Abstract:With the widespread contamination of ochratoxin A (OTA), it is of significant importance for detecting OTA in foods and traditional Chinese medicine (TCM). In this study, a novel label-free fluorescent aptasensor utilizing the interaction between OTA-triggered antiparallel G-quadruplex and (N-methyl-4-pyridy) porphyrin (TMPyP) for the rapid and sensitive determination of OTA was established. The fluorescence of CdTe quantum dots (QDs) could be quenched by TMPyP. In the presence of analyte (OTA), the aptamer could recognize OTA and transform from a random coil to the antiparallel G-quadruplex. The interaction between G-quadruplex and TMPyP could release CdTe QDs from TMPyP, and thus recover the fluorescence of CdTe QDs. Under optimized conditions, the detection limit of the designed aptasensor was 0.16 ng mL-1, with a linear range of 0.2 to 20 ng mL-1. Furthermore, this aptasensor showed high selectivity toward OTA against other structural analogs and other mycotoxins, and was successfully applied in Astragalus membranaceus samples. The presented aptasensor for OTA detection could be a promising tool for the field monitoring of food and TCM.
A label-free aptasensor for ochratoxin a detection with signal amplification strategies on ultrathin micron-sized 2D MOF sheets

Abstract:The increasing requirement for highly sensitive sensors has promoted the development of new functional nanomaterials and various signal amplification strategies. Due to 2 dimensional (2D) materials' excellent features of large surface areas, structural diversity, and abundant exposed accessible active sites, 2D MOF nanosheets exhibit potential applications in biosensing. Here, an ultrathin micron-sized MOF sheets with good dispersibility and high stability were prepared. Furthermore, a novel label-free aptasensor for ochratoxin A (OTA) detection had been developed based on 2D MOF nanosheets. This sensing signal was amplified by signal amplification strategies containing DNA walker machine, enrichment of Mb signal and nanomaterials of 2D MOF nanosheets and Ag NPs. With the help of these signal amplification strategies, the detection of OTA was reached with a low detection limit (0.08 fg mL−1) and a wide linear rage (0.10 fg mL−1 – 1 μg mL−1). Additionally, the aptasensor exhibits excellent OTA sensing performances, for example, good stability, outstanding selectivity, excellent repeatability, and regenerability.

Abstract:A label-free sensing platform is developed based on switching the structure of aptamer for highly sensitive and selective fluorescence detection of ochratoxin A (OTA). OTA induces the structure of aptamer, transforms into G-quadruplex and produces strong fluorescence in the presence of zinc(II)-protoporphyrin IX probe due to the specific bind to G-quadruplex. The simple method exhibits high sensitivity towards OTA with a detection limit of 0.03 nM and excellent selectivity over other mycotoxins. In addition, the successful detection of OTA in real samples represents a promising application in food safety.

Abstract:We report herein a "turn off-on" fluorescent sensor based on ZnCdSe quantum dots (ZnCdSe QDs) and self-assembled zinc porphyrin for sensitive and rapid detection of ochratoxin A (OTA). Using environmentally friendly dodecyl dimethyl betaine as "soft template", zinc 5, 10, 15, 20-tetra(4-pyridyl)-21H-23H-porphine (ZnTPyP) was self-assembled into nanorods (SA-ZnTPyP) through a green process. The OTA detection process involves two steps: (1) the fluorescence of ZnCdSe QDs is significantly quenched by SA-ZnTPyP, and (2) the combination between SA-ZnTPyP and OTA releases ZnCdSe QDs from their quenchers such that the fluorescence is recovered. Under the optimized experimental conditions, the newly developed fluorescent sensor showed a linear response in concentration range 0.5-80 ng mL(-1), with a detection limit of 0.33 ng mL(-1). The excellent applicability of the system has been verified in milk and coffee samples, with relative recoveries between 98.33% and 103.70%. The proposed fluorescent sensor exhibits excellent selectivity and high sensitivity, and thus could be a promising tool for on-site determination of OTA in food safety control and environmental monitoring.

