Abstract:Photothermal therapy (PTT) has attracted attention as a highly effective non-invasive treatment method. However, the high localized temperatures (>50 °C) required for its treatment will inevitably cause damage to the surrounding normal tissues. Therefore, it is important to develop novel and effective strategies to achieve mild photothermal therapy (mPTT). The overexpression of heat shock proteins (HSPs), a widespread heat stress protein, leads to the generation of heat resistance in cancer cells, which seriously affects the therapeutic effect. Thus, inhibiting the expression of HSPs to reduce the heat resistance of tumor cells is expected to enhance the therapeutic effect of mPTT. Here, we successfully synthesized a fluorescent probe bonded with an amphiphilic polypeptide to a cyanine dye and achieved physical encapsulation of the blocker SB705498 through a self-assembly process. SB705498 promotes transient receptor potential vanilloid member 1 (TRPV1) channel blockade that can inhibit the translocation of the heat shock transcription factor 1 (HSF 1) by blocking the influx of calcium and thus affecting the expression of HSPs, which has the potential to enhance the thermotherapy of cancer under mild conditions. In addition, the nanoparticles enabled NIR-II fluorescence imaging with good stability and high photothermal conversion efficiency (48.10 %). Therefore, this study provides a new strategy for realizing precise mPTT(<45 °C) guided by NIR-II imaging. STATEMENT OF SIGNIFICANCE: Inhibition of overexpression of heat shock proteins (HSPs) in cancer photothermal therapy (PTT) is expected to enhance the therapeutic effect of mild photothermal therapy (mPTT). In this study, we synthesized a fluorescent probe bonded to cyanine dyes with amphiphilic polypeptides and physically wrapped the blocker SB705498 through a self-assembly process. As a transient receptor potential vanillin 1 (TRPV1) channel blocker, SB705498 inhibits heat shock transcription factor 1 (HSF1) translocation by blocking calcium ion influx, thereby improving mPTT efficacy by inhibiting the expression of HSPs. The nanoparticles also enable NIR-II fluorescence imaging with good stability and high photothermal conversion efficiency (48.10 %). Thus, this study provides a new strategy for NIR-II mPTT.

Abstract:Photothermal therapy is a promising phototherapeutic modality that has been widely studied in cancer therapy. However, because of the influence of heat shock protein (HSP), the therapeutic efficacy of photothermal therapy (PTT) is significantly suppressed. To improve the therapeutic efficacy, different tumor-specific therapeutic modalities have been chosen to combine with PTT. However, most of them rely on endogenous stimuli to trigger combination therapy, which may suffer from the issue of incomplete activation. Herein, we develop a PTT/thermodynamic combination therapeutic nanosystem whose therapeutic process is controlled by an external stimulus, near-infrared (NIR) light. The nanosystem (ADPPTN) is composed of a second NIR (NIR-II) fluorescent semiconducting polymer (SP) (DPPT) as the core, and a carboxyl group-decorated amphiphilic copolymer (PSMA-PEG) as the shell with an azo-containing compound (AIPH) loaded via electrostatic interaction. Under 808 nm laser irradiation, DPPT can generate heat to conduct PTT, while the elevated temperature may further trigger the release of AIPH radicals, conducting thermodynamic therapy (TDT). In addition, the NIR-II fluorescence signal emitted from DPPT can light the tumor. Compared with the nanoparticles without AIPH (DPPTN), ADPPTN has better anticancer efficacy under laser irradiation both in vitro and in vivo. Thus, our study provides an NIR-II fluorescence imaging-guided PTT/TDT combination therapeutic nanosystem for efficient cancer theranostics.

Abstract:To promote the solar-energy cascadeutilization, it is necessaryto increase the thermal effect of irradiation in the catalytic reactions,while simultaneously augmenting the non-thermal effect, so as to fulfillphotothermal coupling. Herein, the non-thermal and thermal effectof light radiation on the surface of In2O3-basedcatalysts are explored and enhanced by the modification of transitionmetals Fe and Cu. Optical characterizations combined with water-splittingexperiments show that Fe doping greatly broadens the radiation responserange and enhances the absorption intensity of semiconductors'intrinsic portion, and Cu doping facilitates the absorption of visible-infraredlight. The concurrent incorporation of Fe and Cu offers synergisticbenefits, resulting in improved radiation response range, carrierseparation and migration, as well as higher photothermal temperatureupon photoexcitation. Collectively, these advantages comprehensivelyenhance the photothermal synergistic water-splitting reactivity. Thecharacterizations under variable temperature conditions have demonstratedthat the reaction temperature exerts a significant influence on theprocess of radiation absorption and conversion, ultimately impactingthe non-thermal effect. The results of DFT calculations have revealedthat the increasing temperature directly impacts the chemical reactionby reducing the energy barrier associated with the rate-determiningstep. These findings shine new light on the fundamental mechanismsunderlying non-thermal and thermal effect, while also imparting significantinsights for photo-thermal-coupled catalyst designing.

