Abstract:With the intended application of engineered nanomaterials (ENMs) in agriculture, accurate assessment the effect of these ENMs on soil microbial communities is especially necessary. Here, maize plants were cultivated in soil amended by SiO2, TiO2, and Fe3O4 ENMs (100 mg kg-1 soil) for four weeks. The impact of ENMs on bacterial community structure of the rhizosphere soil was investigated by using high-throughput sequencing. In addition, metabolites of maize rhizosphere soil were quantified by gas chromatography-mass spectrometry (GC-MS) based metabolomics. We found that the disturbance of ENMs on soil microbes are in the follow of Fe3O4>TiO2>SiO2. Exposure of Fe3O4 ENMs significantly reduced the abundance of nitrogen-fixation related bacteria Bradyrhizobiaceae (from 2.94% to 2.40%) and iron-redox bacteria Sediminibacterium (from 2.15% to 2.07%). Additionally, Fe3O4 ENMs significantly increased populations of Nocardioides (from 1.63% to 1.77%), Chitinophaga sancti (from 1.12% to 2.08%), Pantoea (from 1.31% to 2.22%), Rhizobiumand (from 1.41% to 1.74%) and Burkholderia-Paraburkholderia (from 1.50% to 2.09%), which are associated with carbon cycling and plant growth promoting. This study provides a perspective on the response of rhizosphere microbial community and low molecular weight metabolites to ENMs exposure, providing a comprehensive understanding of the environmental risk of ENMs.

Abstract:Metal(loid) contaminants in food pose a global health concern. This study offers a global analysis of the impact of nanomaterials (NMs) on crop responses to metal(loid) stresses. Our findings reveal that NMs have a positive effect on the biomass production of staple crops (22.8%), while showing inhibitory effects on metal(loid) accumulation in plants (-38.3%) and oxidative damage (-21.6%) under metal(loid) stress conditions. These effects are influenced by various factors such as NM dose, exposure duration, size and composition. Here we introduce a method using interval-valued intuitionistic fuzzy values by integrating the technique for order preference by similarity to an ideal solution and entropy weights to compare the effectiveness of different NM application patterns. These results offer practical insights for the application of NMs in similar multi-criteria decision-making scenarios, contributing to sustainable agriculture and global food safety.

Abstract:Microorganisms colonize plant-associated environments and constitute complex communities aided in key functions for nutrient acquisition, disease suppression and abiotic stress resistance. In this study, we evaluated the variation of root-associated microbiomes of two typical farmland crops, maize (Zea mays L.) and soybean (Glycine max L. Merr.) respond to organochlorine pesticide stress, taking lindane as an example. Results showed that there were promoted but different attenuation rates of residual lindane in rhizosphere soils during maize and soybean growth, and the differential is due to the comprehensive effects of plant characters and microbial activities. Organochlorine pollution did not have significant impact on the microbial diversity and populations in all rhizo-compartments, but mostly stimulated the microbial connectivity. The multistep and decreasing processes for root-associated microbiomes of both maize and soybean were spatially different and mainly dependent on the shaping roles of host plants. These results expand our understandings of the organochlorine influence on the underground ecological system in crop-dependent soils.

Abstract:The rapid development of nanotechnology makes the environmental impact assessment a necessity to ensure the sustainable use of engineered nanomaterials. Here, silver nanoparticles (AgNPs) at 100 mg/kg were added to soils in the absence or presence of cucumber (Cucumis sativa) plants for 60 days. The response of the soil microbial community and associated soil metabolites was investigated by 16S rRNA gene sequencing and gas chromatography-mass spectrometry (GC-MS)-based metabolomics, respectively. The results show that AgNP exposure significantly increased the soil pH in both unplanted and cucumber-planted soils. The soil bacterial community structure was altered upon Ag exposure in both soils. Several functionally significant bacterial groups, which are associated with carbon, nitrogen, and phosphorus cycling, were compromised by AgNPs in both unplanted and cucumber-planted soils. Generally, plants played a limited role in mediating the impact of AgNPs on the bacterial community. Soil metabolomic analysis showed that AgNPs altered the metabolite profile in both unplanted and cucumber-planted soils. The significantly changed metabolites are involved in sugar and amino acid-related metabolic pathways, indicating the perturbation of C and N metabolism, which is consistent with the bacterial community structure results. In addition, several fatty acids were significantly decreased upon exposure to AgNPs in both unplanted and cucumber-planted soils, suggesting the possible oxidative stress imposed on microbial cell membranes. These results provide valuable information for understanding the biological and biochemical impact of AgNP exposure on both plant species and on soil microbial communities; such understanding is needed to understand the risk posed by these materials in the environment.
Metabolomic analysis reveals dose-dependent alteration of maize (Zea mays L.) metabolites and mineral nutrient profiles upon exposure to zerovalent iron nanoparticles

