Abstract:Molecular dynamics simulations have been performed to investigate the effect of acetone solvent on the crystal morphology of RDX. The results show that the growth morphology of RDX crystal in vacuum is dominated by the (111), (020), (200), (002), and (210) faces using the BFDH laws, and (111) face is morphologically the most important. The analysis of surface structures of RDX crystal indicates that (020) face is non-polar, while (210), (111), (002), and (200) faces are polar among which (210) face has the strongest polarity. The interaction between acetone solvent and each RDX crystal face is different, and the order of binding energy on these surfaces is (210) > (111) > (002) > (200) > (020). The analysis of interactions among RDX and acetone molecules reveal that the system nonbond interactions are primary strong van der Waals and electrostatic interactions containing π-hole interactions, the weak hydrogen bond interactions are also existent. The effect of acetone on the growth of RDX crystal can be evaluated by comparing the binding energies of RDX crystalline faces. It can be predicted that compared to that in vacuum, in the process of RDX crystallization from acetone, the morphological importance of (210) face is increased more and (111) face is not the most important among RDX polar surfaces, while the non-polar (020) face probably disappears. The experimentally obtained RDX morphology grown from acetone is in agreement with the theoretical prediction.

Abstract:In this work, a molecular dynamics investigation was performed to study the effect of the solvents acetone and cyclohexanone on the crystal habits of hexogen. The results show that the adsorption energies of hexogen polar faces such as the (002), (210), (111), and (200) faces are larger than that of the nonpolar (020) face. The adsorption interactions of hexogen surfaces with acetone are stronger than those with cyclohexanone. The adsorption interactions consist of van der Waals and electrostatic interactions. The effect of solvents on the crystal growth of hexogen is discussed by comparing the adsorption energy of each crystal face. It may qualitatively predict that if hexogen crystallizes from polar solvents (acetone and cyclohexanone), the morphological importance of polar faces increases, especially in acetone solvent, while the importance of the nonpolar (020) face probably diminishes, even disappears. The theoretical prediction properly explains the different crystal morphologies of hexogen observed experimentally in these two solvents.

Abstract:To obtain crystal products with ideal morphology and better quality, it is important to fully understand and grasp the affecting mechanism of solvents on crystal morphology. In this work, the interactions between solvent/solute molecules and different crystal faces of pyriproxyfen are investigated by a combination of experiments and molecular simulations. It is found that pyriproxyfen crystals grow into a lamellar morphology in methanol and ethanol, while the crystal grows into a three-dimensional shuttle morphology in n-butanol and n-heptane. Molecular simulations reveal that the molecular arrangement of crystal faces makes the alcohol hydroxyl adsorption sites exposed in different degrees, and the (002) face is more sensitive to alcohol hydroxyl than other faces. The adsorption of alcohol hydroxyl groups hinders the growth of crystal planes, so (002) and (102) faces become the main crystal planes in methanol and ethanol, and the lamellar crystal is formed. The developed knowledge of the growth mechanism based on the interaction between the solvent and crystal interface can be conducive to the further optimization of the pyriproxyfen crystal products.

