Abstract:Herein is presented a proof-of-concept study of protease sensing that combines nontoxic silicon quantum dots (SiQDs) with Förster resonance energy transfer (FRET). The SiQDs serve as the donor and an organic dye as the acceptor. The dye is covalently attached to the SiQDs using a peptide linker. Enzymatic cleavage of the peptide leads to changes in FRET efficiency. The combination of interfacial design and optical imaging presented in this work opens opportunities for use of nontoxic SiQDs relevant to intracellular sensing and imaging.

Abstract:Amino terminal Cu(II)- and Ni(II)-binding (ATCUN) motif is included in a protein or peptide with a structural characterization comprising of a free NH2-terminus, a histidine residue in the third position and two intervening peptide nitrogens. We reported a signal-on electrochemical strategy for detection of protease based on the formation of ATCUN-Cu(II) on electrode surface. In the method, an amino residue in the native peptide substrate was replaced with a histidine. Cleavage of the sequence-specific peptide pre-immobilized on an electrode by the protease led to the presence of ATCUN motif and the formation of ATCUN-Cu(II) in the presence of Cu(II). Consequently, a voltammetric signal was observed. Contrarily, inhibition of protease by its potential inhibitor decreased the voltammetric response. To demonstrate the feasibility and sensitivity of our strategy, beta-secretase was tested as a model protease. As a result, a detection limit of 0.08 nmol L-1 was achieved. The half-maximum inhibition value (IC50) of a well-known beta-secretase inhibitor was found to be 22.6 nmol L-1, which is in agreement with the reported value. (C) 2016 Elsevier B.V. Abstract:Cu（Ⅱ） detection is important because it plays crucial role in several biological processes and ecological systems.Fluorescent techniques have attracted more and more attention in Cu（Ⅱ） detection.In this report,we contribute a novel strategy to use fluorescence spectroscopy for Cu（Ⅱ） specific detection.The specificity relies on the fact that,of the many metal cations,only Cu（Ⅱ） can catalyze the hydrolyzation of a-amino acid ester.The novelty originates from the unique aggregation-induced emission（AIE） property of the fluorescent label.We designed a model a-amino acid ester（TPE-Ala） constructed with alanine and tetraphenylethene-functionalized methanol（TPE-methanol）.In comparison with the precursor TPE-Ala, TPE-methanol has lower solubility and is easy to form aggregates in water,thereby displaying a higher fluorescent response.Thus,the Cu（Ⅱ） catalyzed hydrolyzation can be monitored by recording the fluorescence enhancement and fluorescent detection Cu（Ⅱ） is rationally achieved.
A Colorimetric Strategy for Assay of Protease Activity Based on Gold Nanoparticle Growth Controlled by Ascorbic Acid and Cu(II)-coordinated Peptide

Abstract:We developed a colorimetric method for assay of protease activity through the growth of gold nanoparticles (AuNPs) with ascorbic acid (AA) as the reducing agent. The method is based on the difference in the catalytic activity of various Cu2+ species toward AA oxidation. A mutational peptide is employed as the protease substrate, in which only one amino acid residue is replaced by the histidine residue. Specifically, HAuCl4 can be reduced into AuNPs by AA; Cu2+ ion promotes the oxidation of AA under oxygen atmosphere by a redox cycling, thus preventing the formation of AuNPs. Cleavage of the substrate peptide results in the exposure of an amino terminal Cu2+ and Ni2+-binding (ATCUN) motif in the NH2-terminus of one fragment. The resultant ATCUN peptide can sequestrate Cu2+ and thus depress the catalytic oxidation of AA. The consumption of AA is monitored by UV-vis spectra and differential pulse voltammetry. The high extinction coefficient of the generated AuNPs enables the quantitative and sensitive colorimetric analysis of protease. To demonstrate the analytical performances, p-secretase was tested as a model protease. By employing peptide-functionalized magnetic beads, p-secretase in serum was determined with a satisfactory result. This work provides valuable information for designing of novel protease biosensors. Abstract:We have established a real-time and label-free fluorescence turn-on strategy for protease activity detection and inhibitor screening via peptide-induced aggregation-caused quenching of a perylene probe. Because of electrostatic interactions and high hydrophilicity, poly-l-glutamic acid sodium salt (PGA; a negatively charged peptide) could induce aggregation of a positively charged perylene probe (probe 1) and the monomer fluorescence of probe 1 was effectively quenched. After a protease was added, PGA was enzymatically hydrolyzed into small fragments and probe 1 disaggregated. The fluorescence recovery of probe 1 was found to be proportional to the concentration of protease in the range from 0 to 1 mU/ml. The detection limit was down to 0.1 mU/ml. In the presence of a protease inhibitor, protease activity was inhibited and fluorescence recovery reduced. Moreover, we demonstrated the potential application of our method in a complex mixture sample including 1% human serum. Our method is simple, fast and cost effective.

