Abstract:As a kind of commonly-used synthetic materials for many pesticides, thiol compounds, once being leaked, can cause serious harm to the environment and humans. Therefore, the efficient detection of thiol compounds is essential. In this study developed a turn-on fluorescent probe (Cu@Zn-CP) for the highly sensitive fluorescence detection of thiol compounds. The probe was constructed based on a zinc coordination polymer (Zn-CP), whose fluorescence was quenched through the effective doping of Cu2+ ions. After the introduction of methyl thioglycolate (MTC), a rapid fluorescence turn-on response was generated within 90 s with a low detection limit of 23 ppb. Even after being reused for five cycles, the sensor maintains excellent detection performance and demonstrates good recyclability. It can also detect MTC in river water, with a spike recovery rate between 98-103 %. Furthermore, the designed Cu@Zn-CP exhibits good universality for detecting multifarious thiol compounds, including L-cysteine, glutathione, monothioglycerol, and 2-hydroxy-1-ethanethiol. This result provides a potential recyclable fluorescent sensor for thiol compounds.
Colorimetric and Ratiometric Fluorescent Chemosensor Based on Diketopyrrolopyrrole for Selective Detection of Thiols: an Experimental and Theoretical Study

Abstract:A colorimetric and ratiometric fluorescent thiol probe was devised with diketopyrrolopyrrole (DPP) fluorophore. The probe gives absorption and emission at 523 and 666 nm, respectively. In the presence of thiols, such as cysteine, the absorption and emission band shifted to 479 and 540 nm, respectively. Correspondingly, the color of the probe solution changed from purple to yellow, and the fluorescence changed from red to yellow. The emission intensity at 540 nm was enhanced by 140-fold. The Stokes shift of probe 1 (107 nm) is much larger than the unsubstituted DPP fluorophore (56 nm). Mass spectral analysis demonstrated that besides the expected Michael addition of thiols to the C═C bonds, the CN groups of the malonitrile moieties also react with thiols to form 4,5-dihydrothiazole structure. Probe 1 was used for fluorescence imaging of intracellular thiols. In the presence of thiols, both the green and red channel of the microscopy are active. With removal of the intracellular thiols, signal can only be detected through the red channel; thus, ratiometric bioimaging of intracellular thiols was achieved. The ratiometric response of probe 1 was rationalized by DFT calculations. Our complementary experimental and theoretical studies will be useful for design of ratiometric/colorimetric molecular probes.
Target-activated and Ratiometric Photochromic Probe for "Double-Check" Detection of Toxic Thiols in Live Cells

Abstract:Photochromic molecules can achieve reversible isomerization upon alternate light irradiations, which offers a great opportunity to improve the precision of analytes detection and imaging in complicated biological environments. Previous reported photochromic probe exhibited only mono-color switching and an initially fluorescence-ON state that may cause high background signal and impose an adverse impact on the desired sensing precision. To overcome this set-back, we developed a novel photochromic probe with an analyte-activation mode for ratiometric sensing of toxic thiols in both real water samples and live cells. The dynamic dual-fluorescence signal is released only after the fast and selective cleavage of the 2,4-dinitrophenyl sulfonate by the targeted thiophenol derivatives. Consequently, a “double-check” with synchronized dual-fluorescence blinking for analyte detection is successfully employed upon alternate light triggers with rapid response ( k =7.2×10 −2 s −1 ), high sensitivity (LOD=6.1 nM) as well as selectivity of thiophenol derivatives over other common thiol species (e.g., GSH, Cys and Hcy). The photochromic probe was successfully introduced to the fast and on-site detection of highly toxic thiophenols in real waste water samples. Moreover, by using confocal laser-scanning microscopy (CLSM) and flow cytometric analysis, the potential applications of this ratiometric photochromic probe for trace toxic thiol sensing in live cells are examined.

Abstract:A new ratiometric fluorescent probe is designed and prepared based on the concept of supramolecular encapsulation and dye competition. This supramolecular probe is based on two commercially-available dyes, one common guest and a simple-to-synthesize host. Fluorescence spectroscopy confirms that the supramolecular probe is capable of detecting thiols quantitatively with a broad linear region in phosphate buffered saline or fetal bovine serum. Mechanistic study shows a reaction between thiol specie and the guest to alter the distribution of encapsulated dyes. The supramolecular probes are demonstrated to quantitatively detect extracellular biological thiols by plate reader, which shows it keeps its effectiveness in complex buffered systems.

