Abstract:During the process of enzymatic hydrolysis of cellulose,the conversion of cellobiose to glucose is restricted due to the deficiency of cellobiase in the cellulase system.The accumulation of the hydrolysis product,cellobiose and glucose,caused severe feedback inhibition on the cellulase catalytic reaction.An effective solution was presented to resolve the problems.The spores containing cellobiase from Asperillus niger ZU-07 and the cells of Lactobacillus delbrium were entrapped together into calcium alginate gels to form a coimmobilized cell system,which was used to convert cellulosic hydrolysate to lactic acid.Analysis indicated that the cellobiose in the cellulosic hydrolysate could be hydrolyzed to glucose quickly by the cellobiase in the coimmobilized cells,and that the glucose could be converted to lactic acid effectively by the immobilized L.delbrueckii cells.The optimum reaction temperature of the coimmobilized cells was 48 ℃,and the produced lactic acid concentration by the coimmobilized cells reached 41.71 g/L after 48 h synergetic reaction in the cellulosic hydrolysate containing 49.40 g/L reducing sugar.The coimmobilized cells were quite stable and efficient,and could be repeatedly used for at least 12 batches.
Synergetic Saccharification and Lactic Acid Fermentation by Coimmobilized Cells Using Cellulosic Materials

Abstract:In order to improve the conversion efficiency of lactic acid from cellulose,the spores containing cellobiase with high activity from Aspergillus niger ZU-07 and the cells of Lactobacillus delbrium were embedded together into calcium alginate gel beads,and the formed coimmobilized cell system was coupled with the enzymatic hydrolysis of cellulosic material to set up a novel coupling bioreactor.The research results showed that the cellobiose in the hydrolysate was converted to glucose rapidly by the cellobiase in the coimmobilized cells,and the glucose was utilized rapidly by L.delbrium to produce lactic acid.Therefore,the feedback inhibition on cellulase caused by cellobiose and glucose was eliminated effectively.When the hydrolysis temperature and fermentation temperature were controlled at 50℃ and 48℃, respectively,and the volume ratio of the coimmobilized cells to the column reactor was 40%,the produced lactic acid concentration and the yield of lactic acid from cellulose were 55.7 g·L-1 and 91.5%,respectively.Under the fed-batch process,the final concentration of lactic acid and productivity increased to 106.7 g·L-1 and 1.270 g·L-1·h-1,respectively,while the dosage of cellulase per gram substrate reduced by 25%.
Lactic acid production from cellulosic waste by immobilized cells of Lactobacillus delbrueckii

Abstract:Industrial waste corn cob residue (from xylose manufacturing) without pretreatment was hydrolyzed by cellulase and cellobiase. The cellulosic hydrolysate contained 52.4 g l −1 of glucose and was used as carbon source for lactic acid fermentation by cells of Lactobacillus delbrueckii ZU-S2 immobilized in calcium alginate gel beads. The final concentration of lactic acid and the yield of lactic acid from glucose were 48.7 g l −1 and 95.2%, respectively, which were comparative to the results of pure glucose fermentation. The immobilized cells were quite stable and reusable, and the average yield of lactic acid from glucose in the hydrolysate was 95.0% in 12 repeated batches of fermentation. The suitable dilution rate of continuous fermentation process was 0.13 h −1 , and the yield of lactic acid from glucose and the productivity were 92.4% and 5.746 g l −1 h −1 , respectively. The production of lactic acid by simultaneous saccharification and fermentation (SSF) process was carried out in a coupling bioreactor, the final concentration of lactic acid was 55.6 g l −1 , the conversion efficiency of lactic acid from cellulose was 91.3% and the productivity was 0.927 g l −1 h −1 . By using fed-batch technique in the SSF process, the final concentration of lactic acid and the productivity increased to 107.6 g l −1 and 1.345 g l −1 h −1 , respectively, while the dosage of cellulase per gram substrate decreased greatly. This research work should advance the bioconversion of renewable cellulosic resources and reduce environmental pollution.
Enzymatic Hydrolysis of Corncob and Ethanol Production from Cellulosic Hydrolysate

