Abstract:Nowadays, more and more studies have linked the abnormal expression of active molecules in organelles with the occurrence of diseases, so there is an urgent need to develop tools for detecting active molecules in specific organelles. However, the recognition receptors of most organelle-targeting probes currently developed always remain active, which easily causes them to react with the analyte in the cytoplasm, thus misjudging the role of the analyte in the physiological and pathological processes. Therefore, it is of great significance to develop a new strategy for the design of probes capable of high-fidelity imaging of the analyte in specific organelles. Herein, we propose a new strategy that the activation of recognition receptors that can be triggered by the microenvironment of targeting organelles. Based on this strategy, we develop a novel lysosome-targeting fluorescent probe (Lyso-SO2) for imaging of sulfur dioxide (SO2) with high-fidelity in lysosomes. The inert probe is activated by the acidic environment in the lysosome and then responds quickly (<2 s) and sensitively (LOD = 0.34 μM) to SO2. This paradigm by taking full advantage of the features of the organelle microenvironment provides a promising methodology for developing organelle-targeting probes for high-fidelity imaging.

Abstract:The sulfur dioxide (SO 2 )-associated dynamic interplay between mitochondria (Mito) and endoplasmic reticulum (ER) is essential for intracellular homeostasis regulation. Visualizing such organelles interactions can help elucidate basic pathogenesis. However, it is hampered by challenges in developing quick response, remarkable sensitivity and high selectivity within a single fluorescent probe for simultaneous and distinct tracking of their crosstalk. Herein, we engineered a SO 2 -responsive Mito-ER dual-targeted fluorescent probe (MER-SO 2 ) for dual-color visualization of Mito (red fluorescence) and ER (blue fluorescence) in living cells and in vivo. Under physiological conditions, the benzopyranium cationic structure in MER-SO 2 could target Mito, following fast response to SO 2 (5 s), which converted to a neutral structure, targeting to ER emitting blue fluorescence via functional fluorescence conversion. Probe MER-SO 2 also displayed high sensitivity, favourable specificity (against 24 types of non-targeting analytes). Importantly, such a high-performance probe allowed in-situ and dynamic tracking of Mito-ER interactions in Mito-dysfunction living cells and zebrafish via co-localization. Furthermore, the probe enabled accurate detection of sulphite in food with recoveries rate (96–100%), and incorporation into an agarose-based portable device for SO 2 assay. Taken together, this initial SO 2 -responsive probe represents a promising tool for discriminative tracking of functional Mito-ER interactions in living systems and holds great potential in many other fields such as food engineering.

Abstract:Reactive sulfur species (RSS) include sulfur dioxide (SO2) and various biothiols, such as hydrogen sulfide (H2S), glutathione (GSH), cysteine (Cys), and homocysteine (Hcy). These RSS are closely related to each other through complicated symbiotic networks, and play indispensable roles in multiple pathophysiological processes. Due to their similarity in chemical properties, it is challenging for developing a single fluorescent probe to differentiate them simultaneously. Herein we report the first mitochondria-targetable fluorescent probe NIR-NBD for simultaneous discrimination of Cys/Hcy/H2S, GSH, and SO2 from each other. The probe is characterized of the presence of three different types of electrophilic sites, and displays multiple sets of signal pattern in response to Cys/Hcy/ H2S, GSH, and SO2, including red-green for Cys/Hcy/H2S, red for GSH, and green for SO2. Notably, the probe exhibits excellent selectivity, outstanding sensitivity (detection limit = 3.5 nM) and fast response (within 4 min) toward SO2. Furthermore, the probe has been successfully applied for monitoring mitochondrial SO2 and various biothiols, visualizing the endogenous generation of SO2, and probing into the role of biothiols during apoptosis process.
A remarkable membrane-permeable fluorescent probe for real-time imaging of mitochondrial SO2 with high fidelity during ferroptosis