Abstract:The article describes the design and synthesis of a near-infrared ratiometric fluorescent probe CySS for thiols,the probe CySS has heptamethine cyanine as the reporter,a disulfide group as the thiol reaction site. The properties and application of CySS were studied in detail.The results demonstrate that CySS displays high sensitivity,good selectivity,and pH-independent.Moreover,we have demonstrated that CySS is suitable for imaging thiols in living cells,providing apotentially powerful tool for thiols detection in biological systems.

Abstract:A "turn-on" thiol-responsive fluorescence probe was synthesized and integrated into polymeric nanoparticles for sensing intracellular thiols. There is a photo-induced electron transfer process in the off state of the probe, and this process is terminated upon the reaction with thiol compounds. Configuration interaction singles (CIS) calculation was performed to confirm the mechanism of this process. A series of sensing studies were carried out, showing that the probe-integrated nanoparticles were highly selective towards biological thiol compounds over non-thiolated amino acids. Kinetic studies were also performed to investigate the relative reaction rate between the probe and the thiolated amino acids. Subsequently, the Gibbs free energy of the reactions was explored by means of the electrochemical method. Finally, the detection system was employed for sensing intracellular thiols in cancer cells, and the sensing selectivity could be further enhanced with the use of a cancer cell-targeting ligand in the nanoparticles. This development paves a path for the sensing and detection of biological thiols, serving as a potential diagnostic tool in the future.

Abstract:Thiol-containing amino acids (aminothiols) such as cysteine (Cys) and homocysteine (Hcy) play a key role in various biological processes including maintaining the homeostasis of biological thiols. However, abnormal levels of aminothiols are associated with a variety of diseases. The native chemical ligation (NCL) reaction has attracted great attention in the fields of chemistry and biology. NCL of peptide segments involves cascade reactions between a peptide-a-thioester and an N-terminal cysteine peptide. In this work, we employed the NCL reaction mechanism to formulate a Forster resonance energy transfer (FRET) strategy for the design of ratiometric fluorescent probes that were selective toward aminothiols. On the basis of this new strategy, the ratiometric fluorescent probe 1 for aminothiols was judiciously designed. The new probe is highly selective toward aminothiols over other thiols and exhibits a very large variation (up to 160-fold) in its fluorescence ratio (I458/I603). The new fluorescent probe is capable of ratiometric detection of aminothiols in newborn calf and human serum samples and is also suitable for ratiometric fluorescent imaging of aminothiols in living cells.

Abstract:Biological thiols, such as glutathione, cysteine, and homocysteine, are closely associated with human health and disease. In this work, we demonstrated a general strategy for the design of fluorescent probes for the detection of various biological related thiols. The preferred strategy we employed was to introduce a 3,5-dinitropyridyl moiety to a selected fluorophore via ether linkage to afford fluorescent probe. Once the ether bond was thiolyzed by biothiol, the hydroxy-functionalized fluorescent dye would be released and fluorescence emission restored. Herein, some novel fluorescent probes based on this strategy have been developed for sensing biothiols in living cells. All these probes presented here showed desirable properties, such as high sensitivity, high selectivity and preferred reaction kinetics. Moreover, the probes have been used for imaging of intracellular biothiols with emission colors ranging from green to near-infrared (NIR).

Abstract:On the basis of the protection reaction between ethanethiol and aldehyde, we designed and synthesized two new ratiometric fluorescent chemosensors, 3 and 4, by using intramolecular charge transfer (ICT) as a signaling mechanism. Upon the addition of Hg(2+) ion, both probes displayed apparent luminescence color changes, which could be observed by naked eyes under a UV lamp. Unexpectedly, both chemosensors also gave response to the addition of trace silver ions, making this kind of chemosensors as the first example of ratiometric fluorescent probe that showed dual channel fluorescence for both Hg(2+) and Ag(+). The test strips experiments suggested that 3 and 4 could serve as practical fluorescent probes for rapid detection of Hg(2+) ion.

