Abstract:The transient receptor potential canonical subfamily member 5 (TRPC5) is a non-selective calcium-permeant cation channel. As a depolarizing channel, its function is studied in the central nervous system and kidney. TRPC5 forms heteromultimers with TRPC1, but also forms homomultimers. It can be activated by reducing agents through reduction of the extracellular disulfide bond. Here we present the 2.9 Å resolution electron cryo-microscopy (cryo-EM) structure of TRPC5. The structure of TRPC5 in its apo state is partially open, which may be related to the weak activation of TRPC5 in response to extracellular pH. We also report the conserved negatively charged residues of the cation binding site located in the hydrophilic pocket between S2 and S3. Comparison of the TRPC5 structure to previously determined structures of other TRPC and TRP channels reveals differences in the extracellular pore domain and in the length of the S3 helix. Together, these results shed light on the structural features that contribute to the specific activation mechanism of the receptor-activated TRPC5.

Abstract:Members of the transient receptor potential (TRP) ion channels conduct cations into cells. They mediate functions ranging from neuronally mediated hot and cold sensation to intracellular organellar and primary ciliary signaling. Here we report a cryo-electron microscopy (cryo-EM) structure of TRPC4 in its unliganded (apo) state to an overall resolution of 3.3 angstrom. The structure reveals a unique architecture with a long pore loop stabilized by a disulfide bond. Beyond the shared tetrameric six-transmembrane fold, the TRPC4 structure deviates from other TRP channels with a unique cytosolic domain. This unique cytosolic N-terminal domain forms extensive aromatic contacts with the TRP and the C-terminal domains. The comparison of our structure with other known TRP structures provides molecular insights into TRPC4 ion selectivity and extends our knowledge of the diversity and evolution of the TRP channels.

Abstract:Transient receptor potential (TRP) channels are sensors for a wide range of cellular and environmental signals, but elucidating how these channels respond to physical and chemical stimuli has been hampered by a lack of detailed structural information. Here we exploit advances in electron cryo-microscopy to determine the structure of a mammalian TRP channel, TRPV1, at 3.4 Å resolution, breaking the side-chain resolution barrier for membrane proteins without crystallization. Like voltage-gated channels, TRPV1 exhibits four-fold symmetry around a central ion pathway formed by transmembrane segments 5–6 (S5–S6) and the intervening pore loop, which is flanked by S1–S4 voltage-sensor-like domains. TRPV1 has a wide extracellular ‘mouth’ with a short selectivity filter. The conserved ‘TRP domain’ interacts with the S4–S5 linker, consistent with its contribution to allosteric modulation. Subunit organization is facilitated by interactions among cytoplasmic domains, including amino-terminal ankyrin repeats. These observations provide a structural blueprint for understanding unique aspects of TRP channel function.

Abstract:Structural studies on TRP channels, while limited, are poised for a quickened pace and rapid expansion. As of yet, no high-resolution structure of a full length TRP channel exists, but low-resolution electron cryomicroscopy structures have been obtained for 4 TRP channels, and high-resolution NMR and X-ray crystal structures have been obtained for the cytoplasmic domains, including an atypical protein kinase domain, ankyrin repeats, coiled coil domains and a Ca(2+)-binding domain, of 6 TRP channels. These structures enhance our understanding of TRP channel assembly and regulation. Continued technical advances in structural approaches promise a bright outlook for TRP channel structural biology.

Abstract:TRPC6 and TRPC3 are receptor-activated nonselective cation channels that belong to the family of canonical transient receptor potential (TRPC) channels. They are activated by diacylglycerol, a lipid second messenger. TRPC6 and TRPC3 are involved in many physiological processes and implicated in human genetic diseases. Here we present the structure of human TRPC6 homotetramer in complex with a newly identified high-affinity inhibitor BTDM solved by single-particle cryo-electron microscopy to 3.8 Å resolution. We also present the structure of human TRPC3 at 4.4 Å resolution. These structures show two-layer architectures in which the bell-shaped cytosolic layer holds the transmembrane layer. Extensive inter-subunit interactions of cytosolic domains, including the N-terminal ankyrin repeats and the C-terminal coiled-coil, contribute to the tetramer assembly. The high-affinity inhibitor BTDM wedges between the S5-S6 pore domain and voltage sensor-like domain to inhibit channel opening. Our structures uncover the molecular architecture of TRPC channels and provide a structural basis for understanding the mechanism of these channels.

