Abstract:Facile preparation of water-soluble and fluorescent Ag nanoclusters (NCs) stabilized by glutathione at room temperature is described. Although the glutathione layer was introduced to prevent the silver nanoparticles from decomposition and increase their water solubility, this simple surface optimization resulted in surprisingly high efficiency of selective Hg2+ sensing, where the limit of detection (LOD) was as low as 10-10?M (0.02 ppb, 0.1 nM). This result revealed a simple and practical strategy for Hg2+ detection using fluorescent Ag NCs as sensor probe, with the lowest detecting limits reported to date.

Abstract:We describe a method for the preparation of water-soluble gold nanoclusters (Au-NCs) from chloroauric acid using denatured-casein as both a reducing and stabilizing agent. The resulting Au-NCs were characterized by photoluminescence, UV–vis absorption, and X-ray photoelectron spectroscopies, and by transmission electron microscopy. The Au-NCs have an average diameter of 1.7 ± 0.2 nm and exhibit orange-red fluorescence emission peaking at 600 nm (with a Stokes’ shift as large as 237 nm), a quantum yield of 4.3 %, and good stability over the physiologically relevant range of pH values and ionic strength. Cytotoxicity studies showed the Au-NCs to display negligible effects in terms of altering cell proliferation or triggering apoptosis. Fluorescence imaging of HeLa cancer cells was accomplished by loading such cells with the Au-NCs. The fluorescence of the Au-NCs is found to be strongly quenched by Hg(II) ions, and thus the Au-NCs can be used for detecting and, possibly, imaging of Hg(II). An assay was worked out for the determination of Hg(II), and its limit of detection is 1.83 nM, which is 5.5 times lower than the maximum allowed concentration of Hg(II) in drinking water as defined by the US EPA.

Abstract:Water–soluble fluorescent silver nanoclusters (Ag NCs) were prepared with the assistance of commercially available polyinosinic acid (PI) or polycytidylic acid (PC). The fluorescence of the Ag NCs is effectively quenched by trace mercury(II) ions, which can be applied for their detection. The response of the Ag NCs prepared with PI to Hg(II) ion is linear in the Hg(II) concentration range from 0.05 to 1.0 μM (R 2 = 0.9873), and from 0.5 to 10 μM of Hg(II) (R 2 = 0.9971) for Ag NCs prepared with PC. The detection limits are 3.0 nM and 9.0 nM (at an S/N of 3), respectively. The method is simple, sensitive and fairly selective. Figure Water-soluble fluorescent Ag nanoclusters (NCs) were facilely prepared using commercially available polyinosinic acid or polycytidylic acid. The fluorescence intensity of the as-prepared Ag NCs was effectively quenched by trace Hg 2+ , which was used for the detection of Hg 2+ in water samples with good performance.

Abstract:In this article, we report on the easy synthesis of fluorescent Ag nanoclusters (Ag NCs) and demonstrate their multifunctional use in highly selective sensing Hg2+ and colorimetric detection of H2O2 and glucose. The red emission fluorescent Ag NCs were prepared using polymethacrylic acid sodium salt (PMAA) and polyvinylpyrrolidone (PVP) as templates via a convenient hydrothermal process. The Ag NCs were then used in the sensitive and selective detection of Hg2+ based on the fluorescence quenching behavior. The detection limit was 10 nM in the linear range from 10 nM to 16 mu M. We further demonstrate the novel use of the as-prepared Ag NCs as a dual channel probe for colorimetric and fluorescence turn-on detection of H2O2 and glucose.

Abstract:An easy prepared fluorescence turn-on and colorimetric dual channel probe was developed for rapid assay of Hg(2+) ions with high sensitivity and selectivity by using poly(acrylic acid)-templated silver nanoclusters (PAA-AgNCs). The PAA-AgNCs exhibited weak fluorescence, while upon the addition of Hg(2+) ions, AgNCs gives a dramatic increase in fluorescence as a result of the changes of the AgNCs states. The detection limit was estimated to be 2 nM, which is much lower than the Hg(2+) detection requirement for drinking water of U.S. Environmental Protection Agency, and the turn-on sensing mode offers additional advantage to efficiently reduce background noise. Also, a colorimetric assay of Hg(2+) ions can be realized due to the observed absorbance changes of the AgNCs. More importantly, the method was successfully applied to the determination of Hg(2+) ions in real water samples, which suggests our proposed method has a great potential of application in environmental monitoring.
Surface engineered bimetallic gold/silver nanoclusters for in situ imaging of mercury ions in living organisms