Abstract:A simple fluorescence and electrochemical dual-channel biosensor based on bifunctional Zr(IV)-based metal-organic framework (Zr-MOF) was proposed to detect Ochratoxin A (OTA). The bifunctional Zr-MOF, with photoluminescence properties and enormous electroactive ligands, was exploited to load OTA-specific aptamers for designing signal probes, greatly simplifying the probe-fabrication process and improving sensing reliability. Upon specific recognition of aptamer toward OTA, the anchored probe was released from the sensing interface into the reaction solution. In this circumstance, the increased amount of the signal probe in reaction solution led to an enhanced fluorescence response, while the decreased amount of the signal probe on the sensing interface resulted in a diminished electrochemical response. According to the dual-channel signal change with increasing OTA concentration, the visual fluorescence strategy was established for intuitive OTA detection, and meanwhile, sensitive electrochemical assay with a detection limit of 0.024 pg/mL was also achieved with the help of one-step electrodeposition as a sensing platform. Moreover, the proposed dual-channel assay has been successfully applied to determine OTA levels in corn samples with rapid response, superior accuracy, and high anti-interference capability, providing a promising method for food safety monitoring.

Abstract:Ochratoxin A (OTA) poses significant risks to human health, being potentially nephrotoxic, carcinogenic, genotoxic, and immuno-toxic. In this work, we developed a dual-mode aptasensor for OTA analysis, integrating colorimetric, electrochemical, and smartphone-based detection. The bifunctional Fe-MIL-88 metal-organic framework, acting as both a nanozyme capable of catalyzing the 3,3',5,5'-tetramethylbenzidine substrate and an electrochemical signal amplifier, enabled OTA quantification through current response or changes in color and absorbance intensity. Besides, the spatial confinement effect enhances the local concentration of Fe-MIL-88 signal probes through rolling circle amplification reaction, thereby contributing to a substantial enhancement in sensitivity. The proposed technique is simple, disposable, highly sensitive and selective, enabling OTA detection in the range of 1 fg/mL to 250 ng/mL, with a limit of detection of 0.22 fg/mL (3σ rule). Furthermore, we successfully detected OTA in corn, wheat, and red wine samples, with results good concordance with those obtained using commercial enzyme-linked immunoassay kits.

Abstract:Herein, an autonomous driven DNA walker-based aptasensor is proposed for the detection of Ochratoxin A (OTA). A nanoscale metal-organic framework (MOF) is synthesized and used to load Mn2+, a cofactor for DNAzyme. The DNA walker and Mn2+@MOF are assembled on the Au surface, integrating the DNA walker and metal auxiliary ions. The proposed sensor avoids the addition of metal auxiliary ions of DNAzyme from the external environment, which achieves the fully autonomous driving of DNA walker and facilitates the on-site detection of OTA. In addition, the substrate strands are modified with a newly synthesized tetraferrocene signal marker, further achieving signal amplification. The sensor demonstrates a sensitivity of 0.289 pg/mL and is successfully applied to real food sample analysis.

Abstract:The fabrication of bioreceptor-free method for accurate and sensitive detection of ochratoxin A (OTA) in cereal is critical, but still a significant challenge to mitigate risks to food industries and public health. In this study, a smartphone-ratiometric fluorescence sensor for the ultrasensitive detection of OTA is developed based on a porphyrinic metal-organic framework and silica nanoparticle composite (Zr-MOF/SiNPs) away from the use of antibodies and aptamers. Due to the excellent recognition ability of Zr-MOF and good storage stability of SiNPs, OTA is detected by Zr-MOF/SiNPs with a wide linear range of 0.05–1000 ng/mL and low detection limit of 0.016 ng/mL. Moreover, the red–blue ratio values of the fluorescence images are extracted through the smartphone color recognizer application with a limit of detection of 1.74 ng/mL, lower than the permissible content of OTA in cereal prescribed by World Health Organization. This sensing platform has been successfully applied in maize samples with superior repeatability and satisfactory recoveries, providing a novel way for simple and label-free analysis of OTA in cereal.
Establishment of a Dual-Signal Enhanced Fluorescent Aptasensor for Highly Sensitive Detection of Ochratoxin A

