The Soluble (Pro)Renin Receptor Induces Endothelial Cell Dysfunction By Activating The Nox4/Nf-Kappa B Pathway

Abstract:Objectives: The circulating soluble (pro)renin receptor (sPRR) is increased in patients with several cardiovascular diseases. It is unknown whether sPRR alters vascular endothelial cell function. We tested the hypothesis that elevated sPRR evokes endothelial cell dysfunction. Methods: Primary human umbilical vein endothelial cells (HUVECs) treated (2 h) with a recombinant histidine-tagged sPRR (sPRR-His; 50 nM) exhibited IκBα protein degradation concurrent with nuclear translocation of NF-κB p65 and increased NF-κB p65 transcription factor activity. At 2 h, sPRR-His induced a increase in Nox4 protein and a increase in medium H2O2 as assessed by immunoblotting and ROS-Glo H2O2 assay, respectively. At 3 h of sPRR-His treatment, the mRNA expression of IL-6, IL-8, VCAM-1 and ICAM-1 was increased as assessed by qRT-PCR; ELISA detected a increase in medium IL-6 and IL-8. Moreover, enhanced apoptosis was evidenced by a increase of cleaved caspase 3 protein and a increase in the apoptotic cell number, as assessed by immunoblotting and annexin V-FITC/PI double staining flow cytometry, respectively. In addition, sPRR-His induced a decrease in phospho-eNOS (Ser1177) protein abundance. Results: A Nox1/4 inhibitor GKT137892, Nox4 siRNA, and NADPH oxidase inhibitor apocynin completely abolished sPRR-His-induced NF-κB activation, inflammation, adhesion, and apoptosis, similar to the effect of NF-κB inhibition with PDTC. Conclusion: sPRR-His evokes Nox4-derived ROS production which degrades IκBα to an extent that precipitates nuclear translocation of NF-κB p65 and promotes inflammation, adhesion, apoptosis, and decreases p-eNOS (Ser1177):eNOS. sPRR-His should be considered as a viable therapeutic target in the treatment and / or prevention of cardiovascular disorders.
