Itraq-Based Proteomic Analysis Reveals Mitochondrial 'Damage'-Associated Molecular Patterns Are Involved in Pulmonary Inflammation in Lypopolysaccharide-induced Acute Lung Injury

Abstract:Background: Injury releases mitochondrial 9damage9-associated molecular patterns (mitochondrial DAMPs;MTDs)into the circulation with functionally important immune consequences,and acute respiratory distress syndrome is characterized with uncontrolled lung inflammation. However, the role of MTDs in acute lung injury(ALI) still remains unclear. Methods: Bronchoalveolar lavage fluid (BALF) and serum levels of NADH-ubiquinone oxidoreductase chain 1 (ND-1)and albumin in ARDS patients and healthy volunteers were quantified by ELISA. Meanwhile,Lypopolysaccharide(LPS),MTDs,or combination of them were sprayed into mouse intratracheally via PennCentury sparyer. After 24h, these mouse were sacrificed, the lung tissue were immediately collected. iTRAQ-based proteomic approach was applied to explore the lung tissue protein alterations to investigate the underlying mechanism of MTDs in ALI. Resluts: Serum and BALF levels of ND1 in ARDS patients were much higher than those in healthy volunteers,so was B/S radio. Correlation analysis showed that both serum and BALF levels of ND1 were positively related to B/S. In the proteomic analyses,122 proteins were diferentially expressed between the MTDs treated group and combination of LPS and MTDs treated group. Gene ontology analysis revealed that the diferentially expressed proteins were related to immune response., and pathway enrichment analysis showed that these proteins were enriched in TNF signaling pathway. Conclusions: BALF and serum MTDs levels in ARDS patients was increased.MTDs may be involved in the pathogenesis of ARDS by amplifying infammation via TNF signaling pathway.