Abstract:A robust and versatile dual-signal enhanced fluorescent aptasensor was developed for ochratoxin A (OTA) detection based on fluorescence resonance energy transfer between 5-carboxyfluorescein (FAM) and Super Green I (SG) fluorophores as the donor and graphene oxide (GO) nanosheet as the acceptor. Abundant SG probes were adsorbed into the FAM-complementary DNA (cDNA)-aptamer double-stranded structure to achieve remarkably enhanced fluorescence responses. Without OTA, the FAM-cDNA-SG conjugates coexisted with GO nanosheets, exhibiting strong fluorescence signals. In the presence of OTA, it was captured by the aptamers to release cDNA-FAM and SG probes, which were adsorbed by GO, leading to OTA-dependent fluorescence quenching. The changed fluorescence intensity was measured for accurate quantitation of OTA. Under optimum conditions, the dual-signal enhanced fluorescent aptasensor realized fascinating sensitivity with a limit of detection of 0.005 ng/mL and a wide concentration range of 0.02-20 ng/mL, as well as high selectivity for OTA over other interfering substances, excellent accuracy with average recoveries of 91.37-116.83% in the fortified malt matrices, and superior reliability and practicability in actual samples. This FAM-cDNA-aptamer-SG/GO nanosheet-based aptasensing platform could be extended to monitor other contaminants or trace molecules in food, environmental, and diagnostic fields by altering the corresponding aptamers.

Abstract:Ochratoxin A (OTA) is a powerful mycotoxin that can cause severe damage to human health, and its detection has attracted considerable attention in the field of food science. We present a robust and facile label-free colorimetric aptasensor for OTA detection using the aptamer-enhanced oxidase-like activity of MnO2 nanoflowers. The catalytic activities of the nanozymes could be improved by adsorption of the aptamers onto the MnO2 nanoflowers due to the increased affinity of the nanoflowers for the chromogenic substrate. The linear range for OTA detection varied from 0.05 to 33.35 ng/mL with a detection limit of 0.069 ng/mL. The limit of detection of the proposed strategy is equivalent to or even better than those of several previous methods. Moreover, the colorimetric aptasensor exhibited good specificity and stability for the analysis of OTA in wheat flour and red wine samples. Therefore, this method appears to have promising applications in the detection of mycotoxins.

Abstract:A label-free fluorescent aptasensor for specific and ultrasensitive monitoring ochratoxin A(OTA) was developed using the specific aptamer of OTA(OSA) as recognition element, an aggregation-induced emission(AIE) molecule(a 9,10-distyrylanthracene with two ammonium groups, DSAI) as a fluorescent probe, and graphene oxide(GO) as a quencher. In the absence of OTA, the AIE probe DSAI and OSA complex(DSAI/OSA) is adsorbed on the GO surface, and the fluorescence of DSAI will be quenched efficiently via the fluorescence resonance energy transfer(FRET) from DSAI to GO. Upon the OTA addition, a more stable complex(OSA-OTA) is formed and released from GO. Meanwhile, DSAI and OSA-OTA can form a new complex(DSAI/OSA-OTA), then the fluorescent signal of DSAI recovers gradually. Therefore, by introducing GO and DSAI, the fluorescence signal of DSAI can be easily turned from “off” to “on” after the addition of OTA, and the ultrasensitive detection of OTA by monitoring the change of the fluorescence signal of DSAI can be readily realized. The detection limit of the assay can reach 0.324 nmol/L with a linear detection range of 10—200 nmol/L. And the aptasensor exhibits high selectivity for OTA against other analogues. Moreover, it has been successfully applied for the detection of OTA in red wine samples.

Abstract:Ochratoxin A (OTA) is a harmful mycotoxin, which is mainly secreted by Penicillium and Aspergillus species. In this work, an electrochemical aptasensor is presented for OTA detection based on Au nanoparticles (AuNPs) modified zeolite imidazolate frameworks (ZIFs) ZIF-8 platform and duplex-specific nuclease (DSN) triggered hybridization chain reaction (HCR) signal amplification. G-quadruplex-hemin assembled HCR nanowire acted as a nicotinamide adenine dinucleotide (NADH) oxidase and an HRP-mimicking DNAzyme. Besides, thionine (Thi) was enriched as a redox probe for signal amplification in this pseudobienzyme electrocatalytic system. Under the optimal conditions, the analytical response ranged from 1 to 107 fg ml−1 with a detection limit of 0.247 fg ml−1. Furthermore, the aptasensor was proven to be applied in real wheat samples with a recovery between 96.8 and 104.2%, illustrating the potential prospects in practical detection.
Construction of a Label-free Electrochemical Ochratoxin Aptasensor Based on Pt Nanoparticles@ metal-organic Framework Nanomimetic Enzyme

