Abstract:A new method for the rapidly screening of lipase inhibitors from extracts of traditional Chinese medicine using lipase-immobilized magnetic beads coupled with high-performance liquid chromatography-mass spectrometry(LC-MS) was proposed. After the lipase-immobilized magnetic beads were prepared, they were incubated with total flavonoid of Fructus aurantii to fish the ligands of lipase. Then, the eluent of magnetic beads' screening was analyzed by LC-MS. Four compounds, as naringin, hesperidin, neohesperidin and poncirin, were identified as the potential lipase inhibitors. Furthermore, the in vitro inhibitory activities against lipase of four compounds were tested. The results showed that hesperidin, neohesperidin and poncirin exhibit lipase inhibitory activities, while poncirin was firstly reported to be a lipase inhibitor. Our research showed that active compounds could be rapidly screened from extracts of traditional Chinese medicine via lipase-immobilized magnetic beads coupled with LC-MS.
Efficient Screening of Pancreatic Lipase Inhibitors from Rheum Palmatum by Affinity Ultrafiltration-High-performance Liquid Chromatography Combined with High-Resolution Inhibition Profiling

Abstract:Introduction: Pancreatic lipase is one of the most important key targets in the treatment of obesity. Inhibition of pancreatic lipase can effectively reduce lipid absorption and treat obesity and other related metabolic disorders.Objectives: The goal of this study is the efficient screening of pancreatic lipase inhibitors in the root and rhizome of Rheum palmatum using affinity ultrafiltration-high-performance liquid chromatography (AUF-HPLC) combined with high-resolution inhibition profiling (HRIP).Methods: Potential pancreatic lipase ligands and pancreatic lipase inhibitors in ethyl acetate fraction of R. palmatum were screened using AUF-HPLC and HRIP, respectively. All screened compounds were identified by HPLC- quadrupole time-of-flight (Q-TOF)/MS. Eight compounds were screened out by both AUF-HPLC and HRIP, and six compounds were screened out by either AUF-HPLC or HRIP alone. The pancreatic lipase inhibitory activities of all screened compounds were verified by enzyme inhibition assay and molecular docking.Results: Five new potent pancreatic lipase inhibitors were discovered, namely procyanidin B5 3,3 '-di-O-gallate (IC50 = 0.06 +/- 0.01 mu M), 1,6-di-O-galloyl-2-O-cinnamoyl-beta-D-glucoside (IC50 = 12.83 +/- 0.67 mu M), 1-O-(1,3,5-trihydroxy)phenyl-2-O-galloyl-6-O-cinnamoyl-beta-D-glucoside (IC50 = 17.84 +/- 1.33 mu M), 1,2-di-O-galloyl-6-O-cinnamoyl-beta-D-glucoside (IC50 = 18.39 +/- 1.52 mu M), and 4-(4 '-hydroxyphenyl)-2-butanone-4'-O-beta-D-(2"-O-galloyl-6"-O-cinnamoyl)-glucoside (IC50 = 2.91 +/- 0.40 mu M). It was found that procyanidin B5 3,3 '-di-O-gallate showed higher pancreatic lipase inhibitory activity than the positive control orlistat (IC50 = 0.12 +/- 0.02 mu M).Conclusion: The combination of affinity ultrafiltration-high-performance liquid chromatography (AUF-HPLC) and high-resolution inhibition profiling (HRIP) could reduce the risk of false-negative screening and missed screening and could achieve more efficient screening of bioactive compounds in complex natural products.
Enzyme immobilized on polyamidoamine-coated magnetic microspheres for α-glucosidase inhibitors screening from Radix Paeoniae Rubra extracts accompanied with molecular modeling.

