Abstract:The paper presents a novel strategy to identify analytical markers of traditional Chinese medicine preparation (TCMP) rapidly via direct analysis in real time mass spectrometry (DART-MS). A commonly used TCMP, Danshen injection, was employed as a model. The optimal analysis conditions were achieved by measuring the contribution of various experimental parameters to the mass spectra. Salvianolic acids and saccharides were simultaneously determined within a single 1-min DART-MS run. Furthermore, spectra of Danshen injections supplied by five manufacturers were processed with principal component analysis (PCA). Obvious clustering was observed in the PCA score plot, and candidate markers were recognized from the contribution plots of PCA. The suitability of potential markers was then confirmed by contrasting with the results of traditional analysis methods. Using this strategy, fructose, glucose, sucrose, protocatechuic aldehyde and salvianolic acid A were rapidly identified as the markers of Danshen injections. The combination of DART-MS with PCA provides a reliable approach to the identification of analytical markers for quality control of TCMP.

Abstract:The increasing demand for rapid and sensitive roadside identification of trace illegal drugs drives the development of high-performance miniature mass spectrometric instrumentation and methodology. Here, we report a synchronized flash-thermal-desorption purging and ion injection (SFTDPI) method to increase the sensitive and rapid screening of volatile and nonvolatile illegal drugs for miniature ion trap mass spectrometry (ITMS). The flash-thermal desorption could reach 290 degrees C in 2.5 s, which could achieve efficient vaporization of nonvolatile noscapine with boiling point at 565 degrees C. ITMS using discontinuous atmospheric pressure interface (DAPI) has an ion utilization ratio of less than 1%, the synchronized purging for flash-thermal desorption and ion injection with DAPI could accurately control the time interval along with the desorption, gas purging, ionization and ion injection, and the sample utilization ratio increases more than five times. The miniature SFTDPI-ITMS presents good performance: (1) more than 60 times improvement in sensitivity was achieved compared to the previously reported thermal-desorption acetone-assisted photoionization ion trap mass spectrometer for nonvolatile drugs, and the minimum detectable quantity reaches 50 pg for fentanyl. (2) Ten kinds of mixing drugs with boiling point difference of 300 degrees C can be simultaneously identified within 3 s under a single analysis. SFTDPI-ITMS was deployed at the roadside checkpoint of Sino-Burmese border; fentanyl in the captured encapsulated powder and the suspected opiate had been successfully identified.

Abstract:A thermal desorption low temperature plasma (TD-LTP) ionization source was developed for the direct, rapid and sensitive detection of pesticides by mass spectrometry without complex sample pretreatment. A thermal desorption sampler was added in front of the plasma generator, and the analyte was collected on a polytetrafluoroethylene (PTFE) swab and desorbed into gas molecules, and then gas molecules were transported to the plasma generator by carrier gas to be ionized. The utilization of thermal desorption sampler helped to transform the interaction between gas phase plasma and sample from gas-solid or gas-liquid to gas-gas, which greatly increased the sensitivity and stability especially for non-volatile sample (e.g. pesticides) compared with conventional LTP ionization source. The TD-LTP ionization source was coupled to a homemade rectilinear ion trap mass spectrometry. The operation parameters of the TD-LTP ionization source were optimized and the characteristic ions of 12 different pesticides were successfully obtained and investigated. Then the TD-LTP ionization source was connected with commercial ACQUITY TQD mass spectrometer, and the pesticide residue level in broomcorn was evaluated.