Abstract:Ion channel heteromers display distinct electrophysiological and ligand-binding properties, allowing precise modulation of cellular functions in specific tissues. Among the TRPC subtypes, the TRPC1/4/5 heteromers are the most abundant isoform in the human brain and represent a primary target for treating anxiety and depression. However, the structural organization of TRPC1/4/5 heteromers remain unclear, limiting the advancement of research on these channels. Here, we report the cryo-EM structure of TRPC1/5 heterotatramer at a resolution of 2.84 angstrom. It consists of an asymmetric structure with three TRPC5 subunits and one TRPC1 subunit. Further structural and functional analysis revealed that the TRPC1 subunit contributes to an asymmetrical ion conduction pathway, a characteristic that differentiates it from TRPC5 homomers. The unique features of the TRPC1 pore loop and its interface interactions with TRPC5 were shown to play crucial roles in modulating the ion conduction properties and gating mechanisms of the heteromeric complex. In addition, we identified key residues within the gating regions that influence channel activation and ion permeability, elucidating the structural basis for the functional divergence between the TRPC1/C5 heterotetramer and TRPC5 homotetramer. Notably, TRPC5 residues were found to dominate gating mechanisms, underscoring the complex interplay between subunit composition and functional outcomes in TRPC channels. Our findings provide novel insights into the structural and functional dynamics of TRPC1/C5 heteromers, paving the way for targeted therapeutic strategies in TRPC5-related disorders.

Abstract:Transient receptor potential (TRP) ion channels have a crucial role as cellular sensors, mediating diverse physical and chemical stimuli. The formation of heteromeric structures expands the functionality of TRP channels; however, their molecular architecture remains largely unknown. Here we present the cryo-electron microscopy structures of the human TRPC1/TRPC4 heteromer in the apo and antagonist-bound states, both consisting of one TRPC1 subunit and three TRPC4 subunits. The heteromer structure reveals a distinct ion-conduction pathway, including an asymmetrically constricted selectivity filter and an asymmetric lower gate, primarily attributed to the incorporation of TRPC1. Through a structure-guided electrophysiological assay, we show that both the selectivity filter and the lower part of the S6 helix participate in deciding overall preference for permeating monovalent cations. Moreover, we reveal that the introduction of one lysine residue of TRPC1 into the tetrameric central cavity is enough to render one of the most important functional consequences of TRPC heteromerization: reduced calcium permeability. Our results establish a framework for addressing the structure–function relationship of the heteromeric TRP channels.

Abstract:Transient receptor potential canonical (TRPC) channels constitute a group of receptor-operated calcium-permeable nonselective cation channels of the TRP superfamily. The seven mammalian TRPC members, which can be further divided into four subgroups (TRPC1, TRPC2, TRPC4/5, and TRPC3/6/7) based on their amino acid sequences and functional similarities, contribute to a broad spectrum of cellular functions and physiological roles. Studies have revealed complexity of their regulation involving several components of the phospholipase C pathway, G(i) and G(o) proteins, and internal Ca2+ stores. Recent advances in cryogenic electron microscopy have provided several high-resolution structures of TRPC channels. Growing evidence demonstrates the involvement of TRPC channels in diseases, particularly the link between genetic mutations of TRPC6 and familial focal segmental glomerulosclerosis. Because TRPCs were discovered by the molecular identity first, their pharmacology had lagged behind. This is rapidly changing in recent years owning to great efforts from both academia and industry. A number of potent tool compounds from both synthetic and natural products that selective target different subtypes of TRPC channels have been discovered, including some preclinical drug candidates. This review will cover recent advancements in the understanding of TRPC channel regulation, structure, and discovery of novel TRPC small molecular probes over the past few years, with the goal of facilitating drug discovery for the study of TRPCs and therapeutic development. Exploring Functional Roles of TRPV1 Intracellular Domains with Unstructured Peptide-Insertion Screening.

Abstract:TRPV1 is a polymodal nociceptor for diverse physical and chemical stimuli that interact with different parts of the channel protein. Recent cryo-EM studies revealed detailed channel structures, opening the door for mapping structural elements mediating activation by each stimulus. Towards this goal, here we have combined unstructured peptide-insertion screening (UPS) with electrophysiological and fluorescence recordings to explore structural and functional roles of the intracellular regions of TRPV1 in mediating various activation stimuli. We found that most of the tightly packed protein regions did not tolerate structural perturbation by UPS when tested, indicating that structural integrity of the intracellular region is critical. In agreement with previous reports, Ca2+-dependent desensitization is strongly dependent on both intracellular N- and C-terminal domains; insertions of an unstructured peptide between these domains and the transmembrane core domain nearly eliminated Ca2+-dependent desensitization. In contrast, channel activations by capsaicin, low pH, divalent cations, and even heat are mostly intact in mutant channels containing the same insertions. These observations suggest that the transmembrane core domain of TRPV1, but not the intracellular domains, is responsible for sensing these stimuli.