Abstract:In this study, a method for direct screening and identification of α-glucosidase inhibitors (AGIs) from extracts of natural products was established based on polyamidoamine (PAMAM) coated magnetic microspheres. A facile route to synthesize the magnetic PAMAM was employed and α-glucosidase was successfully covalently attached to its surface through cross linking of glutaraldehyde. Using the enzyme-loaded magnetic microspheres, potential inhibitors were fished out from crude extracts directly, followed by structure confirmation. The inhibitory activities of the screened components were further investigated by the enzyme-loaded magnetic microspheres. The Fe3O4 @PAMAM@α-Glu microspheres displayed favorable dispersibility, fast magnetic separation, large enzyme binding amount (42.9 μg•mg−1) and high enzyme activity. Moreover, the α-glucosidase on the surface of PAMAM coating maintained high storage stability and remarkable reusability. Taking advantage of specific interaction of the α-glucosidase with AGIs, the materials could selectively capture a known AGI (+)-catechin under the interference of an inactive compound salicylic acid, with a binding capacity as high as 15.4%. Additionally, using the Fe3O4 @PAMAM@α-Glu microspheres in the inhibition assay, the enzymatic reaction could be stopped by magnetic separation instead of the traditional addition of Na2CO3 solution, which not only eliminated the disturbance of termination reagent to the results, but also reused the immobilized α-glucosidase. The screening and inhibitory activity verification of potential ligands in Radix Paeoniae Rubra (“Chi-shao” in Chinese) extracts were achieved by using Fe3O4 @PAMAM@α-Glu microspheres, demonstrating practical applicability of our method. Therefore, the magnetic PAMAM-based screening approach could be a feasible and alternative strategy for discovering enzyme inhibitors from natural product extracts.

Abstract:It is integral to establish methods to investigate inhibitory activity and screen inhibitors to α-glucosidase (α-Glu) from complex natural products. Here, α-Glu immobilized paper-based bioreactor platform was firstly developed for simultaneous ligand screening and visual ratiometric evaluating inhibitory activity. N-terminus of α-Glu was anchored on paper with chitosan and glutaraldehyde as cross-linking reagents. Smaller Michaelis constant (K m) for immobilized α-Glu (10.64 mM) than free α-Glu (15.63 mM) indicated higher catalytic activity. Sample (5 μL) was dropped on α-Glu immobilized paper for incubation, potent inhibitors would complex with α-Glu, while unbound compounds could be washed off. By chromatographic analyzing eluents from α-Glu and inactive α-Glu immobilized paper, potent inhibitors could be screened out. Simultaneously, activity of immobilized α-Glu was evaluated by fabricating visual ratiometric fluorescent system using ascorbic acid, Cu2+, and o-phenylenediamine without the interference of complex matrix. The developed platform was validated by model mixtures (positive control acarbose, IC50 0.88 mg/mL; and negative control gallic acid, negligible inhibition at 5 mg/mL), and applied for real crude extracts. α-Glu inhibitory activities were visually detected for Duzhong leaves and navel orange peels (43.6 % and 26.0 % inhibition at 10 mg/mL), and four flavonoids were screened, identified, and verified as α-Glu inhibitors. Further, molecular docking studies revealed the possible inhibitory mechanisms, which came from the significant affinity interaction by forming hydrogen bonds. Results suggested the convenience, specificity, reliability, and efficiency of the proposed method.

Abstract:Identifying medicinally relevant compounds from natural resources generally involves the tedious work of screening plants for the desired activity before capturing the bioactive molecules from them. In this work, we created a paper-based ligand fishing platform to vastly simplify the discovery process. This paper-based method exploits the enzymatic cascade reaction between α -glucosidase (GAA), glucose oxidase (GOx), and horseradish peroxidase (HRP), to simultaneously screen the plants and capture the GAA inhibitors from them. The designed test strip could capture ligands in tandem with screening the plants, and it features a very simply operation based on direct visual assessment. Multiple acylated flavonol glycosides from the leaves of Quercus variabilis Blume were newly found to possess GAA inhibitory activities, and they may be potential leads for new antidiabetic medications. Our study demonstrates the prospect of the newly discovered GAA ligands as potential bioactive ingredients as well as the utility of the paper-based ligand fishing method.

Abstract:α-Glucosidase plays important roles in the digestion and absorption of carbohydrates in the small intestine. The inhibition of α-glucosidase is regarded as a potential way to treat diabetes. We established an approach to screening α-glucosidase inhibitors from medicinal plants using enzyme-coated magnetic bead. Using 1-(3-dimethyl-aminopropyl)-3-ethylcarbodiimide and N-hydroxysuccinimide as reaction reagents, α-glucosidase was immobilized on the magnetic beads by covalent linkage. The conjugation of α-glucosidase to the magnetic beads was characterized using scanning electron microscope and X-ray diffractometer. The proposed approach was applied in fishing potential α-glucosidase inhibitors from extract of Morus alba, a Chinese medicinal plant. The structures of potential active compounds were identified via liquid chromatography-mass spectrometry and nuclear magnetic resonance. The results demonstrated that two flavonoids (isoquercitrin and astragalin) could bind to α-glucosidase, which was confirmed via conventional α-glucosidase inhibitory assay. Our findings suggested that enzyme-coated magnetic beads may be suitable for discovering active compounds from medicinal plants.