Abstract:Chemical sensing for the sensitive and reliable detection of mercury(II) ions (Hg2+) is of great importance in environmental protection, food safety, and biomedical applications. Due to the bio-enrichment property of Hg2+ in organisms, it is particularly meaningful to develop an effective tool that can in situ and rapidly monitor the level of Hg2+ in living organisms. In this work, we report ligand functionalized gold-silver bimetallic nanoclusters with bright red fluorescence as intracellular probes for imaging Hg2+ in living cells and zebrafish. The bimetallic nanoclusters of DTT-GSH@Au/AgNCs (DG-Au/AgNCs) with strong fluorescence that benefited from the synergistic effect of Au and Ag atoms were obtained through a one-pot synthesis method, incorporating glutathione (GSH) and dithiothreitol (DTT) as the reducers and functionalized ligands. Attractively, the bright red fluorescence of DG-Au/AgNCs could be rapidly and selectively quenched by Hg2+ within 1 min with a very low detection limit of 1.01 nM. Additionally, DG-Au/AgNCs had a great advantage in the detection of Hg2+ in living cells and zebrafish owing to its notably strong red fluorescence at 665 nm, which could avoid effectively auto-fluorescence interference from the organism. Such easily prepared bimetallic fluorescent nanoclusters would be expected to provide a noninvasive and sensitive approach in the detection of heavy metals in situ for environmental protection.

Abstract:An easily prepared fluorescent Ag nanoclusters (Ag NCs) probe for the sensitive and selective detection of Hg(2+) and Cu(2+) ions was developed here. The Ag NCs were synthesized by using polymethacrylic acid sodium salt as a template via a convenient hydrothermal process. The as-prepared fluorescent Ag NCs were monodispersed, uniform and less than 2 nm in diameter, and can be quenched in the presence of mercury (Hg(2+)) or copper (Cu(2+)) ions. Excellent linear relationships existed between the quenching degree of the Ag NCs and the concentrations of Hg(2+) or Cu(2+) ions in the range of 10 nM to 20 μM or 10 nM to 30 μM, respectively. By using ethylenediaminetetraacetate (EDTA) as the masking agent of Cu(2+), Hg(2+) was exclusively detected in coexistence with Cu(2+) with high sensitivity (LOD = 10 nM), which also provided a reusable detection method for Cu(2+). Furthermore, the different quenching phenomena caused by the two metals ions such as changes in visible colour, shifts of UV absorbance peaks and changes in size of Ag NCs make it easy to distinguish between them. Therefore the easily synthesized fluorescent Ag NCs may have great potential as Hg(2+) and Cu(2+) ions sensors.
Sensitive detection of mercury (II) ion using wave length-tunable visible-emitting gold nanoclusters based on protein-templated synthesis

Abstract:In this study, a simple, facile, green, and versatile strategy was developed for the synthesis of water-soluble and well-dispersed fluorescent gold nanoclusters (Au NCs) with wave length-tunable emissions using protein as capping agent and template. The resultant Au NCs exhibited excellent characteristics such as large Stokes shift, good stability, and high resistance to photobleaching. They showed strong emission at 680 nm, with a quantum yield of 4.5%. Heavy metal ion (Hg 2+ ) could quench the fluorescence of Au NCs efficiently and thus the as-prepared Au NCs were successfully developed as “turn-off” fluorescent probes for the detection of Hg 2+ sensitively and selectively. The lowest concentration to quantify mercuric ions could be as low as 1 nM and the fluorescence intensity of the Au NCs was proportional to the concentrations of Hg 2+ over the range 8 × 10 −8 M to 1 × 10 −5 M ( R = 0.9982). To validate the practicality of this probe, real water samples spiked with Hg 2+ were determined with RSD values less than 3%. And the results demonstrated that the developed probes worked well in environmental samples. The action mechanism between the Au NCs and Hg 2+ was explored as well.
A Selective Nanoprobes for Hg2+ based on Silver Nanoclusters with Red Fluorescence Emission

