Abstract:Graphene is a potential candidate for applications in biomedical field. It is inevitable that graphene is in contact with the ubiquitous bacterial environment. More attention has been paid to the antimicrobial activity of graphene derivatives (graphene oxide, reduced graphene oxide) than the interaction between graphene and bacteria. Herein, we explore interaction between graphene micron-sheet and bacteria from micro (gene expression) and macro (colonies) perspectives. Results demonstrate that graphene micron-sheet accelerates the biofilm forming thus promoting pathogen expansion toward both Gram-negative bacteria E. coli and Gram-positive bacteria S. aureus. The graphene micron-sheet acts as a "habitat" for increasing bacterial attachment and biofilm forming. For E. coli, graphene micron-sheet, firstly changes the integrity of periplasmic and outer membrane components, then makes membrane-associated and cell division genes increased, and finally promotes bacterial proliferation; For S. aureus, graphene micron-sheet can accelerate biofilm forming and develop bacterial expansion owing to the regulation of the quorum-sensing system and global regulatory proteins. The work can shed new light on the range of possible mode of actions, developing a better understanding of the capabilities of graphene micron-structures.

Abstract:Graphene, a unique two-dimensional single-atom-thin nanomaterial with exceptional structural, mechanical, and electronic properties, has spurred an enormous interest in many fields, including biomedical applications, which at the same time ignites a growing concern on its biosafety and potential cytotoxicity to human and animal cells. In this review, we present a summary of some very recent studies on this important subject with both experimental and theoretical approaches. The molecular interactions of graphene with proteins, DNAs, and cell membranes (both bacteria and mammalian cells) are discussed in detail. Severe distortions in structures and functions of these biomacromolecules by graphene are identified and characterized. For example, the graphene is shown to disrupt bacteria cell membranes by insertion/cutting as well as destructive extraction of lipid molecules directly. More interestingly, this cytotoxicity has been shown to have implications in de novo design of nanomedicine, such as graphene-based band-aid, a potential 'green' antibiotics due to its strong physical-based (instead of chemical-based) antibacterial capability. These studies have provided a better understanding of graphene nanotoxicity at both cellular and molecular levels, and also suggested therapeutic potential by using graphene's cytotoxicity against bacteria cells.

Abstract:Examining interactions between nanomaterials and cell membranes can expose underlying mechanisms of nanomaterial cytotoxicity and guide the design of safer nanomedical technologies. Recently, graphene has been shown to exhibit potential toxicity to cells; however, the molecular processes driving its lethal properties have yet to be fully characterized. We here demonstrate that graphene nanosheets (both pristine and oxidized) can produce holes (pores) in the membranes of A549 and Raw264.7 cells, substantially reducing cell viability. Electron micrographs offer clear evidence of pores created on cell membranes. Our molecular dynamics simulations reveal that multiple graphene nanosheets can cooperate to extract large numbers of phospholipids from the membrane bilayer. Strong dispersion interactions between graphene and lipid-tail carbons result in greatly depleted lipid density within confined regions of the membrane, ultimately leading to the formation of water-permeable pores. This cooperative lipid extraction mechanism for membrane perforation represents another distinct process that contributes to the molecular basis of graphene cytotoxicity.

Abstract:To explore the biocompatibility of graphene oxide (GO) with proteins, we investigated its effects on conformation and activity of catalase, by using molecular spectroscopic techniques such as UV–visible absorption, fluorescence, circular dichorism (CD) and fourier transform infrared (FT-IR) spectroscopy, and conducting in vitro enzymatic activity assay. CD combined with FT-IR and UV–visible spectra measurements indicated that GO induced the decrease of α-helical content and the increase of β-sheet structure in catalase, resulting in the loosening and unfolding of the protein skeleton. The internal hydrophobic amino acids (mainly tryptophan) in the protein were exposed, and their characteristic fluorescence was distinctly quenched, according to synchronous fluorescence and fluorescence emission together with UV–visible spectra analyses. The results showed that the tertiary and secondary structure of catalase is changed by GO. The conformational changes of the protein were also found to show a dependence on concentration and time of GO treatment. Moreover, the activity assay indicated that catalase activity is inhibited by GO. The study demonstrated that GO could cause negative effects on proteins, and the biosafety of its application may worth further investigation.