Abstract:A colorimetric method has been established for α-glucosidase activity assay and its inhibitor screening. The method is based on the specific recognition between 1,4-phenylenediboronic acid (PDBA) and 4-aminophenyl-α- d -glucopyranoside (pAPG), which may induce aggregation of pAPG-functionalized gold nanoparticles (AuNPs) to achieve color change of the test solution. Because pAPG is the substrate of α-glucosidase, the aggregation of AuNPs will be influenced by α-glucosidase since there is no coordination reactivity between PDBA and 4-aminobenzene, the hydrolyzed product of pAPG catalyzed by the enzyme. Therefore, a simple and easily-operated colorimetric method for the assay of α-glucosidase activity can be developed. Under the optimized experimental conditions, the ratios of absorbance at a wavelength of 650 nm to that at 520 nm vary linearly with the α-glucosidase activity within a range from 0.05 to 1.1 U/mL with a lowest detection limit of 0.004 U/mL. Moreover, using the proposed method, the inhibition effect of gallic acid and quercetin on α-glucosidase activity can be tested with IC 50 values of 1.16 mM and 1.82 μM, respectively. Thus, the method has a great potential not only for the detection of α-glucosidase activity, but also for the screening of its inhibitors.

Abstract:This study constructed a cellulose membrane (CM)-based enzymatic colorimetric biosensor with high storage, long-term stability, and rapid screening. CM was oxidized by sodium periodate (OCM), and the C2-C3 position hydroxyl group (–OH) on the CMs was oxidized to the aldehyde group (–CHO). The amino group (–NH2) of α-glu and the –CHO on OCM immobilized α-glu reacted through a simple Schiff base reaction to obtain Schiff-based CM (SCM), which could screen AGIs rapidly within only 80 sec. The screening mechanism was that 4-Nitrophenyl α-D-glucopyranoside (PNPG) under the catalysis of α-glu produces yellow 4-Nitrophenyl (PNP), which through physical absorption immobilized on SCM immediately renders the SCM colorless to yellow. When acarbose was added, it was competitively bound to the active site of α-glu so that α-glu could not catalyze PNPG to produce PNP, thereby reducing the color intensity of SCM. SCM inherited CM's highly loose interpenetrating porous morphology was exhibited by FESEM results, providing many channels and sites for immobilizing α-glu. The VLOD of SCM is 5 × 10−4 mM in the linear range of acarbose from 0 to 0.6 mM, and the IC50 was 16.77 μM. After 120 days of storage at 4 °C, the color intensity was lost less than 10 %.

Abstract:Agrimonia pilosa Ledeb (APL) is a significant source of inhibitors for α-glucosidase, which is an essential target enzyme for the treatment of type 2 diabetes, cancer and acquired immune deficiency syndrome. Ligand fishing is a suitable approach for the highly selective screening of bioactive substances in complex mixtures. Yet it is unable to conduct biomedical imaging screening, which is crucial for real-time identification. In this case, a bioanalytical platform combining magnetic fluorescent ligand fishing and in-situ imaging technique was established for the screening and identification of α-glucosidase inhibitors (AGIs) from APL crude extract, utilizing α-glucosidase coated CuInS2/ZnS-Fe3O4@SiO2 (AG-CIZSFS) nanocomposites as extracting material and fluorescent tracer. The AG-CIZSFS nanocomposites prepared through solvothermal and crosslinking methods displayed fast magnetic separation, excellent fluorescence performance and high enzyme activity. The tolerance of immobilized enzyme to temperature and pH was stronger than that of free enzyme. Prior to proof-of-concept with APL crude extract, a number essential parameters (glutaraldehyde concentration, immobilized time, enzyme amount, reaction solution pH, incubation temperature, incubation time, percentage of methanol in eluen, elution times and eluent volume) were optimized using an artificial test mixture. The fished ligands were identified by UPLC-MS/MS and their biological activities were preliminarily evaluated by real-time cellular morphological imaging of human colon carcinoma (HCT-116) cells based on confocal laser scanning microscope (CLSM). Their α-glucosidase inhibitory activities were further verified and studied by classical pNPG method and molecular docking. The isolated compounds exhibited significant α-glucosidase inhibitory activities with a IC50 value of 11.57 µg·mL-1. Six potential AGIs including tribuloside, ivorengenin A, tormentic acid, 1β, 2β, 3β, 19α-Tetra hydroxyurs-12-en-28-oic acid, corosolic acid and pomolic acid were ultimately screened out and identified from APL crude extracts. The proposed approach, which combined highly specific screening with in-situ visual imaging, provided a powerful platform for discovering bioactive components from multi-component and multi-target traditional Chinese medicine (TCM).

