Abstract:Chiral N-oxide salen ligands and their corresponding Al complexes were synthesized. Notably, the catalytic activity and asymmetric induction ability of the bifunctional N-oxide salen-Al for asymmetric cyanosilylation were compared with that of bi-component catalyst system including chiral pyrrolidine salen–Al complex and Ph3PO. Interestingly, introducing N-oxide group in salen unit could enhance the activity and enantioselectivity of bifunctional catalyst without further adding Ph3PO as co-catalyst and inverse configurations were generated from the two systems. In addition, excellent yields and high ee values could be obtained under milder conditions in bifunctional catalyst system compared with that of bi-component catalyst system.

Abstract:An enantioselective primary amine-catalyzed total N-selective nitroso aldol reaction (N-NA) was achieved through the oxidation of primary aromatic amines to the corresponding nitrosoarenes catalyzed by selenium reagents and 30% H2O2. Abstract:Highly enantioselective alpha-arylation of simple ketones has been achieved by chiral primary amine catalyzed asymmetric retro-Claisen cleavage of beta-diketones. This mild organocatalytic strategy enables the construction of alpha-aryl tertiary carbon stereocenters in good yields and excellent enantioselectivities (up to 98 % ee) with the para-quinone monoimines as aryl sources. Furthermore, oxidative catalytic asymmetric alpha-arylation has also been realized with free p-aminophenols.

Abstract:A general phase-transfer catalyst (PTC) mediated enantioselective alkylation of N-acylsulfenamides is reported. Essential to achieving high selectivity was the use of the triethylacetyl sulfenamide protecting group along with aqueous KOH as the base under biphasic aqueous conditions to enable the reaction to be performed at -40 °C. With these key parameters, enantiomeric ratios up to 97.5:2.5 at the newly generated chiral sulfur center were achieved with an inexpensive cinchona alkaloid derived PTC. Broad scope and excellent functional group compatibility was observed for a variety of S-(hetero)aryl and branched and unbranched S-alkyl sulfenamides. Moreover, to achieve high selectivity for the opposite enantiomer, a pseudoenantiomeric catalyst was designed and synthesized from inexpensive cinchonidine. Given that sulfoximines are a bioactive pharmacophore of ever-increasing interest, selected product sulfilimines were oxidized to the corresponding sulfoximines with subsequent reductive cleavage affording the free-NH sulfoximines in high yields. The utility of the disclosed method was further demonstrated by the efficient asymmetric synthesis of atuveciclib, a phase I clinical candidate for which only chiral HPLC separation had previously been reported for isolation of the desired (R)-sulfoximine stereoisomer.

One of these catalysts was shown to promote the asymmetric Michael addition reaction of 1,3-diphenylpropane-1,3-dione to nitroolefins, affording the desired products in good yields (up to 95%) and enantioselectivities (up to 98% ee).

Abstract:The first highly enantioselective direct alpha-amination of aryl ketones was reported to be catalyzed by organic primary amines derived from cinchona alkaloids. Excellent enantioselectivities (88-99% ee) have been achieved for a broad spectrum of aryl ketones. The presence of 4 A molecular sieves was of great assistance for the high conversions and enantiocontrol.

Abstract:The enantio and diastereoselective synthesis of biaryl atropisomerism with a sulfur stereogenic center is a significant challenge for current chemical synthesis due to the lack of effective synthetic strategies. Herein, we report an efficient method for the construction of chiral biaryl sulfilimines possessing both axial and sulfur-central chiralities via copper/chiral cobalt anion catalyzed sulfur-arylation of sulfenamides with cyclic diaryliodonium salts under mild conditions, gaining various chiral sulfilimines in high yields (up to 99%) and excellent stereoselectivities (up to 98% ee, >20:1 dr). Moreover, the stereoselective sulfur-arylation was performed on a gram scale, and the corresponding products were transformed into various valuable chiral biaryl sulfoximine and their derivatives.

Abstract:A novel direct enantioselective α-alkylation of secondary acyclic amines with simple ketones by combining photocatalysis and lipase catalytic promiscuity is described. Various β-amino ketones were synthesized from secondary acyclic amines in good yields of up to 89% with moderate enantio- and diastereoselectivities (up to 89:11 er and 93:7 dr) under mild conditions with oxidant- and cofactor-free. This novel protocol has the characteristics of wide substrate scope, excellent functional group tolerance, and simple operation.