Abstract:In this study, a highly selective fluorescent sensor (E)-2((2-(benzo[d]thiazol-2-yl)quinolin-8-yl)oxy)-N'-((7-(diethylamino)-2-oxo-2H-chromen-3-yl)methylene)acetohydrazide (TQC) was synthesized from 2methylquinolin-8-ol and 4-(diethylamino)-2-hydroxybenzaldehyde and its structure was characterized by NMR, C-13 NMR. E.51-HR-MS and density functional theory (NT) calculation. Sensor TQC showed an obvious "on-off-on" fluorescence response to Cu2+ and PPi in a DMSO/HEPES (3:2 v/v, pH - 7.4) buffer system. The detection limits of sensor TQC were 0.06 mu M to Cu2+ and 0.01 mu M to PPi. In addition, sensor TQC showed a 1:1 binding stoichiometry to Cu2+ and TQC-Cu2+ complex showed a 2:1 binding stoichiometry to PPi. The optimum pH range of sensor TQC and TQC-Cu2+ was 3-8. Further studies demonstrated that sensor TQC could be made into test paper strips for the qualitative of Cu2+ and PPi and showed sequentially "on-off-on" fluorescent bio-imaging of Cu2+ and PPi in HeLa cells. Abstract:Aliphatic biothiols, including cysteine, homocysteine, and glutathione, are associated with a wide range of biological functions. Hence, the selective detection of intracellular thiols has significance for investigating cellular function. Ratiometric fluorescent probes are very useful in the biological sciences because they allow measurement at two emission wavelengths, providing a built-in correction for environmental effects. In this study, a benzoxazine–hemicyanine based probe (1) was synthesized and characterized. The probe shows quick colorimetric and ratiometric response toward biothiols in the presence of other biomolecules and it functions in a wide pH range exhibiting large shifts in both absorption and fluorescence spectra. The detection mechanism, which was induced by the addition of mercaptan toward the activated double bond in the probe, was verified using 1H NMR and mass spectrometry analysis. The photophysical properties changes that occurred upon the reaction of the probe were found to be induced by the variation of intermolecular charge transfer, which was confirmed by theoretical calculations based on density functional theory (DFT). The fluorescence imaging of intracellular thiols was also examined with excellent performance.

Abstract:A new compound, N-(4-(1,5-diphenyl-4,5-dihydro-1H-pyrazol-3-yl)phenyl)-acrylamide (probe L), was designed and synthesized as a highly sensitive and selective fluorescent probe for recognizing and detecting thiol from other amino acids. On being mixed with thiol in buffered DMSO:HEPES = 1:1 solution at pH 7.4, the probe exhibited the blue emission at 474 nm. This probe is very sensitive and displayed a linear fluorescence off-on response to thiol. The fluorescence emission of the probe is pH independent in the physiological pH range. Living cell imaging of HeLa cells confirmed its cell permeability and its ability to selectively detect thiol in cells. A Water-Soluble Conjugated Polymer with Pendant Disulfide Linkages to Peg Chains: A Highly Efficient Ratiometric Probe with Solubility-Induced Fluorescence Conversion for Thiol Detection

Abstract:We investigated a water-soluble conjugated polymer (WSCP) with pendant disulfide linkages to poly(ethylene glycol) (PEG) chains, which is a highly efficient ratiometric probe with solubility-induced fluorescence conversion for thiol detection. This WSCP was doped with a low-bandgap fluorophore, 1,4-dithienyl benzothiadiazole (DBT), and was modified with PEGs by disulfide linkages to increase its water solubility. The free probe exhibited good solubility in aqueous solution (28 mg/mL) and showed purple fluorescence because of the low doping ratio of DBT. The separation of water-soluble PEG chains from the conjugated backbone induced by the cleavage of the disulfide linkages would lead to a significant decrease of the water solubility of the probe. The combined utilization of scanning electron microscopy, dynamic light scattering, and fluorescence spectrophotometer further confirmed that decreased solubility produced an aggregation of the hydrophobic conjugated backbone and subsequently increased fluorescence resonance energy transfer efficiency from the conjugated backbone to DBT which manifested as fluorescence conversion from purple to red. The fluorescence ratiometry (I-628/I-420) of the probe varied from the lowest value of 0.095 to 1.15 (12-fold maximum enhancement). The detection limit was 2.56 mu g/mL (0.021 mM). The WSCP probe was confirmed to be a good sensing material with high selectivity for thiols by examining various biological molecules. We also successfully achieved the imaging of intracellular thiols in HeLa cell. Considering that the disulfide could be replaced by other cleavable linkages, such a fluorescence ratiometry induced by decreased solubility could be utilized for detecting other chain-cleavable biomolecules, which would contribute to the development of new probes based on conjugated polymers.

Abstract:A selective phosphorescent biothiols probe is synthesized based on Ir(III) complex 1, which has 2,2'-biquinoline as the N^N ligand for realizing the satisfied two-photon absorption cross-section and two-functionalized 2-phenylpyridine ligands with an α,β-unsaturated ketone moiety as the thiol reaction site. The one- and two-photon optical properties of 1 are investigated through UV-vis absorption spectrum and photoluminescence spectrum. This Ir(III) complex can act as an excellent one- and two-photon excited "OFF-ON" phosphorescent probe for biothiols based on the 1,4-addition of biothiol to α,β-unsaturated ketones. Moreover, one- and two-photon-induced luminescent imagings of biothiols in living cells are also realized. Furthermore, the experiments of time-resolved photoluminescence technique and fluorescence lifetime imaging microscopy demonstrate that 1 is able to detect biothiols in the presence of strong background fluorescence. In addition, probe 1 is adsorbed into the shell of mesoporous silica nanoparticles with core-shell structure to form a nanoprobe, which can realize the ratiometric detection of biothiols in absolute water solution and living cells based on two phosphorescent signals.