α-Glucosidase immobilization on the metal–organic framework composite membrane for enzyme inhibitor screening

Abstract:In this study, a novel screening approach is presented, merging capillary electrophoresis analysis with an enzymatic assay employing immobilized α-glucosidase. A metal–organic framework composite membrane was devised for α-glucosidase immobilization. Employing polyvinylidene fluoride (PVDF) as the substrate material, it was functionalized with dopamine, creating a polydopamine composite layer. This layer acted as a covalent linker, anchoring Zn 2+ , which subsequently chelated with the organic ligand 2-ethylimidazole, leading to the self-assembly of a three-dimensional porous framework ZIF-8. This structure facilitated the in situ encapsulation of α-glucosidase. Characterization techniques including scanning electron microscopy, Fourier transform infrared-attenuated total reflectance spectroscopy, X-ray photoelectron spectroscopy, and X-ray diffraction validated the composite material's structure. A thorough investigation of various parameters affecting immobilization was conducted. The immobilized α-glucosidase exhibited enhanced pH tolerance, thermal stability, and remarkable reusability, retaining 79.3% of its original activity over 10 cycles, with a batch-to-batch reproducibility of 5.9% ( n = 5). Its Michaelis–Menten constant ( K m ) was determined to be 2.85 mM. Application of this immobilized enzyme in screening potential inhibitors from 13 traditional Chinese medicines revealed Sanguisorba radix as the most potent inhibitor. This study underscores the potential of this immobilized enzyme screening strategy for identifying enzyme inhibitors in traditional Chinese medicines.

Abstract:In this study, a ligand fishing method for the screening of α‐glucosidase inhibitors from Ginkgo biloba leaf was established for the first time using α‐glucosidase immobilized on the magnetic metal‐organic framework. The immobilized α‐glucosidase exhibited enhanced resistance to temperature and pH, as well as good thermal stability and reusability. Two ligands, namely quercitrin and quercetin, were screened from Ginkgo biloba leaf and identified by ultra‐high performance liquid chromatography‐tandem mass spectrometry. The half‐maximal inhibitory concentration values for quercitrin and quercetin were determined to be 105.69 ± 0.39 and 83.49 ± 0.79 μM, respectively. Molecular docking further confirmed the strong inhibitory effect of these two ligands. The proposed approach in this study demonstrates exceptional efficiency in the screening of α‐glucosidase inhibitors from complex natural medicinal plants, thus exhibiting significant potential for the discovery of antidiabetic compounds.

Abstract:Herein, a fluorometric and colorimetric dual-mode assay platform used for α-glucosidase (α-Glu) activity sensing based on aggregation-induced emission enhancement (AIEE) of AuNCs was developed for the first time. The quantum yield (QY) and fluorescence lifetime of AuNCs were successfully ameliorated by Ce 3+ -triggered AIEE (Ce@AuNCs). Subsequently, on the basis of the inner filter effect (IFE) and dynamic quenching effect (DQE) between 2,6-dichlorophenolindophenol (DCIP) and Ce@AuNCs as well as the reduction of DCIP by ascorbic acid (AA) generated from α-Glu-catalyzed hydrolysis of L -ascorbic acid-2-O-α- D -glucopyranosyl (AA2G), the marriage of fluorometric and colorimetric modes applied for α-Glu activity monitoring was achieved. Besides, the feasibility of this dual-mode sensing system was confirmed by the assays versus potential interfering substances and in real samples. In particular, this system was further applied to evaluate natural α-Glu inhibitors (AGIs) including luteolin, apigenin, and hesperidin. Overall, the multi-mode optical sensor newly designed here has the potential for the accurate discovery of natural anti-diabetes drugs and the therapy of diabetes.