Abstract:Enzymatic hydrolysis of corncob and ethanol fermentation from cellulosic hydrolysate were investigated. After corncob was pretreated by 1% H2SO4 at 108°C for 3h, the cellulosic residue was hydrolyzed by cellulase from Trichoderma reesei ZU-02 and the hydrolysis yield was 67.5%. Poor cellobiase activity in T. reesei cellulase restricted the conversion of cellobiose to glucose, and the accumulation of cellobiose caused severe feedback inhibition to the activities of β-1,4-endoglucanase and β-1,4-exoglucanase in cellulase system. Supplementing cellobiase from Aspergillus niger ZU-07 greatly reduced the inhibitory effect caused by cellobiose, and the hydrolysis yield was improved to 83.9% with enhanced cellobiase activity of 6.5CBUg−1 substrate. Fed-batch hydrolysis process was started with a batch hydrolysis containing 100gl−1 substrate, with cellulosic residue added at 6 and 12h twice to get a final substrate concentration of 200gl−1. After 60h of reaction, the reducing sugar concentration reached 116.3gl−1 with a hydrolysis yield of 79.5%. Further fermentation of cellulosic hydrolysate containing 95.3gl−1 glucose was performed using Saccharomyces cerevisiae 316, and 45.7gl−1 ethanol was obtained within 18h. The research results are meaningful in fuel ethanol production from agricultural residue instead of grain starch.

Abstract:Various types of lignocellulosic wastes extensively used in biofuel production were provided to assess the potential of EXLX1 as a cellulase synergist. Enzymatic hydrolysis of natural wheat straw showed that all the treatments using mixtures of cellulase and an optimized amount of EXLX1, released greater quantities of sugars than those using cellulase alone, regardless of cellulase dosage and incubation time. EXLX1 exhibited different synergism and binding characteristics for different wastes, but this can be related to their lignocellulosic components. The cellulose proportion could be one of the important factors. However, when the cellulose proportion of different biomass samples exhibited no remarkable differences, a higher synergism of EXLX1 is prone to occur on these materials, with a high proportion of hemicellulose and a low proportion of lignin. The information could be favorable to assess whether EXLX1 is effective as a cellulase synergist for the hydrolysis of the used materials. Binding assay experiments further suggested that EXLX1 bound preferentially to alkali pretreated materials, as opposed to acid pretreated materials under the assay condition and the binding preference would be affected by incubation temperature.
Valorization of residual lignin from corncob residues into thermosensitive lignin-based "molecular glues" for recycling cellulase

Abstract:The cost of enzymolysis is a major bottleneck for the industrialisation of lignocellulosic enzymatic hydrolysis technology, and recycling cellulase can reduce this cost. Herein, a sulfobetaine prepolymer (CPS) with terminal chlorine was grafted onto enzymatic hydrolysis residual lignin (EHL) from corncob to construct thermosensitive lignin-based "molecular glues" (lignin-based sulfobetaine polymers, L-CPS) that were used to recover and recycle cellulase. L-CPS2 (1.0 g/L) was added to the corncob residue (CCR) enzymolysis system (50 °C, pH 4.5). After hydrolysis, L-CPS2 co-precipitated with cellulase through hydrophobic binding when cooling to 25 °C. This co-precipitation decreased the amount of cellulase by 40 %. In summary, a thermally responsive lignin-based molecular glue was constructed for green recycling of cellulase, providing a new approach to decreasing the cost of lignocellulosic enzymolysis and high value utilisation of industrial lignin.

Abstract:Natural lignocellulose is used as raw material to produce chemicals through biological transformation. The accessibility of cellulase to substrate was also one of the limiting factors of industrial production. Polyethylene glycol (PEG) can be used as additive in enzymatic hydrolysis of lignocellulose. In this study, enzymatic activity on simultaneous or non-simultaneous addition of PEG 4000 was investigated, and the partly delignified rice straw, the rice straw and filter paper were used as substrates, respectively. Enzyme activity was characterized by reducing sugar concentration in supernatant which was quantified through 3,5-dinitrosalicylic acid (DNS) method. Addition of PEG has been proven to facilitate enzymatic hydrolysis of lignocellulosic materials. Furthermore, PEG had the positive effect on hydrolytic enzyme activity of pure cellulose materials without lignin. Changes in lignocellulose materials have been observed by inverted microscope and Scanning electron microscope (SEM), and no chemical changes were shown by Fourier transform infrared spectroscopy (FTIR). The promotion of PEG on enzymatic hydrolysis of pure cellulose materials may be due to its loose physical structure and similar phenomenon in natural lignin materials. PEG loosens the physical structure of lignocellulose, thus facilitating enzymatic hydrolysis. This may be a new idea to optimize the lignocellulosic enzymatic hydrolysis process.