Abstract:Significance In biomedical and environmental applications, as well as manufacture and disposal, the interaction of graphene-based nanomaterials (GBNs) with living cells is inevitable and sometimes crucial. While the cytotoxic properties of GBNs are well established, the mechanisms behind the cytotoxicity remain controversial. In this study, we first utilize a magnetic field to form films with aligned graphene oxide (GO), showing that the alignment of sharp GO edges plays a crucial role in the antibacterial activity. We then demonstrate using model systems that GO unequivocally induces physical disruption of lipid bilayers and that oxidation stems from a direct electron transfer mechanism. Altogether, our results elucidate the physicochemical, edge-based cytotoxicity of GBNs, while providing guidance for the design of engineered surfaces using GBNs.

Abstract:Biofilm contamination presents a significant threat to public health, the food industry, and aquatic/marine-related applications. In recent decades, although various methods have emerged to combat biofilm contamination, the intricate and persistent nature of biofilms makes complete eradication challenging. Therefore, innovative alternative solutions are imperative for addressing biofilm formation. Instead of solely focusing on the eradication of mature biofilms, strategically advantageous measures involve the delay or prevention of biofilm formation on surfaces. Quorum sensing, a communication system enabling bacteria to coordinate their behavior based on population density, plays a pivotal role in biofilm formation for numerous microbial species. Materials possessing antibiofilm properties that target quorum sensing have gained considerable attention for their potential to prevent biofilm formation. This Review consolidates recent research progress on the utilization of materials with antiquorum sensing properties for combating biofilm formation. These materials can be categorized into three distinct types: (i) antibiofilm nanomaterials, (ii) antibiofilm surfaces, and (iii) antibiofilm hydrogels with antiquorum sensing capabilities. Finally, the Review concludes with a brief discussion of current challenges and outlines potential avenues for future research.
Bi-functional quercetin/copper nanoparticles integrating bactericidal and anti-quorum sensing properties for preventing the formation of biofilms

Abstract:Biofilms formed by pathogenic bacteria present a persistent risk to human health. While the eradication of matured biofilms remains a formidable challenge, delaying or preventing their formation, which is coordinately regulated by quorum sensing (QS), presents a simpler and more advantageous strategy. Quercetin, a naturally occurring compound with anti-QS properties, has the potential to act as an antibiofilm agent. However, it is plagued by certain inherent drawbacks, including poor water solubility and limited bioavailability. Furthermore, solely blocking QS is not enough to prevent biofilm formation because it lacks bactericidal properties. To address these difficulties, we fabricated a bi-functional nanoparticle through the co-assembly of quercetin and copper ions in a facile manner. The resulting quercetin/copper nanoparticles (QC NPs) demonstrated minimal cytotoxicity and hemolysis in vitro. In response to the low pH of microenvironments that were populated by bacteria colonies, the QC NPs underwent disassembly, releasing copper ions and quercetin. The former exterminated bacteria by disrupting the integrity of the cell membrane, while the latter disrupted the processes involved in QS that are responsible for the biofilm by downregulating the expression of specific genes, effectively preventing the formation of biofilms by both Gram-negative Pseudomonas aeruginosa and Gram-positive Staphylococcus aureus. In addition, QC NPs were integrated into a bacterial cellulose membrane to provide antibiofilm properties. The composite membrane was proven to be highly effective in inhibiting biofilm formation in vitro and also demonstrated the ability to reduce inflammatory responses and accelerate the healing of bacteria-infected wounds in vivo. Overall, the bi-functional QC NPs hold great potential for use in addressing the challenges associated with the management of bacterial biofilms.

