THE APOLIPOPROTEIN E GENE AFFECTS THE DEFAULT MODE NETWORK ACROSS THE ALZHEIMER’S DISEASE SPECTRUM

Abstract:Previous studies that investigated the effects of apolipoprotein E (APOE) ε4 status on default mode networks (DMN) and its disruption due to the presence of a disease such as Alzheimer's disease (AD). However, no studies have investigated DMN connectivity across the Alzheimer's disease spectrum of APOE population. The aim of this study was to exploring DMN functional connectivity(FC) changes of APOE population might provide valuable evidence for better understanding the development of AD. Magnetic resonance imaging data and neuropsychological test scores from 169 subjects (CN = 46, EMCI = 52, LMCI = 41, AD=30) were obtained from the Alzheimer's Disease Neuroimaging Initiative. Posterior cingulate cortex seed-based FC analysis was used to obtain the DMN patterns. To analyze the interaction between the ApoE genotype, and disease on the DMN, a whole-brain voxel-wise mixed ANCOVA with ApoE genotype (ε4 carriers and non-carriers), disease status (control, EMCI, LMC or AD) as fixed factors was performed, controlling for age, gender, education, and gray matter volumes (using the full factorial option in SPM8). Mediation analysis was performed to further examine whether DMN FC strength could mediate the effects of APOE genotype on MMSE in patients. the trend for changed DMN FC strength was opposite in ε4 carriers compared to ε4 non-carriers across the Alzheimer's disease spectrum patients. Specifically, the increases in posterior cingulate cortex FC with the right precuneus, insular, fusiform were positively correlated with the changes in mini-mental state examination (MMSE) in ε4 non-carriers across the Alzheimer's disease spectrum patients. Furthermore, Conditional analysis revealed that connectivity strength regulates the effects of APOE genotype on cognitive function across the Alzheimer's disease spectrum patients.