Abstract:<p>The acquisition during biomass saccharification of elevated levels of fermentable sugars with lower cellulase concentration is central to ensuring an economically viable and industrially relevant hydrolytic process. Thus, using a new cellulase preparation (LT4) at low cellulase loading (2 mg protein/g dried substrate), this study assessed the possible boosting effect of integrating accessory enzymes and additives on high-solids hydrolysis of sugarcane bagasse via fed-batch feeding. Hydrolysis which commenced with initial 8% solids loading and subsequent substrate feeding of 4% solids at 6 h, 18 h, and 24 h respectively, proved optimal for the 20% high-solids saccharification producing 158 g/L total sugars and 83% glucose yield after 72 h with the combined optimized additives and accessory enzymes. The results obtained indicate that the integration of accessory enzymes and additives offers a benignant approach to minimizing the enzyme load and cost of high solids saccharification of lignocellulosic heteropolymers while also boosting enzyme hydrolytic performance.</p>

Abstract:Porous particle characteristics of lignocellulosic biomass and the water state distribution surrounding particles determine enzyme/solute transfer, while inherently affecting the liquefaction and hydrolysis reactions. For submillimeter biomass particles (<500 μm), deep eutectic solvent pretreatment tended to create additional macropores (∼4%, pore volume of 4.3 mL/g) and micropores (∼3%, pore area of 2.26 m 2 /g) at the extragranular and cell wall scales, simultaneously with reducing particle size by 15%. The pretreated biomass exhibited preferential rapid liquefaction over sugar production, leading to a ∼70% reduction in particle size only after 3 h hydrolysis. This further increased particle porosity (∼85% after 12 h hydrolysis) with the transition from large surface-pores to interior micropores. The increases in the tissue/cell-scale pore size and intraparticle area had greater significance for improving hydrolysis performance than particle size reduction. Additionally, particle liquefaction released constrained water in capillary pores and enhanced slurry fluidity; capillary water and free water became main transfer carriers during the hydrolysis. Free water activity was positively related to sugar production ( r = 0.80–0.85), and integrating it with particle component and porosity allowed for accurate prediction of hydrolysis yields ( R 2 = 0.98–0.99). Inspired by particle liquefaction, a short-time feeding strategy was proposed to reconstruct high-solid hydrolysis.

Abstract:A sensitive UCST‐type PSDM can be used to significantly improve the lignocellulosic enzymatic hydrolysis. After enzymatic hydrolysis, it can also be used to efficiently recycle cellulase from hydrolyzate, and the process is sustainable. PSDM can efficiently recover cellulase through hydrophobic coprecipitation at low temperatures, which provides a new approach for reducing the lignocellulosic enzymatic hydrolysis cost, and separating protein. Enzymatic strengthening and cellulase recovery are effective ways for improving the economy of lignocellulosic hydrolysis. A series of temperature‐responsive quaternary ammonium salt type enzymatic hydrolysis promoters (PSDM) were obtained by aqueous‐solution free radical polymerization. PSDM dissolved under lignocellulosic enzymatic hydrolysis conditions (50°C). After hydrolysis, it co‐precipitated with cellulase when cooling. PSDM had significantly improved hydrolysis and recycling enzyme properties. The efficiency of high‐solid substrate enzymatic hydrolysis of corncob residues improved to 1.2 times, and 50% of the cellulase amount was saved when adding 1.5 g/L of PSDM1 to the system. Hydrophobic binding may be the main driving force for co‐precipitation between PSDM and cellulase. In this work, regulation of glucose yield and cellulase consumption were realized by adjusting the ratio of positive and negative charges in PSDM. This work provides a new idea for reducing the cost of technology for this lignocellulosic sugar platform.