Abstract:Pt NP50_141n-MOF nanocomposite material with peroxidase-like properties was used as electrode substrate, arid a label-free electrochemical aptasensor was constructed by screen printing electrode for the detection of ochratoxin ( OTA). Utilizing the nanomimetic enzyme characteristics of Pt NPs@Mn-MOF, Pt NPs@Mn-MOF was used as an electrode substrate to capture OTA aptamer chain and catalyze the reduction of F1202 to generate current response signal. The introduction of ()TA can obdurate the cataL,lic active sites of the nanomimetic enzyme, which leads to the decrease of current signal. In the range of 0.01 300 ng/mL, the current response value decreases gradually with the :Increase of OTA concentration. The current response signal was detected by the chronoamperometry, so the quantitative detection of OTA was realized indirectly. In addition, the aptasensor can be detected through a small U-disk type electrochemical workstation, which can be connected to a computer or a mobile phone for real-time detection. It has high detection sensitivity and good reproducibility, and the detection limit is as low as 3.33 pg/mL ( S/N=3). The constructed aptasensor can be used for OTA detection in real corn samples, and shows potential application in mycotoxin detection.

Abstract:In this study, a novel aptasensor based on Aptamer/NH2 Janus particles is developed for the detection of Ochratoxin A(OTA). By coating gold on the hemispherical surface of the aminated polystyrene particles, Ochratoxin A aptamer is immobilized on the surface of the gold layer for selective identification and the other hemispherical able to bind to Glassy carbon electrode via peptide bond. Under optimum conditions, the sensor exhibited a wide dynamic range of OTA concentration from 1 x 10(-5) nM to 10 nM, and the detection limit is 3.3 x 10(-3) pM on condition of acceptable stability and reproducibility. The sensors were showed excellent performance in the detection of OTA in red wine sample with recoveries between 95.7% and 100.18%, which studied by the standard addition spiking technique. This work provides a new idea and method for preparing immune electrochemical sensors and is expected to be used for the OTA detection in red wine sample analysis.

Abstract:In this paper, an ultrasensitive photoelectrochemical aptasensor based on amplified signal strategy and competitive mechanism for ochratoxin A (OTA) detection was developed. The prepared photoactive composite material (BiFeO3/Bi2S3) possessed excellent separation ability of photogenerated electron-hole pair and effectively inhibited the recombination of hole and pair, resulting in significantly enhanced photocurrent intensity. The amplified signal strategy was achieved by adding methylene blue (MB), which facilitated electron transfer and improved the photoelectric conversion efficiency. In the presence of OTA, the aptamer can be recognized and captured, the complementary DNA (cDNA) and MB were released from the aptasensor electrode, a decreased photocurrent signal appeared, thus achieving quantitative detection for OTA. Under optimal conditions, the aptasensor exhibited a wide linear relationship range from 10 5 to 10 13 g/mL and a low detection limit of 0.0135 fg/mL (3s/S). Simultaneously, the proposed photoelectrochemical aptasensor also showed excellent accuracy, stability, precision and selectivity.
A Sensitive Electrochemical Aptasensor for Ochratoxin A Detection Based on Double Cascade Amplification Strategy with Nb.BbvCI-Driven DNA Walker and Porous Co3O4 Nanozyme

Abstract:Developing a strategy for the determination of trace ochratoxin A (OTA) in food is very important to ensure food safety. Here, based on a tripod probe, an electrochemical aptasensing platform for detected OTA sensitively was designed, which used the 3D DNA walker by endonuclease Nb.BbvCI powered and the nanozyme of porous Co3O4 nanodisks loaded Pt nanoparticles (Pt NPs@Co3O4) as a cascaded signal amplifying device. With OTA as a model target, the swing-arm probe of DNA walker was triggered. Next, probe on the magnetic beads was cleaved and released by Nb.BbvCI to realize the first signal amplification. After that, the partial sequence of the released probe is well assembled with triple-strand DNA by strand reaction and the other sequence exposed as a capture probe was combined with the Pt NPs@Co3O4. It can efficiently make the decomposition of H2O2, bring about significant current, and the signal was amplified again. The designed electrochemical aptasensor demonstrated outstanding detection linearity in the OTA concentration range of 1 pg/mL to 100 ng/mL, with a detection limit of 0.194 pg/mL (S/N = 3), with excellent reproducibility and high specificity. It can be used to find OTA in food. This research supplies a potential split-new avenue for the sensitive detection of electrochemical aptasensor.

