A13710 Calcineurin Dephosphorylates and Regulates the Ubiquitin Ligase Kelch-like 3

Abstract:Objectives: Kelch-like 3 (KLHL3) is a component of an E3 ubiquitin ligase that regulates blood pressure (BP) by targeting With-No-Lysine (WNK) kinases for degradation. Previously, we reported that angiotensin II inactivates KLHL3 by protein kinase C-mediated phosphorylation at S433 (KLHL3-S433-P) (Shibata et al. PNAS 2014), resulting in increased salt reabsorption and BP elevation. The identity of a phosphatase regulating KLHL3-S433-P was unknown. Given that calcineurin inhibitors (CNIs) cause hypertension, we postulated that calcineurin dephosphorylates KLHL-S433-P. Methods: Active calcineurin and purified KLHL3 were incubated in vitro, and KLHL3-S433-P levels were analyzed by Western blotting using phospho-specific antibody (Ishizawa et al. BBRC 2016). We also analyzed whether calcineurin affects KLHL3-S433-P in cell culture by using tacrolimus, a CNI, and siRNA knockdown of calcineurin A. In vivo, we evaluated the levels of KLHL3-S433-P in mice that received subcutaneous injection of tacrolimus. Results: In in vitro phosphatase assay, calcineurin markedly reduced KLHL3-S433-P levels. The results were also confirmed by malachite green phosphate assay. In HEK cells expressing KLHL3, activation of calcineurin by ionomycin, a calcium ionophore, induced a significant decrease in KLHL3-S433-P, which was blocked by tacrolimus and by calcineurin A knockdown. In addition, tacrolimus significantly reduced WNK4 ubiquitination and increased WNK4 levels associated with the increase in KLHL3-S433-P, confirming the functional significance. Moreover, in vivo, we found that KLHL3-S433-P levels were significantly increased in the kidney of mice that received tacrolimus. Conclusion: These data indicate that calcineurin dephosphorylates KLHL3-S433-P, and provide insights into the mechanism of CNI-induced BP elevation.
