au nanorods with POF, we can easily realize fiber SERS sensor. Long-period fiber gratings are fabricated on single mode polymer optical fiber with 120 um period and 50% duty cycle. The polarization characteristic of this kind of birefringent grating is studied. Theoretical analysis shows this type grating has two separated polarization peaks, and it will be advantageous in SPR sensing applications.

Abstract:This paper presents a sensor that leverages a cloverleaf hollow fiber (CHF) for optofluidic surface-enhanced Raman spectroscopy (SERS) detection within the fiber. Unlike other microstructured fibers, this design features four air holes, providing a larger contact area between the test liquid and the enhanced substrate at the fiber core, which simplifies liquid injection. By taking advantage of this unique structure, silver nanoparticles were modified inside the fiber to serve as the Raman enhancement substrate. The Raman signals are excited via the evanescent field, enabling highly sensitive SERS detection. Rhodamine 6G (R6G) was used as the target analyte, achieving a detection limit of 10 -11 M, an enhancement factor (EF) of 1.59×10 8 , and a relative standard deviation (RSD) of 4.8%, demonstrating both high sensitivity and reproducibility in fiber-based detection. Additionally, SERS detection of enrofloxacin solution was carried out, further validating the fiber’s capability in detecting antibiotic substances with excellent stability. The successful detection achieved with this fiber underscores its significant potential for applications in biomedical, agricultural, food safety, and environmental monitoring.

Abstract:To fabricate a new surface-enhanced Raman scattering (SERS)-active optical fiber sensor, the design and preparation of SERS-active sensing layer is one of important topics. In this study, we fabricated a highly sensitive three-dimensional (3D) SERS-active sensing layer on the optical fiber terminal via in situ polymerizing a porous polymer material on a flat optical fiber terminal through thermal-induced process, following with the photochemical silver nanoparticles growth. The polymerized polymer formed a 3D porous structure with the pore size of 0.29–0.81μm, which were afterward decorated with abundant silver nanoparticles with the size of about 100nm, allowing for higher SERS enhancement. This SERS-active optical fiber sensor was applied for the determination of 4-mercaptopyridine, crystal violet and maleic acid The enhancement factor of this SERS sensing layer can be reached as about 108. The optical fiber sensor with high sensitive SERS-active porous polymer is expected for online analysis and environment detection.

Abstract:Surface enhanced Raman spectroscopy(SERS) can largely boost the Raman signal, and thus broaden the manifold potential applications of SERS. According to the current limitations on generality and efficiency, this work designed and developed a transparent, flexible and freestanding substrate and its preparation, conservation and operation procedures. The substrate consists of a polymer supporter and embedded metal nanostructures, which can be used for direct and on-line measurements of samples in various morphologies and circumstances. The detection limit of a flexible SERS(Ag) substrate is lower than 1 pmol/L.

Abstract:In this paper, a surface-enhanced Raman scattering (SERS) spectral probe based on a polymethylmethacrylate (PMMA) polymer fiber is produced by femtosecond laser ablation and photoreduced deposition of silver nanoparticles. It shows that a surface grating structure enabled by femtosecond laser ablation could further improve the SERS signal by around four times larger than the ordinarily roughened structure. By varying femtosecond laser pulse energy and scan surface period in the fabrication process, the enhancement factor of the SERS signal is optimized with laser scan surface period of around 5 μm at 1.2 μJ femtosecond laser pulse energy for a PMMA fiber probe. In particular biochemical analysis, much softer SERS probes are preferred, and our polymer SERS fiber probe shows an alternative method for in situ measurement of the SERS signal and further fabrication of polymer SERS microchip.