Abstract:"One-pot" method was used to prepare the good water-soluble,high stable silver nanoclusters (AgNCs)with red emission which was protected by chicken egg white (CEW) with sodium borohydride as reducer.The fluorescence-based sensing behavior by CEW@AgNCs towards metal ions was investigated in buffered aqueous media.Among the metal ion studied,the fluorescence intensity of CEW@AgNCs could be effectively quenched by Hg2+.The fluorescence intensity (F0-F)/F0 of the CEW@AgNCs showed a linear relation over the concentration of Hg2+ ranges of 9.99～79.37μmol/L.Therefore,CEW@AgNCs has been used as a probe to develop a rapid,highly selective and sensitive method for Hg2+ detection.In addition,the proposed method was also used to analyze the Hg2+ of real water samples,indicating that it would be widely broaden the application.

Abstract:Fluorescent gold nanoclusters (AuNCs) were synthesized using a drug target bacterial enoyl-ACP reductase (FabI) as a template. The physical and chemical properties of the AuNCs were studied by UV-vis absorption, fluorescence, X-ray photoelectron spectroscopy and TEM. The AuNCs-FabI conjugate was prepared by in situ reduction of tetrachloroaurate in the presence of FabI. The conjugated particles were loaded onto nylon membranes by taking advantage of the electrostatic interaction between the negatively charged AuNCs@FabI and the nylon film which is positively charged at pH 7.4. This results in the formation of a test stripe with sensor spots that can be used to detect Hg(II) ion in the 1 nM to 10 μM concentration range. The test stripes are simple, convenient, selective, sensitive, and can be quickly read out with bare eyes after illumination with a UV lamp.

Abstract:In this paper, a facile approach for preparation of AuAgS/Ag2S nanoclusters was developed. The unique AuAgS/Ag2S nanoclusters capped with biomolecules exhibit interesting excellent optical and catalytic properties. The fluorescent AuAgS/Ag2S nanoclusters show tunable luminescence depending on the nanocluster size. The apoptosis assay demonstrated that the AuAgS/Ag2S nanoclusters showed low cytotoxicity and good biocompatibility. Therefore, the nanoclusters can be used not only as a probe for labeling cells but also for their photocatalytic activity for photodegradation of organic dye. Moreover, a highly selective and sensitive assay for detection of mercury including Hg2+ and undissociated mercury complexes was developed based on the quenching fluorescent AuAgS/Ag2S nanoclusters, which provides a promising approach for determining various forms of Hg in the mercury-based compounds in environment. These unique nanoclusters may have potential applications in biological labeling, sensing mercury, and photodegradation of various organic pollutants in waste water.

Abstract:A novel approach for preparation of graphitic carbon nitride nanosheets (CNNS) from stripping graphitic carbon nitride by strong acid and ultrasonic technology was demonstrated in this study for the first time. Transmission electron microscopy (TEM) was employed to characterize the surface morphology. Atomic force microscope (AFM) was carried out to characterize the thickness of nanosheets. X-Ray diffraction (XRD) was performed to estimate the lattice structure. X-Ray photoelectron spectroscopy (XPS) was carried out to characterize the surface composition and element analysis. Fourier transform infrared spectroscopy (FT-IR) was allowed to identify the functional groups. The as-synthesized CNNS exhibited excellent emission property as well as excitation-independent emission behavior, and fluorescence quantum yields could reach approximately 12.53%. Mercury ion (Hg2+) can make a result of quenching the significant intensity of fluorescence of CNNS by formation of a covalent bond between empty orbital of Hg2+ and the p electrons of N (turn-off). Moreover, the addition of the L-cysteine (L-Cys) can enhance the intensity of fluorescence of the CNNS-Hg2+ system through the thiol group of L-Cys anchored with Hg2+ and drag it from the surface of CNNS (turn-on). The CNNS was consequently functioned as a fluorescence probe towards "off-on" detection of Hg2+ and L-Cys with high sensitivity and selectivity. Moreover, the fluorescent probe was applied to detect tap water and well water with satisfactory results.
Synthesis of Fluorescent Α-Chymotrypsin A-functionalized Gold Nanoclusters and Their Application to Blot-Based Technology for Hg2+detection