Abstract:Solution crystallization is an important separation unit operation in active pharmaceutical ingredient production. Solvent is one of the important factors affecting crystal morphology. How to select/design suitable solvents is still one of the most urgent problems in the crystallization field. In this paper, a framework for crystallization solvent design based on the developed quantitative prediction model of crystal morphology is proposed. First, molecular dynamics is used to predict the crystal morphology in solvents. Next, solvent descriptors are selected by stepwise regression method. Then, the quantitative relationship between crystal aspect ratio and solvent descriptors is developed. Subsequently, Computer-Aided Molecular Design method is integrated with the developed quantitative prediction model. The crystallization solvent design problem is expressed as a Mixed-Integer Non-Linear Programming model with maximum yield as the objective function, which is solved by decomposition algorithm. Abstract:Flaky crystals are fragile, rendering them unsuitable for various applications including use, processing, storage and transportation. This work investigates the growth and morphology regulation mechanisms of flaky crystals using benzanilide as a model material. Initially, block-like crystals are prepared by solvent screening and cooling crystallization with its morphology greatly improved. Subsequently, growth kinetics of (111), (1–10) and (020) faces are established with crystal growth experiments. The crystal growth of (001) face is also determined to follow a two-dimensional nucleation model with the microscopic surface analysis using atomic force microscopy. The changes of the surface integration resistance and the growth step height are regarded as kinetic mechanisms of the growth behavior and crystal morphology modulation by solvent and supersaturation. Finally, the molecular mechanism of the modulation of the solvent in crystal morphology is revealed base on crystal surface structure analysis and molecular dynamics simulations. The weak solvent-interface polar interactions facilitate the incorporation of benzanilide molecules into the weak polarity (001) face, leading to the surface roughening and increased crystal thickness when employing strongly polar solvents such as acetonitrile. Unlike the former, the relatively weak hydrogen bonding interactions between solvents and strong polarity (020) face, and the higher diffusion ability of molecules promote the rapid growth and disappearance of (020) face in ethanol and acetonitrile with strong hydrogen bond formation ability. This study can contribute to a deeper understanding of the solvent effect on crystal morphology and provide guidance for optimizing crystallization conditions to obtain shape-tailored crystal products.

Abstract:Low sensitivity and highly energetic materials (LSHEMs) are currently the most promising high-energy materials due to their structural stability. However, many problems exist due to their abundant hydrogen bonding (HB) in the structure, such as low solubility and difficulty controlling the crystallization process. In this paper, aiming at regulating the crystal morphology of LSHEMs, 4,4′,5,5′-tetranitro-1H,1′H-[2,2′-biimidazole]-1,1′-diamine is used as a model substance to explore the mechanism of controlling the crystal morphology in solvents. The molecular structure, crystal structure, and HB sites on crystal faces were investigated by molecular electrostatic potential surface, Hirshfeld surface, and HB analyses, and it was found that the HB density [δ(HB)] of faces is of the order (011) > (11–1) > (110) > (100) in crystal or solution, determining the tendency of interaction with polar solvents. Then, using molecular dynamics simulation and the modified attachment energy model, we found that the attachment energy of crystal faces and the predicted crystal morphologies in different solvents were determined by HB sites on faces and strongly correlated with the solvent polarity. The experimental morphologies were consistent with the trend predicted including the aspect ratio trend, which confirmed our theoretical speculation. This work provides an effective method of choosing solvents for morphology customization of LSHEMs, which will help guide the morphology control and realize the industry application of LSHEMs.

Abstract:Cocrystallization is a helpful method for explosives design. However, lack of understanding of the cocrystallization mechanism leads to inefficiency in cocrystal preparation. Therefore, studying the effects of solvent on cocrystal is of great importance for the efficient application of 2,4,6,8,10,12-hexanitrohexaazaisowurtzitane (CL-20). In this paper, the effect of solvent on cocrystallization is investigated by the CL-20/HMX cocrystal/solvent cluster model, the CL-20/HMX/solvent mixture model, the CL-20/HMX cocrystal/solvent interface model combined with quantum chemistry and molecular dynamic methods. The authors find that the hydrogen bond between CL-20 and 1,3,5,7-tetranitro-1,3,5,7-tetrazocane (HMX) is the strongest and the binding energy of cocrystal and solvent molecules is the weakest in ethyl acetate (EA) solvent, indicating that CL-20 and HMX tend to be combined together and there is less hindrance by solvent molecules. Analysis of the CL-20/HMX/solvent mixture and mass density distribution studies show that the solvent effect has a great influence on the crystal faces and the cocrystallization rate of CL-20 and HMX is the highest in EA solvent. The XRD and SEM characterization results are consistent with the theoretical calculations. The present work on the effects of solvent on CL-20/HMX cocrystals is beneficial for understanding the mechanism of the growth of energetic cocrystal materials. It is helpful in selecting more suitable theoretical and experimental conditions and makes access to excellent cocrystals more efficient.