Abstract:Manipulation of intracellular protein expression is a promising cancer therapeutic approach; however, traditional strategies relying on nonspecific molecular drugs often suffer from limited therapeutic benefits and off-target adverse events. Here, we report a semiconducting polymer nanomanipulator (SPNm) that can photothermally manipulate intracellular protein expression to synergize with its own function of second near-infrared (NIR-II) photothermal therapy (PTT) for thermal sensitization and metastasis-inhibited cancer therapy. SPNm comprises an NIR-II absorbing semiconducting polymer core as a photothermal transduction agent and a lysine-specific histone demethylase 3 A (KDM3A) inhibitor as a downstream effector protein expression manipulator, which are encapsulated into a thermo-responsive lipid shell. Upon 1064 nm laser irradiation, SPNm mediates the photothermal effect to melt the lipid shell for on-demand release of the KDM3A inhibitor to inhibit the demethylation function of KDM3A to upregulate the level of di-methylation of histone H3 lysine 9 (H3K9me2), and in turn simultaneously downregulate the expression of heat-shock protein 90 (HSP90) and cancer cell metastasis-related proteins, consequently inhibiting the thermo-tolerance response and metastasis of cancer cells. Thus, a single treatment of SPNm-mediated two-step NIR-II phototherapy achieves efficient tumor growth suppression at an unprecedented photothermal ablation depth (1 cm) and lung metastasis inhibition. Therefore, this study provides a promising thermal-controlled strategy to synergize phototherapeutics with protein expression manipulation for enhanced cancer treat-ment.(c) 2022 Elsevier Ltd. Abstract:Activatable photodynamic therapy (A-PDT) has attracted great attention in precision medicine, which can be activated by endogenous or exogenous stimuli to selectively produce reactive oxygen species (ROS) at the disease site. Thermal responsive polymers with a lower critical solution temperature (LCST) have normally been utilized for constructing A-PDT system. Herein, we fabricated a photothermal activatable photosensitizer (A-PS) by the combination of thermal responsive porphyrin-containing P(AAm-co-AN-co-TPP)-b-POEGMA amphiphilic block copolymer with an upper critical solution temperature (UCST) of 42 °C and a cyanine dye of IR780. The photoactivity of porphyrin units could be severely inhibited by IR780 due to the fluorescence resonance energy transfer (FRET) from TPP to IR780 during blood circulation process ("OFF" state). After an uptake by A549 cells and then irradiated with 808 nm laser, A-PS nanoparticles were subsequently dissociated owing to the increased local temperature above the UCST of the polymer chains by excellent photothermal conversion of IR780, resulting in the enhanced photoactivity of TPP ("ON" state) and the remarkable antitumor effect. Therefore, the UCST-based A-PS extended the biological application of thermal responsive polymers, which may provide a new insight into the design of smart cancer therapeutic systems.

Abstract:Despite immunotherapy involving immune checkpoint inhibitors (ICIs) have revolutionized cancer therapy, the clinical efficacy is limited due to ICI resistance. Pyroptosis is a gasdermin-mediated programmed cell death that enhances responses to ICIs. However, nontargeted elicitation of pyroptosis may induce systemic side effects and toxicity. Therefore, we reasonably design and construct a tumor-specific prodrug that combines the heat shock protein 90 inhibitor tanespimycin (17-AAG) with the photosensitizer chlorin e6 (Ce6) to induce pyroptosis, by utilizing the high glutathione level in the tumor microenvironment. The released Ce6 and 17-AAG produce reactive oxygen species by laser triggering, which induces gasdermin E-mediated pyroptosis. Furthermore, 17-AAG reduces myeloid-derived suppressor cells and sensitizes tumors to anti-programmed death-1 (PD-1) therapy. Thus, our prodrug strategy achieves tumor-targeted pyroptosis to suppress tumor growth, thereby improving the response to anti-PD-1 therapy and extending the survival of 4T1 breast tumor-bearing mice. Consequently, this pyroptosis-based prodrug represents a novel strategy for enforcing immunogenic photodynamic therapy.