Graphene oxide affects bacteriophage infection of bacteria by promoting the formation of biofilms

Abstract:Graphene oxide (GO) is increasingly used in a range of fields, such as electronics, biosensors, drug delivery, and water treatment, and the likelihood of its release into the environment is increasing correspondingly. GO is involved in the formation of biofilms and leads bacteria to over proliferate, but the effects of GO on bacteriophage infection remain unexplored. We noted bacterial overgrowth in experiments when GO was used to treat the bacterial culture medium, leading us to question whether bacterial proliferation caused by GO affects phage infection of target bacteria. Treating Pseudomonas aeruginosa with GO at a low dosage (0.02 mg/mL) led to biofilm expansion in LB medium. Biofilm formation in the presence of GO affected the ability of bacteriophages to kill bacteria and reproduce. Similarly, the presence of GO deposits increased the ratio of bacteria to phage, providing a favorable environment for bacterial growth. Additionally, increasing the positive electrical charge in the culture environment inhibited the rejection of bacteriophages by negatively charged GO, improving phage reproduction. Finally, adding GO to sewage in imitation field experiments significantly increased the bacterial diversity and richness in the sewage, stimulating a significant increase in the variety and number of bacteria. Collectively, these results indicate that GO hinders phage infection by providing a bacterial refuge. The results of this study provide valuable insights into how GO interacts with bacteriophages to explore the effects on bacterial growth.

Abstract:In this study, a novel nanobiocomposite was synthesized using graphene oxide, lignin, silk fibroin and ZnO and used in biological fields. To synthesize this structure, after preparing graphene oxide by the Hummer method, lignin, silk fibroin, and ZnO nanoparticles (NPs) were added to it, respectively. Also, ZnO NPs with a particle size of about 18 nm to 33 nm was synthesized via Camellia sinensis extract by green methodology. The synthesized structure was examined as anti-biofilm agent and it was observed that the Graphene oxide-lignin/silk fibroin/ZnO nanobiocomposite has a significant ability to prevent the formation of P. aeruginosa biofilm. In addition, due to the importance of the possibility of using this structure in biological environments, its toxicity and blood compatibility were also evaluated. According to the obtained results from MTT assay, the viability percentages of Hu02 cells treated with Graphene oxide-lignin/silk fibroin/ZnO nanobiocomposite after 24, 48, and 72 h of incubation were 89.96%, 89.32%, and 91.28%. On the other hand, the hemolysis percentage of the synthesized structure after 24 h and 72 h of extraction was 9.5% and 11.76% respectively. As a result, the synthesized structure has a hemolysis percentage below 12% and its toxicity effect on Hu02 cells is below 9%.

The Effect of Water Hardness on the Toxicity of Graphene Oxide to Bacteria in Synthetic Surface Waters.

Abstract:Graphene oxide (GO), used in a wide variety of applications, is increasingly being introduced into aquatic environments; this situation calls for research on GO toxicity to assess its environmental risks. In this study, the toxic effect of GO to E.coli was studied before and after its aggregation equilibrium in the synthetic surface waters (the soft water, moderately hard water, and hard water) to reveal the effects of GO aggregation and solution hardness. The cytotoxicity of GO increased with increasing solution hardness while decreased after GO aggregation. The 3 h 50% inhibitory concentration (IC50) values of dispersed GO in the soft water, moderately hard water, and hard water were 12.2 +/- 2.2, 8.5 +/- 1.5, and 4.0 +/- 1.0 mg/L, respectively. After 24 h shaking (aggregation equilibrium) in the synthetic surface waters, the dispersed GO aggregated and the 3 h IC50 values of GO aggregates in the three synthetic waters were 40.3 +/- 6.9, 15.9 +/- 2.2, and 7.5 +/- 1.5 mg/L, respectively. The dispersed GO sheets wrapped E. coli cells and cut the cell membrane, resulting in the disruption of cell membrane and the cell inactivation. With increasing water hardness, the heteroaggregation between GO sheets/aggregates and E. coli cells was enhanced, resulting in the increase of toxic effect. The GO aggregates could also entrap E.coli cells while exhibited limited effect on cell membrane disruption without sharp edges, thereby causing the lower toxic effect compared with the dispersed GO sheets. These outcomes shed new light on the assessment of ecological effects of GO.
Bacterial Surface Disturbances Affecting Cell Function during Exposure to Three-Compound Nanocomposites Based on Graphene Materials