Abstract:Nanoscale zero-valent iron (nZVI) has been widely applied in the environmental field to degrade organic pollutants. The potential risk posed from nZVI on crop species is not well understood and is critical for sustainable application in the future. In this study, maize (Zea mays L.) plants were cultivated in field soils mixed with nZVI at 0, 50, and 500 mg/kg soil for four weeks. Upon exposure to 500 mg/kg nZVI, ICP-MS results showed that Fe accumulated by roots and translocated to leaves was increased by 36% relative to untreated controls. At 50 mg/kg, root elongation was enhanced by 150–200%; at 500 mg/kg, pigments, lipid peroxidation, and polyphenolic levels in leaves were increased by 12, 87 and 23%, respectively, whereas the accumulation of Al, Ca, and P were decreased by 62.2%, 19.7%, and 13.3%, respectively. A gas chromatography–mass spectrometry (GC–MS) based metabolomics analysis of maize roots revealed that antioxidants and stress signaling-associated metabolites were downregulated at 50 mg/kg, but were upregulated at 500 mg/kg. At 50 mg/kg, the content of glutamate was increased by 11-fold, whereas glutamine was decreased by 99% with respect to controls. Interestingly, eight metabolic pathways were disturbed at 50 mg/kg, but none at 500 mg/kg. This metabolic reprogramming at the lower dose represented potential risks to the health of exposed plants, which could be particularly important although no phenotypic impacts were noted. Overall, metabolites analysis provides a deeper understanding at the molecular level of plant response to nZVI and is a powerful tool for full characterization of risk posed to crop species as part of food safety assessment.

Abstract:Soil microbiome is the key driver mediating soil P transformation in agroecosystems. However, the underlying genomic information related to soil P cycling in response to organic C inputs is largely unknown. By using metagenomic sequencing, we investigated the effect of P fertilization and C input (i.e., glucose and lignin) on functional profiles of microbial genes related to P cycling in bulk and rhizosphere soils. Abstract:Metabolomics is an emerging tool to understand the potential implications of nanotechnology, particularly for agriculture. Although molybdenum (Mo) is a known plant micronutrient, little is known of its metabolic perturbations. Here, corn and wheat seedlings were exposed to MoO3 nanoparticles (NPs) and the corresponding bioavailable Mo6+ ion at moderate and excessive levels through root exposures. Physiologically, corn was more sensitive to Mo, which accumulated up to 3.63 times more Mo than wheat. In contrast, metabolomics indicated 21 dysregulated metabolites in corn leaves and 53 in wheat leaves. Five more metabolomic pathways were perturbed in wheat leaves compared to corn leaves. In addition to the overall metabolomics analysis, we also analyzed individual metabolite classes (e.g., amino acids, organic acids, etc.), yielding additional dysregulated metabolites in plant tissues: 7 for corn and 7 for wheat. Most of these were amino acids as well as some sugars. Additional significantly dysregulated metabolites (e.g., asparagine, fructose, reduced glutathione, mannose) were identified in both corn and wheat, due to Mo NP exposure, by employing individual metabolite group analysis. Targeted metabolite analysis of individual groups is thus important for finding additional significant metabolites. Abstract:Based on previously published hydroponic plant, planktonic bacterial, and soil microbial community research, manufactured nanomaterial (MNM) environmental buildup could profoundly alter soil-based food crop quality and yield. However, thus far, no single study has at once examined the full implications, as no studies have involved growing plants to full maturity in MNM-contaminated field soil. We have done so for soybean, a major global commodity crop, using farm soil amended with two high-production metal oxide MNMs (nano-CeO 2 and -ZnO). The results provide a clear, but unfortunate, view of what could arise over the long term: ( i ) for nano-ZnO, component metal was taken up and distributed throughout edible plant tissues; ( ii ) for nano-CeO 2 , plant growth and yield diminished, but also ( iii ) nitrogen fixation—a major ecosystem service of leguminous crops—was shut down at high nano-CeO 2 concentration. Juxtaposed against widespread land application of wastewater treatment biosolids to food crops, these findings forewarn of agriculturally associated human and environmental risks from the accelerating use of MNMs.
Metabolomics of Soybean (glycine Max L.) Response to Co-Exposure of Pyrene and Three Metal Oxide Engineered Nanomaterials