Abstract:Enantioselective α-functionalizations of carbonyl compounds are fundamental transformations for the asymmetric synthesis of organic compounds. One of the more recent developments along this line is in aminocatalysis, which leads to the direct α-functionalization of simple aldehydes and ketones. However, most of the advances have been achieved with linear aldehydes and ketones as substrates. Effective aminocatalysis with α-branched carbonyls, particularly α-branched ketones, has remained elusive. The primary difficulty arises from the space-demanding α-substituent, which impedes iminium/enamine formation. In 2005, synthetic organic chemists revived catalysis using primary amines, which brought new attention to these challenges, because of the conformational flexibility of primary amines. On the basis of early biomimetic studies by Hine, in 2007 we developed the bioinspired chiral primary amine catalysts featuring primary-tertiary diamines. This type of catalyst involves enamine/iminium catalysis, and we could apply this chemistry to all of the major types of ketones and aldehydes. In this Account, we present research from our laboratory that significantly expands aminocatalysis to include α-branched ketones such as β-ketocarbonyls and α-substituted vinyl ketones. Our primary amine catalysis methodology, when used alone or in conjunction with metal catalysts, provides convenient access to both enantiopure α-tertiary and quaternary ketones, structures that are not available via other approaches. Our mechanistic studies showed that acidic additives play the critical role in facilitating catalytic turnover, most likely by shuttling protons during the enamine/iminium tautomerizations. These additives are also critical to induce the desired stereochemistry via ammonium N-H hydrogen bonding. Proton transfer by shuttling is also stereoselective, resulting in enantioselective enamine protonation as observed in the reactions of α-substituted vinyl ketones. In addition, we have carried out density functional theory studies that help to delineate the origins of the stereoselectivity in these reactions.

Abstract:α-Chiral alkyl primary amines are virtually universal synthetic precursors for all other α-chiral N-containing compounds ubiquitous in biological, pharmaceutical, and material sciences. The enantioselective amination of common alkyl halides with ammonia is appealing for potential rapid access to α-chiral primary amines, but has hitherto remained rare due to the multifaceted difficulties in using ammonia and the underdeveloped C(sp3)–N coupling. Here we demonstrate sulfoximines as excellent ammonia surrogates for enantioconvergent radical C–N coupling with diverse racemic secondary alkyl halides (>60 examples) by copper catalysis under mild thermal conditions. The reaction efficiently provides highly enantioenriched N-alkyl sulfoximines (up to 99% yield and >99% ee) featuring secondary benzyl, propargyl, α-carbonyl alkyl, and α-cyano alkyl stereocenters. In addition, we have converted the masked α-chiral primary amines thus obtained to various synthetic building blocks, ligands, and drugs possessing α-chiral N-functionalities, such as carbamate, carboxylamide, secondary and tertiary amine, and oxazoline, with commonly seen α-substitution patterns. Abstract:Chiral <i>α</i>-branched amines are common structural motifs in functional materials, pharmaceuticals, and chiral catalysts. Therefore, developing efficient methods for preparing compounds with these privileged scaffolds is an important endeavor in synthetic chemistry. Herein, we describe an atom-economical, modular method for a nickel-catalyzed enantioselective α-alkenylation of readily available linear <i>N</i>-sulfonyl amines with alkynes to afford a wide variety of allylic amines without the need for exogenous oxidants, reductants, or activating reagents. The method provides a platform for constructing chiral α-branched amines as well as derivatives such as α-amino amides and β-amino alcohols, which can be conveniently accessed from the newly introduced alkene. Abstract:A dual activation strategy integrating primary amine catalysis and Lewis base activation has been developed for an asymmetric alpha-benzylation reaction. Enamines derived from b-ketocarbonyls could react effectively with in situ generated ortho-quinone methides under Lewis base activation in asymmetric alpha-benzylation of beta-ketocarbonyls and a-branched aldehydes. The approach enables the creation of acyclic all-carbon quaternary stereocenters with excellent enantioselectivities and good activity.

Abstract:Enantioselective sulfonylation of α,β-unsaturated carbonyl compounds from sulfur dioxide is achieved via the synergistic merger photoredox and hydrogen bonding catalysis. This approach provides a facile route to enantioenriched sulfones in the absence of transition metals by using DABCO·(SO2)2 as the sulfur dioxide surrogate. With this method, a variety of sulfones bearing a chiral centre in the α-position are prepared in moderate to good yields.

Abstract:We report herein the first example of asymmetric hydroazidation of α-substituted vinyl ketones by using chiral primary amines as the catalysts. A simple chiral primary-tertiary diamine catalyst derived from l-phenylalanine was found to readily promote this aza-Michael addition reaction with enamine protonation as the key stereogenic step, thus enabling the effective synthesis of α-chiral β-azido ketones with good yields and moderate enantioselectivities.