Abstract:Mitochondrial sulfur dioxide (SO2) plays a double-edged role in cells, and the real-time and in situ tracing of its dynamic behaviors to elucidate its complicated functions in detail is of great significance. Here, we developed a simple mitochondria-targeted fluorescent probe ZW for tracing SO2 with good membrane permeability. In probe ZW, the 1-phenylpyrrolidine-decorated benzopyrylium unit is employed as the selective response site for SO2. Besides, it also acts as the main fluorophore for signal conversion. The spectral results displayed that ZW could emit near-infrared (NIR) fluorescence (670 nm) and has a highly sensitive and selective response to SO2 (LOD = 0.19 μM). For biological imaging, compared with the control probe ZE, concentration- and time-dependent results verified that probe ZW has remarkable cell delivery with low concentration (200 nM) and fast response time (3 min). Furthermore, the NIR emission of ZW rendered high-fidelity imaging in living cells. Owing to its positive charge, ZW showed favorable mitochondria-targeting properties by colocalization experiments. Probe ZW could detect SO2 in real-time and in situ with high photostability in cells. Significantly, it has the ability to monitor the changes of endogenous SO2 during ferroptosis.

Abstract:SO2 and its derivatives play an important role in the antioxidation and anticorrosion of food and medicine. In biological systems, abnormal levels of SO2 lead to the occurrence of many biological diseases. Hence, the development of suitable tools for monitoring SO2 in mitochondria is beneficial for studying the biological effect of SO2 in subcellular organelles. In this research, DHX-1 and DHX-2 are fluorescent probes designed on the basis of dihydroxanthene skeletons. Importantly, DHX-1 (650 nm) and DHX-2 (748 nm) show near-infrared fluorescence response toward endogenous and exogenous SO2, which showed advantages of great selectivity, good sensitivity and low cytotoxicity, and the detection limit is 5.6 μM and 4.08 μM of SO2, respectively. Moreover, DHX-1 and DHX-2 realized SO2 sensing in HeLa cells and zebrafish. Moreover, cell imaging demonstrated that DHX-2 with a thiazole salt structure possesses good mitochondria-targeting ability. Additionally, DHX-2 was perfectly achieved by in situ imaging of SO2 in mice.

Abstract:Bioactive small molecules, hypochlorous acid (HClO) and sulfur dioxide (SO2), play vital roles in the maintenance of normal physiological function of living organisms. Once their balance is interrupted, it would cause oxidative stress and various diseases. Therefore, monitoring the change of HClO and SO2 in cellular levels and in vivo is of great significant for exploring their physiological and pathological roles. Herein, we present a dual-site and dual-turn-on fluorescence probe PTBI that could simultaneously monitor HClO and SO2 in mitochondria by two well-separated emission channels with a difference of 115 nm. The probe exhibited fast response (HClO: < 5 s, SO2: <1 min) and low detection limits with 45.8 and 53.2 nM for HClO and SO2, respectively. Using the single fluorescence molecule, we not only realized the detection of the basal and exogenous/endogenous levels of HClO and SO2, but also achieved the monitoring mitochondrial oxidative stress induced by HClO in living cells and zebrafish.

Abstract:The level of sulfur dioxide (SO2) and viscosity in mitochondria play vital roles in various of physiological and pathological processes. Abnormalities in mitochondrial SO2 and viscosity are closely associated with numerous biological diseases. It is of great significance to develop novel fluorescence probe for simultaneous detection of SO2 and viscosity within mitochondria. Herein, we have developed a water-soluble, mitochondrial-targeted and near-infrared fluorescent probe, CMBT, for the simultaneous detection of SO2 and viscosity. The probe CMBT incorporates benzothiazolium salt as a mitochondrial targeting moiety and 7-diethylaminocoumarin as a rotor for viscosity detection, respectively. Based on the prompt reaction between nucleophilic HSO3−/SO32− and the backbone of benzothiazolium salt derivative, probe CMBT displayed high sensitivity and selectivity toward SO2 with a limit of detection as low as 0.17 μM. As viscosity increased, the twisted intramolecular charge transfer (TICT) process was restricted, resulting in fluorescence emission enhancement at 690 nm. Moreover, probe CMBT demonstrated exceptional mitochondrial targeting ability and was successfully employed to image variation of SO2 and viscosity in living cells and mice. The work highlights the great potential of the probe as a convenient tool for revealing the relationship of SO2 and viscosity in biological systems.