Abstract:In this work, a simple and rapid screening strategy was developed combining capillary electrophoresis analysis with enzymatic assay based on immobilized α-glucosidase. For α-glucosidase immobilization, magnetic core-shell metal-organic frameworks composite (Fe3O4@) was fabricated by a step-by-step assembly method, and α-glucosidase was in situ encapsulated in crystal lattice of ZIF-8. The composite was characterized by transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction and vibrating sample magnetometer. After immobilization, α-glucosidase exhibited enhanced tolerance to temperature and pH, and its reusability was greatly improved with 74% of initial enzyme activity after being recycled 10 times. The Michaelis-Menten constant of immobilized enzyme was calculated to be 0.47 mM and its inhibition constant and IC50 for acarbose were 0.57 µM and 0.18 μM, respectively. The immobilized enzyme was subsequently applied to inhibitor screening from 14 TCMs, and Rhei Radix et Rhizoma was screened out. Among the commercially available 10 components presented in Rhei Radix et Rhizoma, gallic acid, (+)-catechin and epicatechin exhibited the strongest inhibitory effect on α-glucosidase. Their binding sites and modes with α-glucosidase were simulated via molecular docking to further verify the inhibition screening assay results. The positive results indicated that the Fe3O4@ screening strategy may provide a new avenue for discovering enzyme inhibitors from TCMs.

Abstract:A fluorescence method was established for a α-glucosidase activity assay and inhibitor screening based on β-cyclodextrin-coated quantum dots. p-Nitrophenol, the hydrolysis product of the α-glucosidase reaction, could quench the fluorescence of β-cyclodextrin-coated quantum dots via an electron transfer process, leading to fluorescence turn-off, whereas the fluorescence of the system turned on in the presence of α-glucosidase inhibitors. Taking advantage of the excellent properties of quantum dots, this method provided a very simple, rapid and sensitive screening method for α-glucosidase inhibitors. Two α-glucosidase inhibitors, 2,4,6-tribromophenol and acarbose, were used to evaluate the feasibility of this screening model, and IC50 values of 24 μM and 0.55 mM were obtained respectively, which were lower than those previously reported. The method may have potential application in screening α-glucosidase inhibitors.
An On-Line Detection System for Screening Small Molecule Inhibitors of Α-Amylase and Α-Glucosidase in Prunus Mume.

Abstract:High-throughput screening of inhibitors from natural products is an efficient approach to target key enzymes in diabetes progression. In this study, an on-line detection system was established for the first time to rapidly screen inhibitors of alpha-amylase and alpha-glucosidase from Prunus mume . Among 28 identified compounds, 26 and 21 compounds showed strong inhibitory effect against alpha-amylase and alpha-glucosidase, respectively. Their inhibitory effects were validated by in vitro enzyme assay and fluorescence quenching which demonstrated that these inhibitors effectively interfered enzyme active sites. The inhibition kinetics suggested that chemical structures are of great importance for interfering the enzyme structures and their microenvironment polarity. Among evaluated compounds, isorhamnetin-3-O-glucoside (19) showed the strongest binding activities to alpha-amylase and alpha-glucosidase (6.34 x 10(6).nmol(-1) and 6.28 x 10(6).nmol(-1) , respectively) by the on-line detection system. Its IC50 values were 0.16 +/- 0.06 and 0.09 +/- 0.01 mu M against alpha-amylase and alpha-glucosidase, respectively. 19 gave a much higher K-i for alpha-amylase (0.1307 mM) than alpha-glucosidase (0.0063 mM), indicating its selectivity towards alpha-glucosidase. This reported method was rapid and reliable to identify prototype inhibitors against key enzymes in diabetes, and thus might serve as a general platform to screen enzyme inhibitors from natural products. Development of High Performance Liquid Chromatography with Immobilized Enzyme Onto Magnetic Nanospheres for Screening Enzyme Inhibitor

Abstract:A novel-immobilized enzyme strategy created by magnetic nanospheres for monitoring enzyme activity and screening inhibitors followed by high performance liquid chromatography (HPLC) has been demonstrated. Through the reaction of the aldehyde groups with amine groups, α-glycosidase was simply and stably immobilized onto magnetic nanospheres by the cross-linking agent glutaraldehyde. In order to profiling the activity of the immobilized α-glucosidase, the natural substrate was hydrolyzed by it and the yield of product was determined by HPLC. Compared with traditional bioassay approach, the prepared immobilized α-glucosidase displays a high activity and stability which allows it to be easily reused for 10 times. Enzyme inhibition assays by known inhibitor glucobay and three candidate traditional Chinese medicines (TCMs) were then investigated using a similar methodology. This assay was able to readily detect the change of the immobilized enzyme activity based on measuring a decrease of product formation using HPLC. The approach is general and offers many attractive advantages including easy product isolation, inexpensive cost, and high efficiency in terms of reagent consumption.
Luminescent solid-state carbon dots synthesized based on the one-step hydrothermal method for the immobilization of α-glucosidase and imaging screening of enzyme inhibitors