Abstract:In this study, a soft microwave plasma torch (SMPT) combined with a mass spectrometer (MS) was used for the first time as an analytical method to detect and analyze various pharmaceutical and personal care products (PPCPs) in aquatic environments without the need for sample pretreatment. For this purpose, ambient SMPT was used to generate plasma for ionizing the analyte molecules. Accordingly, nine PPCPs were identified by the SMPT-MS, and their identification was verified by collision-induced dissociation (CID). The technique’s performance was verified with known PPCP samples, and the limits of detection (LOD) and quantification (LOQ) obtained over a linear range of 50–1 μg/L were 1.56 to 2.81 and 2.07 to 3.62 μg/L, respectively, with the standard addition recovery rate falling between 87.14 and 115.16

Abstract:A direct analytical method based on dual ambient plasma ion source tandem mass spectrometry was used for the simultaneous determination of four sterols in the environment. This technology has very high sensitivity and the method detects the four sterols in methanol-water (1:3) solutions with limits of detection (LOD) and limits of quantification (LOQ) ranging from 1.2 ng/L to 6.9 ng/L and 7.6 ng/L to 10.0 ng/L, respectively. The method was also used to test water quality at three locations within the city and successfully detected all four sterols at very low concentrations. The dual plasma source tandem mass spectrometry technique is extremely simple, rapid, sensitive and highly efficient compared to other traditional methods, providing a useful screening tool for sterols in water.

Abstract:Taste and odor (T&O) compounds are widespread in water environments and have attracted considerable public attention. Nowadays, the standard detections of these chemicals rely mainly on off-line methods such as GC-MS or evaluation by trained analysts' senses. In this study, we report a method for the rapid detection of T&O compounds in water by exploiting a newly invented chemi-ionization source, in combination with headspace vapor measurement at room temperature. The calibrated limits of detection (LODs) of 2-methylbutyraldehyde, methyl tert-butyl ether (MTBE), methyl methacrylate (MMA), 2-isobutyl-3-methyoxypyrazine (IBMP), and 2-isopropyl-3-methoxypyrazine (IPMP) are in the range of 3.5-50.2 ng L-1, and the estimated LODs of 2-methylisoborneol (2-MIB) and geosmin (GSM) are 0.25 and 0.77 ng L-1, respectively. The calibration results reveal that the instrumental LODs for 2-methylbutyraldehyde, MTBE, MMA, beta-cyclocitral, 2-MIB, and GSM are 1-2 orders of magnitude better than the odor thresholds of humans. The accuracy, precision, recovery, and linearity (R-2) of the method are tested. Water samples from city tap water and three rivers in Beijing are assessed using this technique, and the typical T&O compositions are observed with concentrations ranging from 0.2 to 297 ng L-1. The new ultra-sensitive rapid detection method shows comparable sensitivities to the existing off-line technique and displays great potential for real-time detection of T&O pollution in water environments. Abstract:A novel vapor dynamic headspace enrichment device was set up for nontarget screening of volatile organic compounds (VOCs) in drinking water. The main operating parameters of this device, such as length of distillation tube, volume of collected condensate, and choice of absorbent, were optimized. In this device, vapor was utilized as a purge gas and water was utilized as a absorbent. With the help of the device, one liter of water sample could be concentrated to 5 mL and the sensitivity of traditional purge and trap-gas chromatography-mass spectrometry (P&T-GC-MS) could be improved 1 - 2 orders of magnitude. Source and disinfected water samples from a water treatment plant were analyzed with this method. Compared with the traditional P&T-GC-MS analysis without pre-enrichment, the numbers of identified VOCs were improved from 0 to 16 for source water and 5 to 35 for disinfected water samples. It is also shown that there are many halide compounds in VOCs in disinfected water which do not exist in source water.

Abstract:The drug detection technology plays a pivotal role in the domains of pharmaceutical regulation and law enforcement. In this study, we introduce a method that combines thermal desorption corona discharge ionization (TD-CDI) with mass spectrometry for efficient drug detection. The TD-CDI module, characterized by its compact and simple design, enables the separation of analytes within seconds and real-time presentation of one or two analyte peaks on the mass spectrum most of the time, which reduces matrix interference and improves detection performance. Through experimental investigation, we studied the characteristics of TD-CDI for analyte separation and detection, even with the same mass number, and optimized the TD-CDI approach. TD-CDI-MS was employed for the rapid detection of drugs in various traditional medicine, food products, and human samples. Additionally, by utilizing TD-CDI for segmented hair direct analysis, it becomes possible to trace the drug usage cycle of individuals. This underscores the feasibility of the proposed analytical method within the realm of drug detection.