Abstract:Optical fiber Raman and surface-enhanced Raman scattering (SERS) probes hold great promise for in vivo biosensing and in situ monitoring of hostile environments. However, the silica Raman scattering background generated within the optical fiber increases in proportion to the length of the fiber, and it can swamp the signal from the target analyte. While filtering can be applied at the distal end of the fiber, the use of bulk optical elements has limited probe miniaturization to a diameter of 600 µm, which in turn limits the potential applications. To overcome this limitation, femtosecond laser micromachining was used to fabricate a prototype micro-optical filter, which was directly integrated on the tip of a 125 µm diameter double-clad fiber (DCF) probe. The outer surface of the microfilter was further modified with a nanostructured, SERS-active, plasmonic film that was used to demonstrate proof-of-concept performance with thiophenol as a test analyte. With further optimization of the associated spectroscopic system, this ultra-compact microprobe shows great promise for Raman and SERS optical fiber sensing.

Abstract:A combined taper-and-cylinder optical fiber fabricated by simple tube-etching and modified with silver nanoparticles is developed for surface-enhanced Raman scattering (SERS) sensing. It has the advantages of high light transmission efficiency and large interaction areas for light and silver nanoparticles. The detection of rhodamine 6G in remote mode indicates that the sensitivity could realize 10−13 M. The fiber SERS probe with high flexibility and sensitivity shows great potential for molecule detection in various sensing applications.

Abstract:Remote sensing remains a challenge due to its demand for high sensitivity, convenient sampling and rapid response time. Surface enhanced Raman scattering (SERS) spectroscopy is a powerful analytical method for the detection of various samples. Here, aiming at increasing the sensitivity, a novel strategy for the preparation of a SERS probe is demonstrated by using hollow optical fiber tips decorated by layer-by-layer assembly of two kinds of nanoparticles. Specifically, Au@Ag core-shell nanorods and Ag nanospheres with opposite surface charge were assembled layer-by-layer on the tip of hollow optical fibers through electrostatic interaction. Then, much more hotspots are generated due to the close gap between the nanorods and nanospheres in the resultant 3D structure, which can lead to a dramatically enhanced SERS activity of the probe compared with that fabricated by pure silver sphere nanoparticles or nanorods. On the other hand, taking the advantages of the vibration spectroscopic fingerprints property of SERS spectra and the long-distance communication capacity of optical fibers, the remote online detection of biological species including proteins, funguses and cells can be easily achieved within a few minutes. Therefore, such a novel kind of optical fiber-SERS sensor holds great potential for the rapid detection of a wide range of samples due to its superiority of simplicity and high sensitivity.

Fiber Sensors for Molecular Detection

Abstract:The demand on sensors for detecting chemical and biological agents is greater than ever before, including medical, environmental, food safety, military, and security applications. At present, most detection or sensing techniques tend to be either non-molecular specific, bulky, expensive, relatively inaccurate, or unable to provide real time data. Clearly, alternative sensing technologies are urgently needed. Recently, we have been working to develop a compact fiber optic surface enhanced Raman scattering (SERS) sensor system that integrates various novel ideas to achieve compactness, high sensitivity and consistency, molecular specificity, and automatic preliminary identification capabilities. The unique sensor architecture is expected to bring SERS sensors to practical applications due to a combination of 1) novel SERS substrates that provide the high sensitivity and consistency, molecular specificity, and applicability to a wide range of compounds; 2) a unique hollow core optical fiber probe with double SERS substrate structure that provides the compactness, reliability, low cost, and ease of sampling; and 3) an innovative matched spectral filter set that provides automatic preliminary molecule identification. In this paper, we will review the principle of operation and some of the important milestones of fiber SERS sensor development with emphasis on our recent work to integrate photonic crystal fiber SERS probes with a portable Raman spectrometer and to demonstrate a matched spectral filter for molecule identification.
SERS Substrate Based on the Flexible Hybrid of Polydimethylsiloxane and Silver Colloid Decorated with Silver Nanoparticles