Abstract:Copper (II) (Cu2+) and 3-nitropropionic acid (3-NPA) are harmful substances usually found in foods. Therefore, establishing rapid, sensitive, and accurate detection method for both of these substances is significant research work. A pyrene-based fluorescent sensor (PP) was synthesized to enable highly selective and sensitive simultaneous detection of Cu2+ and 3-NPA in food samples. Initially, PP showed weak fluorescence because of the photo–induced electron transfer process (PET). Upon the presence of Cu2+, the emission of PP at 454nm enhanced owing to the coordination between PP and Cu2+ which hindered the PET. Additionally, under 365nm UV irradiation, the color of the PP solution transitions from orange to bright blue, allowing for naked-eye detection. The sensor PP demonstrated high selectivity and sensitivity toward Cu2+ compared to other competitive metal ions, with the detection limit was 0.036μM, within a linear range of 0–20μM. The binding stoichiometry between PP and Cu2+ is determined to be 2:1 using the Job’s plot method, with a binding constant (Ka) of 7.2 ×103M−1/2. Furthermore, at lower pH (in the range 2.55–5.98), the emission of PP was also enhanced due to the protonation of PP which suppresses the PET process. Given its sensitive and linear response to acidic pH, PP exhibits excellent sensing performance for detecting 3-NPA, with a low detection limit of 0.32μM, surpassing previously reported methods in terms of detection sensitivity.

Abstract:Sensor formats have been developed for detecting the activity of proteolytic enzymes based on fluorescent conjugated polymer superquenching. These sensors employ a reactive peptide sequence within a tether linking a quencher to a biotin. The peptide binds to sensors containing colocated biotin-binding protein and fluorescent polymer by means of biotin-biotin binding protein interactions, resulting in a strong quenching of polymer fluorescence. Enzyme-mediated cleavage of the peptide results in a reversal of the fluorescence quenching. These assays for protease activity are simple, sensitive, fast, and have the specificity required for screening chemical libraries for novel protease inhibitors in a high-throughput screening assay environment. These assays have been demonstrated for enterokinase, caspase-3/7, and beta-secretase.

Abstract:Development of sensitive fluorescence "Turn-On" strategies for imaging enzyme activity in living cells is of disease-diagnostic importance but remains challenging. Herein, by employing a click condensation reaction and rational design of a single quenched probe Cys(StBu)-Lys(Gly-Lys(DABCYL)-Gly-Gly-Arg-Arg-Val-Arg-Gly-FITC)-CBT (1), we developed a "smart" dual quenching strategy and applied it to detect intracellular furin activity with enhanced sensitivity. At physiological conditions, 1 was subjected to reduction-controlled condensation reaction to form 1-NPs and its fluorescence intensity further dropped to 1/2.8 of its original. Upon furin cleavage in vitro, the dual quenched 1-NPs had fluorescence "Turn-On" contrast 11-fold more than that of single quenched control probe FITC-Gly-Arg-Val-Arg-Arg-Gly-Gly-Lys(DABCYL)-Gly-OH (1-P). Live cell imaging results indicated that 1 showed fluorescence "Turn-On" contrast 6.3-fold of that of 1-P for sensing intracellular furin activity. We envision that, by replacing the RVRR substrate with other enzyme-cleavable ones, our versatile "smart" dual quenching strategy could be easily adjusted for the detection (or imaging) of other intracellular enzymes' activity with enhanced sensitivity.

Abstract:Cu2+ and S2− play important roles in our environment, so it is beneficial to develop sensors capable of detecting Cu2+ and S2− in biological samples. To accomplish this goal, a new peptide-based fluorescent chemical sensor L (FITC-Ahx-His-Glu-Phe-Cys-NH2) has been designed and synthesized. It can specifically recognize Cu2+ by forming a L-Cu complex which can quench the fluorescence of L. The subsequent addition of S2− can remove Cu2+ from the L-Cu complex, thereby restoring the fluorescence in a process described as an “On-Off-On” cycle. Cu2+ and S2− can be detected by L with extremely high sensitivity and the detection limits is 32 nM and 53 nM respectively. Additionally, the sensor L can detect Cu2+ and S2− at physiological pH conditions. The result of this research is the development of a peptide-based fluorescent chemical sensor L, which can specifically detect Cu2+ and S2− in the environment.