Abstract:Modern pharmacology studies have demonstrated that α-glucosidase is closely associated with a variety of metabolic disorders. Natural herbs have emerged as possible sources of α-glucosidase inhibitors (AGIs) in recent years. Thus far, highly selective screening of biologically active chemicals in complicated mixtures using ligand fishing approaches has not been able to achieve biomedical imaging, which is critical for real-time identification. Hence, this paper presented a novel technique for screening potential AGIs in Scutellaria baicalensis by utilizing fluorescent carbon dots that have been immobilized with α-glucosidase (α-glu-NH 2 -CDs). The synthesized carbon dots materials exhibited excellent fluorescence properties, biocompatibility, and an abundance of functional groups, as demonstrated by the utilization of several techniques. The activity of α-glucosidase covalently linked to the material under ideal immobilization conditions was 86.81% of that of the free form. The immobilized α-glucosidase also exhibited great temperature stability, pH stability and reusability according to an examination of its enzymatic characteristics. As a result, three ligands were identified as potential AGIs, i.e., baicalein, wogonin and scullcapflavone II by UPLC-MS/MS. The molecular docking and enzyme inhibition test indicated that the isolated compounds exhibited significant α-glucosidase inhibitory activities. Moreover, the proliferation inhibitory activity of the screened ligands on HCT-116 cells was further verified morphologically using in situ imaging technique. These results strongly suggested that the established method, which combined ligand fishing with in situ visual imaging, not only screen and separate the potential AGIs in Scutellaria baicalensis quickly and effectively, but also provide an effective platform for screening bioactive compounds on different receptors.

Abstract:A highly sensitive colorimetric sensing strategy based on enzyme@metal-organic framework (GAA@Cu-MOF) and IrO2/MnO2 nanocomposite was exploited innovatively for screening of α-glucosidase (GAA) inhibitors. IrO2/MnO2 nanocomposite exhibits excellent oxidase-mimicking activity which can directly catalyze the oxidation of 3,3,5,5,-tetramethylbenzidine (TMB) into a blue product with an absorption maximum at 652 nm. And GAA@Cu-MOF can decompose L-ascorbic acid-2-O-α-D-glucopyranosyl (AAG) to ascorbic acid (AA). The produced AA can destroy the IrO2/MnO2 nanocomposite and reduce its oxidase-like activity. However, the generation of AA is restricted when GAA inhibitors are added to the system, which allows the oxidase-like activity of the IrO2/MnO2 nanocomposite to be maintained. In view of this, a method for screening of GAA inhibitors was developed. In addition to enhancing the stability of GAA, the method can also effectively avoid the potential interference of H2O2 in the screening process of GAA inhibitors, which helps to improve the sensitivity of the method. Therefore, highly sensitive determination for acarbose and ascorbic acid are achieved with detection limits of 6.27 nM and 1.23 μM, respectively. The proposed method was successfully applied to screen potential GAA inhibitors from oleanolic acid derivatives.

Abstract:The detection of α-glucosidase (AGLU) inhibitors is critical for the screening of anti-diabetic drugs. In this study, we first demonstrate a liquid crystal (LC)-based assay to detect anti-diabetic drugs. When the solution of non-ionic surfactant dodecyl α-D-glucopyranoside (DDG) is introduced onto the LCs, self-assembled monolayers are formed at the aqueous/LC interface, which induces the perpendicular orientation of LC molecules at the interface. Accordingly, the LCs show a dark image. However, when a mixture of AGLU and DDG is introduced onto the LCs, a bright image is observed due to enzymatic hydrolysis of DDG by AGLU, which prevents formation of the surfactant monolayers and results in the planar or tilted orientation of LC molecules at the interface. Using the LC-based AGLU assay, the detection of three AGLU inhibitors that are popular anti-diabetic drugs including acarbose, migliol, and voglibose is demonstrated. In addition, the linear detection ranges and the detection limits of these drugs are also determined. This method provides a simple and powerful strategy to rapidly and accurately detect AGLU inhibitors, which is very promising in the applications of screening anti-diabetic drugs.