Abstract:TRPV2 is a member of the Transient Receptor Potential (TRP) family of non-selective cation channels. It is heat-sensitive, and has been implicated in mechano-sensation and regulation of cardiovascular function. To understand better the structural basis for TRPV2 function, we have expressed full-length and engineered variants of TRPV2 in budding yeast, and have purified the proteins for single-particle electron cryomicroscopy. The purified proteins are largely monodisperse when analyzed by size-exclusion chromatography, and yield robust Ca2+ flux when reconstituted into defined proteoliposomes. Preliminary structural studies by cryo-EM suggest structural differences between TRPV2 and TRPV1 that may help to explain their different gating properties. Abstract:In mammalians, transient receptor potential mucolipin ion channels (TRPMLs) exhibit variable permeability to cations such as Ca2+, Fe2+, Zn2+, and Na+ and can be activated by the phosphoinositide PI(3,5)P2 in the endolysosomal system. Loss or dysfunction of TRPMLs has been implicated in lysosomal storage disorders, infectious diseases, and metabolic diseases. TRPML2 has recently been identified as a mechanosensitive and hypotonicity-sensitive channel in endolysosomal organelles, which distinguishes it from TRPML1 and TRPML3. However, the molecular and gating mechanism of TRPML2 remains elusive. Here, we present the cryo-EM structure of the full-length mouse TRPML2 in lipid nanodiscs at 3.14 Å resolution. The TRPML2 homotetramer structure at pH 7.4 in the apo state reveals an inactive conformation and some unique features of the extracytosolic/luminal domain and voltage sensor-like domain that have implications for the ion-conducting pathway. This structure enables new comparisons between the different subgroups of TRPML channels with available structures and provides structural insights into the conservation and diversity of TRPML channels. These comparisons have broad implications for understanding a variety of molecular mechanisms of TRPMLs in different pH conditions, including with and without bound agonists and antagonists.

Abstract:Identification of previously unknown ion channels is always a challenging work though the transmembrane protein-coding genes in human genome has been comprehensively annotated for years. Despite being the gold standard of functional assay for ion channels, electrophysiological recordings are often accompanied by electrical noise, leak conductance and background currents of the membrane system. These unwanted signals, if not treated properly, lead to mischaracterization of proteins with seemingly unusual ion-conducting properties. In recent ten years, the technical revolution of cryo-electron microscopy (cryo-EM) have greatly advanced our understanding on the structures and gating mechanisms of various ion channels, and also raised concerns about the pore-forming ability of some previously identified channel proteins. In this review, we summarize cryo-EM findings on ion channels with molecular identities recognized or disputed in recent ten years, and discuss current knowledge of proposed channel proteins awaiting cryo-EM analyses. We also present a classification of ion channels according to their architectures and evolutionary relationships, and discuss the possibility and strategy of identifying more ion channels by analyzing structures of transmembrane proteins of unknown function.
A First Look into the 3D Structure of the TRPV2 Channel by Single-Particle Cryo-Em