Abstract:Metal oxide engineered nanomaterials (ENMs) offer a range of opportunities for sustainable agriculture. Despite ENMs acting as vectors of nutrients to plants, it remains unclear whether they promote the uptake of contaminants, such as polycyclic aromatic hydrocarbons (PAHs), by plants. The present work revealed that ENMs (SiO2 nanoparticles, nSiO2; CeO2 nanoparticles, nCeO2; 100 mg L-1) significantly enhanced the accumulation of Pyr by 18.4-34.8% in soybean (Glycine max L.) tissues over 15-d hydroponic cultivation. Metabolomics analyses suggested the metabolic disturbance occurred in both Pyr-only exposure (910.3 +/- 87.6 mg kg-(1) Pyr) and the co -exposure of Pyr and ENMs (804.5 +/- 52.1 - 1226.6 & PLUSMN; 12.0 mg kg? 1 Pyr). In addition, higher Pyr contents induced more notable reprogramming of multiple metabolic processes in soybean, such as C/N cycle. Further statistical analyses indicated that more than 70% metabolic permutations were induced by Pyr, which was more profound than ENMs. The results in this study highlight the potential risks and specific molecular mechanisms of emergent ENMs if co-exposure with ubiquitous PAHs. This study also promotes further research into the responsible and sustainable development of nanotechnology in the agriculture.

Assessing the Impacts of Cu and Mo Engineered Nanomaterials on Crop Plant Growth Using a Targeted Proteomics Approach

Abstract:In this study, we investigated the effects of molybdenum (Mo)-based nanofertilizer and copper (Cu)-based nanopesticide exposure on wheat through a multifaceted approach, including physiological measurements, metal uptake and translocation analysis, and targeted proteomics analysis. Wheat plants were grown under a 16 h photoperiod (light intensity 150 μmol·m-2·s-1) for 4 weeks at 22 °C and 60% humidity with 6 different treatments, including control, Mo, and Cu exposure through root and leaf. The exposure dose was 6.25 mg of element per plant through either root or leaf. An additional low-dose (0.6 mg Mo/plant) treatment of Mo through root was added after phytotoxicity was observed. Using targeted proteomics approach, 24 proteins involved in 12 metabolomic pathways were quantitated to understand the regulation at the protein level. Mo exposure, particularly through root uptake, induced significant upregulation of 16 proteins associated with 11 metabolic pathways, with the fold change (FC) ranging from 1.28 to 2.81. Notably, a dose-dependent response of Mo exposure through the roots highlighted the delicate balance between nutrient stimulation and toxicity as a high Mo dose led to robust protein upregulation but also resulted in depressed physiological measurements, while a low Mo dose resulted in no depression of physiological measurements but downregulations of proteins, especially in the first leaf (0.23 < FC < 0.68) and stem (0.13 < FC < 0.68) tissues. Conversely, Cu exposure exhibited tissue-specific effects, with pronounced downregulation (18 proteins involved in 11 metabolic pathways) particularly in the first leaf tissues (root exposure: 0.35 < FC < 0.74; leaf exposure: 0.49 < FC < 0.72), which indicated the quick response of plants to Cu-induced stress in the early stage of exposure. By revealing the complexities of plants' response to engineered nanomaterials at both physiological and molecular levels, this study provides insights for optimizing nutrient management practices in crop production and advancing toward sustainable agriculture.

Abstract:A staggering number of nanomaterials-based products are being engineered and produced commercially. Many of these engineered nanomaterials (ENMs) are finally disposed into the soil through various routes in enormous quantities. Nanomaterials are also being specially tailored for their use in agriculture as nano-fertilizers, nano-pesticides, and nano-based biosensors, which is leading to their accumulation in the soil. The presence of ENMs considerably affects the soil microbiome, including the abundance and diversity of microbes. In addition, they also influence crucial microbial processes, such as nitrogen fixation, mineralization, and plant growth promoting activities. ENMs conduct in soil is typically dependent on various properties of ENMs and soil. Among nanoparticles, silver and zinc oxide have been extensively prepared and studied owing to their excellent industrial properties and well-known antimicrobial activities. Therefore, at this stage, it is imperative to understand how these ENMs influence the soil microbiome and related processes. These investigations will provide necessary information to regulate the applications of ENMs for sustainable agriculture and may help in increasing agrarian production. Therefore, this review discusses several such issues.