Abstract:Theranostic nanoagents are promising for precision medicine. However, biodegradable nanoagents with the ability for photoacoustic (PA) imaging guided photothermal therapy (PTT) are rare. We herein report the development of biodegradable semiconducting polymer nanoparticles (SPNs) with enhanced PA and PTT efficacy for cancer therapy. The design capitalizes on the enzymatically oxidizable nature of vinylene bonds in conjunction with polymer chemistry to synthesize a biodegradable semiconducting polymer (DPPV) and transform it into water-soluble nanoparticles (SPNV). As compared with its counterpart SPN (SPNT), the presence of vinylene bonds within the polymer backbone also endows SPNV with a significantly enhanced mass absorption coefficient (1.3-fold) and photothermal conversion efficacy (2.4-fold). As such, SPNV provides the PA signals and the photothermal maximum temperature higher than SPNT, allowing detection and photothermal ablation of tumors in living mice in a more sensitive and effective way. Our study thus reveals a general molecular design to enhance the biodegradability of optically active polymer nanoparticles while dramatically elevating their imaging and therapeutic capabilities.

Abstract:Photothermal therapy (PTT) has attracted tremendous attention due to its noninvasiveness and localized treatment advantages. However, heat shock proteins (HSPs) associated self‐preservation mechanisms bestow cancer cells thermoresistance to protect them from the damage of PTT. To minimize the thermoresistance of cancer cells and improve the efficacy of PTT, an integrated on‐demand nanoplatform composed of a photothermal conversion core (gold nanorod, GNR), a cargo of a HSPs inhibitor (triptolide, TPL), a mesoporous silica based nanoreservoir, and a photothermal and redox di‐responsive polymer shell is developed. The nanoplatform can be enriched in the tumor site, and internalized into cancer cells, releasing the encapsulated TPL under the trigger of intracellular elevated glutathione and near‐infrared laser irradiation. Ultimately, the liberated TPL could diminish thermoresistance of cancer cells by antagonizing the PTT induced heat shock response via multiple mechanisms to maximize the PTT effect for cancer treatment.

Abstract:Nowadays, the photothermal therapy (PTT) has received widespread attention and research by rapidly killing tumors with local high temperature. However, due to the irregular edges of tumor and the blurred boundary between normal and necrotic tissues, the desirable treatment cannot be achieved by the single PTT, and excessive heat will cause serious inflammation in local tissues. Herein, an injectable composite hydrogel is prepared by the oxidized hyaluronic acid (OHA) and hydroxypropyl chitosan (HPCS) via the imine bonds, which is employed as the delivery substrate for functional substances. In the gel medium, the mesoporous polydopamine (MPDA) nanoparticles are incorporated as the high efficiency photothermal agent and a reservoir of DOX, which can achieve the good photothermal conversion performance and pulsed drug release. Besides, the addition of the curcumin-cyclodextrin host-guest inclusion complex (CUR@NH2-CD) in the composite hydrogel could reduce the inflammation caused by PTT. The composite hydrogel shows favorable the Hepa1-6 tumor inhibition in vivo by virtue of the comprehensive effect of the admired photothermal efficacy of MPDA, chemotherapy of DOX and anti-inflammatory of CUR. It can be predicted that the composite hydrogel has a broad prospect in the field of comprehensive therapy for tumor.

Abstract:Tumoral thermal defense mechanisms considerably attenuate the therapeutic outcomes of mild-temperature photothermal therapy (PTT). Thus, developing a simple, efficient, and universal therapeutic strategy to sensitize mild-temperature PTT is desirable. Herein, we report self-delivery nanomedicines ACy NPs comprising a near-infrared (NIR) photothermal agent (Cypate), mitochondrial oxidative phosphorylation inhibitor (ATO), and distearoylphosphatidylethanolamine-polyethylene glycol 2000 (DSPE-PEG2000), which have a high drug-loading efficiency that can reverse tumoral thermal resistance, thereby increasing mild-temperature PTT efficacy. ACy NPs achieved targeted tumor accumulation and performed NIR fluorescence imaging capability in vivo to guide tumor PTT for optimized therapeutic outcomes. The released ATO reduced intracellular ATP levels to downregulate multiple heat shock proteins (including HSP70 and HSP90) before PTT, which reversed the thermal resistance of tumor cells, contributing to the excellent results of mild-temperature PTT in vitro and in vivo. Therefore, this study provides a simple, biosafe, advanced, and universal heat shock protein-blocking strategy for tumor PTT.