Abstract:Combating pathogenic microorganisms in an era of ever-increasing drug resistance is crucial. The aim of the study was to evaluate the antibacterial mechanism of three-compound nanocomposites that were based on graphene materials. To determine the nanomaterials’ physicochemical properties, an analysis of the mean hydrodynamic diameter and zeta potential, transmission electron microscope (TEM) visualization and an FT-IR analysis were performed. The nanocomposites’ activity toward bacteria species was defined by viability, colony forming units, conductivity and surface charge, cell wall integrity, ATP concentration, and intracellular pH. To ensure the safe usage of nanocomposites, the presence of cytokines was also analyzed. Both the graphene and graphene oxide (GO) nanocomposites exhibited a high antibacterial effect toward all bacteria species (Enterobacter cloacae, Listeria monocytogenes, Salmonella enterica, and Staphylococcus aureus), as well as exceeded values obtained from exposure to single nanoparticles. Nanocomposites caused the biggest membrane damage, along with ATP depletion. Nanocomposites that were based on GO resulted in lower toxicity to the cell line. In view of the many aspects that must be considered when investigating such complex structures as are three-component nanocomposites, studies of their mechanism of action are crucial to their potential antibacterial use.

Influences of graphene on microbial community and antibiotic resistance genes in mouse gut as determined by high-throughput sequencing.

Abstract:Graphene is a promising candidate as an antibacterial material owning to its bacterial toxicity. However, little information on influence of graphene on gut microbiota is available. In this study, mice were exposed to graphene for 4 weeks, and high-throughput sequencing was applied to characterize the changes in microbial community and antibiotic resistance genes (ARGs) in mouse gut. The results showed that graphene exposure increased biodiversity of gut microbiota, and changed their community. The 1 mu g/d graphene exposure had higher influences on the gut microbiota than 10 mu g/d and 100 mu g/d graphene exposures, which might be due to higher aggregation of high-level graphene. The influence of graphene on gut microbiota might attribute to that graphene could induce oxidative stress and damage of cell membrane integrity. The results were verified by the increase of ratio of Gram-negative bacteria. Outer membrane of Gram-negative bacteria could reduce the membrane damage induced by graphene and make them more tolerance to graphene. Further, we found that graphene exposure significantly increased the abundance and types of ARGs, indicating a potential health risk of graphene. Effects of Low-Level Engineered Nanoparticles on the Quorum Sensing of Pseudomonas Aeruginosa PAO1

Abstract:The toxicity of engineered nanoparticles (ENPs) on bacteria has aroused much interest. However, few studies have focused on the effects of low-level ENPs on bacterial group behaviors that are regulated by quorum sensing (QS). Herein, we investigated the effects of nine ENPs (Ag, Fe, ZnO, TiO2, SiO2, Fe2O3, single-wall carbon nanotubes (SWCNTs), graphene oxide (GO), and C60) on QS in Pseudomonas aeruginosa PAOl. An ENP concentration of 100 μg L−1 did not impair bacterial growth. However, concentrations of 100 μg L−1 of Ag and GO ENPs induced significant increases in 3OC12–HSL in the culture and significantly promoted protease production and biofilm formation of PAO1. C4–HSL synthase and its transcription factors were less sensitive to 100 μg L−1 Ag and GO ENPs compared with 3OC12-HSL. Fe ENPs induced a significant increase in the 3OC12–HSL concentration, similar to Ag and GO ENPs. However, Fe ENPs did not induce any significant increase in protease production or biofilm formation. Different size distributions, chemical compositions, and aggregation states of the ENPs had different effects on bacterial QS. These whole circuit indicators could clarify the effects of ENPs on bacterial QS. This study furthers our understanding of the effects of low-level ENPs on bacterial social behaviors.
Nano-enabled Quenching of Bacterial Communications for the Prevention of Biofilm Formation