Abstract:Polycyclic musk tonalide (AHTN) is an emerging organic pollutant which has been frequently detected in drinking water and source water in recent years.The gas chromatography-mass spectrometry (GC-MS) is usually used as the detection method for AHTN.While,there are some problems for GC-MS.So a simple and rapid high performance liquid chromatography (HPLC) method was developed to overcome the deficiency of GC-MS.The sample analysis could be completed with HPLC method within 5 min.Assisted with enrichment techniques such as solid phase extraction,the detection limit can reach to ng/L level.In the new detection process,aqueous AHTN sample is mixed with acetonitrile in a certain ratio,which improves the separation efficiently of AHTN in liquid chromatography column and eliminate the influence of membrane filtration on the recovery of samples.By comparison,when samples prepared with pure water and actual water were determined,it was found that the relative errors for the two methods were less than 2.0％ and the relative deviation of the two methods was less than 1.0％.Thus,the developed HPLC method can be used as an alternative method for detecting AHTN in water.This method provides reference to the determination of other organic compounds with low water solubility and weak polarity.

Abstract:This work proposed a novel strategy based on quadruplex stable isotope derivatization for sensitive, accurate, speedy and green determination of panaxadiol (PD) and panaxatriol (PT). This strategy integrated with ultrasound-assisted dispersive liquid-liquid microextraction, microwave-assisted derivatization, and magnetic graphene oxide dispersive solid phase extraction as efficient sample pretreatment techniques, coupling with ultra-high performance liquid chromatography tandem mass spectrometry detection in multiple reaction monitoring mode for the analytical purpose of high-throughput, sensitivity, selectivity, green and accuracy. Quadruplex mass spectrometry sensitizing derivatization reagents, 3-N-(D-0-/D-3-methyl-, and D-0-/D-5-ethyl-)-2'-carboxyl chloride rhodamine 6 G, were designed, synthesized and applied for the high-throughput derivatization of hydroxyl-containing PD and PT for the first time. The analogue derivatives of PD and PT standards using 3-N-propyl-2'-carboxyl chloride rhodamine 6 G as derivatization reagent were employed as the internal standards for the mass spectrometry quantitation to minimize matrix effect. All rhodamine 6 G derivatives of PD and PT were easily and selectively adsorbed by magnetic graphene oxide for efficient magnetic dispersive solid phase extraction before injection. Parameters of (micro-) extractions, derivatization and liquid chromatography-mass spectrometry conditions were all optimized in details. Method validation results consisting of linearity, accuracy, precision, repeatability, matrix effect, stability and limits of detection (LODs, 0.02 ng/mL for PD and 0.03 ng/mL for PT) were determined. The analytical performance of this method benefited from efficient integration of suitable sample pretreatment techniques and characteristic structure of rhodamine 6 G with four mass tags in derivatization reagents. The developed and validated method has been applied to the quantitation of PD and PT for the quality assessment of related foodstuffs and traditional Chinese medicinal materials. Abstract:Abstract A novel strategy for the direct analysis of non-conjugated steroids in water using paper spray mass spectrometry (PS-MS) has been developed. PS-MS was used in the identification and quantification of non-conjugated (free) steroids in fish tank water samples. Data shown herein indicates that individual amounts of free steroids can be detected in aqua as low as; 0.17 ng/μL, 0.039 ng/μL, 0.43 ng/μL, 0.0076 ng/μL for aldosterone, corticosterone, cortisol, and β -estrone, respectively, and with an average relative standard deviation of ca. < 10% in the positive ion mode using PS-MS/MS. Direct detection of free steroids in a raw water mixture, from aquaculture, without prior sample preparation is demonstrated. The presence of free steroids released in fish water samples was confirmed via tandem mass spectrometry using collision-induced dissociation. This approach shows promise for rapid and direct water quality monitoring to provide a holistic assessment of non-conjugated steroids in aqua .