Abstract:To have a better understanding on molecular mechanism of the crystal morphology manipulation, the effect of solvents on morphology of crystals was investigated by using sulfamerazine (SMR) as model compound, through combination of experimental investigations and molecular simulations. The modified attachment energy model was employed to build the crystal morphology model in different solvents. Hirshfeld surfaces and 2-D fingerprint plots were used to investigate the solute-solute interactions, and molecular dynamics simulation was employed to investigate the interactions between solvent molecules and different crystal faces. Function of factors such as surface structure, roughness and the radial distribution function were also considered and discussed. The results suggested these factors are important to crystal habits since they could affect the adsorption competition between the solute and solvent molecules. It was found that the method developed in this work could be used to predict the morphology of SMR and could guide the selection of crystallization solvents for obtaining target morphology.

XO-PBC: An accurate and efficient method for molecular crystals.

Abstract:In this work, we propose the XO-PBC method, which combines the eXtended ONIOM method (XO) with the periodic boundary condition (PBC) for the description of molecular crystals. XO-PBC tries to embed a finite cluster cut out from the solid into the periodic environment, making it feasible to employ advanced molecular quantum chemistry methods, which are usually prohibitively expensive for direct PBC calculations. In particular, XO-PBC utilizes the results from force calculations to design the scheme to fragment the molecule when crystals are made of large molecules and to select cluster model systems automatically consisting of dimer up to tetramer interactions for embedding. By applying an appropriate theory to each model, a satisfactory accuracy for the system under study is ensured, while a high efficiency is achieved with massively parallel computing by distributing model systems onto different processors. A comparison of the XO-PBC calculations with the conventional direct PBC calculations at the B3LYP level demonstrates its accuracy at substantially low cost for the description of molecular crystals. The usefulness of the XO-PBC method is further exemplified, showing that XO-PBC is able to predict the lattice energies of various types of molecular crystals within chemical accuracy (< 4 kJ/mol) when the doubly hybrid density functional XYG3 is used as the target high level and the periodic PBE as the basic low level. The XO-PBC method provides a general protocol that brings the great predictive power of advanced electronic structure methods from molecular systems to the extended solids.

Abstract:Rationalization of the relationship between solvents and crystal habit is of great importance to the development of crystal engineering. In this paper, the impact of different solvents on vanillin habit was investigated. The results show that the crystal habit of vanillin is closely related to the type of solvent, it was found that vanillin crystals were improved to the short and thick plate in organic acid, and the antisolvent water elongates crystals to needles, in contrast, alkane solvents could not change its habit. The molecular assembly preference of vanillin in solution and crystal was investigated by proton nuclear magnetic resonance, electrostatic potential map, noncovalent interaction plot, and crystal structure analysis. The results suggest that vanillin molecules prefer to assemble by a head-to-tail motif via the intermolecular H-bond. The underpinning mechanism of the solvent effect on the vanillin habit was further rationalized by crystal surface analysis and molecular adsorption analysis. “Half-parallel-half-intercepted” and “all-intercepted” H-bonding patterns were observed on the lateral and capped faces, respectively. Different adsorption preferences of propionic acid, water, and n-hexane on the lateral and capped faces were observed via adsorption locator analysis, the adsorbed solvent molecules might result in interference on the crystal growth of corresponding faces, and hence modify the crystal habit.

Abstract:In this work, we were devoted to explore the effect of solvent polarities on the excited-state intramolecular proton transfer (ESIPT) process of 1-acetamido-4-hydroxyanthraquinone (AcHAQ) in three different polarity solvents (acetonitrile, chloroform, and cyclohexane) based on the density functional theory (DFT) and time-dependent DFT (TD-DFT) methods, and thereby regulating the distribution ratio between the dual excited-state isomers (enol and keto). The calculated geometrical parameters and infrared (IR) vibrational spectra have confirmed the excited-state intramolecular hydrogen bond (IHB) strengthening mechanism. Natural bond orbital (NBO) population analysis indicates that the intramolecular charge transfer (ICT) around IHBs has enhanced IHB, thereby triggering the ESIPT reaction. In addition, results obtained from the scanned potential energy curve (PEC) manifest that ESIPT process prefers to occur along the O 3 -H 2 ... O 1 IHB and energy barriers corresponding to the proton transfer in ACN are the lowest among all the studied solvents.

