Abstract:The synthesis of ROS-responsive polymers has attracted much attentions in recent years. Herein, we report a series of amphiphilic methoxy poly(ethylene glycol)-b-poly(carbonate-selenide)s (mPEG45-b-poly(MSe)m, m=12, 24, 36, 48, 60) by introducing selenide groups onto the backbone via one-pot lipase-catalyzed ring-opening polymerization (ROP), the synthesized amphiphilic polymers with enhanced ROS-responsiveness can self-assemble to form nanoparticles. Nuclear Magnetic Resonance (NMR), Gel Permeation Chromatography (GPC) and Fourier Transform Infrared (FTIR) are employed to confirm the structure of the polymers. Besides, the oxidation process of mPEG45-b-poly(MSe)m nanoparticles is also clarified. Ultraviolet-visible Spectrophotometer (UV-Vis), Dynamic Light Scattering (DLS) and Transmission Electron Microscopy (TEM) are further utilized to demonstrate the turbidity, morphology and size changes under oxidization, proving that the selenide groups endow the mPEG45-b-poly(MSe)m nanoparticles with enhanced ROS-response properties. Therefore, the above studies highlight the synthesized ROS-responsive polymers and provided a potential material for further applications.

Abstract:The presence of too many reactive oxygen species (ROS) will damage cells. To eliminate ROS, a nanocomposite film containing diselenide bonds is fabricated via the layer-by-layer assembly method, using adamantane-terminated diselenide block copolymer micelles and β-cyclodextrin-modified nanoparticles as building blocks. This film possesses potential stability under the normal physiological condition and can eliminate different kinds of ROS.

Abstract:Selenoxide elimination reaction has been widely used in the field of organic synthesis. However, few studies have been conducted to apply this reaction in biodegradable nanomedicine. In this work, the selenoxide elimination reaction was used for cancer treatment via producing excess cellular reactive oxygen species (ROS) for the first time. The β-seleno diesters and porphyrin derivates containing nanoparticle could be responsive to the intracellular ROS and produce acrylates through the elimination reaction. The acrylates would further deplete intracellular GSH in tumor cells and finally improved the anticancer activity in the mice tumor model. Different from traditional ROS-responsive nanomedicine, the elimination product of this reaction could regenerate cytotoxic ROS and specifically disturb the redox balance of tumor cells. This work would provide attractive avenues for the development of therapeutic strategies against cancer via synthesis of well-designed biodegradable polymers.
ROS-Responsive Chalcogen-Containing Polycarbonates for Photodynamic Therapy.

Abstract:Reactive oxygen species (ROS)-responsive polymers have attracted attention for their potential in photodynamic therapy. Herein, we report the ROS-responsive aliphatic polycarbonates prepared by the ring-opening polymerization (ROP) of three six-membered cyclic carbonate monomers with ethyl selenide, phenyl selenide or ethyl telluride groups. Under catalysis of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU), all three monomers underwent the controlled anionic ROP, showing a feature of equilibrium polymerization due to the bulky effect of 5,5-disubstituents. With PEG macroinitiator, three series amphiphilic block copolymers were prepared. They could form spherical nanoparticles of ∼100 nm, which were stable in neutral phosphate buffer but dissociated rapidly under triggering of H2O2. We studied the H2O2-induced oxidation profiles of selenide- or telluride-containing small molecules by 1H NMR and revealed the factors that affect the oxidation kinetics and products. On this basis, the oxidative degradation mechanism of the copolymer nanoparticles has been clarified. Under the same oxidative condition, the telluride-containing nanoparticle degraded with the fastest rate while the phenyl selenide-based one degraded most slowly. These ROS-responsive nanoparticles could load photosensitizer chlorin e6 (Ce6) and anticancer drug doxorubicin (DOX). Under red light irradiation, Ce6-sensitized production of 1O2 that triggered the degradation of nanoparticles, resulting in an accelerated payload release. In vitro cytotoxicity assays demonstrate that the nanoparticles coloaded with DOX and Ce6 exhibited a synergistic cell-killing effect to MCF-7 cells, representing a novel responsive nanoplatform for PDT and/or chemotherapy.