Abstract:Similar to nitric oxide (NO) molecule, carbon monoxide (CO) molecule and hydrogen sulfide (H 2 S) molecule, sulfur dioxide (SO 2 ) is also considered to be another important endogenous signaling molecule. SO 2 derivatives are widely used as food antioxidants and antimicrobial agents. However, SO 2 is also a toxic environmental pollutant and intake of elevated levels of sulfites can lead to diarrhea, low blood pressure and allergic and asthmatic reactions in susceptible people. The specific physiological effects and transformation processes of SO 2 and its derivatives are not well understood due to the limitations of existing detection methods. Therefore, it is an urgent need to develop highly sensitive, high-resolution, real-time in situ assays. Herein, we specifically designed a ratiometric near-infrared fluorescent probe CQT for detecting SO 2 derivatives. After the reaction of CQT with HSO 3 − /SO 3 2− , a gradual decrease in red fluorescence at 662 nm accompanied by an enhanced blue fluorescence at 489 nm, reaching a detection range of 2.8–80 μM. In addition, we found that CQT had high energy transfer efficiency, large Stokes shift and low detection limit, which meant that CQT could sensitively monitor small changes of SO 2 concentration in cells and accurately target lysosomes.

Abstract:Analysis of subcellular organelles (e.g., a cytoplasm membrane and mitochondria) during cellular processes can provide particularly useful information for our understanding of cell chemistry and biology. For this purpose, fluorescent probes capable of dynamically imaging multiple organelles in a simultaneous and selective manner are highly demanded, yet such probes are scarcely reported due to the challenges in molecular design. In this study, we developed a dual-colored aggregation-induced emission (AIE) probe TPNPDA-C12 with twisted intramolecular charge transfer (TICT) to visualize the membrane and mitochondria of the same cells through distinct fluorescence channels simultaneously. We also successfully used the probe to monitor and distinguish the dynamic changes of the organelles during cell apoptosis and necrosis induced by reactive oxygen species (ROS) and cytotoxins.

Abstract:Background: Mitochondria are the main sites of reactive sulfur species (RSS) production in living cells. RSS in mitochondria play an important role in physiological and pathological processes of life. In this study, a dual-labeling probe that could simultaneously label the mitochondrial membrane and matrix was designed to quantitatively detect RSS of mitochondria in living cells using nano-level super-resolution imaging. Methods: A fluorescent probe CPE was designed and synthesized. The cytotoxicity of CPE was determined and co-localization of CPE with a commercial mitochondrial probe was analyzed in HeLa cells. Then, the uptake patterns of CPE in HeLa cells at different temperatures and endocytosis levels were investigated. The staining characteristics of CPE under different conditions were imaged and quantitated under structured illumination microscopy. Results: A fluorescence probe CPE reacting to RSS was developed, which could simultaneously label the mitochondrial membrane with green fluorescence and the mitochondrial matrix with red fluorescence. CPE was able to demonstrate the mitochondrial morphology and detect the changes of RSS in mitochondria. With the increase of mitochondrial RSS concentration, the light of the red matrix will be quenched. Conclusion: CPE provides a strategy for the design of probes and an attractive tool for accurate examination to changes of mitochondrial morphology and RSS in mitochondria in living cells at the nanoscale.

Abstract:In this report, a mitochondria-targeted probe based on an efficient platform constructed by pyridinium-coumarin and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF) with high selectivity and sensitivity toward SO2 derivatives was presented. As far as we know, pyridinium-coumarin is first used as a donor moiety to construct a FRET system. Compared with our previous work, it showed better solubility, higher biocompatibility and faster response speed. It also presented low detection limit (80.5 nM), excellent energy transfer efficiency (98.2%) and low cytotoxicity. Moreover, the probe could detect exogenous and endogenous bisulfite in living cells as well as in mitochondria successfully.

Abstract:A dual-response fluorescence probe (1), namely 2-(3-cyano-5,5-dimethyl-4-{2-[1-(2-morpholin-4-yl-ethyl)-2-oxo-1,2-dihydro-quinolin-3-yl]-vinyl}-5H-furan-2-ylidene)-malononitrile, was synthesized for detecting SO2 derivatives and viscosity by integrating 3-formyl-2-quinolinone with a terminal morpholine and 2-dicyanomethylene-3-cyano-4,5,5-trimethyl-2,5-dihydrofuran (TCF). This probe shows a ratiometric response towards HSO3− with highly selective and sensitive response (detection limit 0.37 μM), fast response (within 30 s), and a wide working pH range (4.0–11.0). Also, the probe exhibited a significant fluorescence enhancement response to viscosity. Importantly, the as-synthesized probe was successfully applied to image HSO3− and viscosity in lysosome of living HeLa cells lysosome and in zebrafish. In addition, the probe was also used to monitor HSO3− in real water samples.