Abstract:Polymorph discrimination for a molecular crystal has long been a challenging task, which, nonetheless, is a major concern in the pharmaceutical industry. In this work, we have investigated polymorph discrimination on three different molecular crystals, tetrolic acid, oxalic acid, and oxalyl dihydrazide, covering both packing polymorphism and conformational polymorphism. To gain more understanding, we have performed energy decomposition analysis based on many-body expansion, and have compared the results from the XO-PBC method, that is, the eXtended ONIOM method (XO) with the periodic boundary condition (PBC), with those from some commonly used dispersion corrected density functional theory (DFT-D) methods. It is shown here that, with the XYG3 doubly hybrid functional chosen as the target high level to capture the intra- and short-range intermolecular interactions, and the periodic PBE as the basic low level to take long range interactions into account, the XO-PBC(XYG3:PBE) method not only obtains the correct experimental stability orderings, but also predicts reasonable polymorph energy ranges for all three cases. Our results have demonstrated the usefulness of the present theoretical methods, in particular XO-PBC, while highlighted the importance of a better treatment of different kinds of interactions to be beneficial to polymorph control.
Morphology prediction of dihydroxylammonium 5,5′-bistetrazole-1,1′-diolate (TKX-50) crystal in different solvent systems using modified attachment energy model

Abstract:In order to theoretically study the growth morphology of dihydroxylammonium 5,5′-bistetrazole-1,1′-diolate (TKX-50) crystal in different solvent systems, crystal–solvent models were established, and then molecular dynamics (MD) methods were adopted as a means to simulate particle motion. Modified attachment energy (MAE) model was employed to calculate the growth morphology of TKX-50. The simulation results demonstrate that COMPASS force field and RESP charge are suitable for molecular dynamics simulation of TKX-50. The morphologically dominant growth surfaces of TKX-50 in vacuum are (0 2 0), (0 1 1), (1 1 –1), (1 0 0) and (1 2 0), respectively. In water (H2O) and N, N-dimethylformamide (DMF) solvents, the (1 1 –1) face is the largest in the habit face, the growth rate of (0 2 0) face becomes faster. With the increase of temperature, the aspect ratios of TKX-50 crystal in DMF solvent increase, and the areas of the (1 2 0) faces decrease. In ethylene glycol /H2O mixed solvent system with volume ratio of 1/1, aspect ratio of TKX-50 is relatively small. In formic acid /H2O mixed solvents with different volume ratios (1/4, 1/3, 1/2, 1/1 and 2/1), aspect ratio of TKX-50 is relatively small when volume ratio is 1/2.

Accurate Predictions of the Nmr Parameters in Organic and Biological Crystallines

Abstract:Theoretical predictions are helpful for the spectroscopic identification of complicated organic and biological systems. For nuclear magnetic resonance (NMR) parameters, however, the chemical shift and quadrupole coupling constant (QCC) of the solid crystals are considerably affected by hydrogen bonding and van der Waals interactions from neighboring molecules and the crystal lattice leading to significant spectroscopic differences compared to isolated monomer molecules. Therefore, it is necessary to take these two factors into account for the precise predictions of chemical shifts and QCCs of solid crystals. L-alanylglycine dipeptide and nitrobenzene were selected as model crystals to demonstrate these effects. Here, the chemical shielding (CS) and QCC data were calculated based on the periodic structure model. The incorporation of intermolecular hydrogen bonding and crystal lattice effects by periodic models was found to be crucial in obtaining reliable predictions of CS and QCC values and rendering more explicit spectroscopic assignments for solid organic and biological systems.

Abstract:Predicting the probability (ability) of materials to form expected regular atomistic arrangement (single crystal) is highly desired to judge the applicability of the newly designed materials, and a theoretical model is developed for this purpose. The model that has been proved successful in predicting the ability of single-component materials to form single crystals is further extended to two-component materials. The prediction can be easily implemented via common ab initio calculations, and may be widely used in general materials design.