Abstract:Increasing in cases involving drugs of abuse leads to heavy burden for law enforcement agencies, exacerbating demand for rapid screening technique. In this study, atmospheric pressure ionization technologies including direct analysis in real time (DART) ion source coupled to a time-of-flight mass spectrometer (DART-TOF-MS)as well asdopant-assisted positive photoionization ion mobility spectrometry (DAPP-IMS) without radioactivity were utilized together as the powerful analytical tool for the rapid screening and identification of 53 abused drugs.The limits of detection (LOD) were 0.05-2μg/mL when using DART-TOF-MS and 0.02-2μg when using DAPP-IMS which could satisfy the actual requirement in forensic science laboratory. The advantages of this method included fast response, high-throughput potential, high specificity, and minimal sample preparation. A screening library of reduced mobility (K0), accurate mass of informative precursor ion ([M+H]+) and fragment ions was established respectively by employing a bench-top DAPP-IMS and TOF-MS in-source collision induced dissociation (CID) mode. Then the standardized screening procedure was developed with criteria for the confirmation of positive result. A total of 50 seized drug samples provided by local forensic laboratory we reanalyzed to testify the utility of the method. This study suggests that a method combing DART-TOF-MS and DAPP-IMS is promising for the rapid screening and identification of abused drugs with minimal sample preparation and absence of chromatography.

Abstract:Synchronized discharge ionization (SDI) was previously developed for hand-held mass spectrometers with discontinuous atmospheric pressure interfaces. The function of SDI has been demonstrated for analysis of volatile organic compounds in air at high sensitivity, which is attributed to the fact that ions were produced next to the ion trap mass analyzer inside the vacuum manifold. In this study, a simple sampling device was designed and fitted to a hand-held mass spectrometer to characterize its potential in direct analysis of low-volatility chemicals on surfaces. Nine chemicals of vapor pressures ranging from 10(-4) to 10(-8) Torr (at room temperature), including pesticides, illicit drugs, and explosives, were selected to evaluate and demonstrate the analytical capability of the designed system. Compounds of vapor pressures below 10(-7) Torr, such as tetryl, cocaine, and tetrahydrocannabinol (THC), have been successfully detected. Direct analysis of pesticides from fruit and explosives from a large surface area has also been demonstrated. Tandem mass analysis was performed, which helped to confirm the analyte identity as well as to improve the signal-to-noise ratio (S/N).

Abstract:Bile acids, sterols, and eicosanoids serve as important chemical categories in mammalian urine and, sometimes, as primary diagnostic markers for diverse diseases, whereas phenols, usually derived from the diet, are usually observed as annoying interferents in pharmacokinetic and metabolic profiling of phenolic derivatives. Precise analysis of these substances widely suffers from instability and trace distributions, as well as errors and uncertainties resulting from tedious sample preparation procedures. Herein, directly simultaneous determination of phenols, bile acids, sterols, and eicosanoids, in particular those trace ones, in mammalian urinary matrices was attempted using large volume direct injection-online solid phase extraction-ultra high performance liquid chromatography-polarity switching tandem mass spectrometry (LVDI-online SPE-UHPLC-psMS/MS). Large volumes (500 mu L) of liquid-state samples were directly loaded onto an online SPE column via ten consecutive injections. The SPE column was responsible for accumulating targeted components at the loading phase (-4.5 to 5.0 min), while transmitting those trapped analytes into a Waters HSS T3 column at the elution phase (5.0-27.0 min). Phase switching was accomplished using an electronic 6-port/2-channel valve. Analyte detection of all analytes, 28 ones in total, was performed using both positive and negative multiple reaction monitoring modes. Various method validation assays demonstrated the developed method to be extremely sensitive (most limits of quantitation lower than 0.3 ng mL(-1)), precise (all RSDs of intra-and inter-day variations lower than 15%) and accurate (recoveries ramped from 80.4% to 120.0% with RSDs lower than 20%). Significant variations occurred for urine samples from different species, as well as amongst urinary matrices from different individuals within the same group. The findings proved that the developed LVDI-online SPE-UHPLC-psMS/MS method takes advantage of detecting the trace components and preserving those mutable substances from degradation, thus offering a meaningful tool for widely targeted monitoring substances in biofluids.