Abstract:Biothiols, such as cysteine (Cys) and homocysteine (Hcy), play very crucial roles in biological systems. Abnormal levels of these biothiols are often associated with many types of diseases. Therefore, the detection of Cys (or Hcy) is of great importance. In this work, we have synthesized an excellent OFF-ON phosphorescent chemodosimeter 1 for sensing Cys and Hcy with high selectivity and naked-eye detection based on an IrIII complex containing a 2,4-dinitrobenzenesulfonyl (DNBS) group within its ligand. The OFF-ON phosphorescent response can be assigned to the electron-transfer process from IrIII center and C boolean AND N ligands to the DNBS group as the strong electron-acceptor, which can quench the phosphorescence of probe 1 completely. The DNBS group can be cleaved by thiols of Cys or Hcy, and both the 3MLCT and 3LC states are responsible for the excited-state properties of the reaction product of probe 1 and Cys (or Hcy). Thus, the phosphorescence is switched on. Based on these results, a general principle for designing OFF-ON phosphorescent chemodosimeters based on heavy-metal complexes has been provided. Importantly, utilizing the long emission-lifetime of phosphorescence signal, the time-resolved luminescent assay of 1 in sensing Cys was realized successfully, which can eliminate the interference from the short-lived background fluorescence and improve the signal-to-noise ratio. As far as we know, this is the first report about the time-resolved luminescent detection of biothiols. Finally, probe 1 has been used successfully for bioimaging the changes of Cys/Hcy concentration in living cells.

Abstract:A new series of intramolecular-charge-transfer (ICT) molecules (compounds 1, 2, and 3) were synthesized by attaching various electron-donating thiophenes groups to a triphenylamine backbone with an aldehyde group as the electron acceptor. Based on the protection reaction between ethanethiol and aldehyde, the corresponding dithioacetals (compounds S1, S2, and S3) were prepared to serve as novel colorimetric and fluorescent chemosensors for Hg2+ ions. Also, compound S1 was further utilized to construct the chemical-reaction-based conjugated polymer probe (PS1) towards Hg2+ ions. In the presence of as little as 10 nM Hg2+, compound PS1 displayed an apparent change in the fluorescent intensity. The sensing processes were revealed to be mediated by ICT, as confirmed by time-dependent DFT calculations. Furthermore, compound S1 was successfully applied to microscopic imaging for the detection of Hg2+ in HeLa cells with ratiometric fluorescent methods.

Abstract:In this work, a new reversible colorimetric and fluorescent probe for sequential recognition of copper ions and biothiols is synthesized easily. Based on the chelation-enhanced fluorescence quenching (CHEQ) effect, this probe shows high sensitivity and selectivity towards Cu2+, which can be detected by the naked eye. And this experimental phenomenon can also be realised upon addition of biothiols, restoring their initial fluorescence intensity. This probe also features a very high response speed (less than 5 seconds) and a large stokes shift (178 nm) toward Cu2+ and biothiols. Moreover, the detection limit for Cu2+ and biothiols is as low as 7.34 nM and 10.3 nM, respectively. Additionally, this ON-OFF-ON-type fluorescence recognition cycle can be repeated more than 5 times by addition of Cu2+ and biothiols in turn. Particularly, this 1-Cu2+ ensemble is further successfully applied for GSH detection in living cells.

Abstract:A 1,8-naphthalimide-Cu(II) ensemble was rationally designed and synthesized as a new turn-on fluorescent probe utilizing the 'chemosensing ensemble' method for detections of thiols (Cys, Hcy and GSH) with high selectivity over other alpha-amino acids at pH 7.4 in organic aqueous media (EtOH/HEPES, v/v = 9: 1). The recognition mechanism was attributed to the remove Cu(II) from the 1,8-naphthalimide-Cu(II) ensemble by thiols and the release of flurescence of ligand 1. Remarkable fluorescence enhancements were therefore observed in the sensing process of thiols by the 1,8-naphthalimide-Cu(II) ensemble. Furthermore, the 1,8-naphthalimide-Cu(II) ensemble was successfully applied to the fluorescence imaging of thiols in CHO cells with high sensitivity and selectivity. (C) 2013 Elsevier Ltd.