Abstract:The chalcogen-containing polyesters represent an important branch of reactive oxygen species (ROS)-responsive biodegradable polymers. Herein, we report the synthesis and oxidation properties of three ROS-responsive poly(epsilon-caprolactone) (PCL)-type polyesters. Three new -substituted caprolactone-type monomers: -ethylthio caprolactone (M1), -phenylthio caprolactone (M2), and -phenylseleno caprolactone (M3) were synthesized. The controlled ring-opening polymerizations (ROP) of these three monomers were demonstrated with benzyl alcohol as the initiator and stannous octoate as the catalyst at 130 degrees C, affording three homopolymers (P1-P3) with low dispersities. M1 showed a faster ROP rate than M2 and M3. Using PEG as the macroinitiator, three kinds of amphiphilic block copolymers (PS1, PS2, and PSE) that could form micellar nanoparticles in water were also prepared. The oxidation profiles of these block copolymer nanoparticles in neutral phosphate buffer were investigated by H-1 NMR, Nile red fluorescence, laser light scattering, and TEM. We found that the PS1 nanoparticles could sufficiently swell and finally became loosely associated water soluble polymers due to the H2O2-induced hydrophobic-hydrophilic transition from ethyl thioether to its sulfoxide. In contrast, the PS2 nanoparticles degraded very slowly owing to the strong hydrophobic nature of the phenyl thioether. Though the PSE nanoparticles responded to H2O2 much faster than both PS1 and PS2 because of the superior sensitivity of the selenide, the oxidatively produced block copolymer remained amphiphilic and the nanoparticles could not be dissociated completely. These results were discussed based on the results of H2O2-induced oxidation of three model compounds (T1-T3). All the copolymers showed negligible cytotoxicity with the polymer concentration up to 0.5 mg mL(-1), and PS1 has potential as a ROS-responsive vehicle for photodynamic therapy.
Controllable Radical Polymerization of Selenide Functionalized Vinyl Monomers and Its Application in Redox Responsive Photonic Crystals

Abstract:Selenium-containing monomer (p-phenylseleno) styrene (p-PhSeSt) is polymerized by reversible addition-fragmentation chain transfer polymerization. Polymer, (P(p-PhSeSt)), with controlled molecular weight and narrow molecular weight is obtained. The selenide moiety in obtained P(p-PhSeSt) can be selectively oxidized to selenoxide or selenone groups by H2 O2 or NaClO, respectively. These oxidized groups can be further reduced to selenide by Na2 S2 O4 . The structure changing of polymers during such redox cycle is characterized by nuclear magnetic resonance, X-ray photoelectron spectroscopy, and size exclusion chromatography. Properties, such as thermal performance, glass transition temperature, water contact angles, and refractive indices, of the resulting polymers are systematically investigated before and after oxidation. In addition, SiO2 inverse opal photonic crystal (IOPC) is fabricated by sacrificial polymer colloidal template method. Owing to changes of the RIs of P(p-PhSeSt) after selective oxidation, the predictable change of PC bandgap as a redox-responsive PC sensor is successfully realized, which provides new perspectives for modulating photonic crystals.

Abstract:A selenium-containing amphiphilic block copolymer (PEG-PUSe-PEG) with a hydrophobic polyselenide block and two hydrophilic poly(ethylene glycol) (PEG) blocks was synthesized via polymerization of toluene diisocyanate (TDI) with monoselenide-containing diols and subsequent termination with PEG monomethylether. PEG-PUSe-PEG is able to self-assemble in aqueous solution to form block copolymer aggregates. Interestingly, it was found that the aggregates have a good oxidation-responsiveness and undergo a structural dissociation in a mild oxidative environment (such as 0.1% H2O2 v/v) due to the unique sensitivity of selenide groups in presence of oxidants. Compared with the sulfide analogue PEG-PUS-PEG, PEG-PUSe-PEG is more sensitive to oxidants. It is anticipated that selenium-containing block copolymer aggregates may find application in the field of drug delivery systems.

Abstract:Stimuli-responsive polymersomes formed by amphiphilic block copolymers have attracted substantial attention as smart and robust containers for drug delivery and nano/microreactors. Biosourced amphiphilic diblock copolypeptoids were developed that can self-assemble into oxidation-responsive unilamellar vesicles. These vesicles can burst under the action of reactive oxygen species which can be the hydrogen peroxide or the singlet oxygen produced by light-activation of a photosensitizer with spatiotemporal control. Polysarcosine (PSar, also called poly(N-methyl glycine)) was selected as the hydrophilic block because of its resistance to protein adsorption and low toxicity, similar to poly(ethylene glycol) (PEG). We designed and synthesized poly(N-3-(methylthio)propyl glycine) as the hydrophobic block. Its polyglycine backbone is the same as that of PSar, and especially, its hydrophobic N-substituents, thioether side chains, can be oxidized to hydrophilic sulfoxides. These oxidation-responsive polymersomes entirely based on N-substituted poly(amino acid)s were biocompatible as confirmed by cell viability tests and may find applications in drug delivery, biosensing, biodetection, and nano/microreactors.