Abstract:We report measurements of two dimensional radial fingering in viscoelastic concentrated suspensions (pastes) of colloidal particles, in small surface tension conditions. The displacement efficiency, the average finger width and the fractal dimension for growth have been measured in a broad range of paste viscosity and driving pressure. The growth process appears to be fractal in all the experimental conditions, with fractal exponents, D G , covering the whole range from 1.3 at very small tip velocity to 2.0 at very high tip velocity. The displacement efficiency, δ, defined as the volume fraction of paste displaced by water when water flows out of the paste, is almost zero at very low velocity, and seems to tend toward an asymptotic value close to 0.25 at high velocity, δ is almost entirely dominated by the velocity dependence of the tip-splitting cascade. Abstract:Rationale: Drug abuse or dependence results in a series of social problems, including crime and traffic accidents. Spray-inlet microwave plasma torch tandem mass spectrometry (MPT-MS/MS) was developed and used for the direct detection of such drugs in liquid solutions. Methods: Drug sample solutions were directly sprayed into the flame of an MPT by a sampling pump and the ions produced by Penning ionization and ion-molecule reactions were guided into a quadrupole time-of-flight (QTOF) tandem mass spectrometer for mass analysis. The MPT was operated at 40 W and 2.45 GHz in a 700 mL/min argon flow both for the inner and middle plasma. Results: Intact quasi-molecular and molecular ions of various drugs were successfully characterized by spray-inlet MPT-MS/MS. The analysis of one sample was finished within 30 s. Furthermore, the method exhibited excellent efficiency, precision and sensitivity, and the limits of detection and limits of quantification of the samples in methanol were in the range of 5.25-60.0 and 17.5-200 ng g-1 , respectively. Excellent linearities with coefficients of determination (R2 ) of 0.9627-0.9980 were verified in the range 0.05-50 μg g-1 . Four different beverages purchased locally were also analyzed with spray-inlet MPT-MS/MS, and caffeine was directly determined in two of the beverages. By adding six standard drug samples to sport drinks (each drug was 1 μg g-1 ) and Chinese spirit (each drug was 0.1 μg g-1 ), all the drugs except for caffeine were detected successfully. Conclusions: This study indicates that spay-inlet MPT-MS/MS is an effective method for direct and rapid identification of drug solutions, and it has substantial potential for fast and sensitive drug residue detection.
Quantitative analysis of Phthalate Esters by in-situ thermal desorption atmospheric pressure photoionization mass spectrometry using a dopant as the internal standard

Abstract:Phthalate Esters (PAEs) are a group of chemicals widely used in plastic products, and they are also known as endocrine disruptors that can adversely affect the hormonal balance in humans. Since many food-contacting items, such as lunch boxes, fresh keeping film, disposable cups, are made of plastic, the PAEs can easily enter the human body through the polluted foods or drinks. Traditional analytical techniques involve laborious sample pretreatment and low-throughput analysis, so it is necessary to develop techniques that can rapidly and quantitatively detect these compounds. In this article, a miniaturized ion trap mass spectrometer (ITMS) was established firstly, then, an in-situ thermal desorption atmospheric pressure photoionization (in-situ TD-APPI) was designed, and the dopant was explored using for improving the sensitivity and as internal standard (IS) simultaneously in the analytical process. The in-situ TD-APPI presents good performance: (1) More than 20 times improvement in sensitivity compared with Nano-ESI for PAEs; (2) 12 kinds of phthalates were quantified with using acetone as the IS, and the LODs and LOQs ranges were found to be 0.02–1 μg/mL and 0.02–100 μg/mL, respectively. The technique was also applied to the quantification of mixed solutions, and the results were validated by LC/MS. Finally, six categories of food-contacting items containing PAEs were analyzed with superficial content (ng/cm2) ranging from 0 to 6.20 ng/cm2. These results demonstrate the potential of rapid quantification of analysis of PAEs in practical plastic food-contacting items by in-situ TD-APPI.
Rapid Detection of Active Pharmaceutical Ingredients in Drugs by Microwave Plasma Atmospheric Pressure Desorption Ionization Mass Spectrometry