Abstract:The TRPV2 channel is a ∼385 kDa integral membrane protein complex that functions as a high-threshold thermosensor and is involved in calcium signaling during inflammation and nerve injury. TRPV2 is constituted of four subunits, each with six transmembrane (TM) spanning helices with a pore loop between the fifth and sixth TM helices. To date, little is understood about the complex biophysical properties or the 3D architecture of the TRPV2 channel. We used single particle electron cryo-microscopy (cryo-EM) to solve the 3D structure of the tetrameric TRPV2 channel. To obtain sufficient quantities of purified channel proteins for structural and functional analysis, we used heterologous expression in the budding yeast, S. cerevisiae and immunoaffinity chromatography to purify TRPV2. The purified channel was vitrified in the presence of amphipathic polymer, amphipol 8-35, which was used to substitute for solubilizing detergent in TRPV2 preparations. This procedure allowed us to achieve optimal ice thickness and uniform particle distribution in the cryospecimen. Vitrified TRPV2 samples were imaged using an FEI Polara electron cryo-microscope operated at 300 keV at liquid-nitrogen temperature. Images of the channel were acquired on a 4k x 4k CCD camera (TVIPS) at 59,000 nominal magnification of microscope using low-dose mode and a defocus range of 2-5 μm. The 3D reconstruction of TRPV2 was generated with 20,608 particle images extracted from 431 CCD frames, and image processing was performed using EMAN2. The resulting structure was validated using tilt-pair analysis, and is interpretable to ∼15 Å based on "gold standard" resolution criterion. Abstract:Transient receptor potential canonical channels (TRPCs) are calcium-permeable, nonselective cation channels. Human TRPC channels contain six functional members: TRPC1, and TRPC3-7, and they are widely distributed in many tissues. These channels are involved in numerous physiological processes, including regulating myogenic tone and blood pressure, endothelial barrier function regulation, hypoxic pulmonary vasoconstriction, neuronal function, and kidney function. They are also associated with several diseases, including traumatic brain injury, pathologic cardiac hypertrophy, atherosclerosis, the development of cancer, and diabetes. Specifically, gain-of-function mutations of the TRPC6 gene in humans cause focal segmental glomerular sclerosis (FSGS), a chronic kidney disease for which there is no effective treatment. Consequently, TRPC channels serve as drug targets for several diseases, including FSGS and anxiety. Recent developments in single-particle cryo-EM permit the determination of the structures of multiple TRPC channel members (TRPC3, TRPC4, TRPC5, and TRPC6) in multiple states at near-atomic resolutions. These structures demonstrate that TRPC channels are tetramers composed of two layers: The upper transmembrane domain (TMD) and the lower intracellular domain (ICD). The ICD layer consists of ankyrin repeat domains (ARD), linker helices domains (LHD), the TRP helix, and C terminal helices. The four protomers' voltage sensor-like domains (VSLD) and pore domains form the TMD layer. ICD has a bell-like shape with a large central chamber and caps below TMD. The structures reveal both the inhibitory calcium binding site in ICD and the activating calcium binding site in TMD. Calcium binding to the inhibitory site results in the closure of the ICD and the restriction of ion flow through the ICD, whereas calcium dissociation from the inhibitory site results in the opening of the ICD and the unrestricted flow of ions through the ICD. Calcium binding to the activating site increases its activity. Additionally, the binding pockets and binding poses of several small molecule modulators of TRPC channels are identified. Their binding sites are distributed across three distinct regions of TMD. One of the pockets partially overlaps the endogenous activating lipid diacylglycerol-binding site, while another partially overlaps the activating calcium-binding site. In conclusion, these structural findings combined with functional analysis, particularly site-directed mutagenesis data, reveal how intracellular calcium concentrations bi-directionally modulate the TRPC channel, how small molecules bind and tune the activity of TRPC channels, and how the gain-of-function mutations found in FSGS patients over-activate the intracellular calcium signaling of kidney podocytes via the TRPC6 channel in a positive feedback manner. These developments lay the groundwork for a more in-depth mechanistic dissection of TRPC channels. They would also facilitate the discovery of drugs that target TRPC channels to treat associated diseases.

Abstract:Inositol 1,4,5-trisphosphate receptors (IP(3)Rs) are ubiquitously expressed intracellular Ca2+ channels and the major mediators of cellular Ca2+ signals generated by the release of Ca2+ ions from intracellular stores in response to a variety of extracellular stimuli. Despite established physiological significance and proven involvements of IP3R channels in many human diseases, detailed structural basis for signal detection by these ion channels and their gating remain obscure. Recently, single particle electron cryomicroscopy (cryo-EM) has yielded a longawaited near-atomic resolution structure of the entire fulllength type 1 IP3R. This structure provided exciting mechanistic insights into the molecular assembly of IP3R, revealing the pronounced structural conservation of Ca2+ release channels and raising many fundamental and controversial questions on their activation and gating. Here we summarize the major technological advances that propelled our cryo-EM analysis of IP3R to near-atomic resolution and discuss what the future holds for structural biology of Ca2+ release channels.

Abstract:TRPM4 is a calcium-activated, phosphatidylinositol-4,5-bisphosphate (PtdIns(4,5)P-2)-modulated, non-selective cation channel that belongs to the family of melastatin-related transient receptor potential (TRPM) channels. Here we present the electron cryo-microscopy structures of the mouse TRPM4 channel with and without ATP. TRPM4 consists of multiple transmembrane and cytosolic domains, which assemble into a three-tiered architecture. The N-terminal nucleotide-binding domain and the C-terminal coiled-coil participate in the tetrameric assembly of the channel; ATP binds at the nucleotide-binding domain and inhibits channel activity. TRPM4 has an exceptionally wide filter but is only permeable to monovalent cations; filter residue Gln973 is essential in defining monovalent selectivity. The S1-S4 domain and the post-S6 TRP domain form the central gating apparatus that probably houses the Ca2+- and PtdIns(4,5)P-2-binding sites. These structures provide an essential starting point for elucidating the complex gating mechanisms of TRPM4 and reveal the molecular architecture of the TRPM family.