Abstract:Biofilm formation is a major threat to industry, the environment and human health. While killing of embedded microbes in biofilms may inevitably lead to the evolution of antimicrobial resistance (AMR), catalytic quenching of bacterial communications by lactonase is a promising antifouling approach. Given the shortcomings of protein enzymes, it is attractive to engineer synthetic materials to mimic the activity of lactonase. Herein, an efficient lactonase-like Zn-Nx -C nanomaterial was synthesized by tuning the coordination environment around zinc atoms to mimic the active domain of lactonase for catalytical interception of bacterial communications in biofilm formation. The Zn-Nx -C material could selectively catalyze 77.5 % hydrolysis of N-acylated-L-homoserine lactone (AHL), a critical bacterial quorum sensing (QS) signal in biofilm construction. Consequently, AHL degradation downregulated the expression of QS-related genes in antibiotic resistant bacteria and significantly prevented biofilm formation. As a proof of concept, Zn-Nx -C-coated iron plates prevented 80.3 % biofouling after a month exposure in river. Overall, our study provides a nano-enabled contactless antifouling insight to avoid AMR evolution by engineering nanomaterials for mimicking the key bacterial enzymes (e.g., lactonase) functioning in biofilm construction.
Molecular Machinery Responsible for Graphene Oxide's Distinct Inhibitory Effects towards Pseudomonas aeruginosa and Staphylococcus aureus Pathogens

Abstract:Graphene oxide flakes are considered as potential inhibitors for different pathogenic bacteria. However, the efficacy of inhibition changes for different types and strains of bacteria. In this work, we examine Pseudomonas aeruginosa and Staphylococcus aureus, two common hospital-acquired infections, which react quite differently to graphene oxide flakes. The minimum inhibitory tests yield two distinct outcomes: stopped proliferation for S. aureus versus almost no effect for P. aeruginosa. Integrating our experimental evidence with molecular dynamics simulations, we elucidate the molecular machinery involved, explaining the behavior we see in scanning electron microscopy images. According to our simulations, the peptidoglycan network, the outermost layer of S. aureus, is completely entangled with the flakes, acting as a hunting ground, which consequently results in the inhibition of the pathogen itself. Lipopolysaccharides, the outermost layer of P. aeruginosa, on the other hand, resist interacting with the flakes. Lipopolysaccharides make no effective contacts, and thus no effective inhibition of the pathogen takes place. Likewise, the electron microscopy images show complete coverage and wrapping of S. aureus. In contrast, for P. aeruginosa, barely any bacteria are spotted with any flakes on top except for some loosely half-covered cases. As we did not observe any damaged bacteria in our images, we exclude the knife-cutting inhibition mechanism and suggest the wrapping and trapping mechanism for S. aureus for our flakes' rather large size (average area of 0.05 μm2). Antibacterial Activity of Graphite, Graphite Oxide, Graphene Oxide, and Reduced Graphene Oxide: Membrane and Oxidative Stress.

Abstract:Health and environmental Impacts of graphene-based materials need to be thoroughly evaluated before their potential applications. Graphene has strong cytotoxicity toward bacteria. To better understand its antimicrobial mechanism, we compared the antibacterial activity of four types of graphene-based materials (graphite (Gt), graphite oxide (GtO), graphene oxide (GO), and reduced graphene oxide (rGO)) toward a bacterial model Escherichia coli. Under similar concentration and incubation conditions, GO dispersion shows the highest antibacterial activity, sequentially followed by rGO, Gt, and GtO. Scanning electron microscope (SEM) and dynamic light scattering analyses show that GO aggregates have the smallest average size among the four types of materials. SEM images display that the direct contacts with graphene nanosheets disrupt cell membrane. No superoxide anion (O-2(center dot-)) induced reactive oxygen species (ROS) production is detected. However, the four types of materials can oxidize glutathione, which serves as redox state mediator in bacteria. Conductive rGO and Gt have higher oxidation capacities than insulating GO and GtO. Results suggest that antimicrobial actions are contributed by both membrane and oxidation stress. We propose that a three-step antimicrobial mechanism, previously used for carbon nanotubes, is applicable to graphene-based materials. It includes initial cell deposition on graphene-based materials, membrane stress cared by direct contact with sharp nanosheets, and the ensuing superoxide anion-independent oxidation. We envision that physicochemical properties of graphene-based materials, such as density of functional groups, size, and conductivity, can be precisely tailored to either reducing their health and environmental risks or increasing their application potentials.