Abstract:Hydrogen sulfide (H2S) is a versatile modulator in mitochondria and involved in numerous diseases caused by mitochondrial dysfunction. Therefore, many efforts have been made to develop fluorescent probes for mitochondrial H2S detection. However, these cationic small molecule probes are inapplicable for in vivo imaging because of the shallow tissue penetration and poor biostability. Herein, a ratiometric upconversion luminescence nanoprobe with the acid-activated targeting strategy is developed for detecting and bioimaging of mitochondrial H2S. The merocyanine TPAMC modified upconversion nanophosphors, acting as the targeting and response component, are encapsulated into a pH-sensitive husk, composed of DSPE-PEG and poly(L-histidine)-b-PEG, which improved the nanoprobe's stability during transport in vivo. Under lysosomal pH, the PEG shell is interrupted and the targeting sites are exposed to further attach to mitochondria. Taking advantage of the LRET process between TPAMC and UCNPs, ratiometric detection of mitochondrial H2S can be achieved with high selectivity and sensitivity. Cellular testing reveals the precise targeting to mitochondria via a lysosme delivery process. Importantly, the nanoprobe can be used for monitoring of mitochondrial H2S levels in living cells and colon cancer mouse models.

Abstract:Characterizing the long-term nanometer-scale interactions between lysosomes and mitochondria in live cells is essential for understanding their functions but remains challenging due to limitations of the existing fluorescent probes. Here, we develop cell-permeable organic fluorescent probes for lysosomes with excellent specificity and high photostability. We also use an existing Atto 647N dye with high brightness and excellent photostability to achieve specific labeling of mitochondria in live cells. Using these probes, we obtain dual-color structured illumination microscopy (SIM) images of dynamic physical lysosome-mitochondrion interactions in live cells at an ~90-nm resolution over a long time course of ~13 min. We successfully record the consecutive dynamic processes of lysosomal fusion and fission, as well as four types of physical lysosome-mitochondrion interactions by super-resolution imaging. Our probes provide an avenue for understanding the functions and the dynamic interplay of lysosomes and mitochondria in live cells.

Abstract:SO2 derivatives maintain a certain balance and play an important role in many metabolic processes. However, excessive ingestion of them can lead to serious complications of various diseases. Therefore, a rapid and precision detection of SO2 derivatives with high selectivity and sensitivity would be an advance for their bio-analytic studies. Accordingly, a novel deep red and two-photon fluorescent SO2 derivatives probe (DRQ) was developed here for the first time. The probe showed fast ratio response rate (within 5 s), excellent sensitivity (the detection limit is 103 nM for red channel and 17 nM for green channel), and outstanding selectivity toward SO2 derivatives in 10 mM PBS buffer. Moreover, the ratiometric probe DRQ displays a 99 nm blue-shift in emission upon addition of SO2 derivatives. Intriguingly, the probe can be made into test papers for real-time monitoring of SO2 derivatives. Moreover, it can be also applied for visual ratio imaging of SO2 derivatives in cellular mitochondria and zebrafish under two-photon absorption (green channel) and one-photon absorption (red channel).

Abstract:Abstract Mitochondria–lysosome interactions are essential for maintaining intracellular homeostasis. Although various fluorescent probes have been developed to visualize such interactions, they remain unable to label mitochondria and lysosomes simultaneously and dynamically track their interaction. Here, we introduce a cell-permeable, biocompatible, viscosity-responsive, small organic molecular probe, Coupa, to monitor the interaction of mitochondria and lysosomes in living cells. Through a functional fluorescence conversion, Coupa can simultaneously label mitochondria with blue fluorescence and lysosomes with red fluorescence, and the correlation between the red–blue fluorescence intensity indicates the progress of mitochondria–lysosome interplay during mitophagy. Moreover, because its fluorescence is sensitive to viscosity, Coupa allowed us to precisely localize sites of mitochondria–lysosome contact and reveal increases in local viscosity on mitochondria associated with mitochondria–lysosome contact. Thus, our probe represents an attractive tool for the localization and dynamic tracking of functional mitochondria–lysosome interactions in living cells.

