Abstract:Targeting cyclooxygenase-2 (COX-2) for molecular imaging is an attractive approach applicable for its overexpression in inflammation and many malignancies. Herein, for monitoring COX-2, we synthesize a specific COX-2 probe celecoxib-MPA probe (CMP), based on celecoxib and a water-soluble near-infrared dye dye ICG-Der-02 (MPA). Its high affinity for binding to COX-2 is verified by molecular docking, dynamics simulation and inhibition assay. At cellular level, CMP selectively accumulates in cytoplasm of COX-2-positive cells. in vivo assays, probe guided-imaging in inflamed or cancerous tissues confirms that CMP can bind to the locally endogenic COX-2 and exhibit intense fluorescence. Importantly, we further prove the targeting specificity of CMP as the fluorescence is significantly reduced by blocking COX-2 active site through preinjection with celecoxib. The results suggest that the probe CMP, with favorable hydrophilic property, good biocompatibility, long-term observation, excellent targeting ability and optical imaging capability, could serve as a promising probe for real-time monitoring COX-2 in inflammation and tumor.
Development of Rofecoxib-Based Fluorescent Probes and Investigations on Their Solvatochromism, AIE Activity, Mechanochromism, and COX-2-Targeted Bioimaging.

Abstract:Cyclooxygenase-2 (COX-2) fluorescent probes are promising tools for early cancer diagnosis. Traditionally, COX-2 probes were designed by connecting two parts, a fluorophore and a COX-2 binding unit, via a flexible linker. Herein, a new class of COX-2-specific fluorescent probes have been developed by one-step modification from rofecoxib by an integrative approach to combining the fluorophore and binding units into one. Among them, several new rofecoxib analogues not only retained their COX-2 binding ability but also exhibited attractive fluorescent properties, such as tunable Blue-Red emission, solvatochromism, AIE behavior and mechanochromism. Notably, the emission of 2a16 can be switched between greenish-yellow in the crystalline state to red-orange in the amorphous state by grinding and fuming treatments. Furthermore, the highly fluorescent compound 2a16 (Фf = 0.94 in powder) displayed much stronger fluorescent imaging of COX-2 in HeLa cancer cells overexpressing COX-2 than RAW264.7 normal cells with minimal expressing of COX-2. Most importantly, 2a16 can light up human cancer tissues from adjacent normal tissues with much brighter fluorescence by targeting COX-2 enzyme. These results illustrate the potential of 2a16 as a new red fluorescent probe for human cancer imaging in clinical applications.

Abstract:Identifying cancer cells and quantifying cancer-related events in particular organelles in a rapid and sensitive fashion are important for early diagnosis and for studies on pathology and therapeutics of cancers. Herein a smart "off-on" cyclooxygenase-2-specific fluorescence probe (ANQ-IMC-6), able to report the presence of cancer cells and to image Golgi-related events, has been designed and evaluated. Cyclooxygenase-2 (COX-2) has been used as imaging target in the probe design, since this enzyme is a biomarker of virtually all cancer cell lines. In the free state in aqueous solution, ANQ-IMC-6 mainly exists in a folded conformation where probe fluorescence is quenched through photoinduced electron transfer between the fluorophore acenaphtho[1,2-b]quinoxaline (ANQ) and the recognition group, indomethacin (IMC). Fluorescence is turned on, by restraining the photoinduced electron transfer, when ANQ-IMC-6 is forced to adopt the unfolded state following binding to COX-2 in the Golgi apparatus of cancer cells. ANQ-IMC-6 provides high signal-to-background staining and has been successfully used to rapidly differentiate cancer cells from normal cells when using flow cytometry and one- and two-photon fluorescence microscopic imaging. Furthermore, ANQ-IMC-6 may be able to visualize dynamic changes of the Golgi apparatus during cancer cell apoptosis, with possible application to early diagnosis.