Abstract:Currently, some concerns regarding the potential toxicity of nanoparticles (NP) on the environment have emerged. The effect of ZnO, TiO2, and Fe2O3 NP on corn (Zea mays L.), common beans (Phaseolus vulgaris L.), nanobioremediation of polycyclic aromatic hydrocarbons (PAH), and soil organisms from agricultural or forest soils was studied at laboratory, greenhouse, and land level. The samples were analyzed by X-ray diffraction (XRD), field emission scanning electron microscopy with X-ray energy dispersion spectrometry (FESEM-EDS), scanning electron microscopy with energy-dispersive X-ray spectroscopy (SEM-EDS) gas chromatography (GC), ultra-performance liquid chromatography coupled to mass spectrometry (UPLC-MS) and Fourier transform infrared spectrometry with attenuated total reflectance (FTIR-ATR). ZnO-NP did not harm the mycorrhizal root colonization but, the presence of ZnO-NP decreased the degradation of PAH. The synthesis of metabolites from corn was more affected by the PAH than by ZnONP. FTIR spectra showed that NP affected the synthesis of compounds from specific functional groups in common bean plants. Fe2O3-NP were attached to the body of forestsoil organisms and significantly increased the concentration of Fe in their body, while TiO2-NP changed the morphological tissue of roots and stems of common bean as witnessed by micrographs of longitudinal and cross-sections. The NP used in this research significantly changed some response variables on the experiments carried-out at laboratory, greenhouse, and land level.

Abstract:Properly understanding the fundamental interactions between engineered nanoparticles (NPs) and plants is crucial for nano-enabled agriculture. In this study, Fe and Fe3O4 (magnetite), which are naturally occurred nanosized crystals and minerals, were foliar applied to 4-week-old maize plants for 10 days to evaluate their impact on plant photosynthesis and growth. Hill reaction of isolated maize leaf chloroplasts was carried out to determine the performance of two Fe-based NPs on photosynthetic activities at cell level. Meanwhile, gas chromatography-mass spectrometry (GC-MS) based metabolomics was used to explore the deep insight into the interaction between Fe-based NPs and maize plants. Results showed that maize leaf net photosynthesis rate and chlorophyll content were significantly increased by Fe NPs for 19.9% and 19.3%; and Fe3O4 NPs for 27.5% and 26.1%, respectively. Accordingly, plant biomass has been significantly increased by Fe and Fe3O4 NPs by 31.8% and 34.6%, respectively. Metabolomics revealed that both Fe-based NPs induced metabolic reprogramming in maize leaves. The biosynthesis of some compatible solutes and antioxidant compounds were inhibited. In addition, exposure to Fe-based NPs tentatively shut down some energy consuming pathways, such as photorespiration, alanine metabolism, branch chainamino acid biosynthesis. The trade-off of energy consuming pathways might be alternative explanation for the enhanced photosynthesis. The results of this study exhibited the promising potential for Fe-based NPs to be used in nano-enabled agriculture to promote plant growth. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Due to their unique properties, copper-based nanopesticides are emerging in the market. Thus, understanding their effect on crop plants is very important. Metabolomics can capture a snapshot of cellular metabolic responses to a stressor. We selected maize and cucumber as model plants for exposure to different doses of Cu(OH)2 nanopesticide. GC-TOF-MS-based metabolomics was employed to determine the metabolic responses of these two species. Results revealed significant differences in metabolite profile changes between maize and cucumber. Furthermore, the Cu(OH)2 nanopesticide induced metabolic reprogramming in both species, but in different manners. In maize, several intermediate metabolites of the glycolysis pathway and tricarboxylic acid cycle (TCA) were up-regulated, indicating the energy metabolism was activated. In addition, the levels of aromatic compounds (4-hydroxycinnamic acid and 1,2,4-benzenetriol) and their precursors (phenylalanine, tyrosine) were enhanced, indicating the activation of shikimate-phenylpropanoid biosynthesis in maize leaves, which is an antioxidant defense-related pathway. In cucumber, arginine and proline metabolic pathways were the most significantly altered pathway. Both species exhibited altered levels of fatty acids and polysaccharides, suggesting the cell membrane and cell wall composition may change in response to Cu(OH)2 nanopesticide. Thus, metabolomics helps to deeply understand the differential response of these plants to the same nanopesticide stressor.
Combined metagenomic and metabolomic analyses reveal that Bt rice planting alters soil C-N metabolism