Abstract:Various flexible SERS sensors have attracted widespread concern in performing the direct identification of the analytes adsorbed on arbitrary surfaces. Here, a sample method was proposed to integrate plasmonic nanoparticles into polydimethylsiloxane (PDMS) to fabricate flexible substrate for the decoration of silver nanoparticles (AgNPs). The flexible SERS sensor based on AgNPs/AgNPs-PDMS offers highly sensitive Raman detection with enhancement factor up to 8.3 × 109, which can be attributed to the integrative effects from both the increase of the light absorption of the embedded AgNPs in PDMS substrate and the EM enhancement from the adjacent top-top, bottom-bottom and top-bottom AgNPs. After undergoing the cyclic mechanical deformation, the SERS substrate still maintains high mechanical stability and stable SERS signals. However, upon stretching the flexible substrate, there was an amusing phenomenon that SERS signals can be highly increased, which results from that the reduction of lateral nanogaps between top and bottom of the PDMS boundary strengthens the trigger of the plasmon coupling as demonstrated by the simulated result. This result reveals that the tuning and the coupling of the electromagnetic fields can be effectively controlled by the macroscopic mechanical solicitation. That will have an important significance for practical applications in strain-dependent sensors and detectors.

Abstract:Surface-enhanced Raman scattering （SERS） provides important applications in diverse fields.In order to achieve high-precision SERS detection of trace molecules,current research focuses on how to increase the density of hot spots and the number of analyte molecules in the detection area.An ultrafast laser can rapidly construct large-area micro/nano-structures on material surfaces.It is important for the commercial preparation of highperformance SERS sensors.In this paper,the ultrafast laser preparation of high-performance SERS sensors is introduced from the aspect of the density of hot spots and the number of analyte molecules in the detection region.Ultrafast lasers enable both"bottom-up"and"top-down"processing.In particular,the superhydrophobic surface prepared by the ultrafast laser is one of the most effective methods to achieve the enrichment of analyte molecules.Finally,a prospect for the development of laser-prepared SERS substrates is provided.
Simple and Low‐Cost Plasmonic Fiber‐Optic Probe as SERS and Biosensing Platform

Abstract:As an emerging and promising paradigm of nanophotonic "lab‐on‐a‐chip" devices, plasmonic fiber‐optic (FO) probes with nanopatterns suffer from high cost and a complex fabrication process, which keep them from becoming the preferred choice for most optical fiber sensing applications. In this paper, a nanopatterned FO probe is demonstrated to be a surface‐enhanced Raman spectroscopy substrate and a plasmonic biosensor through theoretical simulations and experiments. The probe is assembled with imprinted nanocavities on the tip of the optical fiber and fabricated through a low‐cost and facile method. The FO probe overcomes the shortcomings of conventional optical fiber sensors that have nanoscale patterns and structures, such as requiring a complex and time‐consuming procedure, expensive specialized instruments, and experienced professional operation. Through investigating its surface sensitivity and the plasmonic coupling effect between Au nanoparticles and the nanocavity, this probe is found to have practical applications in the detection of biochemical molecules. The proposed FO probe can be produced under common experimental conditions, and its operation is simple and understandable, which serves as a new strategy for fabricating nanostructures on fiber tips.

Lab-on-Fiber Sensors with Ag/Au Nanocap Arrays Based on the Two Deposits of Polystyrene Nanospheres

Abstract:Surface-enhanced Raman spectroscopy (SERS) can boost the pristine Raman signal significantly which could be exploited for producing innovative sensing devices with advanced properties. However, the inherent complexity of SERS systems restricts their further applications in rapid detection, especially in situ detection in narrow areas. Here, we construct an efficient and flexible SERS-based Lab-on-Fiber (LOF) sensor by integrating Ag/Au nanocap arrays obtained by Ag/Au coating polystyrene nanospheres on the optical fiber face. We obtain rich "hot spots" at the nanogaps between neighboring nanocaps, and further achieve SERS performance with the assistance of laser-induced thermophoresis on the metal film that can achieve efficiency aggregation of detected molecules. We achieve a high Raman enhancement with a low detection limitation of 10−7 mol/L for the most efficient samples based on the above sensor. This sensor also exhibits good repeatability and stability under multiple detections, revealing the potential application for in situ detection based on the reflexivity of the optical fiber.