Abstract:Reactive oxygen species (ROS) play key roles in many physiological processes, such as cell signaling and host innate immunity. However, when they are overproduced, ROS may damage biomolecules in vivo and cause diseases such as cardiovascular or neurodegenerative diseases, cancer, and so forth. Oxidative stress is usually implicated in various inflammatory tissues, representing an important target for the development of various therapeutic strategies. Therefore, various probes for the in vitro detection of ROS or the in vivo diagnosis of the oxidative stress-relevant diseases have been developed. Oxidation-responsive polymers have also attracted great interest due to their potential applications in biomedical fields. In this feature article, we summarize six types of oxidation-responsive polymers based on different oxidation-responsive motifs. Poly(propylene sulfide)s, selenium-based polymers, aryl oxalate- and phenylboronic ester-containing polymers are discussed in detail, while poly(thioketal)s and proline-containing polymeric scaffolds are briefly introduced.

Abstract:In this paper we report the synthesis and study of a series of side-chain selenium-containing amphiphilic poly(ethylene oxide-b-acrylic acid) block copolymers PEO-b-PAA-Se. These block copolymers can self-assemble in aqueous solution and form spherical micellar aggregates. The selenide group of PEO-b-PAA-Se can change into hydrophilic selenoxide under mild oxidation of 0.1% hydrogen peroxide, leading to the disassembly of the spherical micellar aggregates. Small compounds like Nile Red can be encapsulated into the micellar aggregates and show fast release upon the addition of 0.1% hydrogen peroxide. More interestingly, the oxidation state of selenoxide can be reversed to selenide under reduction of Vitamin C, thus recovering the spherical aggregates. The reversible oxidation and reduction process shows good reversibility and can be repeated at least 7 times. It is hoped that this reversible redox controlled system may function as nanocontainers for potential application in the areas such as responsive drug delivery and anti-oxidation studies.
Degradable multisegmented polymers synthesized by consecutive radical addition-coupling reaction of α,ω-macrobiradicals and nitroso compound

Abstract:A consecutive radical addition-coupling reaction induced by spin-trapping agent is applied to produce degradable multisegmented polymer using alpha,omega-dibromo polymer as a precursor. The macroradical generated by single electron transfer process catalyzed by Cu/PMDETA from alpha,omega-dibromo polymer can be efficiently captured by 2-methyl-2-nitrosopropane (MNP), which results in nitroxide radical. The in situ formed nitroxide radical immediately undergoes cross-coupling reaction with polymeric radical, generating block polymer bridged with alkoxyamine moiety. The consecutive radical addition-coupling reaction generates multisegmented polymer via step-growth mechanism. Different multisegmented polymers have been prepared from alpha,omega-dibromo-PS, PtBA, and PtBA-PS-PtBA. The block number of multisegmented polymers can be tailored by varying the feed ratio of alpha,omega-dibromo precursor to MNP. The multisegmented polymer can be degraded in the presence of hydrogen atom donor or air, and the molecular weight distribution transformed back into shape of its original precursor as it is conjugated by alkoxyamine moieties. Polyurethane with Β‐selenocarbonyl Structure Enabling the Combination of Plastic Degradation and Waste Upcycling

Abstract:AbstractDegradable polymers offer a promising solution to mitigate global plastic pollution, but the degraded products often suffer from diminished value. Upcycling is a more sustainable approach to upgrade polymer waste into value‐added products. Herein, we report a β‐selenocarbonyl‐containing polyurethane (SePU), which can be directly degraded under mild conditions into valuable selenium fertilizers for selenium‐rich vegetable cultivation globally, enabling both plastic degradation and waste upcycling. Under oxidation condition, this polymer can be easily and selectively degraded via selenoxide elimination reaction from mixed plastic waste. The degraded product can serve as effective selenium fertilizers to increase selenium content in radish and pak choi. The SePU exhibits excellent mechanical properties. Additionally, we observed the formation of spherulites‐like selenium particles within the materials during degradation for the first time. Our research offers a successful application of selenoxide elimination reaction in the field of plastic degradation for the first time, endowing plastics with both degradability and high reusable value. This strategy provides a promising solution to reduce pollution and improve economy and sustainability of plastics.

Abstract:Synthetic reactive oxygen species (ROS)-responsive biomaterials have emerged as a useful platform for regulating critical aspects of ROS-induced pathologies and can improve such hostile microenvironments. Here, we report a series of new hyperbranched poly(β-hydrazide ester) macromers (HB-PBHEs) with disulfide moieties synthesized via an "A2 + B4" Michael addition approach. The three-dimensional structure of HB-PBHEs with multiacrylate end groups endows the macromers with rapid gelation capabilities to form (1) injectable hydrogels via cross-linking with thiolated hyaluronic acid and (2) robust UV-cross-linked hydrogels. The disulfide-containing macromers and hydrogels exhibit H2O2-responsive degradation compared with the counterparts synthesized by a dihydrazide monomer without disulfide moieties. The cell viability under a high ROS environment can be well-maintained under the protection of the disulfide containing hydrogels.