Abstract:The interface interaction between DMSO solution and HMX crystal was simulated by molecular dynamics (MD) method, and the morphology of HMX in DMSO solvent was predicted through the occupancy model and attachment energy (AE) model. The intramolecular and intermolecular interactions in HMX crystals were dominated by O···H/H···O contacts, accounting for 63.0%. The S value and the adsorption position of DMSO on different crystal planes of HMX have been compared, which showed that (1 0 − 1) has a deep cavity for the adsorption of solvent molecules. The density of DMSO in the z direction of the different planes showed that (0 1 1) has the largest density peak and (0 2 0) has the smallest density peak. The DMSO solvent had an attraction effect with each crystal plane of HMX. The absolute value of Eint was sorted as follows: (1 1 0) > (1 0 1) > (0 2 0) > (1 0 − 1) > (0 1 1). The DMSO solvent affected the attachment energy of each crystal plane of HMX. In our simulation system, the prediction results of the occupancy model were the most consistent with the experimental values.

Abstract:In the field of pharmaceutical crystallization, the success of polymorphic manipulation is vital to the performance of pharmaceutical formulations, and the choice of solvents might affect polymorphic outcomes directly. To better understand the relationship between molecular structure in solution and in the crystal, the solvent effect on molecular conformational evolution and polymorph control of cimetidine (CIM) was investigated. Three polymorphs (A, B, and D) of cimetidine (CIM) were prepared, and it was found that conformers selected in crystals could significantly affect crystal packing and polymorph stability. Two-dimensional nuclear Overhauser effect spectroscopy and quantum chemical calculation results reveal that conformer A is the dominant molecular conformer, although conformational distribution is strongly solvent-dependent. Furthermore, Fourier transform infrared spectroscopy and solvation free energy calculation results show that the interaction strength of CIM with solvents increases in the order isopropanol ≈ acetonitrile < methanol < ethylene glycol, affecting the difficulty of desolvation. The results imply that the interaction strength of CIM with solvents may affect the difficulty of desolvation, conformational rearrangement, and final polymorphic outcome. In the end, the potential mechanism of conformational evolution and polymorphic manipulation of CIM was presented.

Abstract:As widely acknowledged, the realm of novel organic molecules boasting extraordinary attributes pertaining to excited-state intramolecular proton transfer (ESIPT) has emerged as a captivating subject matter. In this context, our primary focus lies in delving into the excited-state behaviour exhibited by 2-benzoxazol-2-yl-4-triethylsilanylethynyl-phenol (BY4TP), an alluring derivative derived from 2-(2-hydroxyphenyl)benzoxazole (HBO). Relying on the examination of four distinct aprotic solvents with varying degrees of polarities, we can unequivocally affirm that solvent polarity exerts a profound influence on the intricate interplay of hydrogen bonding interactions, charge redistribution and reorganisation, as well as associated ESIPT phenomena by light. Through meticulous comparison and precise measurement of reaction barriers across diverse solvent environments, our groundbreaking findings indicate that lowly polar solvents serve as efficacious facilitators for promoting the occurrence of the ESIPT reaction in BY4TP fluorophore. By considering atomic-electronegativity-regulated hydrogen bonding effects and excited state behaviours for BY4TP-S and BY4TP-Se, we also present low atomic electronegativity with Se substation promotes ESIPT reaction. We ardently anticipate that this study will provide invaluable insights into the behaviour exhibited by BY4TP upon excitation and under the influence of solvent polarity and atomic electronegativity, while simultaneously paving new avenues for future research endeavours and applications encompassing novel HBO derivatives.

Abstract:In this work, a novel type of quercetin solvate of dimethylformamide (QDMF) was studied. The crystal structure was solved using single crystal X-ray diffraction and analysed using synthon analysis and other particle informatics tools ( e.g. , solvate analyser). The thermal behaviour and thermodynamic stability of the QDMF were studied experimentally using Raman spectroscopy, differential scanning calorimetry and thermogravimetric analysis. A clear relationship between the two-step desolvation behaviour of QDMF and the type, strength and directionality of the main bulk synthons characterizing the QDMF structure was observed. Additionally, the attachment energy model was used to predict the QDMF morphology, together with face-specific topology and chemical nature of each of the dominant {001}, {110}, and {200}. The {200} facet was found to be significantly rougher than the other two; whereas, the {110} was characterized by a higher percentage of exposed DMF molecules compared to the other two facets. Specific scanning electron microscopy and contact angle measurements were used to experimentally detect differences among the three facets and validate the modelling results.