Abstract:BackgroundThe efficiency and cost of current lignocellulosic enzymes still limit the large-scale production of cellulosic ethanol in industry. Residual lignin after pretreatment severely depresses the activity of polysaccharide hydrolases and the h ydrolysis of holocellulose. If we include in hydrolase mixture construction the ligninase involved in lignin degradation, which mainly includes laccase, manganese peroxidases (MnP) and lignin peroxidase (LiP), it is feasible that this could greatly improve the fermentable sugars yield.ResultsThe psychrophilic lignocellulosic enzymes system of Cladosporium cladosporioides Ch2-2 including ligninase and polysaccharide hydrolases was suitable for selective delignification and efficient saccharification of biomass with wide thermal adaptability. The purified laccase was optimally active at 15°C and pH 3.5, exhibiting high thermostability over a broad range of temperatures (between 4 and 40°C). In addition, manganese-independent peroxidase (MIP), a special type of ligninase with the capacity to oxidize dimethyl phthalate (DMP) in the absence of H2O2 and Mn2+, was optimally active at 20°C and pH 2.5, exhibiting high thermostability over a broad range of temperatures (4 and 28°C), while depressed completely by Fe2+ and essentially unaffected by EDTA. Synergy between Ch2-2 crude enzymes and commercial xylanase obviously enhanced biomass hydrolysis, which could take the place of expensive commercial cellulase mixture. The maximum value of synergistic degree reached 4.7 at 28°C, resulting in 10.1 mg/mL reducing sugars.ConclusionsThe psychrophilic enzymes system of C. cladosporioides Ch2-2 with a different synergistic mechanism has huge potential for the enhancement of biomass hydrolysis at mesophilic and low temperatures. The application scope of the lignocellulosic enzyme cocktail could be greatly enlarged by optimizing the operation conditions specific to the characteristics of ligninase.

Optimization of Cellulase Mixture for Efficient Hydrolysis of Steam-Exploded Corn Stover by Statistically Designed Experiments.

Abstract:To improve the enzymatic hydrolytic efficiency and reduce production cost, a statistically designed experimental approach was used to optimize the composition of cellulase mixture so as to maximize the amount of glucose produced from steam-exploded corn stover (SECS). Using seven purified enzymes (cellobiohydrolases, Cel7A, Cel6A, Cel6B; endoglucanases, Cel7B, Cel12A, Cel61A; and beta-glucosidase) from Trichoderma viride T 100-14 mutant strain, a multi-enzyme mixture was constituted after screening and optimization. The final optimal composition (mol%) of the multi-enzyme mixture was Cel7A (19.8%), Cel6A (37.5%), Cel6B (4.7%), Cel7B (17.7%), Cel12A (15.2%), Cel61A (2.3%) and beta-glucosidase (2.8%). The subsequent verification experiments followed by glucose assay together with scanning electron microscopy (SEM) observation confirmed the validity of the models. The multi-enzyme mixture displayed a high performance in converting the cellulosic substrate (SECS). The amount of glucose produced (15.5mg/ml) was 2.1 times as that of the crude cellulase preparation. The results indicated that the optimized cellulase mixture is an available and efficient paradigm for the hydrolysis of lignocellulosic substrate. The enhanced cellulolytic activity displayed by the constructed cellulase mixture could be used as an effective tool for producing bioethanol efficiently from cellulose.
Enhancing for Bagasse Enzymolysis via Intercrystalline Swelling of Cellulose Combined with Hydrolysis and Oxidation

Abstract:To overcome the biological barriers formed by the lignin-carbohydrate complex for releasing fermentable sugars from cellulose by enzymolysis is both imperative and challenging. In this study, a strategy of intergranular swelling of cellulose combined with hydrolysis and oxidation was demonstrated. Pretreatment of the bagasse was evaluated by one bath treatment with phosphoric acid and hydrogen peroxide. The chemical composition, specific surface area (SSA), and pore size of bagasse before and after pretreatment were investigated, while the experiments on the adsorption equilibrium of cellulose to cellulase and reagent reuse were also performed. Scanning electron microscopy (SEM) and high-performance liquid chromatography (HPLC) were employed for microscopic morphology observations and glucose analysis, respectively. The results showed that pretreated bagasse was deconstructed into cellulose with a nanofibril network, most of the hemicellulose (~100%) and lignin (~98%) were removed, and the SSA and void were enlarged 11- and 5-fold, respectively. This simple, mild preprocessing method enhanced cellulose accessibility and reduced the biological barrier of the noncellulose component to improve the subsequent enzymolysis with a high glucose recovery (98.60%).