Abstract:Sulfur-containing polypeptides, capable of reactive oxygen species (ROS)-responsive structural change, are one of the most important building blocks for the construction of polypeptide-based drug delivery systems. However, the relatively low ROS sensitivity of side-chain thioethers limits the biomedical applications of these polypeptides because they usually require a high concentration of ROS beyond the pathological ROS level in the tumor microenvironment. Herein, we report the design and synthesis of a seleniumcontaining polypeptide, which undergoes random coil-to-extended helix and hydrophobic-to-hydrophilic transitions in the presence of 0.1% H2O2, a concentration that is much lower than the ROS requirement for thioether. ROS-responsive micelles were thus prepared from the amphiphilic copolymer consisting of the hydrophilic poly(ethylene glycol) (PEG) segment and hydrophobic selenopolypeptide segment and were used to encapsulate doxorubicin (DOX). The micelles could be sensitively dissociated inside tumor cells in consequence of ROS-triggered oxidation of side-chain selenoether and structural change of the micelles, thereby efficiently and selectively releasing the encapsulated DOX to kill cancer cells. This work provides an alternative design of ROS-responsive polypeptides with higher sensitivity than that of the existing sulfur-containing polypeptides, which may expand the biomedical applications of polypeptide materials.

Abstract:Herein, the lipase-catalyzed ring-opening copolymerization of the diethylene diselenide carbonate dimer (SeC) with its analogue macrocycle (1,3,12,14-Tetraoxacyclodocosane-2,13-dione, TDC) was explored, and the small-ring carbonate (TMC) was used as control. The structure and composition of the diselenide-functionalized long-chain polycarbonates were confirmed by NMR and FTIR. The 13C NMR and 77Se NMR showed that the copolymers observed random sequence. Moreover, the kinetic process of TDC + SeC copolymerization demonstrated a faster polymerization rate and higher conversion of SeC in comparison with TMC + SeC. Besides, the copolymers exhibited ROS-responsive behaviors when exposed to the oxidizing environment owing to the diselenide groups, which was confirmed by UV–vis, DLS and TEM. Therefore, the ROS responsiveness facilitated potential application for the diselenide-functionalized copolymers.

Abstract:The diselenide bond has attracted intense interest for drug delivery systems (DDSs) for tumor chemotherapy, owing to it possessing higher redox sensitivity than the disulfide one. Various redox-responsive diselenide-containing carriers have been developed for chemotherapeutics delivery. However, the premature drug leakage from these DDSs was significant enough to cause toxic side effects on normal cells. Here, a pH/redox co-triggered degradable polyprodrug was designed as a drug self-delivery system (DSDS) by incorporating drug molecules as structural units in the polymer main chains, using a facile one-pot two-step approach. The proposed PDOX could only degrade and release drugs by breaking both the neighboring acid-labile acylhydrazone and the redox-cleavable diselenide conjugations in the drug's structural units, triggered by the higher acidity and glutathione (GSH) or reactive oxygen species (ROS) levels in the tumor cells. Therefore, a slow solubility-controlled drug release was achieved for tumor-specific chemotherapy, indicating promising potential as a safe and efficient long-acting DSDS for future tumor treatment.
Dual Redox Responsive Coassemblies of Diselenide-Containing Block Copolymers and Polymer Lipids.

Abstract:A general approach is reported to fabricate a stimuli responsive system via coassembly of diselenide-containing block copolymers with polymer lipids, which integrates the stimuli-responsiveness of diselenide chemistry and the biocompatibility of polymer lipids. By using dynamic light scattering, transmission electron microscopy, and zeta potential analyzer, coassembly behavior of these two kinds of polymers and responsiveness of coassemblies have been investigated. These coassemblies can exhibit redox-responsiveness inheriting from the diselenide-containing block copolymers. In the presence of low concentration of hydrogen peroxide or glutathione, the coassemblies can be disrupted.

Abstract:Herein, novel multi-responsive injectable polyester hydrogels were reported based on the diselenide-containing poly(ε-caprolactone) copolymers ((mPEG-PCL-Se)2). The (mPEG-PCL-Se)2 solution remained a free-flowing state at ambient temperature but spontaneously turned into a semisolid hydrogel upon heating to physiologic temperature. The phase transition temperature was examined to be dependent on the composition and aqueous concentration of the copolymers. More importantly, the thermo-responsive hydrogels were endowed with oxidation and reduction-triggered degradation by the incorporation of diselenide groups. Accordingly, the degradation of poly(ε-caprolactone)-based hydrogels was greatly improved and the rate of degradation was well regulated by the concentration of hydrogen peroxide (H2O2) or glutathione (GSH). This superior stimuli-responsive degradation could lead to an enhanced drug release of encapsulated drug (Doxorubicin, DOX). Thus the oxidation and reduction-triggered degradable diselenide-containing poly(ε-caprolactone) hydrogels would offer great potential for the controlled drug delivery.