Abstract:COX-2 fluorescent probes are promising tools for cancer diagnosis. Such probes have been conventionally designed by conjugating a fluorophore to COX-2 inhibitors through lengthy synthetic processes. Herein, a type of fluorescent probe for COX-2 imaging has been developed using a single-step process from rofecoxib. In total, six rofecoxib analogues were designed using this unique strategy. Several analogues retained comparative COX-2 targeting activity of rofecoxib and also exhibited attractive fluorescent properties, which were investigated using a combination of experimental and theoretical approaches. The most potent analogue, 2a1, displayed strong fluorescent imaging of COX-2 in HeLa cells overexpressing COX-2 compared to Raw 264.7 cells and celecoxib-treated HeLa cells that expressed low levels of COX-2. Notably, our studies indicate that 2a1 can differentiate human cancer tissue from adjacent tissue with much brighter fluorescence either in histological section or cultured 3D organoids. These results illustrate the potential of 2a1 as a COX-2 near infrared fluorescent probe for human cancer imaging in clinical settings.
A Hypoxia-Activated Near Infrared Fluorescent Probe for Cyclooxygenase-2 and in Vivo Imaging for Tumor and Inflammation

Abstract:Tumor is one of the most serious diseases, and its early diagnosis is of great significance for improving the prognosis and patient survival rate. Herein, aiming at the detection of cyclooxygenase-2 (COX-2), which plays an important role in tumorigenesis, we synthesized a NIR fluorescent probe YZP1, by using near infrared fluorescent dye Cy5.5 as the fluorophore and indomethacin (IMC) as the targeting group. Due to the quenching effect of azo group, the fluorescence of YZP1 was quenched. Under hypoxia condition and the reduction of Cytochrome P450 reductase, the azo bond of YZP1 was broken, and the fluorescence quantum yield increased from 0.0028 to 0.125. Furthermore, when HeLa cells were co-incubated with YZP1 under hypoxia condition, remarkable red fluorescence were observed, and the ratio of fluorescence intensity between hypoxic and normoxic cells could reach 9 folds. After treated with the celecoxib, an inhibitor of COX-2, of different concentrations, the fluorescence will be weakened gradually, which verify the binding of YZP1 and COX-2. Finally, by using mouse inflammation model and tumor xenograft model, the in vivo imaging ability of YZP1 was also demonstrated. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:In this concept, we present a comprehensive study on the development and application of COX‐2‐specific fluorescent probes for cancer imaging and diagnosis. Targetting cancer cells and measuring cancer‐related activities in specific organelles quickly and accurately are crucial factors for early diagnosis and research on cancer pathology and treatment. This concept explores a variety of probes which include BDIMC1‐2, TPI‐IMC, YZP1, IQ‐1, ANQ‐IMC‐6, Niblue‐C6‐IMC, AZB‐IMC2, CMP, CF3‐Fluorocoxib, IBPC1, CoxFluor, etc. and each one demonstrates unique mechanisms and high selectivity towards COX‐2 enzymes. These probes were designed to enhance fluorescence upon binding to COX‐2 which enable precise visualization of tumor and inflamed tissues. The research emphasizes the importance of COX‐2 as a biomarker in cancer diagnostics, particularly in identifying cancer stem cells and inflamed tissues. This concept highlights the potentiality of these probes in non‐invasive imaging techniques which offering significant advancements in cancer diagnosis and monitoring. The in vivo and in vitro experiments, including applications in mouse models and human tissue samples, confirm the efficacy of these probes in providing detailed imaging for clinical and research applications.

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.
Fluorescence Discrimination of Cancer from Inflammation by Molecular Response to COX-2 Enzymes

Abstract:Accurate identification of cancer from inflammation and normal tissue in a rapid, sensitive, and quantitative fashion is important for cancer diagnosis and resection during surgery. Here we report the use of cyclooxygenase-2 as a marker for identification of cancer from inflammation and the design of a novel smart COX-2-specific fluorogenic probe (NANQ-IMC6). The probe's fluorescence is "turned on" in both inflammations and cancers where COX-2 is overexpressed. Intriguingly, the fluorescent emission is quite different at these two sites with different expression level of COX-2. Hence, NANQ-IMC6 can not only distinguish normal cells/tissues from cancer cells/tissues but also distinguish the latter from sites of inflammation lesions by the different fluorescence recognition of NANQ-IMC6 for COX-2 enzymes. Following spraying with the NANQ-IMC6 solution, cancerous tissue, inflamed tissues, and normal tissues can be accurately discriminated in vivo by the unaided eye using a hand-held ultraviolet lamp emitting at 365 nm. So the probe may have potential application varying from cancer inflammation diagnosis to guiding tumor resection during surgery.