Abstract:A novel fluorescence "turn-on" assay for protease activity based on controlled aggregation of a tetraphenylethene probe is reported. A tetraphenylethene derivative (TPE-2+) with two positive charges is used in this study. TPE-2+ has no emission in an aqueous solution. Heparin is an anionic polymer, which can induce aggregation of TPE-2+. Strong aggregation-induced fluorescence emission (AIE) is observed. Histone is a cationic protein, it binds with heparin via electrostatic attractive interactions, which hinders the binding of TPE-2+ to heparin. TPE-2+ is released, and a decreased emission signal is detected. Upon the addition of a protease, histone is hydrolyzed into small fragments, which weakens its competitive binding to heparin. Free heparin induces aggregation of TPE-2+. A turn-on emission signal is recorded. An efficient and convenient fluorescence assay for protease activity is therefore established. Abstract:1. By applying the soft and hard acids and bases (HSAB) theory to amino acids, a novel peptide chemical sensor L has been designed that can simultaneously detect Zn2+, Cu2+, and S2−. 2. L exhibits minimal biological toxicity and holds potential as a sensor for detecting Zn2+, Cu2+, and S2− in biological systems. A novel peptide‐based chemical fluorescence sensor L (Dansyl‐His‐Pro‐Thr‐Cys‐NH2) was designed and synthesized. This sensor exhibits an "On–Off–On" detection cycle to detect Cu2+, Zn2+, and S2− in solution. According to the chelation‐enhanced fluorescence (CHEF) mechanism, when Zn2+ is present, the fluorescence is significantly enhanced and a blue shift occurs, representing a "Turn‐On" phase of the fluorescence detection mode. Because copper ions (Cu2+) have a paramagnetic quenching sensing mechanism, the fluorescence of L quenches rapidly with the formation of the L‐Cu system, representing the "Turn‐Off" phase. The subsequent introduction of S2− to the L‐Cu system results in the recovery of the L‐fluorescence, thereby representing the second "Turn‐On" phase. As a peptide molecule, the sensor L has several advantages over other types of sensors, including water solubility, high sensitivity, and good biocompatibility, with a very low detection limit. The detection lines of Zn2+ and Cu2+ are 97 nM (R = 0.993) and 75 nM (R = 0.995), respectively. Additionally, the sensor does not exhibit any obvious cell toxicity. These results indicate that this peptide chemiluminescent sensor has the potential to be applied in in vivo detection.

Abstract:Labeling of peptides and proteins with fluorescent dyes is a key step in functionalizing these structures for a wide array of biological assays. However, coupling strategies of such dyes have not been optimized for the most common compounds, while this step is typically the most precious and costly of the whole synthesis. We searched for the best conditions for attachment of the most widely used fluorescent dyes such as 6-carboxyfluorescein, Rhodamine B, and BODIPY-FL to peptides, where amino terminal Cu(II) and Ni(II) binding site (ATCUN) peptides were used as a model system. Surprisingly, conventional methods of dye attachment proved to not be satisfactory and yielded poor efficiency results. We have discovered that when labeling primary amines on peptides, the uncommon synthesis of activated pentafluorophenol (PFP) esters is the most efficient strategy, expedited by microwave irradiation. Coupling to secondary amines is achieved most efficiently through conventional coupling reagents such as HATU and PyBOP. Furthermore, we have employed our fluorescently labeled ATCUN peptides in studies for Cu(II) and Ni(II) sensing, showing that changing the fluorophore does not significantly affect the fluorescence quenching process and discovering the optimal linker length between the ATCUN core and the dye, expanding the repertoire of fluorophores that can be used in this application.
Electrochemical Immunosensors with Protease As the Signal Label for the Generation of Peptide-Cu(ii) Complexes As the Electrocatalysts Toward Water Oxidation

Abstract:This work reported that protease can be used as the signal label to amplify the signal of electrochemical immunosensors. Carbon nanotube (CNT) was used as the carrier of recognition antibody and signal label. The nanolabel was captured by sensor electrode via a sandwich-type immunoreaction. Protease on the surface of nanolabel can catalyze the hydrolysis of peptide substrate to liberate an amino terminal Cu(II)- and Ni(II)-binding (ATCUN) peptide. The released ATCUN peptide can bind with Cu(II) to generate an electrocatalyst which exhibits high electrocatalytic ability for water oxidation. Thus, the signal was amplified again by the redox cycling between ATCUN-Cu(II) and H2O. The immunosensor exhibits low background current because of the poor redox activity of peptide substrate and the high oxidation overpotential of H2O molecule. Based on the signal amplification of protease and redox cycling, the target concentration as low as 10 pg/mL can be readily measured with prostate-specific antigen (PSA) as the model analyte. The proposed immunosensor was applied to determine PSA in human serum samples with satisfactory results.