Abstract:Blot-based technology is widely used in biomedical research, serving as a remarkably efficient platform for biomolecule recognition and detection. In this report, highly fluorescent gold nanoclusters have been synthesized under mild conditions by using a proteolytic enzyme, α-chymotrypsin A (CTRA), as both the stabilizing and reducing agents. The synthesized AuNCs@CTRA was characterized by various techniques including UV-vis absorption, fluorescence, X-ray photoelectron spectroscopy and TEM. The fluorescent AuNCs@CTRA is fairly stable and responsive to mercury ions with high selectivity and sensitivity. These protein capped nanoclusters were electrophoresed on an SDS-PAGE gel and transferred to a cellulose membrane. Mercury ions can specifically quench the red fluorescent AuNCs@CTRA band and selectively stop the green band formation on the membrane through inhibition of the peroxidase mimic activity of AuNCs@CTRA toward the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) substrate in a concentration dependent manner. Therefore, using a blot-technology based system, we demonstrated the operation of the AuNCs@CTRA–cellulose hybrid material for mercury ion visual sensing that can be dually read out under UV light (fluorometric) and the naked eye (colorimetric). This approach also has potential for use in other blot-technology based applications.

Abstract:Silver nanoclusters (AgNCs) were first coated with bovine serum albumin (BSA) and then encapsulated into porous metal-organic frameworks of ZIF-8 by the protein-mediated biomineralization process. Unexpectedly, the fluorescence intensities of the yielded AgNCs-BSA@ZIF-8 nanocomposites were discovered to be continuously enhanced during each of the BSA coating and ZIF-8 encapsulation steps. Compared to common AgNCs, greatly improved photostability and storage stability of AgNCs could also be expected. More importantly, having benefited from the ZIF-8 shells, the prepared nanocomposites could possess the specific accumulation and sensitive response to Cu2+ ions, resulting in the rational quenching of their fluorescence intensities. Moreover, AgNCs-BSA@ZIF-8 nanocomposites were coated onto the hydrophobic arraying slides toward a microdots array-based fluorimetric method for the fast and sensitive evaluation of Cu2+ ions. It was discovered that the developed fluorimetric strategy could ensure the high-throughput analysis of Cu2+ ions in wide pH range, and especially some harsh and high-salt media. It can allow for the detection of Cu2+ ions in blood with the concentrations ranging from 4.0 × 10-4 to 160 μM, thus serving as a new copper detection candidate to be widely applied in clinical test, food safety, and environmental monitoring fields.

Abstract:In this work, we designed a new fluorescent oligonucleotides-stabilized silver nanoclusters (DNA/AgNCs) probe for sensitive detection of mercury and copper ions. This probe contains two tailored DNA sequence. One is a signal probe contains a cytosine-rich sequence template for AgNCs synthesis and link sequence at both ends. The other is a guanine-rich sequence for signal enhancement and link sequence complementary to the link sequence of the signal probe. After hybridization, the fluorescence of hybridized double-strand DNA/AgNCs is 200-fold enhanced based on the fluorescence enhancement effect of DNA/AgNCs in proximity of guanine-rich DNA sequence. The double-strand DNA/AgNCs probe is brighter and stable than that of single-strand DNA/AgNCs, and more importantly, can be used as novel fluorescent probes for detecting mercury and copper ions. Mercury and copper ions in the range of 6.0-160.0 and 6-240 nM, can be linearly detected with the detection limits of 2.1 and 3.4 nM, respectively. Our results indicated that the analytical parameters of the method for mercury and copper ions detection are much better than which using a single-strand DNA/AgNCs. (C) 2014 Elsevier B.V. Abstract:Ag nanoclusters (AgNCs) have attracted considerable attention due to their excellent photoluminescence, low toxicity, good photostability, and biocompatibility, which make them wide applications in the fields of chemical sensing, bioimaging, and single molecule microscopy. Here, we presented a facile approach to a new water-soluble, intense fluorescent AgNCs with yellow-emission under quasi-physiological conditions (pH 6. 6), which are stabilized by a sequence of DNA chimera via a chemical reduction method, and the DNA chimera with nanocluster templating sequence and PrPc aptamer acts as both a ligand to stabilize the metal core and a specific protein binding ligand. The fluorescent AgNCs probes can be used not only for the detection of cellular prion protein (PrPc) but also for PrPc imaging.