Abstract:Precision-targeted detection of cancer in a rapid and sensitive fashion remains a significant challenge in prevention, management, and in particular, treatment. Herein, we have designed and synthesized a unique cyclooxygenase-2 (COX-2) fluorescence probe, dimethylamine-9,10-distyrylanthracene-indomethacin (NDSA-IMC), that could visualize the site of highly expressed COX-2 in the Golgi apparatus of cancer cells, using 9,10-diatyrylanthracene derivative as the luminous unit and a selective inhibitor, indomethacin (IMC), as the recognizing moiety for COX-2. In an aqueous solution, the free state NDSA-IMC showed a weak emission in the absence of COX-2 but enhanced emission in the presence of the enzyme due to the restriction of intramolecular motion of aggregation-induced emission-active NDSA-IMC when bound to COX-2. Cell imaging and flow cytometry experiments indicated that NDSA-IMC could discriminate between cancer and normal cells and visualize the Golgi apparatus of cancer cells via specific targeting of COX-2. Therefore, NDSA-IMC might potentially detect early cancer lesions and ultimately mitigate the population of cancer burden in society. [GRAPHICS] .

Abstract:A novel cyclooxygenase-2 (COX-2) targeted H 2 S-activated cancer-specific fluorescent probe, namely, COX2-H 2 S , was designed and synthesized, with naphthalimide as the fluorophore and indomethacin as the targeting group. This H 2 S-sensing probe was developed to differentiate tumor cells from normal cells and was tested in living cells, Caenorhabditis elegans ( C. elegans ), and zebrafish. The probe could successfully be used for imaging endogenous and exogenous H 2 S in living cells, demonstrating high sensitivity and specificity and strong anti-interference. COX2-H 2 S had the ability to not only discern cancer cells from normal cells but also specifically recognize 9L/lacZ cells from other glioblastoma cells (U87-MG and LN229). It could also be successfully applied for the fluorescent live imaging of H 2 S in both C. elegans and zebrafish.

Abstract:Combining imaging and treatment of cancer into one molecule has become an efficient strategy for studies on pathology and cancer therapeutics. Herein, a novel highly selective and sensitive green-emitting probe (APLN), which contained a fluorophore (Naphthalimide), a linker, and a COX-2 kinase inhibitor (aspirin) has been designed and evaluated. Cyclooxygenase-2 (COX-2), as one of the typical rate-limiting enzymes, has been utilized as imaging and therapeutic target since this enzyme is overexpressed in various cancer cell lines. Aspirin, a nonsteroidal anti-inflammatory drug (NSAID), has been reported to reduce the incidence and risk of cancers by down-regulating the expression of COX-2 in cancer cells. The APLN probe has been proved to successfully label the COX-2 enzyme to identify tumors and significantly inhibit tumor growth in a xenotransplantation model of nude mice. Therefore, this enzyme-targeted fluorescence probe (APLN) is a potential reagent for in situ imaging and fluorescent-visible cancer therapy.

Abstract:Cyclooxygenase-2 (COX-2) expression is up-regulated in inflammatory tissues and many premalignant and malignant tumors. Assessment of COX-2 protein in vivo, therefore, promises to be a powerful strategy to distinguish pathologic cells from normal cells in a complex disease setting. Herein, we report the first redox-activatable COX-2 probe, fluorocoxib Q (FQ), for in vivo molecular imaging of pathogenesis. FQ inhibits COX-2 selectively in purified enzyme and cell-based assays. FQ exhibits extremely low fluorescence and displays time- and concentration-dependent fluorescence enhancement upon exposure to a redox environment. FQ enters the cells freely and binds to the COX-2 enzyme. FQ exhibits high circulation half-life and metabolic stability sufficient for target site accumulation and demonstrates COX-2-targeted uptake and retention in cancer cells and pathologic tissues. Once taken up, it undergoes redox-mediated transformation into a fluorescent compound fluorocoxib Q-H that results in high signal-to-noise contrast and differentiates pathologic tissues from non-pathologic tissues for real-time in vivo imaging.
Celecoxib Conjugated Fluorescent Probe for Identification and Discrimination of Cyclooxygenase-2 Enzyme in Cancer Cells.