Abstract:Cathepsin L (CTL) is a cysteine protease demonstrating upregulated activity in many disease states. Overlapping substrate specificity makes selective detection of CTL activity difficult to parse from its close homologue CTV and the ubiquitous CTB. Current probes of CTL activity have limited applications due to either poor contrast or extra assay steps required to achieve selectivity. We have developed a fluorogenic probe, CTLAP, which displays good selectivity for CTL over CTB and CTV while exhibiting low background fluorescence attributed to intrinsic self‐quenching mechanisms. CTLAP achieves optimum CTL selectivity in the first 10 min of incubation suggesting that CTLAP is amenable for rapid detection of CTL, even in the presence of competing cathepsins.

Abstract:Currentprobes for proteases detection are susceptible tophotobleachingandinefficientin cell penetration. Also it is not applicable for multiplexed detection. Anovel probe based on quantum dot (QD), nanogold, substrate peptide and EGF is studied as the sensor for intracellular proteases detection. Due to the less fluorescence overlapping and the excellent propertiesagainst photobleaching of QDs, the strong photo quenching capacities of nanogolds, and the high transfer efficiency of EGF, this probe can be employedforlong time multiplexed detection, which was difficult for conventional probes. A comprehensive analysis on how to effectively design a fluorescent probe based on substrate peptide containing cleavage sites was first presented. Furthermore, the detection was achieved based on fluorescence polarization. The sensitivity was improved based on the concentration change of the fluorescent molecules, which has not been effectively utilized before.

Abstract:<p>Characterization of the protein-peptide interactions are a critical for understanding the functions and signal pathways of proteins. Herein, a new finding of universal terminal protection that protein bind specifically with peptide and provide a protective coating to prevent peptide hydrolysis in the presence of peptidase. On the basis of this mechanism, we first reported a novel label-free fluorescence biosensor strategy that utilizes the protection of specific terminal protein on peptide-templated gold nanocluster (AuNCs) beacon for the detection of proteins. The fluorescence quenching of peptide-templated AuNCs can be effectively inhibited with increasing concentration of the specific protein, exhibiting a satisfactory sensitivity and selectivity toward protein with the detection limit of MDM2 and gp120 are 0.0019 U/mL and 0.0012 U/mL, respectively. The developed label-free fluorescence biosensor strategy provides new ideas to detect and screen protein for analyzing protein-peptide interaction in biomedical applications.</p>

Abstract:Trypsin is a serine protease playing a key role in regulating pancreatic exocrine function and can be applied as a marker for the diagnosis of pancreatitis. In this work, a convenient and sensitive fluorescent assay was developed toward trypsin. Hydrogen peroxide slowly oxidized a non-fluorescent o-phenylenediamine (OPD) into a fluorescent product 2,3-diaminophenothiazine (DAP) under the catalytic from copper ions. After the introduction of bovine serum albumin (BSA), the combination of BSA with copper ions formed a peroxidase mimic and significantly accelerated the reaction rate. As an efficient protease, trypsin cleaved the lysine and arginine residues in BSA. This destroyed the binding between Cu 2+ and BSA, and brought in a reduction of the catalytic effect. The accompanying decrease in fluorescence provided a response to trypsin in the range of 0.01–600 ng/mL, with a detection limit of 0.007 ng/mL. The scheme had a good selectivity and was successfully applied to the detection of real samples.

Abstract:Herein, a novel assay has been developed for monitoring PreScission protease (His‐PSP) mediated enzyme cleavage of ATTO 590 labeled peptide substrate (ATTO‐LEV). This novel method is based on combining the use of capillary electrophoresis and fluorescence detection (CE‐FL) to dynamically monitor the enzyme cleavage activity. A multivalent peptide substrate was first constructed by immobilizing His‐tagged ATTO 590 labeled peptide substrate (ATTO‐LEVH6) onto the surface of CdSe/ZnS quantum dots (QDs). Once successfully immobilized, the novel multivalent peptide substrate resulted in the Förster resonance energy transfer (FRET) from QDs to ATTO 590. The ATTO‐LEVH6‐QD assembly was then incubated with His‐PSP to study the proteolytic cleavage of surface bound ATTO‐LEVH6 by CE‐FL. Our data suggests that PreScission‐mediated proteolytic cleavage is enzyme concentration‐ and incubation time‐dependent. By combining capillary electrophoresis, QDs and FRET, our study herein not only provides a new method for the detection and dynamically monitoring of PSP enzyme cleavage activity, but also can be extended to the detection of many other enzymes and proteases.