Abstract:We developed a fluorescent aptasensor for label-free detection of ochratoxin A (OTA) based on TdT- and DNA polymerase-assisted multisite strand displacement amplification. This aptasensor exhibits good specificity and high sensitivity with a limit of detection (LOD) of 0.18 ng mL-1, and it can be further applied for the accurate quantification of OTA in complex real samples, holding promising applications in the field of food safety.

Abstract:A one-step electrochemical aptasensor using the thiol- and methylene blue- (MB-) dual-labeled aptamer modified gold electrode for determination of ochratoxin A (OTA) was presented in this research. The aptamer against OTA was covalently immobilized on the surface of the electrode by the self-assembly effect and used as recognition probes for OTA detection by the binding induced folding of the aptamer. Under the optimal conditions, the developed electrochemical aptasensor demonstrated a wide linear range from 0.1 pg mL(-1) to 1000 pg mL(-1) with the limit of detection (LOD) of 0.095 pg mL(-1), which was an extraordinary sensitivity compared with other common methods for OTA detection. Moreover, as a practical application, this proposed electrochemical aptasensor was used to monitor the OTA level in red wine samples without any special pretreatment and with satisfactory results obtained. Study results showed that this electrochemical aptasensor could be a potential useful platform for on-site OTA measurement in real complex samples. (C) 2012 Elsevier B.V. All rights reserved.
A Non-Enzyme and Non-Label Sensitive Fluorescent Aptasensor Based on Simulation-Assisted and Target-Triggered Hairpin Probe Self-Assembly for Ochratoxin a Detection

Abstract:The monitoring and control of mycotoxins has caused widespread concern due to their adverse effects on human health. In this research, a simple, sensitive and non-label fluorescent aptasensor has been reported for mycotoxin ochratoxin A (OTA) detection based on high selectivity of aptamers and amplification of non-enzyme hybridization chain reaction (HCR). After the introduction of OTA, the aptamer portion of hairpin probe H1 will combine with OTA to form OTA-aptamer complexes. Subsequently, the remainder of the opened H1 will act as an initiator for the HCR between the two hairpin probes, causing H1 and H2 to be sequentially opened and assembled into continuous DNA duplexes embedded with numerous G-quadruplexes, leading to a significant enhancement in fluorescence signal after binding with N-methyl-mesoporphyrin IX (NMM). The proposed sensing strategy can detect OTA with concentration as low as 4.9 pM. Besides, satisfactory results have also been obtained in the tests of actual samples. More importantly, the thermodynamic properties of nucleic acid chains in the monitoring platform were analyzed and the reaction processes and conditions were simulated before carrying out biological experiments, which theoretically proved the feasibility and simplified subsequent experimental operations. Therefore, the proposed method possess a certain application value in terms of monitoring mycotoxins in food samples and improving the quality control of food security.

Abstract:In this work, a novel electrochemical aptasensor for the detection of ochratoxin A (OTA) was developed using a three-dimensionally ordered macroporous (3DOM) reduced graphene oxide–Au nanoparticles–molybdenum disulfide (RGO-AuNPs-MoS2) composite material as the sensing interface and Au nanoparticles–carbon-coated ferroferric oxide (AuNPs-Fe3O4@C) composite as the signal amplifier. The 3DOM RGO-AuNPs-MoS2 composite exhibited excellent conductivity and catalytic activity. The 3DOM RGO-AuNPs-MoS2 film was electrodeposited onto the Au electrode surface to immobilize DNA capture probe (CP), which was then hybridized with DNA helper strands (HS) and OTA aptamer (OPT) to form a Y-shaped structure. In the presence of OTA, the OPT was released from the electrode and then combined with AuNPs-Fe3O4@C containing the signal DNA1-metheylene blue (S1-MB) and DNA2-metheylene blue (S2-MB). The current response coming from MB was proportional with the OTA concentration. Under optimal conditions, the linear range of the aptasensor was 1 fg/mL to 0.1 μg/mL, with a detection limit as low as 0.56 fg/mL. The aptasensor was also used to detect OTA in rice and wheat samples, and the results were in agreement with those obtained by liquid chromatography-mass spectrometry (HPLC-MS).