Abstract:Cyclooxygenase-2 (COX-2) is an enzyme overexpressed in most types of cancers and has been used for an excellent targetable biomarker. Celecoxib is an effective inhibitor of COX-2, used in anti-inflammation. Herein we report a one and two-photon fluorescence probe (NP-C6-CXB) for COX-2, based on the conjugation of naphthalamide with Celecoxib, by using flexible hexylene linker. NP-C6-CXB is nonfluorescent in buffer solution and normal cells, while it shows bright fluorescence in solutions and cancer cells in the presence of COX-2 with an excellent selectivity. Interestingly, NP-C6-CXB can discriminate cancer cells (MCF-7) from normal cells (COS-7) in the single culture medium under confocal microscopy. Due to the selective binding affinity of NP-C6-CXB with a COX-2 enzyme, the intensity is proportional to the level of COX-2 enzyme in cancer cells. In vivo and in vitro experiments proved that NP-C6-CXB is a potential tool for identification of tumor and might be used in surgical resection of COX-2 expressed tumors.

Abstract:Detecting and imaging of ultratrace cyclooxygenase-2 in living biosystems could provide much important valuable information for the diagnosis and intervention of cancer. Molecular probes, whose fluorescent signals are generated by cyclooxygenase-2, hold great potential for identification and enumeration of cyclooxygenase-2 in living biosystems. Although quite a few fluorescent probes have been reported for cyclooxygenase-2, the use fluorogenic probe with the excellent two-photon properties for the determination of ultratrace cyclooxygenase-2 has been scarce. Herein, an "off-on" fluorescence probe (BTDAN-COX-2), able to report and image the presence of ultratrace cyclooxygenase-2 in living biosystems, has been designed and evaluated. In order to improve sensitivity and specific selectivity of probe for ultratrace cyclooxygenase-2, BTDAN-COX-2 employed cyclooxygenase-2's inhibitor as recognition group, because it is a classical and efficient recognition group for cyclooxygenase-2. A polarity-sensitive naphthalene derivative (BTDAN) as fluorophore was introduced into the molecule to enhance two-photon properties of BTDAN-COX-2. In the absent of cyclooxygenase-2, BTDAN-COX-2 mainly exists in a folded conformation where probe fluorescence is quenched through photoinduced electron transfer between the fluorophore and the recognition group. Under the condition of existence of cyclooxygenase-2, fluorescence of probe is turned on, because photoinduced electron transfer between the fluorophore and the recognition group is restrained. BTDAN-COX-2 provides high signal-to-background staining for the ultratrace cyclooxygenase-2 and has been successfully used to rapidly detect and image ultratrace cyclooxygenase-2 in living biosystems.

Abstract:Carbon monoxide (CO) can be endogenously produced in mammalian cells and regarded as an essential cell signaling molecule in the body. And it is closely related with a range of physiological and pathological processes. Although some fluorescence probes for tracking CO are reported, near-infrared (NIR) fluorescence probe for CO with a large Stokes shift remains scarce. Herein, a NIR fluorescence probe named DCX-CO is designed and synthesized. In the probe, the allyl formate group is chosen as recognizing moiety and benzopyranonitrile-xanthene-based dye is selected as fluorophore. Upon the addition of CO, the probe shows NIR emission at 762 nm with a large Stokes shift (164 nm). Moreover, the probe displays high sensitivity for CO with 16-fold fluorescence enhancement and excellent selectivity to CO over other potential interference in biological sample. Based on the low cytotoxicity, the probe can be used for the detection of exogenous and endogenous CO in HepG2 cells and HCT116 cells. In addition, taking advantage of NIR emission with a large Stokes shift, DCX-CO is applied for monitoring CO in vivo.

Abstract:Carbon monoxide (CO) is a significant gasotransmitter that naturally modulates inflammatory responses. Visualization of CO in situ would help to reveal its physiological/pathological functions. Unfortunately, most existing CO fluorescent probes show aggregation-caused quenching (ACQ) properties. Herein, we report the reaction-based fluorescent probe (BTCV-CO) with aggregation-induced emission (AIE) characteristics for CO detection and imaging. This ratiometric AIE probe showed excellent stability, high sensitivity (detection limit of 30.8 nM), and superior selectivity. More importantly, this CO-responsive AIE probe could be facilely designed and easily obtained by two-step synthesis with high yield, providing an easy-to-handle AIE toolbox for real-time visualization of CO in a living system.

Abstract:The first Golgi-localized cyclooxygenase-2 (COX-2)-specific near-infrared (NIR) fluorescent probe, Niblue-C6-IMC, able to detect cancer cells, was designed. Importantly, Niblue-C6-IMC preferentially labeled the tumors in a mouse tumor model with deep tissue penetration capacity. It may be a promising molecular tool for guiding tumor resection during surgery.