Abstract:We developed a new mitochondria-targeted ratiometric fluorescent probe (named PPA) for SO2 derivatives (HSO3-/SO32-) with a large (pseudo) Stokes shift (209 nm) based on FRET platform in which water-soluble (E)-4-(4-(dimethylamino)styryl)-1-alkylpyridin-1-ium was chosen as acceptor moiety and phenothiazine oxide as donor moiety. Probe PPA showed specific response toward HSO3-/SO32- over H2S, GSH, Cys and Hcy. The recognition group underwent a nucleophilic addition reaction with HSO3-/SO(3)(2- )to give rise to clear dual-emission signal changes in enhanced blue channel and subdued red channel under a single wavelength excitation. Moreover, probe PPA was successfully used for the detection of endogenous HSO3-/SO32- and bioimaging of mitochondria-targeted in living cells.
Phospholipid-Biomimetic Fluorescent Mitochondrial Probe with Ultrahigh Selectivity Enables in Situ and High-Fidelity Tissue Imaging

Abstract:In situ and directly imaging mitochondria in tissues instead of isolated cells can offer more native and accurate information. Particularly, in the clinical diagnose of mitochondrial diseases such as mitochondrial myopathy, it is a routine examination item to directly observe mitochondrial morphology and number in muscle tissues from patients. However, it is still a challenging task because the selectivity of available probes is inadequate for exclusively tissue imaging. Inspired by the chemical structure of amphiphilic phospholipids in mitochondrial inner membrane, we synthesized a phospholipid-biomimetic amphiphilic fluorescent probe (Mito-MOI) by modifying a C18-alkyl chain to the lipophilic side of carbazole-indolenine cation. Thus, the phospholipid-like Mito-MOI locates at mitochondrial inner membrane through electrostatic interaction between its cation and inner membrane negative charge. Simultaneously, the C18-alkyl chain, as the second targeting group, is deeply embedded into the hydrophobic region of inner membrane through hydrophobic interaction. Therefore, the dual targeting groups (cation and C18-alkyl chain) actually endow Mito-MOI with ultrahigh selectivity. As expected, high-resolution microscopic photos showed that Mito-MOI indeed stained mitochondrial inner membrane. Moreover, in situ and high-fidelity tissue imaging has been achieved, and particularly, four kinds of mitochondria and their crystal-like structure in muscle tissues were visualized clearly. Finally, the dynamic process of mitochondrial fission in living cells has been shown. The strategy employing dual targeting groups should have reference value for designing fluorescent probes with ultrahigh selectivity to various intracellular membranous components.
Dual-Fluorophore and Dual-Site Multifunctional Fluorescence Sensor for Visualizing the Metabolic Process of GHS to SO2 and the SO2 Toxicological Mechanism by Two-Photon Imaging

Abstract:As a momentous gas signal molecule, sulfur dioxide (SO2) participates in diverse physiological activities. Excess SO2 will cause an apparent decrease in the level of intracellular glutathione (GSH), thereby damaging the body's antioxidant defense system. In addition, endogenous SO2 can be generated from GSH by reacting with thiosulfate (S2O32-) and enzymatically reduced to cysteine (Cys), a synthetic precursor of GSH. In view of their close correlation, a two-photon (TP) mitochondria-targeted multifunctional fluorescence sensor Mito-Na-BP was rationally designed and synthesized for detecting SO2 and GSH simultaneously. Under single-wavelength excitation, the sensor responded to GSH-SO2 and SO2-GSH continuously with blue-shifted and green fluorescence-enhanced signal modes, respectively, not just to GSH (enhanced) and SO2 (quenched) at 638 nm with a completely converse response tendency. Given its favorable spectral performance (high sensitivity, superior selectivity, and fast response rate) at physiological pH, Mito-Na-BP has been successfully applied in monitoring the level fluctuation of GSH affected from high-dose SO2 and visualizing in real time the metabolic process of GSH to SO2 by TP imaging. It is expected that this research will provide a convenient and efficient tool for elucidating intricate relationships of GSH and SO2 and facilitate further exploration of their functions in biomedicine.