Abstract:As a complex microbial aggregate, biofilm is a group behavior of bacterial ability to adapt to the environment. Bacteria produce biofilm substrates that enhance their tolerance to stress and cause microbial infections. Biofilm infection is usually closely related to virulence, pathogenicity, and even life-threatening to immunocompromised patients. Therefore, studying bacterial biofilm generation and regulatory mechanisms has become one of the most important fields. It is well known that biofilm formation involves group behavior and relies on complex regulation of quorum sensing (QS). A series of small molecule compounds such as indole, AI-2 (autoinducer-2), AHL (N-acyl-homoserine lactone), AIP (auto-inducing peptide), and DSF (diffusible signal factor) are widely available intraspecific or interspecific signaling molecules, with regulatory functions on a wide range of physiological activities of bacteria, including biofilm formation. Given that various bacteria employ QS mechanisms to regulate biofilm formation, inhibition of QS becomes a promising potential strategy for the treatment of bacterial infections. Here, we describe how bacterial intraspecific and interspecific signaling molecules regulate the mechanism of biofilm formation and dispersion. This may contribute to anti-biofilm active molecules and provide ideas or directions for studies on controlling bacterial infections by inhibiting biofilm formation through QS.

Lateral Dimension-Dependent Antibacterial Activity of Graphene Oxide Sheets.

Abstract:Graphene oxide (GO) is a promising precursor to produce graphene-family nanomaterials for various applications. Their potential health and environmental impacts need a good understanding of their cellular interactions. Many factors may influence their biological interactions with cells, and the lateral dimension of GO sheets is one of the most relevant material properties. In this study, a model bacterium, Escherichia coli ( E. coli ), was used to evaluate the antibacterial activity of well-dispersed GO sheets, whose lateral size differs by more than 100 times. Our results show that the antibacterial activity of GO sheets toward E. coli cells is lateral size dependent. Larger GO sheets show stronger antibacterial activity than do smaller ones, and they have different time- and concentration-dependent antibacterial activities. Large GO sheets lead to most cell loss after 1 h incubation, and their concentration strongly influences antibacterial activity at relative low concentration (<10 μg/mL). In contrast, when incubating with small GO sheets up to 4 h, the inactivation rate of E. coli cells continues increasing. The increase of small GO sheet concentration also results in persistent increases in their antibacterial activity. In this study, GO sheets with different lateral sizes are all well dispersed, and their oxidation capacity toward glutathione is similar, consistent with X-ray photoelectron spectroscopy and ultraviolet-visible absorption spectroscopy results. This suggests the lateral size-dependent antibacterial activity of GO sheets is caused by neither their aggregation states, nor oxidation capacity. Atomic force microscope analysis of GO sheets and cells shows that GO sheets interact strongly with cells. Large GO sheets more easily cover cells, and cells cannot proliferate once fully covered, resulting in the cell viability loss observed in the followed colony counting test. In contrast, small GO sheets adhere to the bacterial surfaces, which cannot effectively isolate cells from environment. This study highlights the importance of tailoring the lateral dimension of GO sheets to optimize the application potential with minimal risks for environmental health and safety.

Abstract:Clarifying the mechanisms of cellular interactions of graphene family nanomaterials is an urgent issue to the development of guidelines for safer biomedical applications and to the evaluation of health and environment impacts. By combining large-scale computer simulations, theoretical analysis, and experimental discussions, here we present a systematic study on the interactions of graphene nanosheets having various oxidization degrees with a model lipid bilayer membrane. In the mesoscopic simulations, we investigate the detailed translocation pathways of these materials across a 56 × 56 nm2 membrane patch which allows us to fully consider the role of membrane perturbation during this process. A phase diagram regarding the transmembrane translocation mechanisms of graphene nanosheets is thereby obtained in the space of oxidization degree and particle size. Then, we propose a theoretical approach to analyze the effects of various initial equilibrium states of graphene nanosheets with membrane on their following cellular uptake process. Finally, we demonstrate that the simulation and theoretical results reproduce some important experimental findings towards the mechanisms of cytotoxicity and antibacterial activity of graphene materials. These results not only provide new insight into the cellular internalization mechanism of graphene-based nanomaterials but also offer fundamental understanding on their physicochemical properties which can be precisely tailored for safer biomedical and environment applications.