Towards Development of Fiber-Optic Surface Enhanced Raman Spectroscopy Probes Using 2-Photon Polymerization for Rapid Detection of Bacteria

Abstract:In this study, a variety of direct laser written surface-enhanced Raman spectroscopy (SERS) micro-structures, designed for bacteria detection, are presented. Various SERS micro-structures were designed to achieve both a high density of plasmonic hot spots and a strong probability of interaction between the hot spots and the target bacterial cells. Twophoton polymerization was used for initial fabrication of the polymeric skeletons of the SERS micro-structures, which were then coated with a 50 nm-thick gold layer via e-beam evaporation. The micro-structures were fabricated on glass coverslips and were assessed using a confocal Raman microscope. To this end, Rhodamine 6G was used as an analyte under 785 nm laser illumination. The optimal SERS micro-structures showed approximately 7×103 enhancement in Raman signal (analytical enhancement factor, AEF) at a wavenumber of 600 cm-1. Real-time detection of E. coli in solution was demonstrated using the fabricated SERS platform with low laser powers and a short acquisition time (785 nm, 5 mW, 50 ms).

Abstract:As a powerful and effective analytical tool, surface-enhanced Raman scattering (SERS) has attracted considerable research interest in the fields of wearable flexible sensing and non-invasive point-of-care testing (POCT) medical diagnosis. In this mini-review, we briefly summarize the design strategy, the development progress of wearable SERS sensors and its applications in this field. We present SERS substrate analysis of material design requirements for wearable sensors and highlight the benefits of novel plasmonic particle-in-cavity (PIC)-based nanostructures for flexible SERS sensors, as well as the unique interfacial adhesion effect and excellent mechanical properties of natural silk fibroin (SF) derived from natural cocoons, indicating promising futures for applications in the field of flexible electronic, optical, and electrical sensors. Additionally, SERS wearable sensors have shown great potential in the fields of different disease markers as well as in the diagnosis testing for COVID-19. Finally, the current challenges in this field are pointed out, as well as the promising prospects of combining SERS wearable sensors with other portable health monitoring systems for POCT medical diagnosis in the future.

Abstract:A broad spectral surface enhanced Raman scattering sensor is developed using the solid core holey photonic crystal fiber with silver nanoparticles cluster. This SERS probe offers an operational excitation wavelength range overlaying visible light and near infrared light. The PCF SERS sensing is demonstrated in the detection of the 4-Mercaptobenzoic acid (10−6 M) solution with 514.5 and 785 nm excitation. In this structure of PCF sensor, the related analysis shows that leakage modes also make an important contribution in the SERS activity not only by the evanescent field way.
Surface-Enhanced Raman Scattering in Silver-Coated Suspended-Core Fiber

Abstract:In this paper, the silver-coated large-core suspended-core fiber (LSCF) probe was fabricated by the dynamic chemical liquid phase deposition method for surface-enhanced Raman scattering (SERS) sensing. The 4-mercaptophenylboronic acid (4-MPBA) monolayer was assembled in the LSCF as the recognition monolayer. Taking advantage of the appropriate core size of the LSCF, a custom-made Y-type optical fiber patch cable was utilized to connect the semiconductor laser, Raman spectrometer, and the proposed fiber SERS probe. The SERS signal is propagated in the silver-coated air channels, which can effectively reduce the Raman and fluorescence background of the silica core. Experiments were performed to measure the Raman scattering spectra of the 4-MPBA in the silver-coated LSCF in a non-enhanced and enhanced case. The experiment results showed that the Raman signal strength was enhanced more than 6 times by the surface plasmon resonance compared with the non-enhanced case. The proposed LSCF for SERS sensing technology provides huge research value for the fiber SERS probes in biomedicine and environmental science. The combination of SERS and microstructured optical fibers offers a potential approach for SERS detection