Abstract:The environmental impacts of genetically modified (GM) plants remain a controversial global issue. To address these issues, comprehensive environmental risk assessments of GM plants is critical for the sustainable development and application of transgenic technology. In this paper, significant differences were not observed between microbial metagenomic and metabolomic profiles in surface waters of the Bt rice (T1C-1, the transgenic line) and non-Bt cultivars (Minghui 63 (the isogenic line) and Zhonghua 11 (the conventional japonica cultivar)). In contrast, differences in these profiles were apparent in the rhizospheres. T1C-1 planting increased soil microbiome diversity and network stability, but did not significantly alter the abundances of potential probiotic or phytopathogenic microorganisms compared with Minghui 63 and Zhonghua 11, which revealed no adverse effects of T1C-1 on soil microbial communities. T1C-1 planting could significantly alter soil C and N, probably via the regulation of the abundances of enzymes related to soil C and N cycling. In addition, integrated multi-omic analysis of root exudate metabolomes and soil microbiomes showed that the abundances of various metabolites released as root exudates were significantly correlated with subsets of microbial populations including the Acidobacteria, Actinobacteria, Chloroflexi, and Gemmatimonadetes that were differentially abundant in T1C-1 and Mnghui 63 soils. Finally, the potential for T1C-1-associated root metabolites to exert growth effects on T1C-1-associated species was experimentally validated by analysis of bacterial cultures, revealing that Bt rice planting could selectively modulate specific root microbiota. Overall, this study indicate that Bt rice can directly modulate rhizosphere microbiome assemblages by altering the metabolic compositions of root exudates that then alters soil metabolite profiles and physiochemical properties. This study unveils the mechanistic associations of Bt plant-microorganism-environment, which provides comprehensive insights into the potential ecological impacts of GM plants.

Abstract:Information about changes in physiological and agronomic parameters through the life cycle of plants exposed to engineered nanoparticles (NPs) is scarce. In this study, corn (Zea mays) plants were cultivated to full maturity in soil amended with either nCeO2 or nZnO at 0, 400, and 800 mg/kg. Gas exchange was monitored every 10 days, and at harvest, bioaccumulation of Ce and Zn in tissues was determined by ICP-OES/MS. The effects of NPs exposure on nutrient concentration and distribution in ears were also evaluated by ICP-OES and μ-XRF. Results showed that nCeO2 at both concentrations did not impact gas exchange in leaves at any growth stage, while nZnO at 800 mg/kg reduced net photosynthesis by 12%, stomatal conductance by 15%, and relative chlorophyll content by 10% at day 20. Yield was reduced by 38% with nCeO2 and by 49% with nZnO. Importantly, μ-XRF mapping showed that nCeO2 changed the allocation of calcium in kernels, compared to controls. In nCeO2 treated plants, Cu, K, Mn, and Zn were mainly localized at the insertion of kernels into cobs, but Ca and Fe were distributed in other parts of the kernels. Results showed that nCeO2 and nZnO reduced corn yield and altered quality of corn.
Zinc Oxide Nanoparticle Exposure Triggers Different Gene Expression Patterns in Maize Shoots and Roots.

Abstract:The potential impacts of environmentally accumulated zinc oxide nanoparticles (nZnOs) on plant growth have not been well studied. A transcriptome profile analysis of maize exposed to nZnOs showed that the genes in the shoots and roots responded differently. Although the number of differentially expressed genes (DEGs) in the roots was greater than that in the shoots, the number of up- or down-regulated genes in both the shoots and roots was similar. The enrichment of gene ontology (GO) terms was also significantly different in the shoots and roots. The "nitrogen compound metabolism" and "cellular component" terms were specifically and highly up-regulated in the nZnO-exposed roots, whereas the categories "cellular metabolic process", "primary metabolic process" and "secondary metabolic process" were down-regulated in the exposed roots only. Our results revealed the DEG response patterns in maize shoots and roots after nZnO exposure.