Abstract:By screening out total nine sugarcane cultivars that contain directly-fermentable sugars, this study examined largely varied hexoses yields from direct enzymatic hydrolysis of bagasse residues. Using two representative sugarcane bagasse substrates, this study sorted out optimal K3PO4 pretreatments by performing classic response surface methodology, which respectively extracted 58% and 63% lignin and significantly reduced cellulose CrI and DP values in two pretreated bagasse residues. These consequently led to remarkably raised cellulose accessibility by 1.4- and 3.6 folds for near-complete biomass enzymatic saccharification with hexoses yields close to 100% (% cellulose). Notably, unlike physical (hot water, steam explosion) and chemical (acid, alkali) pretreatments as previously examined, this optimal pretreatment (6.4% K3PO4, 144.0 °C, 1 h) could not significantly extract arabinose-rich hemicellulose, suggesting that almost all xylose of hemicelluloses was applicable for engineered yeast co-fermentation to achieve maximum bioethanol at 29% (% dry matter) without the necessity of solid-liquid separation, water washing, and detoxification process. Furthermore, this study collected all K3PO4 liquid waste as chemical fertilizer into rice culture for much enhanced plant growth and biomass production up to 1.5 folds. Therefore, this work has proposed a mechanism model to highlight a cost-effective and green-like strategy for high yield of bioethanol with potential value-added bioproducts by using sugarcane bagasse and other lignocellulose residues.

Abstract:Commercially available cellulase cocktails frequently demonstrate high efficiency in hydrolyzing easily digestible pretreated biomass, which often lacks hemicellulose and/or lignin fractions. However, the challenge arises with enzymatic hydrolysis of mildly pretreated lignocellulosic biomasses, which contain cellulose, hemicellulose and lignin in high proportions. This study aimed to address this question by evaluating the supplementation of a commercial cellulolytic cocktail with accessory hemicellulases and two additives (H 2 O 2 and Tween® 80). Statistical optimization methods were employed to enhance the release of glucose and xylose from mildly pretreated sugarcane bagasse. The optimized supplement composition resulted in the production of 304 and 124 mg g −1 DM of glucose and xylose, respectively, significantly increasing glucose release by 84% and xylose release by 94% compared to using only the cellulolytic cocktail. This enhancement might be attributed to a coordinated hemicellulases action degrading hemicellulose, creating more space for cellulase activity, potentially boosted by the presence of H 2 O 2 and Tween® 80. However, the addition of different concentrations of H 2 O 2 in combination with hemicellulase and Tween® 80 did not result a significant difference on sugar release, which could be attributed to the limited range of concentrations studied (5 to 65 μM). The results obtained in this study using the mix of three supplements were also compared to the addition of only hemicellulase and only Tween® 80 to the cellulolytic cocktail. A significant increase in glucose release of 39 and 41%, respectively, was observed when using the optimized combination. For xylose, the increase was 38 and 41%, respectively. This study underscores the substantial potential in optimizing enzyme cocktails for the hydrolysis of mildly pretreated lignocellulosic biomass by using enzymes and additive combinations tailored to the specific biomass composition.

Understanding of promoting enzymatic hydrolysis of combined hydrothermal and deep eutectic solvent pretreated poplars by Tween 80

Abstract:In this study, lignin blockers including non-catalytic protein and surfactants were employed to promote enzymatic digestibility of pretreated poplars. Among them, Tween 80 exhibited the most pronounced facilitation, improving the glucose yield from 26.6% to 99.6% at a low enzyme loading (10 FPU/g glucan), and readily reduced the required cellulase loading by 75%. The underlying mechanism for this remarkable improvement on glucose yields by Tween 80 was elucidated. The impacts of Tween 80 on the enzyme-lignin interaction were explored by quartz crystal microbalance analysis, revealing that the binding rate of Tween 80 on lignin surfaces was 3-fold higher than that of enzyme. More importantly, Tween 80 remarkably decreased the binding capacity and binding rate of enzyme on lignins. Furthermore, the substrate properties dominating the increase in glucose yields with Tween 80 were explored. The results facilitate to understand the underlying mechanism of the promotion of surfactants on enzymatic hydrolysis.