Abstract:A new ionization source, termed microwave plasma atmospheric pressure desorption ionization (MWPAPDI) source, was developed. The stable plasma was generated by microwave plasma torch working at 2450 MHz at atmospheric pressure. Ar was used as working gas. The source was successfully coupled with a Corsair API-TOF time of flight mass spectrometer, and the active pharmaceutical ingredients in drugs were rapidly characterized. The speed was allowed as fast as approximate 360 samples per hour via manual sampling. In the microwave plasma environment, the hydrogen bound moiety of the active ingredients of the drug preparations was directly removed, as a result, the quasi-molecular ions of active component compounds [M+H](+) became the main ions and gave simple mass spectra. Without chemical reagents, MWPAPDI has the feature for direct analysis of samples with high throughput, no pollution and in real time. Therefore, the method is believed to be a useful technique for drug discovery and chemical industry.
Arsenic speciation and effect of arsenate inhibition in a Microcystis aeruginosa culture medium under different phosphate regimes

Abstract:To assess the ecological impact of arsenic pollution during cyanobacterial blooms, As speciation and cyanobacterial growth in phosphate‐modified Microcystis aeruginosa cultures treated with arsenate were investigated under laboratory conditions. Marked growth inhibition was observed when arsenate was added. The inhibition effect of 1 µM arsenate was lower than that of 10 µM arsenate. Increasing phosphate supply (0–175 µM) in the medium decreased the inhibition of As. In the medium, arsenate, arsenite, and dimethylarsenicals (DMA) occurred under phosphate‐deprivation (0 µM) and phosphate‐excess (175 µM) conditions. However, only arsenate and DMA were detected under phosphate‐limited (1 µM) and phosphate‐rich (10 µM) conditions. Moreover, arsenite and DMA concentrations had significantly positive correlation with the biomass production of M. aeruginosa in the existence of phosphate. Arsenic speciation was more significantly affected by phosphate levels than arsenate concentrations. Recovery of total As content in M. aeruginosa culture medium increased with the increasing phosphate supply. The duration of arsenate contamination in the culture of M. aeruginosa had no influence on the variation of As species but affected the concentration of them in the medium under the phosphate‐excess condition. This demonstrated that the effect of M. aeruginosa on As speciation was not related to the duration of As contamination under the phosphate‐excess condition. Environ. Toxicol. Abstract:Current direct analysis methods in mass spectrometry (MS) are predominantly focused on desorbing and ionizing samples in the solid phase. Some sampling difficulties are associated with liquid (solution) or gas samples. The present study has expanded direct MS analysis to solution samples by using the desorption corona beam ionization (DCBI) technique in combination with poly(dimethylsiloxane) (PDMS) substrate sampling. Typically, the PDMS substrate is dipped in water for microextraction of pesticide compounds and then is transferred to an MS ion source for desorption and ionization. This approach improves the detection limit for DCBI and allows more organic compounds in complex mixtures to be identified within seconds. The practical application of this device is demonstrated by identifying five pesticides (acephate, isoprocarb, dimethoate, dichlorvos, and dicofol) in water. The obtained detection limits of pesticides are 1 mu g/L, the measured dynamic ranges are 3 orders of magnitude, the calculated correlation coefficients are between 0.939 and 0.979 at concentration levels of 5-5000 mu g/L, and the repeatabilities defined as a relative standard deviation of five successive injections are in the range of 13-17%. The results indicate that the DCBI technique coupled with PDMS sampling is an excellent method for the analysis of organic pesticides in solution, and it also opens up a new avenue for direct MS studies of solution samples with general importance.