Abstract:Photothermal therapy (PTT) shows great potential in tumor treatment, but the harsh photothermal heating might give rise to irreversible damage to peripheral normal tissues. Combining PTT with the alkyl radicals generated from the thermal degradation of azo initiators should be a good candidate for tumor therapy. Herein, semiconducting polymer nanoparticles (PBT NPs) with good photothermal conversion ability and photothermal stability have been developed as the photothermal agent. Under near-infrared light irradiation, the heat generated by PBT NPs could decompose 2,2′-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride into alkyl radicals, which could enhance the therapeutic effect of PTT under mild conditions. The combination of PTT and alkyl radicals provides a promising strategy to alleviate the side effects caused by hyperthermia and improve the treatment effect of PTT.

Abstract:Photothermal therapy (PTT), one of the most-potent cancer therapeutic strategies known, is highlighted with excessive inflammatory response, while ablating cancer with immunogenic death. This hyperactive immune response may override PTT-triggered immunogenicity, exacerbate skin empyrosis, and incur permanent tissue injury and high-profile tumor regeneration. Therefore, an anticancer balance between pathological and protective immune response is urgently needed for an advanced photothermal therapeutic tactic. Herein, a gas-modulated photothermal immunogenicity strategy is proposed by integrating an amphiphilic-conjugated polymer with a polysulfide-based hydrogen sulfide (H2 S) donor (2,2'-dipyridyl tetrasulfide@CP-PEG) (where CP = conjugated polymer and PEG = poly(ethylene glycol)). The CP is endowed with NIR-II fluorescence capacity and favorable photothermal effect, tracing the tumor for precise therapeutics. The polysulfide donor can release H2 S triggered by intracellular glutathione, which elicits mitochondrial dysfunction and robust anti-inflammation effect. Ultimately, this gas-modulated PTT strategy inhibits tumor growth remarkably and limits the magnitude of PTT-induced proinflammatory tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-1beta (IL-1β) cytokines. Moreover, the regulated inflammation accelerates PTT-induced wound healing. A H2 S-modulated PTT with adaptive immune response is thus recommended as an advanced strategy to cancer therapeutics.

Abstract:Stress‐activated signal inhibition boosted photothermal therapy (PTT): through the integration of specific inhibitors, Hair NPs‐HA@inhibitor can inhibit the typical heat‐activated signals (i.e., HSP90 and p38 MAPK signaling pathway) that are closely related with tumor survival and invasion, leading to the reduced thermoresistance of tumor cells and superior cell killing effect by enhanced PTT. Several stress‐activated signals are highly responsible for the thermal tolerance and attribute to the poor therapeutic outcome of photothermal therapy (PTT). In this study, the naturally human hair‐derived nanoparticles (Hair NPs) with significant photothermal conversion efficacy are explored to combine with stress‐activated signal inhibitors for maximizing the PTT efficiency. During the Hair NPs‐based PTT, two distinct signals are found to be remarkably upregulated. One is the heat‐induced overexpression of HSP90, and the other one is the activation of mitogen‐activated protein kinase (MAPK) signaling pathway. Through the integration of particular inhibitors (17‐allylamino‐17‐demethoxygeldanamycin (17AAG) specifically binding to HSP90 or LY2228820 targeting to p38 MAPK) and the subsequent modification of hyaluronic acid (HA), the resultant Hair NPs‐HA@inhibitor can specifically deliver the inhibitor into tumor cells with minimal side effects on normal tissues, which can specifically block the stressfully activated signals as well as inhibit the expression of their downstream effectors (e.g., MMPs, VEGF) that are closely related with tumor survival and invasion. Without these elements to protect tumor cells or promote tumor progression, the Hair NPs‐induced PTT effect can be much elevated, resulting in a superior antitumor efficacy. These findings represent an effective approach in maximizing PTT effect for fighting against tumor.

Abstract:Although many approaches including operative treatment, chemotherapy, and radiotherapy have been developed to struggle against cancer, they are still insufficient in clinical application owing to the non‐ideal therapeutic efficiency and serious side effects. Photothermal therapy (PTT) is emerging as a novel alternative to the conventional therapies because of minimal invasiveness and high specificity. Conjugated polymers (CPs) with the merits of unique optical properties and biological activities have been extensively investigated as functional photothermal conversion materials. The critical design strategies of CPs with high photothermal conversion efficiency and excellent PTT effect are reviewed in this progress report.