Isolation and Screening of Microorganisms for the Effective Pretreatment of Lignocellulosic Agricultural Wastes

Abstract:Lignocellulosic waste is the most abundant biorenewable biomass on earth, and its hydrolysis releases highly valued reducing sugars. However, the presence of lignin in the biopolymeric structure makes it highly resistant to solubilization thereby hindering the hydrolysis of cellulose and hemicellulose. Microorganisms are known for their potential complex enzymes that play a dominant role in lignocellulose conversion. Therefore, the current study was designed to isolate and screen potential microorganisms for their selective delignification ability for the pretreatment of lignocellulosic biomass. An extensive isolation and screening procedure yielded 36 desired isolates (22 bacteria, 7 basidiomycete fungi, and 7 filamentous fungi). Submerged cultivation of these desired microorganisms revealed 4 bacteria and 10 fungi with potent lignocellulolytic enzyme activities. The potent isolates were identified as Pleurotus, Trichoderma, Talaromyces, Bacillus, and Chryseobacterium spp. confirmed by morphological and molecular identification. The efficiency of these strains was determined through enzyme activities, and the degraded substrates were analyzed through scanning electron microscopy (SEM) and X-ray diffraction (XRD). Among all isolated microbes, Pleurotus spp. were found to have high laccase activity. The cellulose-decomposing and selective delignification strains were subjected to solid-state fermentation (SSF). SSF of field waste corn stalks as a single-carbon source provides Pleurotus spp. better condition for the secretion of ligninolytic enzymes. These isolated ligninolytic enzymes producing microorganisms may be used for the effective pretreatment of lignocellulosic agricultural wastes for the production of high value-added natural products by fermentation.

Abstract:An efficient enzymatic hydrolysis of lignocellulosic biomass into fermentable sugars depends greatly on the pretreatment of raw materials. In this study, a combination of dilute acid pre-extraction and chemicalassisted mechanical refining was used to pretreat wood lignocellulosic biomass for subsequent enzymatic hydrolysis. This work analyzed the surface lignin concentration, specific surface area, crystallinity, fines content, fiber length, and kink index of the resultant pulp substrates and their effects on the enzymatic hydrolysis. The results showed that the combined pretreatment significantly enhanced the enzymatic hydrolysis efficiency, and the maximum glucose conversion yield and glucose concentration were 93.32% and 21.41 g L , respectively. It is found that the surface lignin concentration, specific surface area, and fines content significantly affected the enzymatic hydrolysis.
In Situ Corn Fiber Conversion for Ethanol Improvement by the Addition of a Novel Lignocellulolytic Enzyme Cocktail

Abstract:Corn mashes have high-viscosity and high-sugar characteristics, which hinders yeast-fermentation efficiency and the ethanol yield increase. The excessive viscosity of corn mash is caused by the unutilized cellulose in corn kernel fiber. A novel lignocellulolytic enzymes cocktail with strong substrate specificity was prepared for high-viscosity, high-sugar corn mash. The in situ conversion of corn mashes with novel lignocellulolytic enzymes at the optimum cellulase dosage of 50 FPU/L resulted in about 12% increased ethanol concentration compared with the reference mash at different batch-fermentation scales. Adding the lignocellulolytic enzymes caused the greatest decrease in viscosity of corn mash and residual sugars by 40.9% and 56.3%, respectively. Simultaneously, the application of lignocellulolytic enzymes increased the value of the dried distiller’s grain with solubles (DDGS) by increasing the protein content by 5.51%. The in situ conversion of cellulose can decrease the fermentation broth viscosity and improve the rheological property, thereby improving the ethanol yield. With the same amount of material, the application of the novel enzymes cocktail can enhance the ethanol yield by more than 12%. A quarter of the ethanol yield increase was due to the further hydrolysis of starch, while three quarters to cellulose. Thus, this technology will increase the net revenue of bioethanol industrialization.
